3Serum concentration of sFas and sFasL in healthy HBsAg carriers,chronic viral hepatitis B and C patients显示文摘AIM: To estimate the amount of apoptosis among healthy HBsAg carriers, patients with chronic HBV infection treated with lamivudine and patients with chronic HCV infedJon treated with interferon alpha and ribavirin. Activity of apoptosis was evaluated by serum sFas/sFasL concentration measurement.Moreover dependence between apoptosis and HBV-DNA or HCV-RNA levels was studied.METHODS: Eighty-six persons were included into study: 34 healthy HBsAg carriers, 33 patients with chronic HBV infection and 19 patients with chronic HCV infection. Serum levels of sFas/sFasL were measured by ELISA assay. HBV-DNA and HCV-RNA were measured by RT-PCR assay. Levels of sFas/sFasL were determined before and 2 and 12 wk after therapy in patients with chronic hepatitis B and C infection.HBV-DNA or HCV-RNA was detected before treatment and 6 mo after treatment.RFSULTS: Twenty-four (71%) healthy HBsAg carriers showed HBV-DNA over 10^5/mL, which was comparable to the patients with chronic hepatitis B. Independently from HBV-DNA levels,the concentration of sFas among healthy HBsAg carriers was comparable to healthy persons. Among patients with chronic hepatitis B and C, the concentration of sFas was significantly higher in comparison to healthy HBsAg carriers and healthy persons. In chronic hepatitis B patients the concentration of sFas was decreased during lamivudine treatment. Among chronic hepatitis C patients the concentration of sFas was increased during IFN alpha and ribavirin treatment, sFasL was not detected in control group. Furthermore sFasL oo:urred more frequently in chronic hepatitis C patients in comparison to chronic hepatitis B patients.CONCLUSION: There are no correlations between apoptosis and HBV-DNA levels. However ther is an association between apoptosis and activity of inflammation in patients with chronic HBV infection. 4Effect of pegylated interferon alpha 2b plus ribavirin treatment on plasma transforming growth factor-β1,metalloproteinase-1,and tissue metalloproteinase inhibitor-1 in patients with chronic hepatitis C显示文摘AIM: To evaluate the effect of antiviral treatment on plasma levels of transforming growth factor-β1 (TGF-β1),metalloproteinase 1 (MMP-1), and tissue inhibitor of metalloproteinase-1 (TIMP-1) in patients with chronic hepatitis C.METHODS: TGF-β1, MMP-1, and TIMP-1 plasma concentrations were measured by an enzyme immunoassay in 28 patients, during 48 wk of treatment with pegylated interferon-alpha 2b (PEG-IFN-α2b) plus ribavirin (RBV)and after 24 wk of follow-up. Patients were divided into two groups: responders (R) and non-responders (NR)related to achieved sustained virologic response. Normal values were evaluated in plasma samples of 13 healthy volunteers.RESULTS: Baseline plasma concentrations of TGF-β1and TIMP-1 (30.9±3.7 and 1506±61 ng/mL respectively)measured in all subjects significantly exceeded the normal values (TGF-β1:18.3±1.6 ng/mL and TIMP-1:1102±67 ng/mL). In contrast, pretreatment MMP-1mean level (6.5±0.9 ng/mL) was significantly lower than normal values (11.9±0.9 ng/mL). Response to the treatment was observed in 12 patients (43%). TGF-β1mean concentration measured during the treatment phase decreased to the control level in both groups.However at wk 72, values of NR patients increased and became significantly higher than in R group.TIMP-1 concentrations in R group decreased during the treatment to the level similar to normal. In NR group,TIMP-1 remained significantly elevated during treatment and follow-up phase and significant difference between both groups was demonstrated at wk 48 and 72. MMP-1levels were significantly decreased in both groups at baseline. Treatment caused rise of its concentration only in the R group, whereas values in NR group remained on the level similar to baseline. Statistically significant difference between groups was noted at wk 48 and 72.CONCLUSION: These findings support the usefulness of TGF-β1, TIMP-1, and MMP-1 in the management of chronic hepatitis C. The whole system is fully fiberized, without the use of any free-space optics. 