| 1 | CPSF30-L-mediated recognition of mRNA m^(6)A modification controls alternative polyadenylation of nitrate signaling-related gene transcripts in Arabidopsis显示文摘N6-methyladenosine(m^(6)A),a ubiquitous internal modification of eukaryotic mRNAs,plays a vital role in almost every aspect of mRNA metabolism.However,there is little evidence documenting the role of m^(6)A in regulating alternative polyadenylation(APA)in plants.APA is controlled by a large protein-RNA complex with many components,including CLEAVAGE AND POLYADENYLATION SPECIFICITY FACTOR30(CPSF30).In Arabidopsis,CPSF30 has two isoforms and the longer isoform(CPSF30-L)contains a YT512-B Homology(YTH)domain,which is unique to plants.In this study,we showed that CPSF30-L YTH domain binds to m^(6)A in v itro.In the cpsf30-2 mutant,the transcripts of many genes including several important nitrate signaling-related genes had shifts in polyadenylation sites that were correlated with m^(6)A peaks,indicating that these gene transcripts carrying m^(6)A tend to be regulated by APA.Wild-type CPSF30-L could rescue the defects in APA and nitrate metabolism in cpsf30-2,but m^(6)A-binding-defective mutants of CPSF30-L could not.Taken together,our results demonstrated that m^(6)A modification regulates APA in Arabidops is and revealed that the m^(6)A reader CPSF30-L affects nitrate signaling by controlling APA,shedding new light on the roles of the m^(6)A modification during RNA 3-end processing in nitrate metabolism. | Yiteng Hou Jing Sun Baixing Wu Yangyang Gao Hongbo Nie Zhentian Nie Shuxuan Quan Yong Wang Xiaofeng Cao Sisi Li | 2021 | Molecular Plant2021,14,4: | 12 |
| 2 | Human natural killer cells for targeting delivery of gold nanostars and bimodal imaging directed photothermal/photodynamic therapy and immunotherapy显示文摘Objective:To construct a novel nanoplatform GNS@CaCO3/Ce6-NK by loading the CaCO3-coated gold nanostars(GNSs)with Chlorin e6 molecules(Ce6)into human peripheral blood mononuclear cells(PBMCs)-derived NK cells for tumor targeted therapy.Methods:GNS@CaCO3/Ce6 nanoparticles were prepared and characterized by TEM and UV-vis.The cell surface markers and cytokines secretion of NK cells before and after loading the GNS@CaCO3/Ce6 nanoparticles were detected by Flow Cytometry(FCM)and ELISA.Effects of the GNS@CaCO3/Ce6-NK cells on A549 cancer cells was determined by FCM and CCK-8.Intracellular fluorescent signals of GNS@CaCO3/Ce6-NK cells were detected via Confocal laser scanning microscopic(CLSM)and FCM at different time points.Intracellular ROS generation of GNS@CaCO3/Ce6-NK cells under laser irradiation were examined by FCM.The distribution of GNS@CaCO3/Ce6-NK in A549 tumor-bearing mice were observed by fluorescence imaging and PA imaging.The combination therapy of GNS@CaCO3/Ce6-NK under laser irradiation were investigated on tumor-bearing mice.Results:The coated CaC03 shell on the surface of GNSs exhibited prominent delivery and protection effect of Ce6 during the cellular uptake process.The as-prepared multifunctional GNS@CaCO3/Ce6-NK cells possessed bimodal functions of fluorescence imaging and photoacoustic imaging.The as-prepared multifunctional GNS@CaCO3/Ce6-NK cells could actively target tumor tissues with the enhanced photothermal/photodynamic therapy and immunotherapy.Conclusions:The GNS@CaCO3/Ce6-NK shows effective tumor-targeting ability and prominent therapeutic efficacy toward lung cancer A549 tumor-bearing mice.Through fully utilizing the features of GNSs and NK cells,this new nanoplatform provides a new synergistic strategy for enhanced photothermal/photodynamic therapy and immunotherapy in the field of anticancer development in the near future. | Bin Liu Wen Cao Jin Cheng Sisi Fan Shaojun Pan Lirui Wang Jiaqi Niu Yunxiang Pan Yanlei Liu Xiyang Sun Lijun Ma Jie Song Jian Ni Daxiang Cui | 2019 | Cancer Biology & Medicine2019,16,4: | 7 |
