| 1 | Constituents of the anti-asthma herbal formula ASHMI^(TM) synergistically inhibit IL-4 and IL-5 secretion by murine Th2 memory cells,and eotaxin by human lung fibroblasts in vitro显示文摘OBJECTIVE:Anti-asthma herbal medicine intervention(ASHMI ),a combination of three traditional Chinese medicinal herbs developed in our laboratory,has demonstrated efficacy in both mouse models of allergic asthma,and a double-blind placebo-controlled clinical trial in patients with asthma.This study was designed to determine if the anti-inflammatory effects of individual herbal constituents of ASHMI? exhibited synergy. METHODS:Effects of ASHMI and its components aqueous extracts of Lingzhi(Ganoderma lucidum),Kushen(Sophora flavescens) and Gancao(Glycyrrhiza uralensis),on Th2 cytokine secretion by murine memory Th2 cells(D10.G4.1) and eotaxin-1 secretion by human lung fibroblast(HLF-1) cells were determined by measuring levels in culture supernatants by enzymelinked immunosorbent assay.Potential synergistic effects were determined by computing interaction indices from concentration-effect curve parameters. RESULTS:Individual Lingzhi,Kushen and Gancao extracts and ASHMI(the combination of individual extracts) inhibited production of interleukin(IL)-4 and IL-5 by murine memory Th2 cells and eotaxin-1 production by HLF-1 cells.The mean 25%-inhibitory-concentration(IC25) values(mg/mL) for ASHMI,Lingzhi,Kushen and Gancao for IL-4 production were 30.9,79.4, 123,and 64.6,respectively;for IL-5 production were 30.2,263,123.2 and 100,respectively; for eotaxin-1 were 13.2,16.2,30.2,and 25.1,respectively.The IC50 values(mg/mL) for ASHMI,Lingzhi,Kushen and Gancao for IL-4 production were 158.5,239.9,446.7,and 281.8, respectively;for eotaxin-1 were 38.1,33.1,100,and 158.5,respectively.The interaction indices of ASHMI constituents at IC25 were 0.35 for IL-4,0.21 for IL-5 and 0.59 for eotaxin-1.The interaction indices at IC50 values were 0.50 for IL-4 and 0.62 for eotaxin-1 inhibition.Inhibition of IL-5 did not reach IC50 values.All interaction indices were below 1 which indicated synergy. | 5 | RECENT ADVANCES IN RESEARCH AND FORECASTING OF TROPICAL CYCLONE TRACK, INTENSITY, AND STRUCTURE AT LANDFALL显示文摘This review prepared for the fourth International Workshop on Tropical Cyclone Landfall Processes(IWTCLP-4) summarizes the most recent(2015-2017) theoretical and practical knowledge in the field of tropical cyclone(TC) track, intensity, and structure rapid changes at or near landfall. Although the focus of IWTCLPIV was on landfall, this summary necessarily embraces the characteristics of storms during their course over the ocean prior to and leading up to landfall. In the past few years, extremely valuable observational datasets have been collected for TC forecasting guidance and research studies using both aircraft reconnaissance and new geostationary or low-earth orbiting satellites at high temporal and spatial resolution. Track deflections for systems near complex topography such as that of Taiwan and La Réunion have been further investigated, and advanced numerical models with high spatial resolution necessary to predict the interaction of the TC circulation with steep island topography have been developed. An analog technique has been designed to meet the need for longer range landfall intensity forecast guidance that will provide more time for emergency preparedness. Probabilistic track and intensity forecasts have also been developed to better communicate on forecast uncertainty. Operational practices of several TC forecast centers are described herein and some challenges regarding forecasts and warnings for TCs making landfall are identified. | 10 | 重组脊髓灰质炎病毒用于治疗复发胶质母细胞瘤(英文)显示文摘Background The prognosis of patients with recurrent World Health Organization(WHO)grade IV malignant glioma is dismal,and there is currently no effective therapy.We conducted a dose-finding and toxicity study in this population of patients,evaluating convection-enhanced,intratumoral delivery of the recombinant nonpathogenic polio-rhinovirus chimera(PVSRIPO).PVSRIPO recognizes the poliovirus receptor CD155,which is widely expressed in neoplastic cells of solid tumors and in major components of the tumor microenvironment.Methods We enrolled consecutive adult patients who had recurrent supratentorial WHO grade IV malignant glioma,confirmed on histopathological testing,with measurable disease(contrast-enhancing tumor of≥1 cm and≤5.5 cm in the greatest dimension).The study evaluated seven doses,ranging between 107 and 1010 50%tissue-culture infectious doses(TCID50),first in a dose-escalation phase and then in a dose-expansion phase.Results From May 2012 through May 2017,a total of 61 patients were enrolled and received a dose of PVSRIPO.Dose level-1(5.0×107TCID50)was identified as the phase 2 dose.One dose-limiting toxic effect was observed;a patient in whom dose level 5(1010TCID50)was administered had a grade 4 intracranial hemorrhage immediately after the catheter was removed.To mitigate locoregional inflammation of the infused tumor with prolonged glucocorticoid use,dose level 5 was deescalated to reach the phase 2 dose.In the dose-expansion phase,19%of the patients had a PVSRIPO-related adverse event of grade 3 or higher.Overall survival among the patients who received PVSRIPO reached a plateau of 21%(95%confidence interval,11 to 33)at 24 months that was sustained at 36 months.Conclusions Intratumoral infusion of PVSRIPO in patients with recurrent WHO grade IV malignant glioma confirmed the absence of neurovirulent potential.The survival rate among patients who receivedPVSRIPO immunotherapy was higher at 24 and 36 months than the rate among historical controls. | Desjardins A Gromeier M Herndon JE 2nd Beaubier N Bolognesi DP Friedman AH Friedman HS McSherry F Muscat AM Nair S Peters KB Randazzo D Sampson JH Vlahovic G Harrison WT McLendon RE Ashley D Bigner DD | 2018 | 中华神经外科疾病研究杂志2018，17，4： | 2 |

| 14 | Scalable non-mode selective Hermite–Gaussian mode multiplexer based on multi-plane light conversion显示文摘Non-mode-selective(NMS) multiplexers(muxes) are highly desirable for coherent power combining to produce a high-power beam with a shaped profile(wavefront synthesis) from discrete, phase-locked emitters. We propose a design for a multi-plane light conversion(MPLC)-based NMS mux, which requires only a few phase masks for coherently combining hundreds of discrete input beams into an output beam consisting of hundreds of Hermite–Gaussian(HG) modes. The combination of HG modes as a base can further construct a beam with arbitrary wavefront. The low number of phase masks is attributed to the identical zero-crossing structure of the Hadamard-coded input arrays and of the output HG modes, enabling the practicality of such devices. An NMS mux supporting 256 HG modes is designed using only seven phase masks, and achieves an insertion loss of-1.6 d B, mode-dependent loss of 4.7 d B, and average total mode crosstalk of-4.4 d B.
