3Hematopoietic stem cell-derived adipocytes and fibroblasts in the tumor microenvironment显示文摘The tumor microenvironment(TME) is complex and constantly evolving. This is due, in part, to the crosstalk between tumor cells and the multiple cell types that comprise the TME, which results in a heterogeneous population of tumor cells and TME cells. This review will focus on two stromal cell types, the cancerassociated adipocyte(CAA) and the cancer-associated fibroblast(CAF). In the clinic, the presence of CAAs and CAFs in the TME translates to poor prognosis in multiple tumor types. CAAs and CAFs have an activated phenotype and produce growth factors, inflammatory factors, cytokines, chemokines, extracellular matrix components, and proteases in an accelerated and aberrant fashion. Through this activated state, CAAs and CAFs remodel the TME, thereby driving all aspects of tumor progression, including tumor growth and survival, chemoresistance, tumor vascularization, tumor invasion, and tumor cell metastasis. Similarities in the tumorpromoting functions of CAAs and CAFs suggest that a multipronged therapeutic approach may be necessary to achieve maximal impact on disease. While CAAs and CAFs are thought to arise from tissues adjacent to the tumor, multiple alternative origins for CAAs and CAFs have recently been identified. Recent studies from our lab and others suggest that the hematopoietic stem cell, through the myeloid lineage, may serve as a progenitor for CAAs and CAFs. We hypothesize that the multiple origins of CAAs and CAFs may contribute to the heterogeneity seen in the TME. 7A rapid dehydration leaf assay reveals stomatal response differences in grapevine genotypes显示文摘A simple and reliable way of phenotyping plant responses to dehydration was developed.Fully-developed leaves were detached and placed in a closed plastic box containing a salt solution to control the atmospheric water potential in the container.Three hours of dehydration(weight loss of the leaf)was optimal for measuring changes in stomatal response to dehydration.Application of the plant hormone abscisic acid(ABA)prior to leaf detachment decreased the amount of water loss,indicating that the assay was able to detect differences based on a stomatal response to dehydration.Five different Vitis genotypes(V.riparia,V.champinii,V.vinifera cv.Shiraz,V.vinifera cv.Grenache and V.vinifera cv.Cabernet Sauvignon)with known differences in drought tolerance were screened for their dehydration response and the results obtained corresponded to previous reports of stomatal responses in the vineyard.Significant differences in stomatal density along with differences in the amount and rate of water lost indicate differences in dehydration sensitivity among the genotypes screened.Differences in stomatal response to ABA were also detected.Shiraz had the lowest stomatal density and the highest ABA sensitivity among the genotypes screened,yet Shiraz lost the most amount of water,indicating that it was the least sensitive to dehydration.Despite having the highest stomatal density and intermediate stomatal sensitivity to ABA,V.riparia lost the smallest amount of water,indicating that it was the most sensitive to dehydration.The assay presented here represents a simple and reliable phenotyping method for plant responses to leaf dehydration.Daniel W Hopper Ryan Ghan Grant R Cramer 2014Horticulture Research2014,1,1:2

9Appendectomy and Clostridium difficile colitis:Relationships revealed by clinical observations and immunology显示文摘Advances in understanding the interaction between the human immune system and the microbiome have led to an improved understanding of the function of the vermiform appendix as a safe-house for beneficial bacteria in the colon.These advances have been made despite long standing clinical observations that the appendectomy is a safe and effective procedure.However,more recent clinical data show that an appendectomy puts patients at increased risk for recurrent Clostridium difficile(C.difficile)-associated colitis,and probably other diseases associated with an altered microbiome.At the same time,appendectomy does not apparently put patients at risk for an initial onset of C.difficile-associated colitis.These clinical observations point toward the idea that the vermiform appendix might not effectively protect the microbiome in the face of broad spectrum antibiotics,the use of which precedes the initial onset of C.difficile-associated colitis.Further,these observations point to the idea that historically important threats to the microbiome such as infectious gastrointestinal pathogens have been supplanted by other threats,particularly the use of broad spectrum antibiotics.Nathan L Sanders R Randal Bollinger Ryan Lee Steven Thomas William Parker 2013World Journal of Gastroenterology2013,19,34:2
