1Development of Mining Technology and Equipment for Seafloor Massive Sulfide Deposits显示文摘Seafloor massive sulfide(SMS) deposits which consist of Au, Ag, Cu, and other metal elements, have been a target of commercial mining in recent decades. The demand for established and reliable commercial mining system for SMS deposits is increasing within the marine mining industry. The current status and progress of mining technology and equipment for SMS deposits are introduced. First, the mining technology and other recent developments of SMS deposits are comprehensively explained and analyzed.The seafloor production tools manufactured by Nautilus Minerals and similar mining tools from Japan for SMS deposits are compared and discussed in turn. Second, SMS deposit mining technology research being conducted in China is described, and a new SMS deposits mining tool is designed according to the environmental requirement. Finally, some new trends of mining technology of SMS deposits are summarized and analyzed. 3Advances in BeiDou Navigation Satellite System(BDS)and satellite navigation augmentation technologies显示文摘Several noteworthy breakthroughs have been made with the BeiDou Navigation Satellite System(BDS)and other global navigation satellite systems as well as the associated augmentation systems,such as the commissioning of the BDS-3 preliminary system and the successful launch of the first BDS-3 GEO satellite which carries the satellite-based augmentation payload.Presently,BDS can provide basic services globally,and its augmentation system is also being tested.This paper gives an overview of BDS and satellite navigation augmentation technologies.This overview is divided into four parts,which include the system segment technologies,satellite segment technologies,propagation segment technologies,and user segment technologies.In each part,these technologies are described from the perspectives of preliminary information,research progress,and summary.Moreover,the significance and progress of the BeiDou Satellite-based Augmentation System(BDSBAS),low earth orbit augmentation,and the national BeiDou ground-based augmentation system are presented,along with the airborne-based augmentation system.Furthermore,the conclusions and discussions covering popular topics for research,frontiers in research and development,achievements,and suggestions are listed for future research.Rui Li Shuaiyong Zheng Ershen Wang Jinping Chen Shaojun Feng Dun Wang Liwen Dai 2020Satellite Navigation2020,1,1:9

5A high-quality genome sequence of alkaligrass provides insights into halophyte stress tolerance显示文摘Alkaligrass(Puccinellia tenuiflora) is a monocotyledonous halophytic forage grass widely distributed in Northern China. It belongs to the Gramineae family and shares a close phylogenetic relationship with the cereal crops, wheat and barley. Here, we present a high-quality chromosome-level genome sequence of alkaligrass assembled from Illumina, Pac Bio and 10× Genomics reads combined with genome-wide chromosome conformation capture(Hi-C) data. The ~1.50 Gb assembled alkaligrass genome encodes 38,387 protein-coding genes, and 54.9% of the assembly are transposable elements, with long terminal repeats being the most abundant. Comparative genomic analysis coupled with stress-treated transcriptome profiling uncovers a set of unique saline-and alkaline-responsive genes in alkaligrass. The high-quality genome assembly and the identified stress related genes in alkaligrass provide an important resource for evolutionary genomic studies in Gramineae and facilitate further understanding of molecular mechanisms underlying stress tolerance in monocotyledonous halophytes. 8Isobaric Tags for Relative and Absolute Quantificationbased Comparative Proteomics Reveals the Features of Plasma Membrane-Associated Proteomes of Pollen Grains and Pollen Tubes from Lilium davidii显示文摘Mature pollen grains (PGs) from most plant species are metabolically quiescent. However, once pollinated onto stigma, they quickly hydrate and germinate. A PG can give rise to a vegetative cell-derived polarized pollen tube (PT), which represents a specialized polar cell. The polarized PT grows by the tip and requires interaction of different signaling molecules localized in the apical plasma membrane and active membrane trafficking. The mechanisms underlying the interaction and membrane trafficking are not well understood. In this work, we purified PG and PT plasma-membrane vesicles from Lilium davidii Duch. using the aqueous two-phase partition technique, then enriched plasma membrane proteins by using Brij58 and KCl to remove loosely bound contaminants. We identified 223 integral and membrane-associated proteins in the plasma membrane of PGs and PTs by using isobaric tags for relative and absolute quantification (iTRAQ) and 2-D high-performance liquid chromatography-tandem mass spectrometry. More than 68% of the proteins have putative transmembrane domains and/or lipid-modified motifs. Proteins involved in signal transduction, membrane trafficking and transport are predominant in the plasma-membrane proteome. We revealed most components of the clathrin-dependent endocytosis pathway. Statistical analysis revealed 14 proteins differentially expressed in the two development stages: in PTs, six upregulated and eight downregulated are mainly involved in signaling, transport and membrane trafficking. 