2Fusobacterium nucleatum enhances the efficacy of PD-L1 blockade in colorectal cancer显示文摘Given that only a subset of patients with colorectal cancer(CRC)benefit from immune checkpoint therapy,efforts are ongoing to identify markers that predict immunotherapeutic response.Increasing evidence suggests that microbes influence the efficacy of cancer therapies.Fusobacterium nucleatum induces different immune responses in CRC with different microsatellite-instability(MSI)statuses.Here,we investigated the effect of F.nucleatum on anti-PD-L1 therapy in CRC.We found that high F.nucleatum levels correlate with improved therapeutic responses to PD-1 blockade in patients with CRC.Additionally,F.nucleatum enhanced the antitumor effects of PD-L1 blockade on CRC in mice and prolonged survival.Combining F.nucleatum supplementation with immunotherapy rescued the therapeutic effects of PD-L1 blockade.Furthermore,F.nucleatum induced PD-L1 expression by activating STING signaling and increased the accumulation of interferon-gamma(IFN-γ)^(+)CD8^(+)tumor-infiltrating lymphocytes(TILs)during treatment with PD-L1 blockade,thereby augmenting tumor sensitivity to PD-L1 blockade.Finally,patient-derived organoid models demonstrated that increased F.nucleatum levels correlated with an improved therapeutic response to PD-L1 blockade.These findings suggest that F.nucleatum may modulate immune checkpoint therapy for CRC.Yaohui Gao Dexi Bi Ruting Xie Man Li Jing Guo Hu Liu Xianling Guo Juemin Fang Tingting Ding Huiyuan Zhu Yuan Cao Meichun Xing Jiayi Zheng Qing Xu Qian Xu Qing Wei Huanlong Qin 2021Signal Transduction and Targeted Therapy2021,6,12:3

4Oxidative coupling of cinnamic acid derivatives and their radical-scavenging activities显示文摘4-Hydroxycinnamic acid derivatives,including ferulic acid (FA) (1),sinapic acid (SA) (2) and caffeic acid (CA) (3),are widely distributed in the plant kingdom,and undergo oxidative cross-coupling leading to the corresponding dehydrodimers,trimers and even higher oligomers in plants. In order to evaluate the antioxidative ability of these 4-hydroxycinnamic acid derivatives and their oligomers,we synthesized 8-8'-bis-lactone-dimers (8-8'-DiFA (4),8-8'-DiSA (5) and 8-8'-DiCA (6)),as well as a new trimer (7),by the reaction of the corresponding monomers (1-3) with Ag2O in methanol,and assayed their free radical-scavenging activity by the reaction with 2,2-diphenyl-1-picrylhydrazyl radical (DPPH·). 5Anti-diabetic effect of aloin via JNK-IRS1/PI3K pathways and regulation of gut microbiota显示文摘This research aimed to investigate the antidiabetic activity,underlying mechanisms,and gut microbiota regulation of aloin.The insulin-resistant HepG2(IR-HepG2)cell model and the type 2 diabetic(T2D)mouse model were successfully established using dexamethasone and a high-fat high-sucrose diet with low-dose streptozotocin,respectively.Aloin intervention increased glucose consumption and stimulated the activity of hexokinase and pyruvate dehydrogenase in IR-HepG2 cells.Additionally,it diminished the weight loss,reduced fasting blood glucose levels and hemoglobin A1c activity,and promoted glucose tolerance and fasting serum insulin activity in T2D mice.Histopathological analysis of the liver indicated hepatic protection by aloin.Additionally,aloin treatment inhibited the protein expression of c-Jun N-terminal kinases and activated that of IRS1/PI3K/Akt in the liver.Moreover,aloin modulated the bacterial community in the gut by raising the abundance of Bacteroidota and reducing the richness of Firmicutes,Proteobacteria,and Actinobacteriota.Thus,aloin ameliorated IR via activating IRS1/PI3K/Akt signaling pathway and regulating the gut microbiota,and it may be promising candidate as functional food for diabetic therapy.Ruting Zhong Lanbin Chen Yuanyuan Liu Shouxia Xie Sumei Li Bin Liu Chao Zhao 2022Food Science and Human Wellness2022,11,1:2
6Lanthanide-based downshifting layers tested in a solar car race显示文摘The mismatch between the AM1.5G spectrum and the photovoltaic (PV) cells absorption is one of the most limiting factors for PV performance.To overcome this constraint through the enhancement of solar energy harvesting,luminescent downshifting (LDS) layers are very promising to shape the incident sunlight and,thus,we report here the use of Tb^3+- and Eu^3+-doped organic-inorganic hybrid materials as LDS layers on Si PV cells.Electrical measurements on the PV cell,done before and after the deposition of the LDS layers,confirm the positive effect of the coatings on the cell’s performance in the UV spectral region.The maximum delivered power and the maximum absolute external quantum efficiency increased 14% and 27%,respectively.Moreover,a solar powered car race was organized in which the small vehicle containing the coated PV cells presented a relative increase of 9% in the velocity,when compared to that with the uncoated one.Sandra F.H.Correia Ana R.N.Bastos Lianshe Fu Luís D.Carlos Paulo S.André Rute A.S.Ferreira 2019Opto-Electronic Advances2019,2,6:1

8Somatostatin 4 regulates growth and modulates gametogenesis in zebrafish显示文摘Somatostatin(SST)plays important roles in growth and development.In teleost fishes six SST encoding genes(sst1 to sst6)have been identified although few studies have addressed their function.Here we aim to determine the function of the teleost specific sst4 in the zebrafish.A CRISPR/Cas9 sst4 zebrafish mutant with loss of function(sst4−/−)was produced which grew significantly faster and was heavier at the onset of gonadal maturation than the wild type(WT).Consistent with their faster growth,liver igf1,igf2a and igf2b expression was significantly upregulated in the sst4−/−fish compared to the WT.Histological examination of the ovaries and testis indicated that sst4−/−fish had slightly delayed testicular gametogenesis compared to the WT.Significantly lower expression of igf3,amh,insl3,hsd17b3,hsd11b2,hsd20b,cyp11b and cyp17 was consistently observed in the sst4−/−testis.In contrast,the ovaries had lower expression of igf1,igf2a and cyp19a1a but increased expression of igf2b and hsd20b.The gonadotrophin beta subunits(fshb and lhb)in the brain were downregulated indicating the brain-pituitary-gonadal axis was downregulated in the sst4−/−fish and suggesting that the steroid production is compromised in the maturing gonads.In addition,analysis of sst1 and sst3 mRNA levels in sst4−/−fish suggests a dosage compensation effect of sst1 in the brain and liver.Altogether,the results from the zebrafish sst4−/−line support the idea that sst4 is involved in the regulation of igf signalling,somatic growth and reproduction since steroidogenesis and gametogenesis at pubertal onset were compromised.Chenchao Sui Jie Chen Jing Ma Wenting Zhao Adelino V.M.Canário Rute S.T.Martins 2019Aquaculture and Fisheries2019,4,6:1
