Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes, downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. Finally, Kindlin-2 loss increases osteocyte apoptosis and impairs osteocyte spreading and dendrite formation. Thus, we demonstrate an important role of Kindlin-2 in the regulation of bone homeostasis and provide a potential target for the treatment of metabolic bone diseases. | Huiling Cao Qinnan Yan Dong Wang Yumei Lai Bo Zhou Qi Zhang Wenfei Jin Simin Lin Yiming Lei Liting Ma Yuxi Guo Yishu Wang Yilin Wang Xiaochun Bai Chuanju Liu Jian QFeng Chuanyue Wu Di Chen Xu Cao Guozhi Xiao | 2020 | Bone Research2020,8,1: | 8
Potassium Uptake and Transport in Apple Roots Under Drought Stress显示文摘As one of the most important mineral nutrient elements, potassium(K^+) plays an important role in many plant physiological processes and determines both the yield and quality of crops. There are two typical gene families that regulate K^+transport in higher plants, including K^+channels and K^+transporters. However, little is known about how these channels and transporters divide their work in response to drought stress. In this study, the hydroponic experiment was conducted on Malus hupehensis. The K^+content was found to decrease in response to drought stress in M. hupehensis, the aboveground decreased by 34.15% and the underground decreased by 3.97%. Meanwhile, the root morphology change was detected by scanning the root system. Under conditions of drought, the genes encoding K^+transporters were upregulated including MdCHX1.3, MdCHX4.11, MdCHX4.8, MdCHX4.9, Md HKT1, and MdHAK3.2. The net influx of K^+was inhibited by 19.47% with the action of K^+channel inhibitors(CsCl), however a significant decrease(80.99%; P < 0.05) was found in roots exposed to a PM H+-ATPase(orthovanadate) inhibitor by utilizing a non-invasive micro-test technique. The trend of H+efflux was similar to that of K^+. The data suggested that the positive influx of K^+through the transporter accounted for the main K^+uptake under drought stress. These results suggest that we can improve the uptake of K^+by purposely up-regulating specific K^+transporters under drought stress. This process may improve growth, yield, quality, and stress tolerance in apple trees. | Jianguo Qi Simin Sun Lin Yang Mingjun Li Fengwang Ma Yangjun Zou | 2019 | Horticultural Plant Journal2019,5,1: | 6
Clinical manifestations and pathogen characteristics in children admitted for suspected COVID-19显示文摘Coronavirus disease 2019(COVID-19),which is caused by severe acute respiratory syndrome coronavirus-2(SARS-CoV-2),has spread around the world. However, approaches to distinguish COVID-19 from pneumonia caused by other pathogens have not yet been reported. We retrospectively analyzed the clinical data of 97 children with probable COVID-19.A total of 13(13.4%)patients were confirmed positive for SARS-CoV-2 infection by nucleic acid RT-PCR testing, and 41(42.3%)patients were found to be infected with other pathogens.Notably,no pathogen was detected in 43(44.3%)patients.Among all patients,25(25.8%)had familial cluster exposure history, and 52(53.6%)had one or more coexisting conditions.Fifteen(15.5%)patients were admitted or transferred to the PICU.In the 11 confirmed COVID-19 cases,5(45.5%)and 7(63.6%)were positive for IgM and IgG against SARS-CoV-2,respectively.In 22 patients with suspected COVID-19,1(4.5%)was positive for IgG but negative for IgM.The most frequently detected pathogen was Mycoplasma pneumonia(29,29.9%).One patient with confirmed COVID-19 died. Our results strongly indicated that the detection of asymptomatic COVID-19 or coexisting conditions must be strengthened in pediatric patients. These cases may be difficult to diagnose as COVID-19 unless etiologic analysis is conducted.A serologic test can be a useful adjunctive diagnostic tool in cases where SARS-CoV-2 infection is highly suspected but the nucleic acid test is negative. | Xiaofang Cai Hanlan Jiang Simin Zhang Shengying Xia Wenhui Du Yaoling Ma Tao Yu Wenbin Li | 2020 | Frontiers of Medicine2020,14,6: | 2
