1Effects of Biochar and Super Absorbent Polymer on Substrate Properties and Water Spinach Growth显示文摘Most previous researches have focused on biochar application in agricultural soils; however, limited information is available concerning the effects of biochar amendment on greenhouse substrate properties. A greenhouse experiment was conducted to investigate effects of wheat straw biochar(0–160 m L L-1) and super absorbent polymer(SAP, 0.8 g L-1) on physical and chemical properties of a substrate based on spent pig litter compost and the growth of water spinach(Ipomoea aquatica Forsk). Total porosity, water-holding capacity, p H and electrical conductivity(EC) of the substrate significantly increased with increasing biochar rates, especially in the substrate without SAP. The values of p H and EC were significantly lower in the substrate with SAP than those without SAP at the high biochar application rates(100–160 m L L-1). The germination rates of water spinach decreased with increasing biochar rates when biochar was added alone(76.9%–83.7%), whereas the rates increased to 83.6%–85.8% when biochar was added in combination with SAP. Growth parameters of water spinach and nutrient uptake by shoots and roots increased with increasing biochar rates and reached the maximum values at the biochar rate of 100 m L L-1. There were significant cubic relationships between the uptake of nutrients(N, P, and K) and biochar rates, both with and without SAP addition. In order to avoid negative effects on plant growth,the biochar application rate should be controlled at an optimal level(100 m L L-1). 4Effects of Tillage Management on Infiltration and Preferential Flow in a Black Soil, Northeast China显示文摘The impacts of no-tillage (NT) and moldboard plough (MP) managements on infiltration rate and preferential flow were characterized using a combined technique of double-ring device and dye tracer on a black soil (Mollisols) in Northeast China. The objective of this study is to evaluate how tillage practices enhance soil water infiltration and preferential flow in favor of soil erosion control in the study area. The steady infiltration rates under NT management are 1.6 and 2.1 times as high as those under MP management in the 6th and 8th years of the tillage management in place, while the infiltrated water amounts under NT management are 1.4 and 2.0 times as high as those under MP management, respectively. The depth of methylene blue penetrated into NT soil increases from 43cm in the 6th year to 57cm in the 8th year, which are 16cm and 19cm deeper than those in MP soil, respectively. The results of morphologic image show that more biological macro-pores occur in NT soil than in MP soil. These macro-pores play a key role in enhancing preferential flow in NT soil, which in turn promotes water infiltration through preferential pathways in NT soil. 5Homogeneous label-free fluorescent assay of small molecule-protein interactions using protein binding-inhibited transcription nanomachine显示文摘Quantitative analysis of interactions between small molecules and proteins is a central challenge in chemical genetics, molecular diagnostics and drug developments. Here, we developed a RNA transcription nanomachine by assembling T7 RNA polymerase on a small molecule-labeled DNA heteroduplex. The nanomachine, of which the RNA transcription activity can be quantitatively inhibited by protein binding, showed a great potential for small molecule-protein interaction assay. This finding enabled us to develop a novel homogeneous label-free strategy for assays of interactions between small molecules and their protein receptors. Three small molecule compounds and their protein receptors have been used to demonstrate the developed strategy. The results revealed that the protein-small molecule interaction assay strategy shows dynamic responses in the concentration range from 0.5 to 64 nM with a detection limit of 0.2 nM. 6HPLC-DAD data coupled with second-order calibration method applied to food analysis: Simultaneous determination of six benzoylurea insecticides in various fruit samples by selecting time region of chromatogram显示文摘This work presents a novel application of second-order calibration based on self-weighted alternating trilinear decomposition(SWATLD)algorithm for analyzing the HPLC-DAD data.The proposed method makes it possible to simultaneously determine teflubenzuron,hexaflumuron,flufenoxuron,chlorfluazuron,diflubenzuron and benzoylurea in different fruit samples,i.e.pear,apple and banana,in the selected time region of chromatogram.The concentration,elution time and spectral information of these benzoylurea insecticides are selectively extracted from complex matrices even in the presence of unknown interferences.The root-mean-square error of prediction(RMSEP)and figures of merit,including sensitivity(SEN),selectivity(SEL)and limit of detection(LOD)are employed to access the performance of the method.The LODs obtained for these insecticides are within the range 0.017–0.26 ppm in pears,0.039–0.33 ppm in apples,0.041–0.44 ppm in bananas,respectively.Such a chemometrics-based protocol holds great potential to be extended as a promising alternative for more practical applications in food safety and quality monitoring.QING XiangDong WU HaiLong NIE ChongChong LI Yong YAN XiuFang ZHANG XiHua YIN XiaoLi YU RuQin 2013Science China Chemistry2013,56,11:2
7Rapid determination of thiabendazole in orange extract using excitation-emission matrix fluorescence and second-order calibration based on alternating trilinear decomposition/alternating normalization-weighted error algorithms显示文摘A novel approach is proposed for direct quantitative analysis of thiabendazole in the orange extract by using excitation-emission matrix fluorescence coupled with second-order calibration methods based on the alternating trilinear decomposition(ATLD) and the alternating normalization-weighted error(ANWE) algorithms,respectively. The average recoveries of thiabendazole in the orange extract by using ATLD and ANWE with an estimated component number of two were 99.7 ± 3.3% and 103.5 ± 4.1%,respectively. Furthermore,the accuracy of the two algorithms was also evaluated through elliptical joint confidence region(EJCR) tests as well as figures of merit,such as sensitivity(SEN),selectivity(SEL) and limit of detection(LOD).