8Efficiency and safety of lamivudine therapy in patients with chronic HBV infection, dialysis or after kidney transplantation显示文摘AIM: To analyze the effectiveness and safety of lamivudine treatment in patients with chronic HBV infection undergoing hemodialysis or after kidney transplantation, and to study the frequency of tyrosine - methionine - aspartate - aspartate (YMDD) mutation occurrence after lamivudine treatment.METHODS: We analyzed 91 patients with chronic hepatitis B, among whom, 16 patients underwent hemodialysis, 7patients had kidney transplantation and 68 patients had normal function of kidney. The hemodialysis patients were treated by lamivudine 300 mg/wk. Patients after kidney transplantation and patients with normal function of kidney were treated with lamivudine 100 mg/d. Therapy lasted for 12 mo. HBV-DNA, HBsAg, HBeAg and anti-HBe, and antiHCV antibodies were assessed in sera of patients. The analysis was performed before and 6 mo after the end of lamivudine treatment. Before, during and after the lamivudine therapy,the number of erythrocytes, leukocytes, platelets and hemoglobin concentration, ALT and AST activity, as well as bilirubin, urea and creatinine concentrations were analyzed in sera from patients.RESULTS: After the 12-mo lamivudine treatment, elimination of HBV - DNA was observed in 56% patients undergoing hemodialysis and in 53% patients with normal kidney function. Only 1 from 7 (14%) kidney-transplanted patients eliminated HBV-DNA. Furthermore, HBeAg elimination was observed in 36% hemodialysis patients, in 51% patients with normal function of kidneys and in 43% kidneytransplanted patients. Among the patients undergoing dialysis, no YMDD mutation was found after 12 mo of therapy, while it was detected in 9 patients (13%) with normal function of kidney and in 2 kidney-transplanted patients (29%, P<0.006). We did not observe significant side effects of lamivudine treatment in studied patients.CONCLUSION: Effectiveness of lamivudine therapy in dialysis patients is comparable with that in patients withnormal function of kidney. 12Palaeogeographical zonation of gypsum facies:Middle Miocene Badenian of Central Paratethys (Carpathian Foredeep in Europe)显示文摘Studies on Middle Miocene Badenian gypsum in various parts of Central Paratethys,the oldest widespread primary marine gypsum,in western Ukraine,southern Poland and Moravia (Czech Republic) indicate that there are three principal gypsum facies:crystalline gypsum,stromatolitic gypsum and clastic gypsum.The latter typically occurs between crystalline and stromatolitic gypsum and between stromatolitic gypsum and the land.In addition,it is common in channels within gypsum microbialites,and is the main facies during the deposition of the upper part of Badenian gypsum when important bathymetric differences existed within the marginal part of the Carpathian Foredeep Basin,the largest foredeep basin in Europe.Within crystalline gypsum facies,it is observed the overall size of the crystals increases and that the layering declines towards the permanent,stabilized brine body,and thus the giant gypsum intergrowths-non-layered coarse-crystalline selenite is the end-member of gypsum facies continuum.Typically it passes into layered selenites although owing to fluctuations of pycnocline level,some transitional gypsum subfacies may be missing both in the vertical section as well as in particular outcrops.The following important controls on the development of gypsum facies have been identified:pycnocline level fluctuations,brine level fluctuations including brine sheets and floods,rare marine transgressions,pedogenesis leading to 'alabastrine' gypsum development,and rate of inflow of continental water.Tadeusz Marek Peryt 2013Journal of Palaeogeography2013,2,3:2
13Ultrabroadband wavelength-swept source based on total mode-locking of an Yb:CaF_(2)laser显示文摘We present an ultrabroadband, high-speed wavelength-swept source based on a self-modulated femtosecond oscillator. Photonic crystal fiber was pumped by a mode-locked Yb:CaF2 laser, resulting in a strong spectral broadening from 485 to 1800 nm. The pump laser cavity could be realigned in order to achieve total mode-locking of the longitudinal and transverse TEM00 and TEM01 electromagnetic modes. This led to spatial oscillations of the output beam, which induced modulation of the coupling efficiency to the fiber. Due to the fact that nonlinear spectral broadening was intensity dependent, this mechanism introduced wavelength sweeping at the fiber output. The sweeping rate could be adjusted between 7 and 21.5 MHz by changing the geometry of the pump cavity. By controlling the ratio of the transverse mode amplitudes, we were able to tune the sweeping bandwidth, eventually covering both the 1300 nm and 1700 nm bioimaging transparency windows. When compared with previously demonstrated wavelength-swept sources, our concept offers much broader tunability and higher speed. 