| 3 | Synthesis and evaluation of methionine and folate co-decorated chitosan self-assembly polymeric micelles as a potential hydrophobic drug-delivery system显示文摘In this study, an amphiphilic copolymer folate-succinyl-methionine-chitosan-octyl (FSMCO) was successfully synthesized step by step for self-assembling polymeric micelles. The copolymers formed micelle-like nanoparticles by their amphiphilic characteristics and structures were examined by UV-Vis absorption and Fourier transform spectroscopy. The sizes of blank and ICG derivativeloaded micelles measured by dynamic light scattering were about 170 and 140 nm, respectively, which were spherical in shape with an average zeta potential of 10 mV. Further studies on the stability showed that the micellar solutions maintain their sizes at room temperature for 1 month without distinct aggregation or dissociation. ICG derivative was much better photostable after being entrapped by the new carrier. The prepared FSMCO micelles displayed a good drug loading content (11.7%), entrapment efficiency (66.5%) and sustained release rate for the model drug fluorescein. The copolymers demonstrated weeny cytotoxicity toward Bel-7402, L02 and A549 cells when incubated for 2 d. Ligands modified micelles endowed preferable cell targeting capability and beautiful cell inhibition of HCPT-FSMCO on Bel-7402 tumor cells. | 5 | Clinical factors associated with composition of lung microbiota and important taxa predicting clinical prognosis in patients with severe community-acquired pneumonia显示文摘Few studies have described the key features and prognostic roles of lung microbiota in patients with severe community-acquired pneumonia(SCAP).We prospectively enrolled consecutive SCAP patients admitted to ICU.Bronchoscopy was performed at bedside within 48 h of ICU admission,and 16S rRNA gene sequencing was applied to the collected bronchoalveolar lavage fluid.The primary outcome was clinical improvements defined as a decrease of 2 categories and above on a 7-category ordinal scale within 14 days following bronchoscopy.Sixty-seven patients were included.Multivariable permutational multivariate analysis of variance found that positive bacteria lab test results had the strongest independent association with lung microbiota(R2=0.033;P=0.018),followed by acute kidney injury(AKI;R2=0.032;P=0.011)and plasma MIP-1βlevel(R2=0.027;P=0.044).Random forest identified that the families Prevotellaceae,Moraxellaceae,and Staphylococcaceae were the biomarkers related to the positive bacteria lab test results.Multivariable Cox regression showed that the increase inα-diversity and the abundance of the families Prevotellaceae and Actinomycetaceae were associated with clinical improvements.The positive bacteria lab test results,AKI,and plasma MIP-1βlevel were associated with patients’lung microbiota composition on ICU admission.The families Prevotellaceae and Actinomycetaceae on admission predicted clinical improvements. | Sisi Du Xiaojing Wu Binbin Li Yimin Wang Lianhan Shang Xu Huang Yudi Xia Donghao Yu Naicong Lu Zhibo Liu Chunlei Wang Xinmeng Liu Zhujia Xiong Xiaohui Zou Binghuai Lu Yingmei Liu Qingyuan Zhan Bin Cao | 2022 | Frontiers of Medicine2022,16,3: | 2 |

| 7 | Trend Analysis and Intervention Effect Starting Point Detection of COVID-19 Epidemics Using Recalibrated Time Series Models—Worldwide,2020显示文摘Objective:This study aimed to identify a model for short-term coronavirus disease 2019(COVID-19)trend prediction and intervention evaluation.Methods:We compared the autoregressive integrated moving average(ARIMA)model and Holt exponential smoothing(Holt)model on predicting the number of cumulative COVID-19 cases in China.Based on the mean absolute percentage error(MAPE)value,the optimal model was selected and further tested using data from the United States,Italy and Republic of Korea.The intervention effect starting time points and abnormal trend changes were detected by observing the pattern of differences between the predicted and real trends.Results:The recalibrated ARIMA model with a 5-day prediction time span has the best model performance with MAPEs ranged between 2%and 5%.The intervention effects started to show on February 7 in the mainland of China,March 5 in Republic of Korea and April 27 in Italy,but have not been detected in the US as of May 19.Temporary abnormal trends were detected in Korea and Italy,but the overall epidemic trends were stable since the effect starting points.Conclusion:The recalibrated ARIMA model can detect the intervention effects starting points and abnormal trend changes;thus to provide valuable information support for epidemic trend analysis and intervention evaluation. | Shu Li Chen Chen Shuyin Cao Kejia Hu Hao Lei Xiaolin Xu Qinchuan Wang Changzheng Yuan Sicong Wang Sisi Wang Junlin Jia Yuanqing Ye Xifeng Wu | 2021 | China CDC weekly2021,3,20: | 0 |