Tectonic features, genetic mechanisms and basin evolution of the eastern Doseo Basin,Chad显示文摘The features of the unconformity, fault and tectonic inversion in the eastern Doseo Basin,Chad, were analyzed, and the genetic mechanisms and basin evolution were discussed using seismic and drilling data. The following results are obtained.First, four stratigraphic unconformities,i.e.basement(Tg),Mangara Group(T10),lower Upper Cretaceous(T5)and Cretaceous(T4),four faulting stages,i.e.Barremian extensional faults,Aptian–Coniacian strike-slip faults,Campanian strike-slip faults, and Eocene strike-slip faults, and two tectonic inversions,i.e.Santonian and end of Cretaceous, were developed in the Doseo Basin.Second, the Doseo Basin was an early failed intracontinental passive rift basin transformed by the strike-slip movement and tectonic inversion. The initial rifting between the African and South American plates induced the nearly N-S stretching of the Doseo Basin, giving rise to the formation of the embryonic Doseo rift basin. The nearly E-W strike-slip movement of Borogop(F1)in the western section of the Central African Shear Zone resulted in the gradual cease of the near north-south rifting and long-term strike-slip transformation, forming a dextral transtension fault system with inherited activity but gradually weakened in intensity(interrupted by two tectonic inversions). This fault system was composed of the main shear(F1),R-type shear(F2-F3)and P-type shear(F4-F5)faults,with the strike-slip associated faults as branches. The strike-slip movements of F1 in Cretaceous and Eocene were controlled by the dextral shear opening of the equatorial south Atlantic and rapid expanding of the Indian Ocean, respectively. The combined function of the strike-slip movement of F1 and the convergence between Africa and Eurasia made the Doseo Basin underwent the Santonian dextral transpressional inversion characterized by intensive folding deformation leading to the echelon NE-SW and NNE-SSW nose-shaped uplifts and unconformity(T5)on high parts of the uplifts. The convergence between Africa and Eurasia caused the intensive tectonic inversion of Doseo Basin at the end of Cretaceous manifesting as intensive uplift, denudation and folding deformation, forming the regional unconformity(T4)and superposing a nearly E-W structural configuration on the Santonian structures.Third, the Doseo Basin experienced four evolutional stages with the features of short rifting and long depression,i.e.Barremian rifting,Aptian rifting–depression transition,Albian–Late Cretaceous depression, and Cenozoic extinction, under the control of the tectonic movements between Africa and its peripheral plates. | GAO Huahua DU Yebo WANG Lin GAO Simin HU Jie BAI Jianfeng MA Hong WANG Yuhua ZHANG Xinshun LIU Hao | 2023 | Petroleum Exploration and Development2023,50,5: | 0
Detection and classification of breast lesions using multiple information on contrast-enhanced mammography by a multiprocess deep-learning system: A multicenter study显示文摘Objective: Accurate detection and classification of breast lesions in early stage is crucial to timely formulate effective treatments for patients. We aim to develop a fully automatic system to detect and classify breast lesions using multiple contrast-enhanced mammography(CEM) images. Methods: In this study, a total of 1,903 females who underwent CEM examination from three hospitals were enrolled as the training set, internal testing set, pooled external testing set and prospective testing set. Here we developed a CEM-based multiprocess detection and classification system(MDCS) to perform the task of detection and classification of breast lesions. In this system, we introduced an innovative auxiliary feature fusion(AFF)algorithm that could intelligently incorporates multiple types of information from CEM images. The average freeresponse receiver operating characteristic score(AFROC-Score) was presented to validate system’s detection performance, and the performance of classification was evaluated by area under the receiver operating characteristic curve(AUC). Furthermore, we assessed the diagnostic value of MDCS through visual analysis of disputed cases, comparing its performance and efficiency with that of radiologists and exploring whether it could augment radiologists’ performance. Results: On the pooled external and prospective testing sets, MDCS always maintained a high standalone performance, with AFROC-Scores of 0.953 and 0.963 for detection task, and AUCs for classification were 0.909[95% confidence interval(95% CI): 0.822-0.996] and 