1Ultra-highly diluted plant extracts of Hydrastis canadensis and Marsdenia condurango induce epigenetic modifications and alter gene expression profiles in HeLa cells in vitro显示文摘OBJECTIVE: Methylation-specifi c epigenetic process and gene expression profi les of He La cells treated with ultra-high dilutions(HDs) of two plant extracts, Hydrastis canadensis(HC-30) and Marsdenia condurango(Condu-30), diluted 1060 times, were analyzed against placebo 30C(Pl-30) for alterations in gene profi les linked to epigenetic modifi cations.METHODS: Separate groups of cells were subjected to treatment of Condu-30, HC-30, and Pl-30 prepared by serial dilutions and succussions. Global microarray data recorded on Affymetrix platform, using 25-mer probes were provided by i Life Discoveries, India. Slides were scanned with 3000 7G microarray scanner and raw data sets were extracted from Cel(raw intensity) fi les. Analyses of global microarray data profi le, differential gene expression, fold change and clusters were made using Gene Spring GX12.5 software and standard normalization procedure. Before microarray study, concentration of RNA(ng/μL), RIN value and r RNA ratio for all the samples were analysed by Agilant Bioanalyzer 2100. Reverse transcriptase polymerase chain reaction(RT-PCR) and quantitative RTPCR were done for analyzing SMAD-4 expression. Fluorescence-activated cell sorting study was further made to elucidate fate of cells at divisional stages. Methylation-specifi c restriction enzyme assay was conducted for ascertaining methylation status of DNA at specifi c sites.RESULTS: HDs of HC-30 and Condu-30 differentially altered methylation in specifi c regions of DNA and expression profi les of certain genes linked to carcinogenesis, as compared to Pl-30. Two separate cut sites were found in genomic DNA of untreated and placebo-treated He La cells when digested with McrB C, compared to a single cut observed in Condu-30-treated genomic DNA. SMAD-4 gene expression validated the expression pattern observed in microarray profi le. 2Mechanistic data-driven prediction of as-built mechanical properties in metal additive manufacturing显示文摘Metal additive manufacturing provides remarkable flexibility in geometry and component design,but localized heating/cooling heterogeneity leads to spatial variations of as-built mechanical properties,significantly complicating the materials design process.To this end,we develop a mechanistic data-driven framework integrating wavelet transforms and convolutional neural networks to predict location-dependent mechanical properties over fabricated parts based on process-induced temperature sequences,i.e.,thermal histories.The framework enables multiresolution analysis and importance analysis to reveal dominant mechanistic features underlying the additive manufacturing process,such as critical temperature ranges and fundamental thermal frequencies.We systematically compare the developed approach with other machine learning methods.The results demonstrate that the developed approach achieves reasonably good predictive capability using a small amount of noisy experimental data.It provides a concrete foundation for a revolutionary methodology that predicts spatial and temporal evolution of mechanical properties leveraging domain-specific knowledge and cutting-edge machine and deep learning technologies.Xiaoyu Xie Jennifer Bennett Sourav Saha Ye Lu Jian Cao Wing Kam Liu Zhengtao Gan 2021npj Computational Materials2021,,1:1

6Combined effect of Reynolds and Grashof numbers on mixed convection in a lid-driven T-shaped cavity filled with water-Al_2O_3 nanofluid显示文摘Lid-driven mixed convection has been given immense importance due to its wide range of applications. A T-shaped cavity is introduced and pertinent parameters controlling mixed convection phenomenon are analyzed in this paper. Water-Al2O3 nanofluid is considered inside the cavity to augment heat transfer rate. Galerkin weighted residual method of finite element analysis is applied for the numerical simulations. Numerical solution is obtained for different solid volume fractions of nanofluid(?= 0- 0.15), Grashof numbers(Gr= 0.1 ?5 000) and Reynolds numbers(Re= 0.31 ?1 000) in laminar flow regime. Special attention is given on the analysis of flow at the pure mixed convection regime. It is found that Grashof, Reynolds and Richardson numbers along with solid volume fraction of nanofluid have significant effect on heat transfer characteristics inside the cavity. 