| 8 | The Group A Streptococcus Interleukin-8 Protease SpyCEP Promotes Bacterial Intracellular Survival by Evasion of Autophagy显示文摘Autophagy serves an innate immune function in defending the host against invading bacteria,including group A Streptococcus(GAS).Autophagy is regulated by numerous host proteins,including the endogenous negative regulator calpain,a cytosolic protease.Globally disseminated serotypeM1T1 GAS strains associated with high invasive disease potential express numerous virulence factors and resist autophagic clearance.Upon in vitro infection of human epithelial cell lines with representative wild-type GAS M1T1 strain 5448(M1.5448),we observed increased calpain activation linked to a specific GAS virulence factor,the interleukin-8 protease SpyCEP.Calpain activation inhibited autophagy and decreased capture of cytosolic GAS in autophagosomes.In contrast,the serotype M6 GAS strain JRS4(M6.JRS4),which is highly susceptible to host autophagy-mediated killing,expresses low levels of SpyCEP and does not activate calpain.Overexpression of SpyCEP in M6.JRS4 stimulated calpain activation,inhibited autophagy,and significantly decreased bacterial capture in autophagosomes.These paired loss-and gain-of-function studies reveal a novel role for the bacterial protease SpyCEP in enabling GAS M1 evasion of autophagy and host innate immune clearance. | RenéBergmann Giuseppe Gulotta Federica Andreoni Tomoko Sumitomo Shigetada Kawabata Annelies S.Zinkernagel Gursharan S.Chhatwal Victor Nizet Manfred Rohde Satoshi Uchiyama | 2022 | Infectious Microbes & Diseases2022,4,3: | 1 |

| 16 | Long-term survival of salivary streptococci on dental devices made of ethylene vinyl acetate显示文摘Bacterial infection associated with the use of medical or dental devices is a serious concern.Although devices made of ethylene vinyl acetate(EVA) are often used in the oral cavity,there are no established standards for their storage.We investigated bacterial survival on EVA sheets under various storage conditions to establish a standard for hygienic storage of such dental devices.Bacterial counts were evaluated,which showed a significant decrease after washing with sterilized water,mechanical brushing and rinsing,and using Mouthguard Cleaner as compared to untreated samples.In addition,no bacteria were detected on samples stored 2 days or longer in a ventilated environment,whereas they were detected for up to 14 days on samples without any cleaning stored in a closed environment.Bacterial counts for the untreated samples gradually declined,while surviving bacteria on samples treated with sterilized water and mechanical brushing showed a rapid decrease.Additionally,bacterial identification using polymerase chain reaction(PCR) revealed that Streptococcus oralis was dominantly detected on salivary samples after 14 days of storage among both two subjects.For effective hygienic storage of dental devices made of EVA,washing with sterilized water is important to remove absorbed salivary compounds along with storage in a ventilated environment. | Taiji Ogawa Sayaka Yamasaki Mariko Honda Yutaka Terao Shigetada Kawabata Yoshinobu Maeda | 2012 | International Journal of Oral Science2012,4,1: | 0 |
