| 17 | The GYF domain protein CD2BP2 is critical for embryogenesis and podocyte function显示文摘Scaffolding proteins play pivotal roles in the assembly of macromolecular machines such as the spliceosome.The adaptor protein CD2BP2,originally identified as a binding partner of the adhesion molecule CD2,is a pre-spliceosomal assembly factor that utilizes its glycine-tyrosine-phenylalanine(GYF)domain to co-localize with spliceosomal proteins.So far,its function in vertebrates is unknown.Using conditional gene targeting in mice,we show that CD2BP2 is crucial for embryogenesis,leading to growth retardation,defects in vascularization,and premature death at embryonic day 10.5 when absent.Ablation of the protein in bone marrow-derived macrophages indicates that CD2BP2 is involved in the alternative splicing of mRNA transcripts from diverse origins.At the molecular level,we identified the phosphatase PP1 to be recruited to the spliceosome via the N-terminus of CD2BP2.Given the strong expression of CD2BP2 in podocytes of the kidney,we use selective depletion of CD2BP2,in combination with next-generation sequencing,to monitor changes in exon usage of genes critical for podocyte functions,including VEGF and actin regulators.CD2BP2-depleted podocytes display foot process effacement,and cause proteinuria and ultimately lethal kidney failure in mice.Collectively,our study defines CD2BP2 as a non-redundant splicing factor essential for embryonic development and podocyte integrity. | Gesa I.Albert Christoph Schell Karin M.Kirschner Sebastian Schafer Ronald Naumann Alexandra Muller Oliver Kretz Benno Kuropka Mathias Girbig Norbert Hubner Eberhard Krause Holger Scholz Tobias B.Huber Klaus-Peter Knobeloch Christian Freund | 2015 | Journal of Molecular Cell Biology2015,7,5: | 0 |
| 18 | Single-cell protein profiling in microchambers with barcoded beads显示文摘Single-cell profiling provides insights into cellular behaviour that macroscale cell cultures and bulk measurements cannot reveal.In the context of personalized cancer treatment,the profiling of individual tumour cells may lead to higher success rates for therapies by rapidly selecting the most efficacious drugs.Currently,genomic analysis at the single-cell level is available through highly sensitive sequencing approaches.However,the identification and quantification of intracellular or secreted proteins or metabolites remains challenging.Here,we introduce a microfluidic method that facilitates capture,automated data acquisition and the multiplexed quantification of proteins from individual cells.The microfluidic platform comprises 1026 chambers with a volume of 152 pL each,in which single cells and barcoded beads are co-immobilized.We demonstrated multiplexed single-cell protein quantification with three different mammalian cell lines,including two model breast cancer cell lines.We established on-chip immunoassays for glyceraldehyde-3-phosphate dehydrogenase(GAPDH),galectin-3(Gal-3)and galectin-3 binding protein(Gal-3bp)with detection limits as low as 7.0×10^(4),2.3×10^(5)and 1.8×10^(3)molecules per cell,respectively.The three investigated cell types had high cytosolic levels of GAPDH and could be clearly differentiated by their expression levels of Gal-3 and Gal-3bp,which are important factors that contribute to cancer metastasis.Because it employed commercially available barcoded beads for this study,our platform could be easily used for the single-cell protein profiling of several hundred different targets.Moreover,this versatile method is applicable to the analysis of bacteria,yeast and mammalian cells and nanometre-sized lipid vesicles. | Lucas Armbrecht Rafael Sebastian Muller Jonas Nikoloff Petra Stephanie Dittrich | 2019 | Microsystems & Nanoengineering2019,5,1: | 0 |
