| 1 | Does combined therapy of curcumin and epigallocatechin gallate have a synergistic neuroprotective effect against spinal cord injury?显示文摘Systematic inflammatory response after spinal cord injury(SCI) is one of the factors leading to lesion development and a profound degree of functional loss. Anti-inflammatory compounds, such as curcumin and epigallocatechin gallate(EGCG) are known for their neuroprotective effects. In this study, we investigated the effect of combined therapy of curcumin and EGCG in a rat model of acute SCI induced by balloon compression. Immediately after SCI, rats received curcumin, EGCG, curcumin + EGCG or saline [daily intraperitoneal doses(curcumin, 6 mg/kg; EGCG 17 mg/kg)] and weekly intramuscular doses(curcumin, 60 mg/kg; EGCG 17 mg/kg)] for 28 days. Rats were evaluated using behavioral tests(the Basso, Beattie, and Bresnahan(BBB) open-field locomotor test, flat beam test). Spinal cord tissue was analyzed using histological methods(Luxol Blue-cresyl violet staining) and immunohistochemistry(anti-glial fibrillary acidic protein, anti-growth associated protein 43). Cytokine levels(interleukin-1β, interleukin-4, interleukin-2, interleukin-6, macrophage inflammatory protein 1-alpha, and RANTES) were measured using Luminex assay. Quantitative polymerase chain reaction was performed to determine the relative expression of genes(Sort1, Fgf2, Irf5, Mrc1, Olig2, Casp3, Gap43, Gfap, Vegf, NfκB, Cntf) related to regenerative processes in injured spinal cord. We found that all treatments displayed significant behavioral recovery, with no obvious synergistic effect after combined therapy of curcumin and ECGC. Curcumin and EGCG alone or in combination increased axonal sprouting, decreased glial scar formation, and altered the levels of macrophage inflammatory protein 1-alpha, interleukin-1β, interleukin-4 and interleukin-6 cytokines. These results imply that although the expected synergistic response of this combined therapy was less obvious, aspects of tissue regeneration and immune responses in severe SCI were evident. | Jiri Ruzicka Lucia Machova Urdzikova Barbora Svobodova Anubhav G. | 2 | Epigenetic and non-epigenetic mode of SIRT1 action during oocyte meiosis progression显示文摘Background: SIRT1 histone deacetylase acts on many epigenetic and non-epigenetic targets. It is thought that SIRT1 is involved in oocyte maturation;therefore, the importance of the ooplasmic SIRT1 pool for the further fate of mature oocytes has been strongly suggested. We hypothesised that SIRT1 plays the role of a signalling molecule in mature oocytes through selected epigenetic and non-epigenetic regulation.Results: We observed SIRT1 re-localisation in mature oocytes and its association with spindle microtubules.In mature oocytes, SIRT1 distribution shows a spindle-like pattern, and spindle-specific SIRT1 action decreasesα-tubulin acetylation. Based on the observation of the histone code in immature and mature oocytes, we suggest that SIRT1 is mostly predestined for an epigenetic mode of action in the germinal vesicles(GVs) of immature oocytes. | 3 | Antibodies Against Annexin V and Prothrombin,Their Correlation with Other Anti-phospholipid Antibodies in Recurrent Pregnancy Loss显示文摘Objective To study the findings of serum antibodies against annexin V, prothrombin,ph-inositol, ph-acid, ph-ethanolamine, ph-serine, ph-glycerol, cardiolipin, and beta2-glycoprotein I and analyze the trophoblast annexin V receptorsMethods Sera from 156 patients aged 26-41 years with recurrent pregnancy loss (3-7 times) were investigated. Eighty-four fertile healthy women aged 24-38 yearswere included in a control group. ELISA methods were used for detecting a panel of seraanti-phospholipid antibodies. Immunolocalization of annexin V receptors in 143trophoblast specimens of 156 patients was investigated by the immunofluorescencetechnique using Annexin V-FITC, Apoptosis and AnnexinV-CY3 commercial kits.Results Positivity for anti-phospholipid antibodies mainly against ph-serine, ph-ethanolamine, and ph-inositol was found together in 80.8% (126 out of 156 patients),anti-prothrombin antibodies in 12% (18), and anti-annexin V antibodies in 13. 5%(21) women. No significant levels of anti-phospholipid antibodies were found in 6controls. Placenta immunohistopathology also exhibited some changes manifested bythe presence of apoptotic and necrotic cells in trophoblast, and very fewmicrothrombotization in some intervillous spaces.Conclusion Our detailed study demonstrated the prevalence of majority of anti-phospholipid antibodies as a high risk factor for repeated reproductive failure.
