| 3 | Autophagy inhibition by chloroquine sensitizes HT-29 colorectal cancer cells to concurrent chemoradiation显示文摘AIM:To investigate whether the inhibition of autophagy by chloroquine(CQ)sensitizes rectal tumors to radiation therapy(RT)or concurrent chemoradiation(chemoRT).METHODS:In vitro,HCT-116 and HT-29 colorectal cancer(CRC)cell lines were treated as following:(1)PBS;(2)CQ;(3)5-fluorouracil(5-FU);(4)RT;(5)CQ and RT;(6)5-FU and RT;(7)CQ and 5-FU;and(8)5-FU and CQ and RT.Each group was then exposed to various doses of radiation(0-8 Gy)depending on the experiment.Cell viability and proliferative capacity were measured by3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)and clonogenic assays.Clonogenic survivalcurves were constructed and compared across treatment groups.Autophagy status was determined by assessing the LC3-Ⅱto LC3-Ⅰratio on western blot analysis,autophagosome formation on electron microscopy and identification of a perinuclear punctate pattern with GFPlabeled LC3 on fluorescence microscopy.Cell cycle arrest and cell death were evaluated by FACS and AnnexinⅤanalysis.All experiments were performed in triplicate and statistical analysis was performed by the student’s t test to compare means between treatment groups.RESULTS:RT(2-8 Gy)induced autophagy in HCT-116and HT-29 CRC cell lines at 4 and 6 h post-radiation,respectively,as measured by increasing LC3-Ⅱto LC3-Ⅰratio on western blot.Additionally,electron microscopy demonstrated autophagy induction in HT-29 cells24 h following irradiation at a dose of 8 Gy.Drug treatment with 5-FU(25μmol/L)induced autophagy and the combination of 5-FU and RT demonstrated synergism in autophagy induction.CQ(10μmol/L)alone and in combination with RT effectively inhibited autophagy and sensitized both HCT-116 and HT-29 cells to treatment with radiation(8 Gy;P<0.001 and 0.00001,respectively).Significant decrease in clonogenic survival was seen only in the HT-29 cell line,when CQ was combined with RT at doses of 2 and 8 Gy(P<0.5 and P=0.05,respectively).There were no differences in cell cycle progression or Annexin V staining upon CQ addition to RT.CONCLUSION:Autophagy inhibition by CQ increases CRC cell sensitivity to concurrent treatment with 5-FU and RT in vitro,suggesting that addition of CQ to chemoRT improves CRC treatment response. | Caitlin A Schonewolf Monal Mehta Devora Schiff Hao Wu Bruce G Haffty Vassiliki Karantza Salma K Jabbour | 2014 | World Journal of Gastrointestinal Oncology2014,6,3: | 12 |

| 6 | An advanced anaerobic biofilter with effluent recirculation for phenol removal and methane production in treatment of coal gasification wastewater显示文摘An advanced anaerobic biofilter(AF) was introduced for the treatment of coal gasification wastewater(CGW),and effluent recirculation was adopted to enhance phenol removal and methane production.The results indicated that AF was reliable in treating diluted CGW,while its efficiency and stability were seriously reduced when directly treating raw CGW.However,its performance could be greatly enhanced by effluent recirculation.Under optimal effluent recirculation of 0.5 to the influent,concentrations of chemical oxygen demand(COD) and total phenol in the effluent could reach as low as 234.0 and 14.2 mg/L,respectively.Also,the rate of methane production reached 169.0 m L CH_4/L/day.Though CGW seemed to restrain the growth of anaerobic microorganisms,especially methanogens,the inhibition was temporary and reversible,and anaerobic bacteria presented strong tolerance.The activities of methanogens cultivated in CGW could quickly recover on feeding with glucose wastewater(GW).However,the adaptability of anaerobic bacteria to the CGW was very poor and the activity of methanogens could not be improved by long-term domestication.By analysis using the Haldane model,it was further confirmed that high effluent recirculation could result in high activity for hydrolytic bacteria and substrate affinity for toxic matters,but only suitable effluent recirculation could result in high methanogenic activity. | Yajie Li Salma Tabassum Zhenjia Zhang | 2016 | Journal of Environmental Sciences2016,28,9: | 7 |
| 7 | Inhibitory effect of high phenol concentration in treating coal gasification wastewater in anaerobic biofilter显示文摘In this paper, the inhibition of methanogens by phenol in coal gasification wastewater(CGW)was investigated by both anaerobic toxicity tests and a lab-scale anaerobic biofilter reactor(AF). The anaerobic toxicity tests indicated that keeping the phenol concentration in the influent under 280 mg/L could maintain the methanogenic activity. In the AF treating CGW,the result showed that adding glucose solution as co-substrate could be beneficial for the quick start-up of the reactor. The effluent chemical oxygen demand(COD) and total phenol reached1200 and 100 mg/L, respectively, and the methane production rate was 175 m L CH4/g COD/day.However, if the concentration of phenol was increased, the inhibition of anaerobic micro-organisms was irreversible. The threshold of total phenol for AF operation was 200–250 mg/L. The extracellular polymeric substances(EPS) and particle size distribution of anaerobic granular sludge in the different stages were also examined, and the results indicated that the influence of toxicity in the system was more serious than its effect on flocculation of EPS. Moreover, the proportion of small size anaerobic granular sludge gradually increased