| 1 | A novel nonviral nanoparticle gene vector: Poly-L-lysine- silica nanoparticles显示文摘DNA delivery is a core technology for gene structure and function research as well as clinical settings. The ability to safely and efficiently targeted transfer foreign DNA into cells is a fundamental goal in biotechnology. With the development of nanobiotechnology, nanoparticle gene vectors brought about new hope to reach the goal. In our research, silica nanoparticles (SiNP) were synthesized first in a microemulsion system polyoxyethylene nonylphenyl ether (OP-10)/cyclohexane/ammonium hydroxide, at the same time the effects of SiNP size and its distribution were elucidated by orthogonal analysis; then poly-L-lysine (PLL) was linked on the surface of SiNP by nanoparticle surface energy and electrostatically binding; lastly a novel complex nanomate-rial-poly-L-lysine-silica nanoparticles (PLL-SiNP) wasprepared. The analysis of plasmid DNA binding and DNase I enzymatic degradation discovered that PLL-SiNP could bind DNA, and protect it against enzymatic degradation. | 7 | Mid-Holocene Climate Variations Recorded by Palaeolake in Marginal Area of East Asian Monsoon: A Multi-proxy Study显示文摘Traditionally, the mid-Holocene in most parts of China was thought to be warmer with higher precipitation, resulting from a strong Asian summer monsoon. However, some recent researches have proposed a mid-Holocene drought interval of millennial-scale in East Asian monsoon margin areas. Thus whether mid-Holocene was dry or humid remains an open issue. Here, Zhuyeze palaeolake, the terminal lake of the Shiyang River Drainage lying in Asian monsoon marginal areas, was selected for reconstructing the details of climate variations during the Holocene, especially mid-Holocene, on the basis of a sedimentological analysis. Qingtu Lake (QTL) section of 6.92m depth was taken from Zhuyeze palaeolake. Multi-proxy analysis of QTL section, including grain size, carbonate, TOC, C/N and δ13C of organic matter, was used to document regional climatic changes during 9-3 cal ka B.P. The record shows a major environmental change at 9.0-7.8 cal ka B.P., attributed to a climate trend towards warmth and humidity. This event was followed by a typical regional drought event which occurred during 7.8-7.5 cal ka B.P. And a warm and humid climate prevailed from 7.5 to 5.0 cal ka B.P., attributed to the warm/humid Holocene Optimum in this region. After that, the climate gradually became drier. | 15 | Identification of positive cofactor 4 as a diagnostic and prognostic biomarker associated with immune infiltration in hepatocellular carcinoma显示文摘Background and aims:Human positive cofactor 4(PC4)is associated with the development and therapeutic resistance of several malignancies.However,the role of PC4 in hepatocellular carcinoma(HCC)remains obscure.Methods:The expression status of PC4 was explored in Gene Expression Omnibus and The Cancer Genome Atlas datasets.Subsequently,the prognostic and diagnostic significance of PC4 in HCC patients was analyzed.Functional enrichment analyses were conducted to explore biological functions and potential mechanisms.The CIBERSORT algorithm was used for immune infiltration analysis.The risk signature was constructed by LASSO-Cox regression and was validated with the International Cancer Genome Consortium dataset.Quantitative real-time polymerase chain reaction was used to verify the expression levels of all genes.Tumor Immune Dysfunction and Exclusion analysis evaluated immunotherapy response.Finally,using online databases,PC4-related competing endogenous RNA networks were constructed.Results:PC4 levels were significantly upregulated in HCC and positively correlated with the pathological grade and clinical stage.The PC4-high expression group showed worse prognosis.In addition,PC4 could distinguish between tumor and normal tissues with an area under the curve of 0.965.The PC4 level was associated with immune checkpoints and immune cell infiltration.In the training and validation sets,the eight-gene risk signature strongly correlated with HCC patient prognosis.Tumor Immune Dysfunction and Exclusion analysis showed that patients in both the PC4-low and low-risk groups were more likely to benefit from immunotherapy.Finally,an lncRNA/microRNA-101-3p/PC4 network was constructed.Conclusion:We confirmed PC4 as a diagnostic and prognostic biomarker in HCC patients.We also developed and validated an eight-gene risk signature,which will help in clinical decision-making.The competing endogenous RNA network could help explore the regulatory mechanisms of PC4 in HCC. | Liangliang Bai Guan Liu Gang Dou Xiaojun He Chenyu Gong Hongbin Zhang Kai Tan Xilin Du | 2023 | iLIVER2023,2,4: | 0 |
