1Rapid quantification of semen hepatitis B virus DNA by real-time polymerase chain reaction显示文摘AIM: To examine the sensitivity and accuracy of real-time polymerase chain reaction (PCR) for the quantification of hepatitis B virus (HBV) DNA in semen.METHODS: Hepatitis B viral DNA was isolated from HBV carriers' semen and sera using phenol extraction method and QTAamp DNA blood mini kit (Qiagen, Germany). HBV DNA was detected by conventional PCR and quantified by TaqMan technology-based real-time PCR (quantitative polymerase chain reaction (qPCR)). The detection threshold was 200 copies of HBV DNA for conventional PCR and 10 copies of HBV DNA for real time PCR per reaction.RESULTS: Both methods of phenol extraction and QIAamp DNA blood mini kit were suitable for isolating HBV DNA from semen. The value of the detection thresholds was 500 copies of HBV DNA per mL in the semen. The viral loads were 7.5×107 and 1.67×107 copies of HBV DNA per mL in two HBV infected patients' sera, while 2.L4×105 and 3.02×105 copies of HBV DNA per mL in the semen.CONCLUSION: Real-time PCR is a more sensitive and accurate method to detect and quantify HBV DNA in the semen.Wei-Ping Qian Yue-Qiu Tan Ying Chen Ying Peng Zhi Li Guang-Xiu Lu Made C. 2ACSL4 is a predictive biomarker of sorafenib sensitivity in hepatocellular carcinoma显示文摘Sorafenib is the first-line treatment of advanced hepatocellular carcinoma(HCC).However,there is a lack of validated biomarkers to predict sorafenib sensitivity.In this study we investigated the role of ACSL4,a positive-activating enzyme of ferroptosis,in sorafenib-induced cell death and HCC patient outcome.We showed that ACSL4 protein expression was negatively associated with IC50 values of sorafenib in a panel of HCC cell lines(R=−0.952,P<0.001).Knockdown of ACSL4 expression by specific siRNA/sgRNA significantly attenuated sorafenib-induced lipid peroxidation and ferroptosis in Huh7 cells,and also rescued sorafenib-induced inhibition of xenograft tumor growth in vivo.We selected 29 HCC patients with surgery as primary treatment and sorafenib as postoperative adjunct therapy from a hospital-based cohort.A high proportion(66.7%)of HCC patients who had complete or partial responses to sorafenib treatment(according to the revised RECIST guideline)had higher ACSL4 expression in the pretreated HCC tissues,compared with those who had stable or progressed tumor growth(23.5%,P=0.029).Since ACSL4 expression was independent of sorafenib treatment,it could serve as a useful predictive biomarker.Taken together,this study demonstrates that ACSL4 is essential for sorafenib-induced ferroptosis and useful for predicting sorafenib sensitivity in HCC.This study may have important translational impacts in precise treatment of HCC.Ji Feng Pei-zhi Lu Guang-zhi Zhu Shing Chung Hooi Yong Wu Xiao-wei Huang Hui-qi Dai Pan-hong Chen Zhong-jie Li Wen-jing Su Chuang-ye Han Xin-ping Ye Tao Peng Jing Zhou Guo-dong Lu 2021Acta Pharmacologica Sinica2021,42,1:11

6Prognostic value of alcohol consumptionand some other dietary habits for survival in a cohort of Chinese men with lung cancer显示文摘Background:Alcohol consumption and some other dietary habits are thought to be associated with lung cancer incidence.However,the effects of these habits on lung cancer prognosis have been studied rarely.The purpose of this study was to address these gaps in knowledge.Methods:We studied a cohort of 1052 Chinese men in Hong Kong who were diagnosed with primary lung cancer.Cox proportional hazards models were used to determine the prognostic values of consumption of alcohol,fresh fruits or vegetables,meat,and fried or preserved food.Results:Compared with never drinkers,men who drank alcohol 1-3 days per week had a more favorable lung cancer prognosis(hazard ratio[HR]:0.82,95%confidence interval[CI]0.68-0.97);however,this survival advantage was not significant in men who drank alcohol more frequently(HR:0.91,95%CI 0.73-1.14).Compared with men who consumed preserved or fried food only occasionally,men who consumed these foods frequently had a higher risk of lung cancer mortality(HR:1.20,95%CI 1.00-1.42).Conclusions:Occasional consumption of alcohol was a favorable survival factor for Chinese men with lung cancer.However,this survival benefit did not exist for frequent drinkers of alcohol.Chinese men with lung cancer who were frequent consumers of fried or preserved food had a worse prognosis than those who consumed these foods only occasionally.Wentao Li Lap Ah Tse Joseph S.K.Au Kai Shing Yu Feng Wang Ignatius Tak-sun Yu 2017Chinese Journal of Cancer2017,36,7:3
7Brain delivering RNA-based therapeutic strategies by targeting mTOR pathway for axon regeneration after central nervous system injury显示文摘Injuries to the central nervous system(CNS)such as stroke,brain,and spinal cord trauma often result in permanent disabilities because adult CNS neurons only exhibit limited axon regeneration.The brain has a surprising intrinsic capability of recovering itself after injury.However,the hostile extrinsic microenvironment significantly hinders axon regeneration.Recent advances have indicated that the inactivation of intrinsic regenerative pathways plays a pivotal role in the failure of most adult CNS neuronal regeneration.Particularly,substantial evidence has convincingly demonstrated that the mechanistic target of rapamycin(mTOR)signaling is one of the most crucial intrinsic regenerative pathways that drive axonal regeneration and sprouting in various CNS injuries.In this review,we will discuss the recent findings and highlight the critical roles of mTOR pathway in axon regeneration in different types of CNS injury.Importantly,we will demonstrate that the reactivation of this regenerative pathway can be achieved by blocking the key mTOR signaling components such as phosphatase and tensin homolog(PTEN).Given that multiple mTOR signaling components are endogenous inhibitory factors of this pathway,we will discuss the promising potential of RNA-based therapeutics which are particularly suitable for this purpose,and the fact that they have attracted substantial attention recently after the success of coronavirus disease 2019 vaccination.To specifically tackle the blood-brain barrier issue,we will review the current technology to deliver these RNA therapeutics into the brain with a focus on nanoparticle technology.We will propose the clinical application of these RNA-mediated therapies in combination with the brain-targeted drug delivery approach against mTOR signaling components as an effective and feasible therapeutic strategy aiming to enhance axonal regeneration for functional recovery after CNS injury.Ming-Xi Li Jing-Wen Weng Eric S.Ho Shing Fung Chow Chi Kwan Tsang 2022Neural Regeneration Research2022,17,10:2
