5Potential therapeutic effects and applications of Eucommiae Folium in secondary hypertension显示文摘Eucommiae Folium(EF),a traditional Chinese medicine,has been used to treat secondary hypertension,including renal hypertension and salt-sensitive hypertension,as well as hypertension caused by thoracic aortic endothelial dysfunction,a high-fat diet,and oxidized low-density lipoprotein.The antihypertensive components of EF are divided into four categories:flavonoids,iridoids,lignans,and phenylpropanoids,such as chlorogenic acid,geniposide acid and pinoresinol diglucoside.EF regulates the occurrence and development of hypertension by regulating biological processes,such as inhibiting inflammation,regulating the nitric oxide synthase pathway,reducing oxidative stress levels,regulating endothelial vasoactive factors,and lowering blood pressure.However,its molecular antihypertensive mechanisms are still unclear and require further investigation.In this review,by consulting the relevant literature on the antihypertensive effects of EF and using network pharmacology,we summarized the active ingredients and pharmacological mechanisms of EF in the treatment of hypertension to clarify how EF is associated with secondary hypertension,the related components,and underlying mechanisms.The results of the network pharmacology analysis indicated that EF treats hypertension through a multicomponent,multi-target and multi-pathway mechanism.In particular,we discussed the role of EF targets in the treatment of hypertension,including epithelial sodium channel,heat shock protein70,rhoassociated protein kinase 1,catalase,and superoxide dismutase.The relevant signal transduction pathways,the ras homolog family member A(RhoA)/Rho-associated protein kinase(ROCK)and nicotinamide adenine dinucleotide phosphate(NADPH)oxidase/eNOS/NO/Ca^(2+)pathways,are also discussed.Mengyuan Li Yanchao Zheng Sha Deng Tian Yu Yucong Ma Jiaming Ge Jiarong Li Xiankuan Li Lin Ma 2022Journal of Pharmaceutical Analysis2022,12,5:2
6Targeting and signaling of Rho of plants guanosine triphosphatases require synergistic interaction between guanine nucleotide inhibitor and vesicular trafficking显示文摘Most eukaryotic cells are polarized.Common toolbox regulating cell polarization includes Rho gua-nosine triphosphatases(GTPases),in which spatio-temporal activation is regulated by a plethora of reg-ulators.Rho of plants(ROPs)are the only Rho GTPases in plants.Although vesicular trafficking was hinted in the regulation of ROPs,it was unclear where vesicle-carried ROP starts,whether it is dynamically regulated,and which components participate in vesicle-mediated ROP targeting.In addition,although vesicle trafficking and guanine nucleotide inhibitor(GDI)pathways in Rho signaling have been extensively studied in yeast,it is unknown whether the two pathways interplay.Unclear are also cellular and developmental con-sequences of their interaction in multicellular organisms.Here,we show that the dynamic targeting of ROP through vesicles requires coat protein complexⅡand ADP-ribosylation factor 1-mediated post-Golgi trafficking.Trafficking of vesicle-carried ROPs between the plasma membrane and the trans-Golgi network is mediated through adaptor protein 1 and sterol-mediated endocytosis.Finally,we show that GDI and vesicle trafficking synergistically regulate cell polar-ization and ROP targeting,suggesting that the estab-lishment and maintenance of cell polarity is regulated by an evolutionarily conserved mechanism.Fu-Rong Ge Sen Chai Sha Li Yan Zhang 2020Journal of Integrative Plant Biology2020,62,10:2

8ADH1C inhibits progression of colorectal cancer through the ADH1C/PHGDH/PSAT1/serine metabolic pathway显示文摘Colorectal cancer(CRC)is the third most common cancer in men and the second most common cancer in women worldwide.CRC is the second leading cause of cancer-related deaths.Although some progress in the treatment of CRC has been achieved,the molecular mechanism of CRC is still unclear.In this study,alcohol dehydrogenase 1C(ADH1C)was first identified as a target gene closely associated with the development of CRC by the comprehensive application of transcriptomics,proteomics,metabonomics and in silico analysis.The ADH1C mRNA and protein expression in CRC cell lines and tumor tissues was lower than that in normal intestinal epithelial cell lines and healthy tissues.Overexpression of ADH1C inhibited the growth,migration,invasion and colony formation of CRC cell lines and prevented the growth of xenograft tumors in nude mice.The inhibitory effects of ADH1C on CRC cells in vitro were exerted by reducing the expression of PHGDH/PSAT1 and the serine level.This inhibition could be partially reversed by adding serine to the culture medium.These results showed that ADH1C is a potential drug target in CRC.Sha Li Hong Yang Wan Li Jin-yi Liu Li-wen Ren Yi-hui Yang Bin-bin Ge Yi-zhi Zhang Wei-qi Fu Xiang-jin Zheng Guan-hua Du Jin-Hua Wang 2022Acta Pharmacologica Sinica2022,43,10:1