7Production of fermented tea petal decoction with insights into in vitrobiochemical tests,antioxidant assay and GC-MS analysis显示文摘This research work was designed to attempt and propose the first report on production and biochemical characterization of fermented tea flower petal decoction or simply tea petal wine.The tea petal decoction and brewer’s yeast or Saccharomyces cerevisiae were co-cultured for fermentation.Antioxidant activity and chromatographic separation of potential candidates were assessed.Primary investigations for qualitative characters on this fermented broth revealed the presence of steroids,tannin,flavonoids,phenol,cardiac glycosides,coumarin,caffeine etc.Our manufactured fermented broth showed high free radical scavenging activity after 2 months of aging.High DPPH scavenging activities were also observed in solvent fractions of acetone,ethanol and methanol.The antioxidant activity,alcohol percentage and other qualities were seen to be gradually increased during aging.Gas chromatography-mass spectrometry analysis revealed the presence of 44 compounds including many potential antioxidant molecules and other bioactive agents.Hopefully,presence of alcohol with medicinally active compounds and antioxidant activity will make it as acceptable as a good wine and tea flower as economically functional.Soumya Majumder Sumedha Saha Arindam Ghosh Sukanya Acharyya Sahadeb Sarkar Sourav Chakraborty Malay Bhattacharya 2021Food Production, Processing and Nutrition2021,3,1:0
8Livelihood status of small-scale fishermen and determinants of their income: Insights from north-eastern floodplains of Bangladesh显示文摘The small-scale fishermen in the north-eastern floodplains are one of the most vulnerable communities of Bangladesh due to their limited income.This study assessed their livelihood status using a sustainable livelihood framework and employed a multiple regression model to evaluate factors influencing their earnings.Primary data were collected from fishermen’s households in Tanguar Haor and Hakaluki Haor through direct interviews.Results revealed that these households lacked human capital,had poor sanitation facilities and suffered from frequent diseases.Their income is very limited as they lack training on other socioeconomic activities to comple-ment their livelihood grounded on fishing.Hence,their financial capital was very deficient and ranked lowest among other livelihood capitals.Fishermen’s major physical assets were the boats used for group and individual fishing.These small-scale fishermen had limited involvement with local administration,and around half of them were members of various NGOs.Women of these households had low participation in fishing or other income activities.The income of the fishermen was influenced by their age,fish catch per day,training and NGO mem-bership.To increase the earnings and improve the livelihood status of small-scale fishers in the floodplain,this study suggests providing training on complementary income generating activities,improving sanitation facilities and providing additional government and non-government support.Kishor Kumar Tikadar Md.Jahurul Islam Sourav Mohan Saha Md.Mehedi Alam Sanzib Kumar Barman Md.Asadur Rahman 2022Geography and Sustainability2022,3,3:0

10Tree assemblages and diversity patterns in Tropical Juri Forest,Bangladesh显示文摘Juri is a biodiversity-rich primary forest in Bangladesh, which remains ecologically unexplored. We identified tree species and examined the richness, alpha(a)diversity and floristic similarity patterns within the identified communities. Vegetation and environmental data were sampled in 120(0.04 ha) study plots. Tree communities were delimited by two-way indicator species analysis(TWINSPAN). In total, 78 tree species of 35 families and58 genera were identified. TWINSPAN identified six tree communities: A—Tricalysia singularis; B—Kydia calycina-Castanopsis tribuloides; C—Polyalthia simiarum-Duabanga grandiflora; D—Ficus roxburghii; E—Artocarpus lacucha; F—Artocarpus lacucha. Mean richness, Shannon and Gini-Simpson indices were highest for the Polyalthia simiarum-Duabanga grandiflora community, while Ficus roxburghii showed lowest diversity. Significant differences(p = 0.05) in three diversity indices were recorded between Polyalthia simiarum-Duabanga grandiflora and Ficus roxburghii.
