Antidiabetic Effects of Gegen Qinlian Decoction via the Gut Microbiota Are Attributable to Its Key Ingredient Berberine显示文摘Gegen Qinlian Decoction(GQD),a traditional Chinese medicine(TCM)formula, has long been used for the treatment of common metabolic diseases, including type 2 diabetes mellitus. However, the main limitation of its wider application is ingredient complexity of this formula. Thus,it is critically important to identify the major active ingredients of GQD and to illustrate mechanisms underlying its action. Here,we compared the effects of GQD and berberine,a hypothetical key active pharmaceutical ingredient of GQD,on a diabetic rat model by comprehensive analyses of gut microbiota,short-chain fatty acids, proinflammatory cytokines, and ileum transcriptomics. Our results show that berberine and GQD had similar effects on lowering blood glucose levels, modulating gut microbiota, inducing ileal gene expression,as well as relieving systemic and local inflammation. As expected, both berberine and GQD treatment significantly altered the overall gut microbiota structure and enriched many butyrate-producing bacteria, including Faecalibacterium and Roseburia, thereby attenuating intestinal inflammation and lowering glucose.Levels of short-chain fatty acids in rat feces were also significantly elevated after treatment with berberine or GQD.Moreover, concentration of serum proinflammatory cytokines and expression of immune-related genes, including Nfkb1,Stat1,and Ifnrg1,in pancreatic islets were significantly reduced after treatment. Our study demonstrates that the main effects of GQD can be attributed to berberine via modulating gut microbiota. The strategy employed would facilitate further standardization and widespread application of TCM in many diseases. | Xizhan Xu Zezheng Gao Fuquan Yang Yingying Yang Liang Chen Lin Han Na Zhao Jiayue Xu Xinmiao Wang Yue Ma Lian Shu Xiaoxi Hu Na Lyu Yuanlong Pan Baoli Zhu Linhua Zhao Xiaolin Tong Jun Wang | 2020 | Genomics, Proteomics & Bioinformatics2020,18,6: | 29
Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further, there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients. Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer. Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks. The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response, time to progression, disease control rate, and safety. Results:Of 179 patients screened,125 were eligible and received RC48 treatment. The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively. The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety, suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy. | Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen | 2021 | Cancer Communications2021,41,11: | 29

Arbuscular Mycorrhizal Fungal Inoculation Enhances Suppression of Cucumber Fusarium Wilt in Greenhouse Soils显示文摘A pot experiment was conducted to study the plant growth and fruit yields of cucumber (Cucumis sativus L.) on a greenhouse soil with or without inoculation of arbuscular mycorrhizal fungi (AMFs) and Fusarium oxysporum f. sp. cucu- merinum under unsterilized conditions. Two AMF inocula were tested: only one AMF strain Glomus caledonium 90036 and an AMF consortium mainly consisting of Glomus spp. and Acaulospora spp. There were four treatments including no inoculation (control), inoculation with F. oxysporum but without mycorrhizae (FO), inoculation with F. oxysporum and G. caledonium (FO+M1), and inoculation with F. oxysporum and the AMF consortium (FO+M2). Cucumber plants were harvested at weeks 3 and 9 after transplanting. Compared with the control, the FO treatment without AMF inoculation had less biomass both at weeks 3 and 9 (P < 0.05) and had higher incidence of Fusarium wilt and produced no cucumber fruit at week 9. Both FO+M1 and FO+M2 treatments had higher mycorrhizal colonization than the treatments which received no AMF inoculation at week 3 (P < 0.05), but only the FO+M2 treatment elevated plant biomass, decreased the incidence of Fusarium wilt, and improved cucumber yields to the same level as the control at week 9. The results indicated that the AMF consortium could suppress Fusarium wilt of cucumber and, therefore, showed potential as a biological control agent in greenhouse agroecosystems. | HU Jun-Li LIN Xian-Gui WANG Jun-Hua SHEN Wei-Shou WU Shu PENG Su-Ping MAO Ting-Ting | 2010 | Pedosphere2010,20,5: | 23