9A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. 12Rhesus monkeys exhibiting spontaneous ritualistic behaviors resembling obsessive-compulsive disorder显示文摘Obsessive-compulsive disorder(OCD)is a chronic and debilitating psychiatric disorder that affects∼2%–3%of the population globally.Studying spontaneous OCD-like behaviors in non-human primates may improve our understanding of the disorder.In large rhesus monkey colonies,we found 10 monkeys spontaneously exhibiting persistent sequential motor behaviors(SMBs)in individual-specific sequences that were repetitive,time-consuming and stable over prolonged periods.Genetic analysis revealed severely damaging mutations in genes associated with OCD risk in humans.Brain imaging showed that monkeys with SMBs had larger gray matter(GM)volumes in the left caudate nucleus and lower fractional anisotropy of the corpus callosum.The GM volume of the left caudate nucleus correlated positively with the daily duration of SMBs.Notably,exposure to a stressor(human presence)significantly increased SMBs.In addition,fluoxetine,a serotonergic medication commonly used for OCD,decreased SMBs in these monkeys.These findings provide a novel foundation for developing better understanding and treatment of OCD.Rongwei Zhai Geya Tong Zheqin Li Weichen Song Yang Hu Sha Xu Qiqi Wei Xiaocheng Zhang Yi Li Bingbing Liao Chenyu Yuan Yinqing Fan Ge Song Yinyin Ouyang Wenxuan Zhang Yaqiu Tang Minghui Jin Yuxian Zhang He Li Zhi Yang Guan Ning Lin Dan JStein Zhi-Qi Xiong Zhen Wang 2023National Science Review2023,10,11:1

18Apolipoprotein C1 promotes glioblastoma tumorigenesis by reducing KEAP1/NRF2 and CBS-regulated ferroptosis显示文摘Glioblastoma(GBM),a malignant brain tumor,is a world-wide health problem because of its poor prognosis and high rates of recurrence and mortality.Apolipoprotein C1(APOC1)is the smallest of apolipoproteins,implicated in many diseases.Recent studies have shown that APOC1 promotes tumorigenesis and development of several types of cancer.In this study we investigated the role of APOC1 in GBM tumorigenesis.Using in silico assays we showed that APOC1 was highly expressed in GBM tissues and its expression was closely related to GBM progression.We showed that APOC1 protein expression was markedly increased in four GBM cell lines(U251,U138,A172 and U87)compared to the normal brain glia cell lines(HEB,HA1800).In U251 cells,overexpression of APOC1 promoted cell proliferation,migration,invasion and colony information,which was reversed by APOC1 knockdown.APOC1 knockdown also markedly inhibited the growth of GBM xenografts in the ventricle of nude mice.We further demonstrated that APOC1 reduced ferroptosis by inhibiting KEAP1,promoting nuclear translocation of NRF2 and increasing expression of HO-1 and NQO1 in GBM cells.APOC1 also induced ferroptosis resistance by increasing cystathionine beta-synthase(CBS)expression,which promoted trans-sulfuration and increased GSH synthesis,ultimately leading to an increase in glutathione peroxidase-4(GPX4).Thus,APOC1 plays a key role in GBM tumorigenesis,conferring resistance to ferroptosis,and may be a promising therapeutic target for GBM.Xiang-jin Zheng Wen-lin Chen Jie Yi Wan Li Jin-yi Liu Wei-qi Fu Li-wen Ren Sha Li Bin-bin Ge Yi-hui Yang Yi-zhi Zhang Hong Yang Guan-hua Du Yu Wang Jin-hua Wang 2022Acta Pharmacologica Sinica2022,43,11:0
19Fibroblast growth factor 21 inhibits ferroptosis following spinal cord injury by regulating heme oxygenase-1显示文摘Interfering with the ferroptosis pathway is a new strategy for the treatment of spinal cord injury.Fibroblast growth factor 21 can inhibit ferro ptosis and promote neurofunctional recovery,while heme oxygenase-1 is a regulator of iron and reactive oxygen species homeostasis.The relationship between heme oxygenase-1and ferroptosis remains controve rsial.In this study,we used a spinal co rd injury rat model to show that the levels of fibroblast growth factor 21 in spinal co rd tissue decreased after spinal cord injury.In addition,there was a significant aggravation of ferroptosis and a rapid increase in heme oxygenase-1 expression after spinal cord injury.Furthe r,heme oxygenase-1 aggravated fe rroptosis after spinal cord injury,while fibroblast growth factor 21 inhibited fe rroptosis by downregulating heme oxygenase-1.Thus,the activation of fibroblast growth factor 21 may provide a potential treatment for spinal co rd injury.These findings could provide a new potential mechanistic explanation for fibroblast growth factor 21 in the treatment of spinal cord injury.Qi Gu Weiping Sha Qun Huang Jin Wang Yi Zhu Tianli Xu Zhenhua Xu Qiancheng Zhu Jianfei Ge Shoujin Tian Xiaolong Lin 2024Neural Regeneration Research2024,19,7:0