15The Influence of Carburizing Parameters on Carbon Transfer Coefficient显示文摘Definition of coefficient of carbon transfer in European Standard (EN 10052) is presented as: 'Mass of carbon transferred from carburizing medium into the steel, per unit surface area per second for a unit difference between the carbon potential, and actual surface carbon content'. In this paper, a model is presented of carbon transfer from endothermic atmospheres to carbon steel. The carbon transfer coefficient values were determined experimentally by the foil technique and on specimens, taking into account the following parameters: chemical composition of atmospheres, carbon potential, temperature and time of the carburizing process. Some examples of the variation of the carbon transfer coefficient for two steps of the carburizing process, including soaking before quenching, are given, based on results obtained. 16Effect of vitamin E and human placenta cysteine peptidase inhibitor on expression of cathepsins B and L in implanted hepatoma Morris 5123 tumor model in Wistar rats显示文摘AIM: To examine the effectiveness of human placental inhibitors, by injecting vitamin E to rats with transplanted Norris-5123 hepatoma, on the expression of cathepsins B and L in tumor, liver, lung and blood sera after transplantation of Norris 5123 hepatoma.METHODS: Animals were divided into 10 groups receiving three different concentrations of vitamin E and inhibitors along or in combination and compared with negative control (healthy rats) and positive control (tumor rats). Effectiveness of treatment was evaluated with regard to survival time,tumor response and determination of the activities of proteolytic enzymes and their inhibitors using flurogenic substrates.RESULTS: Cathepsins B and L activities were elevated by 16-fold in comparison with negative control tissues, and their endogenous inhibitor activity decreased by 1.2-fold before treatment. In several cases, tumors completely disappeared following vitamin E plus human placental cyteine protease inhibitor (CPI) compared with controls.The number of complete tumor responses was higher when 20 m/kg vitamin E plus 400 μg of CPI was used, i.e.7/10 rats survived more than two mo. 18Metabolomic and elemental profiling of blood serum in bladder cancer显示文摘Bladder cancer(BC)is one of the most frequently diagnosed types of urinary cancer.Despite advances in treatment methods,no specific biomarkers are currently in use.Targeted and untargeted profiling of metabolites and elements of human blood serum from 100 BC patients and the same number of normal controls(NCs),with external validation,was attempted using three analytical methods,i.e.,nuclear magnetic resonance,gold and silver-109 nanoparticle-based laser desorption/ionization mass spectrometry(LDI-MS),and inductively coupled plasma optical emission spectrometry(ICP-OES).All results were subjected to multivariate statistical analysis.Four potential serum biomarkers of BC,namely,isobutyrate,pyroglutamate,choline,and acetate,were quantified with proton nuclear magnetic resonance,which had excellent predictive ability as judged by the area under the curve(AUC)value of 0.999.Two elements,Li and Fe,were also found to distinguish between cancer and control samples,as judged from ICP-OES data and AUC of 0.807(in validation set).Twenty-five putatively identified compounds,mostly related to glycans and lipids,differentiated BC from NCs,as detected using LDI-MS.Five serum metabolites were found to discriminate between tumor grades and nine metabolites between tumor stages.The results from three different analytical platforms demonstrate that the identified distinct serum metabolites and metal elements have potential to be used for noninvasive detection,staging,and grading of BC.Krzysztof Ossolinski Tomasz Ruman Valerie Copie Brian P.Tripet Leonardo B.Nogueira Katiane O.P.C.Nogueira Artur Kołodziej Aneta Płaza-Altamer Anna Ossolinska Tadeusz Ossolinski Joanna Nizioł 2022Journal of Pharmaceutical Analysis2022,12,6:2