Special Section:SARS-CoV-2 Preliminary evidence from a multicenter prospective observational study of the safety and efficacy of chloroquine for the treatment of COVID-19显示文摘Effective therapies are urgently needed for the SARS-CoV-2 pandemic.Chloroquine has been proved to have antiviral effect against coronavirus in vitro. In this study,we aimed to assess the efficacy and safety of chloroquine with different doses in COVID-19.In this multicenter prospective observational study,we enrolled patients older than 18 years old with confirmed SARS-CoV-2 infection excluding critical cases from 12 hospitals in Guangdong and Hubei Provinces.Eligible patients received chloroquinephosphate s00 mg,orally, once(half dose)or twice(full dose)daily.Patients treated with non-chloroquine therapy were included as historical controls. The primary endpoint is the time to undetectable viral RNA.Secondary outcomes include the proportion of patients with undetectable viral RNA by day 10 and 14,hospitalization time, duration of fever, and adverse events.A total of 197 patients completed chloroquine treatment, and 176patients were included as historical controls. The median time to achieve an undetectable viral RNA was shorter in chloroquine than in non-chloroquine(absolute difference in medians-6.0 days;95%CI-6.0 to—4.0).The duration of fever is shorter in chloroquine(geometric mean ratio 0.6;95%CIO.5 to 0.8).No serious adverse events were observed in the chloroquine group.Patients treated with half dose experienced lower rate of adverse events than with full dose.Although randomized trials are needed for further evaluation, this study provides evidence for safety and efficacy of chloroquine in COVID-19 and suggests that chloroquine can be a cost-effective therapy for combating the COVID-19 pandemic. | Mingxing Huang Man Li Fei Xiao Pengfei Pang Jiabi Liang Tiantian Tang Shaoxuan Liu Binghui Chen Jingxian Shu Yingying You Yang Li Meiwen Tang Jianhui Zhou Guanmin Jiang Jingfen Xiang Wenxin Hong Songmei He Zhaoqin Wang Jianhua Feng Changqing Lin Yinong Ye Zhilong Wu Yaocai Li Bei Zhong Ruilin Sun Zhongsi Hong Jing Liu Huili Chen Xiaohua Wang Zhonghe Li Duanqing Pei Lin Tian Jinyu Xia Shanping Jiang Nanshan Zhong Hong Shan | 2020 | National Science Review2020,7,9: | 20
Pretreatment HIV drug resistance in adults initiating antiretroviral therapy in China,2017显示文摘Background:After the scale-up of antiretroviral therapy(ART)for HIV infected people, increasing numbers of patients have pretreatment drug resistance(PDR).In this study, the prevalence of PDR was evaluated in adults initiating antiretroviral therapy in China. Methods:Blood samples were obtained from 1943 patients who initiated antiretroviral therapy(ART)in 2017 from 13 provinces or cities in China.Pol sequences were used to analyze drug resistance and construct transmission networks.Logistic regression model was used to estimate the potential factors associated with PDR.Results:In total,1711 eligible patients(76.0%male;87.8%aged≥25 years)were included,of which 117(6.8%)had PDR.The highest rates of PDR were 12.2%in Liangshan Prefecture of Sichuan and 9.3 and 8.9%in Dehong and Lincang Prefecture of Yunnan.A multivariate logistic regression analysis revealed that PDR was significantly higher among intravenous drug users(adjusted Odds Ratio(aOR)=2.64,95%CI:1.57–4.44)and individuals from Liangshan,Dehong, and Lincang(aOR=2.04,95%CI:1.26–3.30).In total,754 sequences were used to generate 164 transmission networks.Five transmission networks had two or three sequences containing the same mutations, two networks contained subjects from Liangshan, and one network contained subjects from Dehong. Conclusions:Overall, the PDR prevalence was moderate, with a particularly high prevalence in areas with severe HIV epidemics. These results indicate the importance of continuous PDR monitoring in patients initiating antiretroviral therapy. | Rui-Hua Kang Shu-Jia Liang Yan-Ling Ma Shu Liang Lin Xiao Xin-Hui Zhang Hong-Yan Lu Xiao-Qin Xu Shui-Bin Luo Xiao-Guang Sun Lin Chen Jian-Mei He Guo-Hui Wu Ling-Jie Liao Hui Xing Yi-Ming Shao Yu-Hua Ruan | 2020 | Infectious Diseases of Poverty2020,9,3: | 17