0.912(95% CI: 0.840-0.985), respectively. It also achieved higher sensitivity than all senior radiologists and higher specificity than all junior radiologists on pooled external and prospective testing sets. Moreover, MDCS performed superior diagnostic efficiency with an average reading time of 5 seconds, compared to the radiologists’ average reading time of 3.2 min. The average performance of all radiologists was also improved to varying degrees with MDCS assistance. Conclusions: MDCS demonstrated excellent performance in the detection and classification of breast lesions, and greatly enhanced the overall performance of radiologists. | Yuqian Chen Zhen Hua Fan Lin Tiantian Zheng Heng Zhou Shijie Zhang Jing Gao Zhongyi Wang Huafei Shao Wenjuan Li Fengjie Liu Simin Wang Yan Zhang Feng Zhao Hao Liu Haizhu Xie Heng Ma Haicheng Zhang Ning Mao | 2023 | Chinese Journal of Cancer Research2023,35,4: | 0
Chemo-drugs in cell microparticles reset antitumor activity of macrophages by activating lysosomal P450 and nuclear hnRNPA2B1显示文摘Macrophages in tumors(tumor-associated macrophages,TAMs),a major population within most tumors, play key homeostatic functions by stimulating angiogenesis, enhancing tumor cell growth, and suppressing antitumor immunity.Resetting TAMs by simple, efficacious and safe approach(s)is highly desirable to enhance antitumor immunity and attenuate tumor cell malignancy.Previously,we used tumor cell-derived microparticles to package chemotherapeutic drugs(drug-MPs),which resulted in a significant treatment outcome in human malignant pleural effusions via neutrophil recruitments, implicating that drug-MPs might reset TAMs,considering the inhibitory effects of M2 macrophages on neutrophil recruitment and activation. Here,we show that drug-MPs can function as an antitumor immunomodulator by resetting TAMs with M1 phenotype and IFN-βrelease.Mechanistically,drug molecules in tumor MPs activate macrophage lysosomal P450 monooxygenases, resulting in superoxide anion formation, which further amplifies lysosomal ROS production and pH value by activating lysosomal NOX2.Consequently,lysosomal Ca^(2+)signaling is activated, thus polarizing macrophages towards M1.Meanwhile,the drug molecules are delivered from lysosomes into the nucleus where they activate DNA sensor hnRNPA2B1 for IFN-βproduction.This lysosomal-nuclear machinery fully arouses the antitumor activity of macrophages by targeting both lysosomal pH and the nuclear innate immunity. These findings highlight that drug-MPs can act as a new immunotherapeutic approach by revitalizing antitumor activity of macrophages. This mechanistic elucidation can be translated to treat malignant ascites by drug-MPs combined with PD-1 blockade. | Keke Wei Huafeng Zhang Shuaishuai Yang Yuxiao Cui Bingxia Zhang Jincheng Liu Liang Tang Yaoyao Tan Simin Liu Shiqi Chen Wu Yuan Xiao Luo Chen Chen Fei Li Junwei Liu Jie Chen Pingwei Xu Jiadi Lv Ke Tang Yi Zhang Jingwei Ma Bo Huang | 2023 | Signal Transduction and Targeted Therapy2023,8,2: | 0
Improving current and mitigating energy loss in ternary organic photovoltaics enabled by two well-compatible small molecule acceptors显示文摘Ternary organic photovoltaic(OPV)strategy is an effective but facile approach to enhance the photovoltaic performance for single-junction devices.Herein,a series of ternary OPVs were fabricated by employing a wide bandgap donor(PBDB-TF)and two acceptor-donor-acceptor(A-D-A)-type nonfullerene small molecule acceptors(NF-SMAs,called F-2 Cl and 3 TT-OCIC).As the third component, the near-infrared SMA,3 TT-OCIC,has complementary absorption spectrum, narrow bandgap and wellcompatible crystallization property to the host acceptor(F-2 Cl)for efficient ternary OPVs.With these, the optimal ternary devices yield significantly enhanced power conversion efficiency of 15.23%,one of the very few examples with PCE higher than15%other than Y6 systems. This is mainly attributed to the increased short-circuit current density of 24.92 m A cm^(-2) and dramatically decreased energy loss of 0.53 e V.This work presents a successful example for simultaneously improving current, minimizing energy loss and together with modifying the morphology of active layers in OPVs,which will contribute to the further construction of high performance ternary OPVs. | Yanna Sun Huan-Huan Gao Simin Wu Lingxian Meng Xiangjian Wan Mengyang Li Zaifei Ma Ziqi Guo Shitong Li Hongtao Zhang Chenxi Li Yongsheng Chen | 2021 | Science China Chemistry2021,64,4: | 0
