1变性梯度凝胶电泳(DGGE)在微生物生态学中的应用显示文摘由于从环境样品中分离和培养细菌的困难 ,分子生物学方法已发展用来描述和鉴定微生物群落。近年来基于 DNA方法的群落分析得到了迅速的发展 ,如 PCR扩增技术 ,克隆文库法 ,荧光原位杂交法 ,限制性酶切片段长度多态性法 ,变性和温度梯度凝胶电泳法。DGGE已广泛用于分析自然环境中细菌、蓝细菌 ,古菌、微微型真核生物、真核生物和病毒群落的生物多样性。这一技术能够提供群落中优势种类信息和同时分析多个样品。具有可重复和容易操作等特点 ,适合于调查种群的时空变化 ,并且可通过对切下的带进行序列分析或与特异性探针杂交分析鉴定群落成员。DGGE分析微生物群落的一般步骤如下 :一是核酸的提取 ,二是 1 6Sr RNA,1 8S r RNA或功能基因如可容性甲烷加单氧酶羟化酶基因 ( mmo X)和氨加单氧酶 α-亚单位基因 ( amo A)片段的扩增 ,三是通过DGGE分析 PCR产物。DGGE使用具有化学变性剂梯度的聚丙烯酰胺凝胶 ,该凝胶能够有区别的解链PCR扩增产物。由 PCR产生的不同的 DNA片段长度相同但核苷酸序列不同。因此不同的双链 DNA片段由于沿着化学梯度的不同解链行为将在凝胶的不同位置上停止迁移 [5] 。 DNA解链行为的不同导致一个凝胶带图案 ,该图案是微生物群落中主要种类的一个轮廓。 DGGE使用所有生物中保守的基因?马悦欣 Carola Holmstrm Jeremy Webb Staffan Kjelleberg 2003生态学报2003,23,8:100

3抑制次同步谐振的TCSC主动阻尼控制显示文摘采用同步电压反转(synchronous voltage reversal,SVR)触发控制的可控串联补偿电容(thristor controlled series compensator,TCSC)本身不会诱发系统的次同步谐振(sub-synchronous resonance SSR),而且因为其在次同步频率范围内固有的感性阻抗特性,一直以来工程中都把TCSC作为一种缓解SSR的有效措施。虽然采用SVR触发的TCSC可缓解系统SSR的压力,但其本身却无法主动抑制SSR,该文提出一种TCSC的附加控制——主动阻尼控制,通过调节TCSC的触发,可使系统在危险振荡模式下向机组提供正值的电气阻尼,从而更好地抑制SSR。时域及频域的仿真也验证了该主动阻尼控制的有效性。周长春 刘前进 Lennart Angquist Staffan Rudin 2008中国电机工程学报2008,28,10:19
4卵巢浆液性腺癌组织中TK1和Ki-67表达的意义显示文摘目的探讨细胞质胸苷激酶1(cytoplasmic thymidinekinas-1,TK1)、Ki-67在卵巢浆液性腺癌中表达的意义。方法回顾性研究55例经手术治疗的卵巢浆液性腺癌患者的临床病理资料,并运用免疫组化技术观察TK1、Ki-67在卵巢浆液性腺癌中的表达情况及与临床病理参数之间的意义。结果 TK1阳性表达定位于细胞质,阳性率为72.7%。TK1的表达与肿瘤的最大径、复发、pTNM分期、病理分级密切相关(P<0.05)。Ki-67阳性表达定位于细胞核,阳性率为80.0%,Ki-67的表达与肿瘤的复发、pTNM分期、病理分级有关(P<0.05)。Kappa检验显示TK1的表达与卵巢浆液性腺癌复发较一致(k=0.559,P=0.000),且判断复发比Ki-67更为优越。Kaplan-Meier检验显示pTNM分期、肿瘤复发、MDACC分级、Ki-67、TK1表达分别与预后有关(P<0.05)。COX回归多因素分析显示:肿瘤复发是影响卵巢浆液性腺癌患者的独立性预后因素。结论卵巢浆液性腺癌的复发影响患者的预后,TK1对判断卵巢浆液性腺癌是否有复发倾向具有参考价值,且优于Ki-67,初次手术后肿瘤组织免疫组化TK1高表达预示患者存在高复发的风险,提示临床要加强术后的治疗方案,并提高随访频数。刘冲 郭庆 石群立 余波 马恒辉 Staffan Eriksson Ellen He Sven Skog 2011临床与实验病理学杂志2011,27,12:12