Association of Dietary Pattern during Pregnancy and Gestational Diabetes Mellitus: A Prospective Cohort Study in Northern China显示文摘Objective To examine the association of maternal dietary patterns during pregnancy with gestational diabetes mellitus(GDM) in northern China. Methods The dietary intakes of pregnant women were recorded twice by 24‐hour dietary recalls for three days prior to having been diagnosed with GDM, at 5‐15 and 24‐28 gestational weeks, respectively.GDM was diagnosed, and serum glycosylated hemoglobin(HbA1c) was measured at 24‐28 weeks.Dietary patterns were assessed by factor analysis. The association of the dietary pattern with GDM and HbA1c was examined by multiple logistic models. Results Of 753 participants, 64(8.5%) were diagnosed with GDM. Four dietary patterns were identified: Western pattern(dairy, baked/fried food and white meat), traditional pattern(light‐colored vegetables, fine grain, red meat and tubers), mixed pattern(edible fungi, shrimp/shellfish and red meat)and prudent pattern(dark‐colored vegetables and deep‐sea fish). Compared with the prudent pattern, both the Western pattern and the traditional pattern were associated with an increased risk of GDM(a OR = 4.40, 95% CI: 1.58‐12.22; a OR = 4.88, 95% CI: 1.79‐13.32) and a high level of HbA1c(a OR = 12.37,95% CI: 1.47‐103.91; a OR = 26.23, 95% CI: 2.54‐270.74). Compared to the lowest quartile(Q), Q3 of the Western pattern scores and Q3‐Q4 of the traditional pattern scores were associated with a higher risk of GDM.Conclusion The consumption of the Western pattern or the traditional pattern during pregnancy may increase the risk of GDM. | DU Hong Yi JIANG Hong O Karmin CHEN Bo XU Lin Ji LIU Shu Ping YI Jian Ping HE Geng Sheng QIAN Xu | 2017 | Biomedical and Environmental Sciences2017,30,12: | 16
Grape Seed Proanthocyanidin Extract Alleviates Arsenic-induced Oxidative Reproductive Toxicity in Male Mice显示文摘Objective To determine the ability of grape seed proanthocyanidin extract(GSPE) in alleviating arsenic-induced reproductive toxicity. Methods Sixty male Kunming mice received the following treatments by gavage: normal saline solution(control); arsenic trioxide(ATO; 4 mg/kg); GSPE(400 mg/kg); ATO+GSPE(100 mg/kg); ATO+GSPE(200 mg/kg) and ATO+GSPE(400 mg/kg). Thereafter, the mice were sacrificed and weighed, and the testis was examined for pathological changes. Nuclear factor(erythroid-derived 2)-like 2(Nrf2), heme oxygenase 1(HO1), glutathione S-transferase(GST), NAD(P)H dehydrogenase, and quinone 1(NQO1) expression in the testis was detected by real-time PCR. Superoxide dismutase(SOD), glutathione(GSH), total antioxidative capability(T-AOC), malondialdehyde(MDA), 8-hydroxydeoxyguanosine(8-OHd G), and reproductive indexes were analyzed. Results ATO-treated mice showed a significantly decreased sperm count and testis somatic index and activity levels of SOD, GSH, and T-AOC than control group. Compared to the ATO-treated group, ATO +GSPE group showed recovery of the measured parameters. Mice treated with ATO+high-dose GSPE showed the highest level of m RNA expression of Nrf2, HO, NQO1, and GST. Conclusion GSPE alleviates oxidative stress damage in mouse testis by activating Nrf2 signaling, thus counteracting arsenic-induced reproductive toxicity. | LI Shu Gang DING Yu Song NIU Qiang XU Shang Zhi PANG Li Juan MA Ru Lin JING Ming Xia FENG Gang Ling LIU Jia Ming GUO Shu Xia | 2015 | Biomedical and Environmental Sciences2015,28,4: | 14
A comparative study of natural and experimental nano-sized grinding grain textures in rocks显示文摘In three shallow-level fault systems in eastern China, nano-sized (30―100 nm) grinding grain textures are found in the thin-shelled rheological layer representing frictional-viscous high strain field. The surface layer near slipping plane is composed of stacks of grinding grains with high sphericity and uniform nano-sized diameter, while the underlying layer appears as a mixture of irregular ones with diverse diameters. Rock deformation experiments indicate that the grinding grain texture is a common phenomenon in shearing fractures, suggesting a potential transition from sliding friction to rolling one during the rock deformation process. This transition is crucial for many tribological processes in geo- logical fields. | SUN Yan SHU LiangShu LU XianCai LIU