2Rapid quantification of semen hepatitis B virus DNA by real-time polymerase chain reaction显示文摘AIM: To examine the sensitivity and accuracy of real-time polymerase chain reaction (PCR) for the quantification of hepatitis B virus (HBV) DNA in semen.METHODS: Hepatitis B viral DNA was isolated from HBV carriers' semen and sera using phenol extraction method and QTAamp DNA blood mini kit (Qiagen, Germany). HBV DNA was detected by conventional PCR and quantified by TaqMan technology-based real-time PCR (quantitative polymerase chain reaction (qPCR)). The detection threshold was 200 copies of HBV DNA for conventional PCR and 10 copies of HBV DNA for real time PCR per reaction.RESULTS: Both methods of phenol extraction and QIAamp DNA blood mini kit were suitable for isolating HBV DNA from semen. The value of the detection thresholds was 500 copies of HBV DNA per mL in the semen. The viral loads were 7.5×107 and 1.67×107 copies of HBV DNA per mL in two HBV infected patients' sera, while 2.L4×105 and 3.02×105 copies of HBV DNA per mL in the semen.CONCLUSION: Real-time PCR is a more sensitive and accurate method to detect and quantify HBV DNA in the semen.Wei-Ping Qian Yue-Qiu Tan Ying Chen Ying Peng Zhi Li Guang-Xiu Lu Made C. 4ACSL4 is a predictive biomarker of sorafenib sensitivity in hepatocellular carcinoma显示文摘Sorafenib is the first-line treatment of advanced hepatocellular carcinoma(HCC).However,there is a lack of validated biomarkers to predict sorafenib sensitivity.In this study we investigated the role of ACSL4,a positive-activating enzyme of ferroptosis,in sorafenib-induced cell death and HCC patient outcome.We showed that ACSL4 protein expression was negatively associated with IC50 values of sorafenib in a panel of HCC cell lines(R=−0.952,P<0.001).Knockdown of ACSL4 expression by specific siRNA/sgRNA significantly attenuated sorafenib-induced lipid peroxidation and ferroptosis in Huh7 cells,and also rescued sorafenib-induced inhibition of xenograft tumor growth in vivo.We selected 29 HCC patients with surgery as primary treatment and sorafenib as postoperative adjunct therapy from a hospital-based cohort.A high proportion(66.7%)of HCC patients who had complete or partial responses to sorafenib treatment(according to the revised RECIST guideline)had higher ACSL4 expression in the pretreated HCC tissues,compared with those who had stable or progressed tumor growth(23.5%,P=0.029).Since ACSL4 expression was independent of sorafenib treatment,it could serve as a useful predictive biomarker.Taken together,this study demonstrates that ACSL4 is essential for sorafenib-induced ferroptosis and useful for predicting sorafenib sensitivity in HCC.This study may have important translational impacts in precise treatment of HCC.Ji Feng Pei-zhi Lu Guang-zhi Zhu Shing Chung Hooi Yong Wu Xiao-wei Huang Hui-qi Dai Pan-hong Chen Zhong-jie Li Wen-jing Su Chuang-ye Han Xin-ping Ye Tao Peng Jing Zhou Guo-dong Lu 2021Acta Pharmacologica Sinica2021,42,1:11

7复发-缓解型多发性硬化边缘系统白质纤维束定量DTI改变显示文摘目的探讨复发一缓解型多发性硬化(relapsing remitting multiples clerosis,RRMS)患者边缘系统白质纤维束定量弥散张量的改变。方法RRMS患者24例,年龄和性别相匹配的健康志愿者24例为对照进行MRI扫描,获取常规MRI图像和弥散张量成像(DTI)图像,经GE工作站处理后进行测量分析,比较2组边缘系统主要纤维束(扣带束、穹窿束和终纹束)不同部分各向异性指数(FA)值和平均弥散系数(MD)值的变化。结果在考虑年龄和性别因素的影响后,RRMS组患者穹窿柬(F一15.605,P=0.000135)、右侧穹窿/终纹束(F=15.772,P=0.000127)、左侧穹窿/终纹束(F=8.53,P=0.004)的FA值较健康对照组明显降低;RRMS组患者穹窿束(F=13.28,P=0.0004)、右侧终纹束(F=10.943,p=0.002)、右侧穹窿/终纹束(F=7.326,P=0.008)的MD值较健康对照组明显增高;RRMS组患者左/右侧的前、后扣带束、右侧终纹束(F=0.986,P=0.326)、左侧终纹束的FA和对照组比较无明显差异,RRMS组患者左/右侧的前、后扣带束、左侧终纹束、左侧穹窿/终纹束的MD值较健康对照比较差异均无统计学意义。结论DTI可作为反映RRMS边缘系统纤维束病理性改变的较敏感的检测方法。周福庆 Chi-- Shing Zee 龚洪翰 Mark Shiroishi 2010实用放射学杂志2010,26,3:7

