1Controlled drug delivery vehicles for cancer treatment andtheir performance显示文摘Although conventional chemotherapy has been successful to some extent,the main drawbacks of chemotherapy are its poor bioavailability,high-dose requirements,adverse side effects,low therapeutic indices,development of multiple drug resistance,and non-specific targeting.The main aim in the development of drug delivery vehicles is to successfully address these delivery-related problems and carry drugs to the desired sites of therapeutic action while reducing adverse side effects.In this review,we will discuss the different types of materials used as delivery vehicles for chemotherapeutic agents and their structural characteristics that improve the therapeutic efficacy of their drugs and will describe recent scientific advances in the area of chemotherapy,emphasizing challenges in cancer treatments.Sudipta Senapati Arun Kumar Mahanta Sunil Kumar Pralay Maiti 2018Signal Transduction and Targeted Therapy2018,3,1:9

3The crystal structure of the non-liganded 14-3-3σ protein: insights into determinants of isoform specific ligand binding and dimerization显示文摘Seven different, but highly conserved 14-3-3 proteins are involved in diverse signaling pathways in human cells. It isunclear how the 14-3-3σ isoform, a transcriptional target of p53, exerts its inhibitory effect on the cell cycle in thepresence of other 14-3-3 isoforms, which are constitutively expressed at high levels. In order to identify structuraldifferences between the 14-3-3 isoforms, we solved the crystal structure of the human 14-3-3σ protein at a resolutionof 2.8 ? and compared it to the known structures of 14-3-3ζ and 14-3-3τ. The global architecture of the 14-3-3σ foldis similar to the previously determined structures of 14-3-3ζ and 14-3-3τ: two 14-3-3σ molecules form a cup-shapeddimer. Significant differences between these 14-3-3 isoforms were detected adjacent to the amphipathic groove, whichmediates the binding to phosphorylated consensus motifs in 14-3-3-ligands. Another specificity determining region islocalized between amino-acids 203 to 215. These differences presumably select for the interaction with specific ligands,which may explain the different biological functions of the respective 14-3-3 isoforms. Furthermore, the two 14-3-3σmolecules forming a dimer differ by the spatial position of the ninth helix, which is shifted to the inside of the ligandinteraction surface, thus indicating adaptability of this part of the molecule. In addition, 5 non-conserved residues arelocated at the interface between two 14-3-3σ proteins forming a dimer and represent candidate determinants of homo-and hetero-dimerization specificity. 7Clinical laboratory and imaging evidence for effectiveness of agarose-agarose macrobeads containing stem-like cells derived from a mouse renal adenocarcinoma cell population (RMBs) in treatment-resistant, advanced metastatic colorectal cancer:Evaluation of显示文摘Objective: The complexity, heterogeneity and capacity of malignant neoplastic cells and tumors for rapid change and evolution suggest that living-cell-based biological-systems approaches to cancer treatment are merited. Testing this hypothesis, the tumor marker, metabolic activity, and overall survival(OS) responses, to the use of one such system, implantable macrobeads [RENCA macrobeads(RMBs)], in phase I and IIa clinical trials in advanced,treatment-resistant metastatic colorectal cancer(m CRC) are described here.Methods: Forty-eight m CRC patients(30 females; 18 males), who had failed all available, approved treatments,underwent RMB implantation(8 RMB/kg body weight) up to 4 times in phase I and phase IIa open-label trials.Physicals, labs [tumor and inflammation markers, lactate dehydrogenase(LDH)] and positron emission tomography-computed tomography(PET-CT) imaging to measure number/volume and metabolic activity of the tumors were performed pre-and 3-month-post-implantation to evaluate safety and initial efficacy(as defined by biological responses). PET-CT maximum standard uptake value(SUVmax)(baseline and d 90; SUVmax ≥2.5), LDH,and carcinoembryonic antigen(CEA) and/or cancer antigen 19-9(CA 19-9) response(baseline, d 30 and/or d 60)were assessed and compared to OS.Results: Responses after implantation were characterized by an at least 20% decrease in CEA and/or CA 19-9 in75% of patients. Fluorodeoxyglucose(FDG)-positive lesions(phase I, 39; 2 a, 82) were detected in 37/48 evaluable patients, with 35% stable volume and stable or decreased SUV(10) plus four with necrosis; 10, increased tumor volume, SUV. LDH levels remained stable and low in Responders(R)(d 0–60, 290.4–333.9), but increased steadily in Non-responders(NR)(d 0–60, 382.8–1,278.5)(d 60, P=0.050). Responders to RMBs, indicated by the changes in the above markers, correlated with OS(R mean OS=10.76 months; NR mean OS=4.9 months; P=0.0006).Conclusions: The correlations of the tumor marker, tumor volume and SUV changes on PET-CT, and LDH levels themselves, and with OS, support the concept of a biological response to RMB implantation and the validity of the biological-systems approach to m CRC. 8Curcuma angustifolia ameliorates carbon tetrachloride-induced hepatotoxicity in HepG2 cells and Swiss albino rats显示文摘Objective:To determine the antioxidant and hepatoprotective potential of methanol extract of rhizome of Curcuma angustifolia(MECA)against carbon tetrachloride(CCl4)-induced hepatic damage in vitro and in vivo.Methods:DPPH,ABTS and reducing power assays were performed to estimate the antioxidant effect of MECA.In vitro cytotoxicity of MECA against HepG2 cells was evaluated,whereas serum biochemical parameters and levels of antioxidative enzymes were measured in vivo and in vitro.Additionally,histopathological studies were estimated in order to investigate the hepatoprotective efficacy of MECA.Furthermore,GC-MS analysis of the extract was performed to identify the chemical components.Results:MECA exhibited strong antioxidant activity and attenuated CCl4-induced decrease in the viability of HepG2 cells.Additionally,MECA significantly restored the ALT,AST,ALP,TP and albumin level in comparison with the CCl4 group.After pre-treatment with MECA,effects of SOD,CAT and GSH were increased as well as lipid peroxidation amount decreased on CCl4-induced hepatotoxicity in in vitro and in vivo model.Furthermore,histopathological observation confirmed that MECA reduced liver injury induced by CCl4 in rats.GC-MS analysis confirmed the presence of bioactive constituents such as α-tocopherol(12.27%),phytol(7.61%),squalene(3.71%),β-sitosterol(2.19%),eugenol(2.59%),curcumenol(1.20%),β-elemene(1.00%)and eucalyptol(0.89%).Conclusions:MECA contains antioxidant and hepatoprotective constituents such asα-tocopherol,phytol,squalene and eugenol and exerts hepatoprotective effect both in vitro and in vivo.Sudipta Jena Asit Ray Diptirani Rath Ambika Sahoo Subhashree Singh Noohi Nasim Durga Madhab Kar Sanghamitra Nayak 2019Asian Pacific Journal of Tropical Medicine2019,12,9:2