15Overexpression of PvWOX3a in switchgrass promotes stem development and increases plant height显示文摘Switchgrass(Panicum virgatum L.)is an important perennial,noninvasive,tall ornamental grass that adds color and texture to gardens and landscapes.Moreover,switchgrass has been considered a forage and bioenergy crop because of its vigorous growth,low-input requirements,and broad geography.Here,we identified PvWOX3a from switchgrass,which encodes a WUSCHEL-related homeobox transcription factor.Transgenic overexpression of PvWOX3a in switchgrass increased stem length,internode diameter,and leaf blade length and width,all of which contributed to a 95%average increase in dry weight biomass compared with control plants.Yeast one-hybrid and transient dual-luciferase assays showed that PvWOX3a can repress the expression of gibberellin 2-oxidase and cytokinin oxidase/dehydrogenase through apparently direct interaction with their promoter sequences.These results suggested that overexpression of PvWOX3a could increase gibberellin and cytokinin levels in transgenic switchgrass plants,which promotes cell division,elongation,and vascular bundle development.We also overexpressed PvWOX3a in a transgenic miR156-overexpressing switchgrass line that characteristically exhibited more tillers,thinner internodes,and narrower leaf blades.Double transgenic switchgrass plants displayed significant increases in internode length and diameter,leaf blade width,and plant height but retained a tiller number comparable to that of plants expressing miR156 alone.Ultimately,the double transgenic switchgrass plants produced 174%more dry-weight biomass and 162%more solubilized sugars on average than control plants.These findings indicated that PvWOX3a is a viable potential genetic target for engineering improved shoot architecture and biomass yield of horticulture,fodder,and biofuel crops.Ruijuan Yang Zhenying Wu Chen Bai Zhichao Sun Mengqi Wang Yuzhu Huo Hailing Zhang Yamei Wang Huapeng Zhou Shaojun Dai Wenwen Liu Chunxiang Fu 2021Horticulture Research2021,8,1:1

18Genome-wide identification and characterization of ACBP gene family in Populus reveal salinity alkali-responsive profiles显示文摘Acyl-CoA-binding proteins(ACBPs)are important for the transport of acyl groups for macro molecular biosynthesis involved in plant growth,development,and diverse stress(e.g.,cold,drought,salinity,and heavy metals)responses.Here,we report the phylogeny and characteristics of the ACBP family in the woody plant Populus trichocarpa.Eight genes encoding ACBP proteins were identified,and they are distributed on eight chromosomes in P.trichocarpa.These PtACBP genes were divided into four subgroups according to gene structure,conserved motifs and phylogenetic relationship.Promoter analysis revealed that cis-elements were related to stress response,phytohormone response,and physical and reproductive growth regulation.Expression levels of PtACBP genes varied among different organs,with the highest expression in leaves and the lowest in stems.Quantitative real-time PCR(qRT-PCR)analysis showed that under salinity-alkali stresses(i.e.,200 mM NaCl,75 mM Na2CO3,and 100 mM NaHCO3),four(PtACBP1,PtACBP3,PtACBP4 and PtACBP8)of eight PtACBP genes were significantly induced in roots and leaves.These data provide a comprehensive analysis of the ACBPs family in P.trichocarpa,which could be useful for gene function analyses.Yu Chang Xinru Xu Hongxia Zheng Hao Xie Bo Li Sixue Chen Ying Li Shaojun Dai 2023Journal of Forestry Research2023,34,2:0
19Cell type-specific proteomics uncovers a RAF15-SnRK2.6/OST1 kinase cascade in guard cells显示文摘Multicellular organisms such as plants contain various cell types with specialized functions.Analyzing the characteristics of each cell type reveals specific cell functions and enhances our understanding of organization and function at the organismal level.Guard cells(GCs)are specialized epidermal cells that regulate the movement of the stomata and gaseous exchange,and provide a model genetic system for analyzing cell fate,signaling,and function.Several proteomics analyses of GC are available,but these are limited in depth.Here we used enzymatic isolation and flow cytometry to enrich GC and mesophyll cell protoplasts and perform in-depth proteomics in these two major cell types in Arabidopsis leaves.We identified approximately 3,000 proteins not previously found in the GC proteome and more than600 proteins that may be specific to GC.The depth of our proteomics enabled us to uncover a guard cell-specific kinase cascade whereby Raf15and Snf1-related kinase2.6(SnRK2.6)/OST1(open stomata 1)mediate abscisic acid(ABA)-induced stomatal closure.RAF15 directly phosphorylated SnRK2.6/OST1 at the conserved Ser175 residue in its activation loop and was sufficient to reactivate the inactive form of SnRK2.6/OST1.ABAtriggered SnRK2.6/OST1 activation and stomatal closure was impaired in raf15 mutants.We also showed enrichment of enzymes and flavone metabolism in GC,and consistent,dramatic accumulation of flavone metabolites.Our study answers the long-standing question of how ABA activates SnRK2.6/OST1 in GCs and represents a resource potentially providing further insights into the molecular basis of GC and mesophyll cell development,metabolism,structure,and function.Hongliang Wang Yubei Wang Tian Sang Zhen Lin Rongxia Li Weiwei Ren Xin Shen Bing Zhao Xiao Wang Xuebin Zhang Shaoqun Zhou Shaojun Dai Honghong Hu Chun-Peng Song Pengcheng Wang 2023Journal of Integrative Plant Biology2023,65,9:0