| 9 | Performance of Hepatitis Delta Virus(HDV)RNA Testing for the Diagnosis of Active HDV Infection:Systematic Review and Meta-analysis显示文摘Background and Aims:Hepatitis delta virus(HDV)is a defective virus and causes severe liver disease.Several HDV RNA assays have been developed,however the diagnostic efficacy remains unclear.This systematic review and metaanalysis aims to evaluate the diagnostic accuracy of HDV RNA assays to aid in the diagnosis of active hepatitis D.Methods:The PubMed,Embase,and Cochrane Library databases were systematically searched from the beginning to June 31,2022.Information on the characteristics of the literature and data on sensitivity,specificity,and area under curve(AUC)of the receiver operating characteristic(ROC)were extracted.Stata 14.0 was used for meta-analysis of the combined sensitivity,specificity,positive likelihood ratio,and negative likelihood ratio.Results:A total of 10 studies were included in the meta-analysis.The summary sensitivity,specificity,positive likelihood ratio,negative likelihood ratio,and diagnostic odds ratio of HDV RNA assays for HDV diagnosis were 0.92(95%CI:0.87-0.95),0.90(95%CI:0.86-0.93),7.74(95%CI:5.31-11.29),0.10(95%CI:0.06-0.18)and 99.90(95%CI:47.08-211.99),respectively.The AUC of the pooled ROC curve was 0.95(95%CI:0.92-0.96).Conclusions:The results show that HDV RNA assays had high diagnostic performance.However,that is limited by the number and quality of studies.Standard protocols for the development of assays by manufacturers and larger studies on the use of the assays are needed. | Sisi Chen Xiangying Zhang Ling Xu Yuan Tian Zihao Fan Yaling Cao Zhenzhen Pan Yao Gao Sujun Zheng Zhongping Duan Mei Liu Feng Ren | 2023 | Journal of Clinical and Translational Hepatology2023,11,6: | 0 |
| 10 | Efficacy of acupuncture on repair of glial scars in rats with traumatic brain injury显示文摘OBJECTIVE:To explore the underlying mechanism of acupuncture on nerve repair by investigating its effect on the differentiation of glial cells and the repair of glial scars.METHODS:Sprague-Dawley rats were randomly allocated to three groups:normal group,model group,and acupuncture group.Acupuncture was applied at Renzhong(GV26),Baihui(GV20),Fengfu(GV16),Yamen(GV15)and Hegu(LI4)within 12 h after TBI modeling with a frequency of one session per day for 4 weeks.Neurobehavioral assessment,hematoxylin and eosin staining,immunofluorescence detection,and magnetic resonance imaging scanning were performed on days 3,7,14,and 28 after modeling of traumatic brain injury(TBI).RESULTS:Acupuncture promoted the proliferation of glial cells and glial scars at an early stage but inhibited the proliferation of glial cells and glial scars at a late stage.Morphological observations and immunofluorescence histochemistry showed that the morphology of the perilesional cortex in the acupuncture group was improved and the number of neurons was increased when compared with the model group.The lesion size of ipsilateral brain parenchyma in the acupuncture group was smaller than in the model group on days 7,14,and 28(P<0.05)after TBI modeling.CONCLUSIONS:Acupuncture might have a bidirectional regulatory effect on glial scar repair after TBI by promoting the proliferation of glial cells and glial scars to limit the injured area and relieve nerve injury during the early stages,and by inhibiting glial scar hyperplasia to benefit the regeneration and repair of neurons and axons and promote neurological function recovery during the later stages. | ZENG Hai CAO Luxi PANG Zhao ZHAO Sisi WANG Shiqi LIN Zhuowen CHEN Minan LIN Shujun ZHANG Yimin | 2023 | Journal of Traditional Chinese Medicine2023,43,2: | 0 |