from10.2% to 34.6%. | 14 | Automated brain tumor segmentation on multi-modal MR image using SegNet显示文摘The potential of improving disease detection and treatment planning comes with accurate and fully automatic algorithms for brain tumor segmentation.Glioma, a type of brain tumor, can appear at different locations with different shapes and sizes. Manual segmentation of brain tumor regions is not only timeconsuming but also prone to human error, and its performance depends on pathologists’ experience. In this paper, we tackle this problem by applying a fully convolutional neural network SegNet to 3 D data sets for four MRI modalities(Flair, T1, T1 ce, and T2)for automated segmentation of brain tumor and subtumor parts, including necrosis, edema, and enhancing tumor. To further improve tumor segmentation, the four separately trained SegNet models are integrated by post-processing to produce four maximum feature maps by fusing the machine-learned feature maps from the fully convolutional layers of each trained model. The maximum feature maps and the pixel intensity values of the original MRI modalities are combined to encode interesting information into a feature representation.Taking the combined feature as input, a decision tree(DT) is used to classify the MRI voxels into different tumor parts and healthy brain tissue. | 16 | Characterization of antioxidant and antibacterial compounds from aerial parts of Haplophyllum tuberculatum显示文摘The aerial parts of Haplophyllum tuberculatum(H.tuberculatum)have traditionally been used by local communities to treat fever,gastrointestinal disorders,worms,malaria and fractures,how-ever,the responsible ingredients of the selected medicinal plant have not been identified.In the present study,antibacterial and antioxidant ingredients were isolated and characterized from the plant parts of locally grown H.tuberculatum,and their biological activity was evaluated.The powder aerial parts were extracted with methanol and the extract was successively frac-tionated with different polarities of solvents.All polarities of extracts(hexane,chloroform,ethyl acetate,butanol,methanol and water)were used to determine their antibacterial and antioxi-dant activity by established in-vitro method.The disk diffusion and free radical scavenging 2,2-diphenyl-1-picrylhydrazyl(DPPH)methods were used to evaluate antibacterial and antioxidant activities of the prepared extracts and pure isolated compounds at different concentrations against three strains of Gram-positive and three Gram-negative bacteria.The antibacterial activity results showed that all prepared fractions and isolated pure compounds of the plant species gave signif-icant activity against all Gram(+and-)bacterial strains with the range of inhibition 0-20 mm.The antioxidant activity of the prepared extracts at all concentrations also showed substantial activity.Based on the highest activity,ethyl acetate and chloroform extracts were selected for the separation and isolation of antioxidant and antibacterial compounds by thin layer chromatogra-phy(TLC),column chromatography(CC),and preparative thin layer chromatography(PTLIC).The structure of the isolated antioxidant and antibacterial compounds from the ethyl acetate and chloroform extracts were determined on the basis of nuclear magnetic resonance(NMR)and mass spectra.The isolated pure compounds and active extracts could be used as a medicine for various infectious related diseases. | Salma Salim Al-Saeghi Mohammad Amzad Hossain Salem Said Jaroof Al-Touby | 2022 | Journal of Bioresources and Bioproducts2022,7,1: | 2 |

This investigation deals with these two aspects, an electrochemical treatment method of an olive mill wastewater (OMW) under platinized expanded titanium electrodes using a modified Grignard reactor for toxicity removal as well as the exploration of the use of some specific analytical tools to monitor effluent phenolic compounds elimination. The results showed that electrochemical oxidation is able to remove/mitigate the OMW pollution. Indeed, 87% of OMW color was removed and all aromatic compounds were disappeared from the solution by anodic oxidation. Moreover, 55% of the chemical oxygen demand (COD) and the total organic carbon (TOC) were reduced. On the other hand, UV- Visible spectrophotometry, Gaz chromatography/mass spectrometry, cyclic voltammetry and 13 C Nuclear Magnetic Resonance (NMR) showed that the used treatment seems efficaciously to eliminate phenolic compounds from OMW. It was concluded that electrochemical oxidation in a modified Grignard reactor is a promising process for the destruction of all phenolic compounds present in OMW. | 19 | Nonlinear Coordinated Passivation Control of Single Machine Infinite Bus Power System with Static Var Compensator显示文摘We propose a nonlinear coordinated control of the generator excitation and the static var compensator(SVC) in order to enhance the transient stability and voltage regulation of power system by the passivation approach. SVC is installed in the middle of the transmission line of power system and consists of a single machine infinite bus(SMIB) system. The design of the proposed controller consists of two parts. On one hand,the generator excitation controller is designed based on a backstepping controller. On the other hand, the conception of SVC control input is based on the coordinated passivation approach,which can guarantee the asymptotic stability of the closed-loop system.