9Application of flash nanoprecipitation to fabricate poorly water-soluble drug nanoparticles显示文摘Nanoparticles are considered to be a powerful approach for the delivery of poorly watersoluble drugs. One of the main challenges is developing an appropriate method for preparation of drug nanoparticles. As a simple, rapid and scalable method, the flash nanoprecipitation(FNP) has been widely used to fabricate these drug nanoparticles, including pure drug nanocrystals, polymeric micelles,polymeric nanoparticles, solid lipid nanoparticles, and polyelectrolyte complexes. This review introduces the application of FNP to produce poorly water-soluble drug nanoparticles by controllable mixing devices, such as confined impinging jets mixer(CIJM), multi-inlet vortex mixer(MIVM) and many other microfluidic mixer systems. The formation mechanisms and processes of drug nanoparticles by FNP are described in detail. Then, the controlling of supersaturation level and mixing rate during the FNP process to tailor the ultrafine drug nanoparticles as well as the influence of drugs, solvent, anti-solvent, stabilizers and temperature on the fabrication are discussed. The ultrafine and uniform nanoparticles of poorly watersoluble drug nanoparticles prepared by CIJM, MIVM and microfluidic mixer systems are reviewed briefly. 15Overview of the Shenzhen Emergency Medical Service Call Pattern显示文摘BACKGROUND:In Shenzhen,the Emergency Medical Service(EMS) system has been in service since 1997.This study aims to examine the operation of Shenzhen 120 EMS center and to identify the reasons of calling EMS.METHODS:In this retrospective quantitative descriptive study,the data from the Shenzhen 120 EMS registry in 2011 were analyzed.RESULTS:Shenzhen 120 EMS center is a communication command center.When the number of 120 are dialed,it is forwarded to the closest appropriate hospital for ambulance dispatch.In2011,the Shenzhen 120 EMS center received 153 160 ambulance calls,with an average of 420 calls per day.Calling emergency services was mainly due to traffic accidents.Trauma and other acute diseases constituted a majority of ambulance transports.The adult patients aged 15-60 years are the principal users of EMS.There are no recognized 'paramedic' doctors and nurses.The pre-hospital emergency service is under the operation of emergency departments of hospitals.Shenzhen at present does not have specialized pre-hospital training for doctors and nurses in posttrauma management.Moreover,specialized pre-hospital training,financial support,and public health education on proper use of EMS should be emphasized.CONCLUSION:The Shenzhen 120 EMS center has its own epidemiology characteristics.Traumatic injury and traffic accident are the main reasons for calling ambulance service.In-depth study emphasizing the distribution and characteristics of trauma patients is crucial to the future development of EMS.Shuk Man Lo Yi Min Yu Lap Yip Larry Lee Mi Ling Eliza Wong Sek Ying Chair Edward J Kalinowski Tak Shing Jimmy Chan 2012World Journal of Emergency Medicine2012,3,4:4

17Identification of Ca2+ signaling components in neural stem/progenitor cells during differentiation into neurons and glia in intact and dissociated zebrafish neurospheres显示文摘The development of the CNS in vertebrate embryos involves the generation of different sub-types of neurons and glia in a complex but highly-ordered spatio-temporal manner. Zebrafish are commonly used for exploring the development, plasticity and regeneration of the CNS, and the recent development of reliable protocols for isolating and culturing neural stem/progenitor cells(NSCs/NPCs) from the brain of adult fish now enables the exploration of mechanisms underlying the induction/specification/differentiation of these cells. Here, we refined a protocol to generate proliferating and differentiating neurospheres from the entire brain of adult zebrafish. We demonstrated via RT-qPCR that some isoforms of ip3 r, ryr and stim are upregulated/downregulated significantly in differentiating neurospheres, and via immunolabelling that 1,4,5-inositol trisphosphate receptor(IP3 R) type-1 and ryanodine receptor(RyR) type-2 are differentially expressed in cells with neuron-or radial glial-like properties. Furthermore, ATP but not caffeine(IP3 R and RyR agonists, respectively), induced the generation of Ca^(2+) transients in cells exhibiting neuron-or glial-like morphology. These results indicate the differential expression of components of the Ca^(2+) -signaling toolkit in proliferating and differentiating cells. 19Surgical outcomes for unilateral superior oblique palsy in Chinese population: a retrospective study显示文摘AIM: To evaluate the outcome after surgery for unilateral superior oblique(SO) palsy in Chinese.METHODS: The medical records of 39 patients that underwent surgery for unilateral SO palsy between January 2003 and December 2012 at Caritas Medical Centre, Hong Kong, were retrospectively reviewed. All surgeries were performed by a single surgeon. Pre-operative assessments for vertical deviation, cyclo-deviation, and Knapp's classification were obtained to determine the nature and degree of surgical correction.Vertical deviation was measured at 1wk; 1, 6mo and on last follow-up day post-operatively. Cyclo-deviation was measured on last follow-up day post-operatively.RESULTS: During the 10 y period, 39 subjects were recruited. The most common etiology was congenital(94.9%). Knapp's Type III(66.7%) and Type I(12.8%)classifications were the most common subtypes. To treat SO palsy, the most common surgical procedures were:isolated inferior oblique(IO) anteriorization(41.0%),isolated IO myectomy(10.3%), and isolated IO recession(10.3%). At 3.5 ±2.1y post-operatively, the vertical deviation was significantly reduced(15.1 ±6.2 PD versus0.5±1.4 PD, P <0.0001) without significant improvement in cyclo-deviation(P =0.5). Initial vertical deviation was correlated with cyclo-torsion(r =0.4, P =0.007). Those with over-correction had greater initial vertical deviation(19.4±7.2 PD versus 13.2±4.3 PD, P =0.003). After a single operation, 84.6% of subjects achieved a vertical deviation within ±3 PD.· CONCLUSION: The majority of subjects achieved corrected vertical deviation after a single surgery although there was no improvement in cyclo-deviation.Those with over-correction of primary position deviation had greater preoperative vertical deviation and it may be related to simultaneous multiple muscle surgery.Gordon Shing Kin Yau Victor Tak Yau Tam Jacky Wai Yip Lee Theo Tak Kwong Chan Can Yin Fun Yuen 2015International Journal of Ophthalmology(English edition)2015,8,1:4