9The connection of cytoskeletal network with plasma membrane and the cell wall显示文摘The cell wall provides external support of the plant cells, while the cytoskeletons including the microtubules and the actin filaments constitute an internal framework. The cytoskeletons contribute to the cell wall biosynthesis by spatially and temporarily regulating the transportation and deposition of cell wall components. This tight control is achieved by the dynamic behavior of the cytoskeletons, but also through the tethering of these structures to the plasma membrane. This tethering may also extend beyond the plasma membrane and impact on the cell wall, possibly in the form of a feedback loop. In this review, we discuss the linking components between the cytoskeletons and the plasma membrane, and/or the cell wall. 10High level of deoxycholic acid in human bile does not promote cholesterol gallstone formation显示文摘AIM: To study whether patients with excess deoxycholic acid (DCA) differ from those with normal percentage of DCA with respect to biliary lipid composition and cholesterol saturation of gallbladder bile.METHODS: Bile was collected during operation through puncturing into the gallbladder from 122 cholesterol gallstone patients and 46 gallstone-free subjects undergoing cholecystectomy. Clinical data, biliary lipids, bile acid composition,presence of crystals and nucleation time were analyzed.RESULTS: A subgroup of gallstone patients displayeda higher proportion of DCA in bile than gallstone free subjects.By choosing a cut-off level of the 90th percentile, a group of 13 gallstone patients with high DCA levels (mean 50percent of total bile acids) and a large group of 109 patients with normal DCA levels (mean 21 percent of total bile acids)were obtained. The mean age of the patients with high DCA levels was higher than that of the group with normal levels (mean age: 62 years vs45 years) and so was the mean BMI (28.3 vs. 24.7). Plasma levels of cholesterol and triglycerides were slightly higher in the DCA excess groups compared with those in the normal DCA group. There was no difference in biliary lipid composition, cholesterol saturation, nucleation time or occurrence of cholesterol crystals in bile between patients with high and normal levels of DCA.CONCLUSION: Gallstone patients with excess DCA were of older age and had higher BMI than patients with normal DCA. The two groups of patients did not differ with respect to biliary lipid composition, cholesterol saturation, nucleation time or occurrence of cholesterol crystals. 13Investigation of CRISPR/Cas9-induced SD1 rice mutants highlights the importance of molecular characterization in plant molecular breeding显示文摘Although Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR)/CRISPR-associated 9(Cas9)system has been widely used for basic research in model plants,its application for applied breeding in crops has faced strong regulatory obstacles,due mainly to a poor understanding of the authentic output of this system,particularly in higher generations.In this study,different from any previous studies,we investigated in detail the molecular characteristics and production performance of CRISPR/Cas9-generated SD1(semi-dwarf 1)mutants from T2 to T4 generations,of which the selection of T1 and T2 was done only by visual phenotyping for semidwarf plants.Our data revealed not only on-and off-target mutations with small or lager indels but also exogenous elements in T2 plants.All indel mutants passed stably to T3 or T4 without additional modifications independent on the presence of Cas9,while some lines displayed unexpected hereditary patterns of Cas9 or some exogenous elements.In addition,effects of various SD1 alleles on rice height and yield differed depending on genetic backgrounds.Taken together,our data showed that the CRISPR/Cas9 system is effective in producing homozygous mutants for functional analysis,but it may be not as precise as expected in rice,and that early and accurate molecular characterization and screening must be carried out for generations before transitioning of the CRISPR/Cas9 system from laboratory to field.Sukumar Biswas Jiaqi Tian Rong Li Xiaofei Chen Zhijing Luo Mingjiao Chen Xiangxiang Zhao Dabing Zhang Staffan Persson Zheng Yuan Jianxin Shi 2020Journal of Genetics and Genomics2020,47,5:4

17脑胶质瘤组织中胸腺嘧啶核苷激酶1的表达和意义及其与细胞增殖抗原Ki67表达的关系显示文摘目的研究脑胶质瘤组织中胸腺嘧啶核苷激酶1(TK1)的表达和意义,以及其与细胞增殖抗原Ki67表达的关系。方法 202例脑胶质瘤患者,经病理检查进行脑胶质瘤病理分级及组织类型分型。采用免疫组化染色法检测脑胶质瘤组织中TK1和Ki67的表达。比较不同级别及类型患者脑胶质瘤组织中TK1表达的阳性率;分析TK1与Ki67表达的关系。结果本组患者脑胶质瘤按WHO分级:Ⅰ级12例,Ⅱ级85例,Ⅲ级60例,Ⅳ级45例;组织类型分型:少突胶质细胞瘤28例,星形胶质细胞瘤174例。本组患者TK1阳性119例(58.9%);其中良性组(WHOⅠ级)患者无TK1阳性,低度恶性组(WHOⅡ级)阳性31例(36.5%),高度恶性组(WHOⅢ、Ⅳ级)阳性88例(83.8%);三组间TK1阳性率比较差异均有统计学意义(P<0.05~0.001)。星形胶质细胞瘤及复发患者的TK1阳性率分别显著高于少突胶质细胞瘤和初发的患者(均P<0.05)。TK1与Ki67的表达水平呈正相关(r=0.711,P<0.01)。结论恶性程度高、星形胶质细胞瘤及复发的脑胶质瘤患者瘤组织中TK1的表达增高;提示TK1表达可作为脑胶质瘤病理分级、分型及复发的重要参考指标。TK1与Ki67正相关,提示TK1亦可反映脑胶质瘤细胞的增殖情况。朱海青 宋坤 王娟 张巧全 周金宝 石群立 Staffan Eriksson Ellen He Sven Skog 2012临床神经病学杂志2012,25,5:3