Hao ZHANG XiHui K. | 14 | THE EFFECTS OF RADIATION STRESS ON WAVE HEIGHTS AND SEA LEVEL IN THE INTERACTION OF COUPLED WAVE-TIDE-SURGE IN THE COASTAL AREA显示文摘Based on coastal high resolution (2′×2′) coupled wave tide surge interaction numerical model, the effects of radiation stress on wave heights and sea level in the coastal area of Huanghe Delta were studied. By comparisons of simulated and measured wave heights and sea level for two mediately strong weather cases, it is demonstrated that the results simulated by coupled wave tide surge model are closer to the measured and particularly in excellent agreement in the extreme values of the wave heights and set up. This study shows that the radiation stress can increase the wave heights maximally to 67cm and sea level to 40cm. It is also found that there are areas of over 50cm wave height increase and an area of over 20cm sea level increase in the Huanghe Delta coastal area, and this find may be very important in engineering. For this reason, it is suggested that in the practical engineering application, the coupled wave tide surge interaction numerical model should be prefered. | Lin Xiang, Yin Bao shu, Hou Yi jun, Su Jing zhi, Cheng Ming hua Institute of Oceanology, Chinese Academy of Sciences, Qingdao, 266071, China | 2003 | Journal of Hydrodynamics2003,15,1: | 12

Postoperative complications in patients with portal vein thrombosis after liver transplantation:Evaluation with Doppler ultrasonography显示文摘AIM: To study the postoperative complications in patients with preoperative portal vein thrombosis (PVT) undergoing liver transplantation (LT) and to evaluate the complications with Doppler ultrasonography. METHODS: Retrospective studies were performed on 284 patients undergoing LT (286 LT) with respect to pre-and postoperative clinical data and Doppler ultrasonography. According to the presence and grade of preoperative PVT, 286 LTs were divided into three groups: complete PVT (c-PVT), partial PVT (p-PVT) and non-PVT, with 22, 30 and 234 LTs, respectively. Analyses were carried out to compare the incidence of early postoperative complications. RESULTS: PVT, inferior vena cava (IVC) thrombosis, hepatic artery thrombosis (HAT) and biliary complications were found postoperatively. All complications were detected by routine Doppler ultrasonography and diagnoses made by ultrasound were confirmed by clinical data or/and other imaging studies. Nine out of 286 LTs had postoperative PVT. The incidence of the c-PVT group was 22.7%, which was higher than that of the p-PVT group (3.3%, P < 0.05) and non-PVT group (1.3%, P < 0.005). No difference was found between the p-PVT and non-PVT groups (P > 0.25). Of the 9 cases with postoperative PVT, recanalizations were achieved in 7 cases after anticoagulation under the guidance of ultrasound, 1 case received portal vein thrombectomy and 1 case died of acute injection. Ten LTs had postoperative IVC thrombosis. The c-PVT group had a higher incidence of IVC thrombosis than the non-PVT group (9.1% vs 2.6%, P < 0.05); no significant difference was found between either the c-PVT and p-PVT groups (9.1% vs 6.7%, P > 0.5) or between the p-PVT and non-PVT groups (P > 0.25). Nine cases with IVC thrombosis were cured by anticoagulation under the guidance of ultrasound, and 1 case gained natural cure without any medical treatment after 2 mo. HAT was found in 2 non-PVT cases, giving a rate of 0.7% among 286 LTs. Biliary complications were seen in 12 LTs. The incidence of biliary complications in the c-PVT, p-PVT and non-PVT groups was 9.1%, 3.3% and 4.3%, respectively (P > 0.25 for all), among which 2 stenosis led retransplantations and others were controlled by relative therapy. CONCLUSION: C-PVT patients tend to have a higher incidence of PVT and IVC thrombosis than non-PVT patients after LT. The incidence of postoperative complications in p-PVT patients does not differ from that of non-PVT patients. A relatively low incidence of HAT was seen in our study. Doppler ultrasonography is a convenient and efficient method for detecting posttransplant complications and plays an important role in guiding treatment. | Yi-Ping Jia Qiang Lu Shu Gong Bu-Yun Ma Xiao-Rong Wen Yu-Lan Peng Ling Lin Hong-Yan Chen Li Qiu Yan Luo | 2007 | World Journal of Gastroenterology2007,13,34: | 10