12Tree species composition and diversity in montane forests of Garhwal Himalaya in relation to environmental and soil properties显示文摘The montane forests of Garhwal Himalaya were investigated for tree species composition and diversity in relation to environmental and edaphic variables.A stratified random sampling approach was adopted to collect the field data from each forest.A total of 39 tree species belonging to 31 genera and 23 families were recorded from the sampling area(6 forest stands ×10 plots in each).Lauraceae with 5 species was the largest family while Quercus(4 species) emerged as the largest genus.Species-area curve(asymptote) predicted more species than the observed number of species.Cluster analysis has shown that the two Abies pindrow dominant forest stands had the highest similarity in tree species composition.The tree stem density ranged from 540-1170 ha-1,basal area 23.01-55.94 m^2 ha-1,ShannonWiener diversity index 1.69-2.49,evenness index0.42-0.74,beta diversity 0.40-0.82 and similarity index 0.17-0.54 in the six studied forests.Two-way indicator species analysis(TWINSPAN) identified four distinct tree communities in the study area with Daphniphyllum himalayense,Abies pindrow,Quercus oblongata,and Pinus roxburghii as indicator species of corresponding communities.Canonical correspondence analysis(CCA) ascertained the TWINSPAN results and revealed the relation of tree species with environmental and edaphic variables.The phytosociological attributes of tree species varied from one forest stand to another;however,the tree species richness,density,and diversity peaked at intermediate elevation on the north aspect.Findings reveal that tree composition and diversity in the montane forests of Garhwal Himalaya are influenced by both environmental(elevation and slope aspect)and soil variables(texture,water holding capacity,moisture content,available N,exchangeable P,and pH).This study will help in understanding the forest distribution across the western Himalaya,guiding the conservationists and policymakers in carrying out conservation-related management activities,designing the long-term monitoring studies and assessing the effects of ongoing and future climate change and disturbances.Dinesh Singh RAWAT P.TIWARI Sudipta Kumar DAS J.K.TIWARI 2020Journal of Mountain Science2020,17,12:2

14Performance study and kinetic modeling of hybrid bioreactor for treatment of bi-substrate mixture of phenol-m-cresol in wastewater:Process optimization with response surface methodology显示文摘Performance of a hybrid reactor comprising of trickling filter(TF) and aeration tank(AT) unit was studied for biological treatment of wastewater containing mixture of phenol and m-cresol,using mixed microbial culture.The reactor was operated with hydraulic loading rates(HLR) and phenolics loading rates(PLR) between 0.222-1.078m3/(m2·day) and 0.900-3.456kg/(m3·day),respectively.The efficiency of substrate removal varied between 71%-100% for the range of HLR and PLR studied.The fixed film unit showed better substrate removal efficiency than the aeration tank and was more resistant to substrate inhibition.The kinetic parameters related to both units of the reactor were evaluated and their variation with HLR and PLR were monitored.It revealed the presence of substrate inhibition at high PLR both in TF and AT unit.The biofilm model established the substrate concentration profile within the film by solving differential equation of substrate mass transfer using boundary problem solver tool 'bvp4c' of MATLAB 7.1 software.Response surface methodology was used to design and optimize the biodegradation process using Design Expert 8 software,where phenol and m-cresol concentrations,residence time were chosen as input variables and percentage of removal was the response.The design of experiment showed that a quadratic model could be fitted best for the present experimental study.Significant interaction of the residence time with the substrate concentrations was observed.The optimized condition for operating the reactor as predicted by the model was 230mg/L of phenol,190mg/L of m-cresol with residence time of 24.82 hr to achieve 99.92% substrate removal.Sudipta Dey Somnath Mukherjee 2013Journal of Environmental Sciences2013,25,4:2