| 12 | Eliminating nitrogen chemisorption barrier with single-atom supported yttrium cluster via electronic promoting effect for highly efficient ammonia synthesis显示文摘Nitrogen chemisorption is a prerequisite for efficient ammonia synthesis under ambient conditions,but promoting this process remains a significant challenge.Here,by loading yttrium clusters onto a single-atom support,an electronic promoting effect is triggered to successfully eliminate the nitrogen chemisorption barrier and achieve highly efficient ammonia synthesis.Density functional theory calculations reveal that yttrium clusters with abundant electron orbitals can provide considerable electrons and greatly promote electron backdonation to the N2 antibonding orbitals,making the chemisorption process exothermic.Moreover,according to the“hot atom”mechanism,the energy released during exothermic N2 chemisorption would benefit subsequent N2 cleavage and hydrogenation,thereby dramatically reducing the energy barrier of the overall process.As expected,the proof-of-concept catalyst achieves a prominent NH3 yield rate of 48.1μg·h^(−1)·mg^(−1)at−0.2 V versus the reversible hydrogen electrode,with a Faradaic efficiency of up to 69.7%.This strategy overcomes one of the most serious obstacles for electrochemical ammonia synthesis,and provides a promising method for the development of catalysts with high catalytic activity and selectivity. | Yuzhuo Jiang Mengfan Wang Sisi Liu Lifang Zhang Siyi Qian Yufeng Cao Yu Cheng Tao Qian Chenglin Yan | 2023 | Nano Research2023,16,2: | 0 |

| 14 | Authigenic clay minerals and calcite dissolution influence reservoir quality in tight sandstones:Insights from the central Junggar Basin,NW China显示文摘Authigenic clays and calcite cements are important in the development of reservoir tightness and the formation of hydrocarbon sweet spots.We investigated Jurassic low-permeability sandstone reservoirs in the central Junggar Basin,NW China,using petrography,mineralogy,porosity,and permeability assessment,and stable C and O isotope analysis to ascertain the influence of authigenic clays and calcites on reservoir quality.Here,we establish the properties and diagenetic processes of the reservoir sandstones,and construct a generalizable model of reservoir quality.The results show that the sandstones are mainly litharenite and feldspathic litharenite and can be classified into ductile-lithic-rich sandstones and ductile-lithic-poor sandstones according to rock composition.The ductile-lithic-rich sandstones are tight(mean porosity?7.31%;mean permeability?0.08 mD)as a result of intense compaction.In contrast,the ductile-lithic-poor sandstones can be classified into five types according to diagenetic process.The formation of favorable hydrocarbon reservoir properties is closely related to the presence of authigenic clays and dissolution of calcite.In particular,kaolinite fills intergranular pores,thereby blocking pore space and reducing reservoir quality.Chlorite coating resists compaction and limits the formation of quartz overgrowths,thereby preserving pore space and enhancing reservoir quality.Calcite controls reservoir quality through both precipitation and dissolution.Calcite precipitation results in reduced reservoir quality,whereby early calcites that were precipitated in formation water resist compaction and provide the basis for subsequent dissolution and late precipitation,whereas dissolution of calcite in mesodiagenesis improves reservoir quality.A generalized model is formulated by relating diagenetic facies types to depth and porosity,providing a reference for other similar reservoirs.Our data suggest that deeply buried tight sandstones can be exploration prospects under favorable conditions involving the presence of authigenic clays and dissolution of calcite,as in the central Junggar Basin of this study. | Dongdong Hong Jian Cao Tao Wu Sisi Dang Wenxuan Hu Suping Yao | 2020 | Energy Geoscience2020,1,1: | 0 |