Metabolic Pathway of Monounsaturated Lipids Revealed by In-Depth Structural Lipidomics by Mass Spectrometry显示文摘The study of lipid metabolism relies on the characterization of the lipidome, which is quite complex due to the structure variations of the lipid species.New analytical tools have been developed recently for characterizing fine structures of lipids, with C=C location identification as one of the major improvements. In this study,we studied the lipid metabolism reprograming by analyzing glycerol phospholipid compositions in breast cancer cell lines with structural specification extended to the C=C location level.Inhibition of the lipid desaturase,stearoyl-CoA desaturase 1,increased the proportion of n-10 isomers that are produced via an alternative fatty acid desaturase 2 pathway. However, there were different variations of the ratio of n-9/n-7 isomers in C18:1-containing glycerol phospholipids after stearoyl-CoA desaturase 1 inhibition, showing increased tendency in MCF-7 cells,MDA-MB-468 cells, and BT-474 cells, but decreased tendency in MDA-MB-231 cells. No consistent change of the ratio of n-9/n-7 isomers was observed in SK-BR-3 cells. This type of heterogeneity in reprogrammed lipid metabolism can be rationalized by considering both lipid desaturation and fatty acid oxidation, highlighting the critical roles of comprehensive lipid analysis in both fundamental and biomedical applications. | Simin Cheng Donghui Zhang Jiaxin Feng Qingyuan Hu Aolei Tan Zhuoning Xie Qinhua Chen Huimin Huang Ying Wei Zheng Ouyang Xiaoxiao Ma | 2023 | Research2023,,4: | 0
DT-Net:Joint Dual-Input Transformer and CNN for Retinal Vessel Segmentation显示文摘Retinal vessel segmentation in fundus images plays an essential role in the screening, diagnosis, and treatment of many diseases. The acquired fundus images generally have the following problems:uneven illumination, high noise, and complex structure. It makes vessel segmentation very challenging.Previous methods of retinal vascular segmentation mainly use convolutional neural networks on U Network(U-Net)models,and they have many limitations and shortcomings, such as the loss of microvascular details at the end of the vessels. We address the limitations of convolution by introducing the transformer into retinal vessel segmentation. Therefore,we propose a hybrid method for retinal vessel segmentation based on modulated deformable convolution and the transformer, named DT-Net.Firstly,multi-scale image features are extracted by deformable convolution and multi-head selfattention(MHSA).Secondly, image information is recovered, and vessel morphology is refined by the proposed transformer decoder block. Finally, the local prediction results are obtained by the side output layer. The accuracy of the vessel segmentation is improved by the hybrid loss function.Experimental results show that our method obtains good segmentation performance on Specificity(SP),Sensitivity(SE),Accuracy(ACC),Curve(AUC),and F1-score on three publicly available fundus datasets such as DRIVE,STARE,and CHASE_DB1. | Wenran Jia Simin Ma Peng Geng Yan Sun | 2023 | Computers, Materials & Continua2023,76,9: | 0
分化型甲状腺癌患者停用甲状腺素后短期甲减对血脂的影响(英文)显示文摘Objective: The aim of the study was to investigate the effects of short-term overt hypothyroidism on lipids after thyroxine-withdrawal in patients with iatrogenically induced hypothyroidism before radioiodine treatment for differentiated thyroid carcinoma (DTC). Methods: Thirty patients with a history of differentiated thyroid carcinoma on thyroid-stimulating hormone (TSH)-suppressive thyroxine replacement therapy were studied. Blood sample were taken before and 4 weeks after withdrawal of thyroxine substitution. Venous blood was drawn after an overnight fast and analyzed for serum free T4 (FT4), free T3 (FT3), thyroid-stimulating hormone (TSH), total cholesterol (TC), triglycerides (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) and apolipoprotein A1 (ApoA1), apolipoprotein B (ApoB). Fifty healthy people matched for age and gender were controls. Their blood samples were taken only once. Results: After thyroxine-withdrawal, the patients presented with increased serum TSH