8Transcriptomic Profiling of Human Pluripotent Stem Cell-derived Retinal Pigment Epithelium over Time显示文摘Human pluripotent stem cell(h PSC)-derived progenies are immature versions of cells,presenting a potential limitation to the accurate modelling of diseases associated with maturity or age.Hence,it is important to characterise how closely cells used in culture resemble their native counterparts.In order to select appropriate time points of retinal pigment epithelium(RPE)cultures that reflect native counterparts,we characterised the transcriptomic profiles of the h PSC-derived RPE cells from 1-and 12-month cultures.We differentiated the human embryonic stem cell line H9 into RPE cells,performed single-cell RNA-sequencing of a total of 16,576 cells to assess themolecular changes of the RPE cells across these two culture time points.Our results indicate the stability of the RPE transcriptomic signature,with no evidence of an epithelial–mesenchymal transition,and with the maturing populations of the RPE observed with time in culture.Assessment of Gene Ontology pathways revealed that as the cultures age,RPE cells upregulate expression of genes involved in metal binding and antioxidant functions.This might reflect an increased ability to handle oxidative stress as cells mature.Comparison with native human RPE data confirms a maturing transcriptional profile of RPE cells in culture.These results suggest that long-term in vitro culture of RPE cells allows the modelling of specific phenotypes observed in native mature tissues.Our work highlights the transcriptional landscape of h PSC-derived RPE cells as they age in culture,which provides a reference for native and patient samples to be benchmarked against.Grace E.Lidgerwood Anne Senabouth Casey J.A.Smith-Anttila Vikkitharan Gnanasambandapillai Dominik C.Kaczorowski Daniela Amann-Zalcenstein Erica L.Fletcher Shalin H.Naik Alex W.Hewitt Joseph E.Powell Alice Pebay 2021Genomics, Proteomics & Bioinformatics2021,19,2:1

19Three-dimensional dynamic characterization of square-nosed slugging phenomena in a fluidized bed显示文摘Slugging represents one of the major regimes in fluidization,which occurs in small diameter beds with large bed height-to-diameter ratio or in large diameter beds with internals that resemble multiple small diameter fluidized beds.Slug types include round-nosed slug,wall slug and square nosed slug.Studies of the slugs have been mainly focused on round-nosed or wall slugs known as half slug.typically occurring in Geldart group A particle fuidization.The square-nosed slug typically occurring for Geldart group D particles appears to be regarded as simple in its structure.The Electrical Capacitance Volume Tomogra phy(ECVT)imaging of the square-nosed slugging phenomena conducted in this study reveals otherwise.That is the structure of the square nosed slug is,in fact,complex,particularly with respect to its dynamic variation in fluidization.More broadly.this study examines experimentally the hydrodynamic character-istics of the square-nosed fluidization regime.Specifically,simultaneous measurements from multiple ECVT sensors provide non-inv asive,continuous,3.dimensional imaging of the entire flow region of the slugging bed and hence enabling the dynamic characterization of the evolution of the slugs.The analysis of the 3D images reconstructed for real time gas-solid volume fraction profile of the slugging fluidized bed indicates that there are three different zones.namely.the bottom fluidization zone,the gas slug zone,and the solid slug zone,co-existing in the bed.The three zones present different hydrodynamic characteristics during the slug evolution.It is found that varying the gas velocity of the slugging bed mainly varies the maximum length of the gas slug zone,while it only has a minor effect on the lengths of the bottom fluidization zone and solid slug zone.It also has an insignificant efect on the solid volume fraction of the three zones.Yaswanth Pottimurthy Dawei Wang Cody Park Shalin Patil Andrew Tong Liang-Shih Fan 2022Particuology2022,20,8:0
20High-throughput phenotyping by applying digital morphometrics and fluorescence induction curves in seeds to identifying variations:A case study of Annona(Annonaceae)species显示文摘Differences in physical and structural characteristics of seeds may indicate variability within and between plant populations.In the present study,we performed a close characterization of dimension,shape,and tegument delayed chlorophyll fluorescence in seeds obtained from three species of the genus Annona(Annonaceae),i.e.,Annona coriacea,A.montana,A.squamosa.Results showed that studied seeds may be sorted as scalene ellipsoids expressing low values for the seed sphericity.The morphological estimates suggested differences in seed shape for all species.A high correlation was observed between surface area and volume(r^2>99%)for all the three species suggesting that in addition to structural shape.In addition,we also observed very high positive correlations(Rho=1.000,p<0.001)between surface area and arithmetic mean diameterof the seeds for all species.The first principal component(PCA1)of elliptical Fourier descriptors explained most of the variations in morphological structure of the seeds in the three species.Additionally,a less intense tegument delayed chlorophyll fluorescence was observed for A.montana while the highest intensity was recorded for A.squamosa,revealing the potential use of fluorescence spectroscopy in discrimination at the species level by analyzing the frequency domain by means of Fourier Transform spectra as well as the relationship time-frequency of chlorophyll fluorescence.Montcharles S.Pontes Carlos V.Montefusco-Pereira Biswapriya B.Misra Howard L.Ribeiro-Junior Daniela E.Graciano Jaqueline S.Santos Michele A.S.Nobrega Shaline S.L.Fernandes Anderson R.L.Caires Etenaldo F.Santiago 2018Information Processing in Agriculture2018,5,4:0