and low serum FT4 and FT3 levels compared with controls. Serum TG, TC, LDL, HDL, ApoB and Lp(a) increased after thyroxine withdrawal, reaching statistical significant differences with previous evaluation. However, when compared to euthyroid controls, only TC, LDL and ApoB were increased when patients were hypothyroidism. No changes were observed in ApoA1 in patients during thyroxine withdrawal, or when comparing the values observed in patients to those of euthyroid controls. Conclusion: TG, TC, LDL, HDL, ApoB and Lp(a) were increased during short-term overt hypothyroidism. | Hongyu Wu Shuyao Zuo Chao Ma Bin Liu Fengyu Wu Simin Liu Qin Zhang Xifu Lan | 2009 | The Chinese-German Journal of Clinical Oncology2009,8,11: | 0
TC-Fuse: A Transformers Fusing CNNs Network for Medical Image Segmentation显示文摘In medical image segmentation task, convolutional neural networks(CNNs)are difficult to capture long-range dependencies, but transformers can model the long-range dependencies effectively. However, transformers have a flexible structure and seldom assume the structural bias of input data,so it is difficult for transformers to learn positional encoding of the medical images when using fewer images for training. To solve these problems,a dual branch structure is proposed. In one branch,Mix-Feed-Forward Network(Mix-FFN)and axial attention are adopted to capture long-range dependencies and keep the translation invariance of the model.Mix-FFN whose depth-wise convolutions can provide position information is better than ordinary positional encoding. In the other branch, traditional convolutional neural networks(CNNs)are used to extract different features of fewer medical images. In addition, the attention fusion module BiFusion is used to effectively integrate the information from the CNN branch and Transformer branch, and the fused features can effectively capture the global and local context of the current spatial resolution. On the public standard datasets Gland Segmentation(GlaS),Colorectal adenocarcinoma gland(CRAG)and COVID-19 CT Images Segmentation, the F1-score,Intersection over Union(IoU)and parameters of the proposed TC-Fuse are superior to those by Axial Attention U-Net,U-Net,Medical Transformer and other methods. And F1-score increased respectively by 2.99%,3.42%and 3.95%compared with Medical Transformer. | Peng Geng Ji Lu Ying Zhang Simin Ma Zhanzhong Tang Jianhua Liu | 2023 | Computer Modeling in Engineering & Sciences2023,,11: | 0
Classification of Hailstone Trajectories in a Hail Cloud over a Semi-Arid Region in China显示文摘The growth trajectory of hailstones in clouds determines the ground intensity and spatial distribution of hailfall.A systematic study of hail trajectories can help improve the current scientific understanding of the mechanisms by which hail forms in semi-arid regions of China and,in doing so,improve the quality of hail forecasts and warnings and help to prevent and mitigate disasters. In this study, the WRFv3.7.1 model was employed to provide the background field to drive the hailstone trajectory model.Cluster analysis was then used to classify hail trajectories to investigate the characteristics of different types of hail trajectories and the microphysical characteristics of hail formation. The differences in hail trajectories might be mainly due to differences in the background flow fields and microphysical fields of hail clouds in different regions.Comparative analysis revealed that as the maximum particle size of ground hailfall increased, the maximum supercooled cloud water content and the maximum updraft velocity for the formation and growth of hailstone increased. The larger the size when the hailstone reaches its maximum height, the larger the ground hailstone formed.Overall, the formation and growth of hailstone are caused by the joint action of the dynamical flow field and cloud microphysical processes. The physical processes of hailstone growth and main growth regions differ for different types of hail trajectories. Therefore, different catalytic schemes should be adopted in artificial hail prevention operations for different hail clouds and trajectories due to differences in hail formation processes and ground hailfall characteristics. | Xiaoli LIU Kerui MIN Jianren SANG Simin MA | 2023 | Advances in Atmospheric Sciences2023,40,10: | 0
