1Uncompromised NK cell activation is essential for virus-specific CTL activity during acute influenza virus infection显示文摘Natural killer (NK) cells are indispensable components of both the innate and adaptive immune response. However,their precise roles in the cross-talk between innate and adaptive immunity during influenza virus infection remaincontroversial. By comparing NK cell dynamics and activity under a sub-lethal dose and high dose of influenza virusinfection, we showed that influenza virus PR8 directly infected NK cells during natural infection, which wasconsistent with our previous findings obtained from an in vitro investigation of human NK cells. The impairments incytotoxicity and IFN-γ production by spleen NK cells following high-dose infection were accompanied by decreasedvirus-specific killing mediated by cytotoxic T lymphocytes (CTLs). Importantly, the weakened CTL activity could bereversed by adoptive transfer of spleen NK cells harvested from low-dose-infected mice but not healthy donors.Taken together, our data provide direct evidence supporting the contribution of NK cells to antiviral T-cellresponses. 2Effects of Tillage Management on Infiltration and Preferential Flow in a Black Soil, Northeast China显示文摘The impacts of no-tillage (NT) and moldboard plough (MP) managements on infiltration rate and preferential flow were characterized using a combined technique of double-ring device and dye tracer on a black soil (Mollisols) in Northeast China. The objective of this study is to evaluate how tillage practices enhance soil water infiltration and preferential flow in favor of soil erosion control in the study area. The steady infiltration rates under NT management are 1.6 and 2.1 times as high as those under MP management in the 6th and 8th years of the tillage management in place, while the infiltrated water amounts under NT management are 1.4 and 2.0 times as high as those under MP management, respectively. The depth of methylene blue penetrated into NT soil increases from 43cm in the 6th year to 57cm in the 8th year, which are 16cm and 19cm deeper than those in MP soil, respectively. The results of morphologic image show that more biological macro-pores occur in NT soil than in MP soil. These macro-pores play a key role in enhancing preferential flow in NT soil, which in turn promotes water infiltration through preferential pathways in NT soil. 3Pyrrolizidine alkaloids:An update on their metabolism and hepatotoxicity mechanism显示文摘Pyrrolizidine alkaloids(PAs)are among the most hepatotoxic natural compounds that are widely distributed throughout the world.Most PAs are metabolically activated to trigger toxicity.Exposure to herbal medicine containing PAs and food supplements contaminated by PAs is considered to be one of the two main causes of hepatic sinusoidal obstruction syndrome(HSOS),which is a rare hepatic vascular disease with a high mortality rate.PAs-induced HSOS cases have been reported worldwide.However,there is no clinically effective therapy for PAs-induced HSOS,which is partially because the toxic mechanism is not fully understood.This review focuses on updating the information on the metabolism and the molecular mechanisms of PAs hepatotoxicity,including oxidative stress,apoptosis,and dysfunction of bile acid metabolism,and their interactions.Jie Xu Weiqian Wang Xiao Yang Aizhen Xiong Li Yang Zhengtao Wang 2019Liver Research2019,3,3:5
4Validation of the Nanjing Criteria for Diagnosing Pyrrolizidine Alkaloids-induced Hepatic Sinusoidal Obstruction Syndrome显示文摘Background and Aims:Hepatic sinusoidal obstruction syndrome(HSOS)is caused by toxic injury to sinusoidal endothelial cells in the liver.The intake of pyrrolizidine alkaloids(PAs)in some Chinese herbal remedies/plants remains the major etiology for HSOS in China.Recently,new diagnostic criteria for PA-induced HSOS(i.e.PA-HSOS)have been developed;however,the efficacy has not been clinically validated.This study aimed to assess the performance of the Nanjing criteria for PA-HSOS.Methods:Data obtained from consecutive patients in multiple hospitals,which included 86 PA-HSOS patients and 327 patients with other liver diseases,were retrospectively analyzed.Then,the diagnostic performance of the Nanjing criteria and simplified Nanjing criteria were evaluated and validated.The study is registered in www.chictr.org.cn(ID:ChiCTR1900020784).Results:The Nanjing criteria have a sensitivity and specificity of 95.35%and 100%,respectively,while the simplified Nanjing criteria have a sensitivity and specificity of 96.51%and 96.33%,respectively,for the diagnosis of PA-HSOS.Notably,a proportion of patients with Budd-Chiari syndrome(11/49)was misdiagnosed as PA-HSOS on the basis of the simplified Nanjing criteria,and this was mainly due to the overlapping features in the enhanced computed tomography/magnetic resonance imaging examinations.Furthermore,most of these patients(10/11)had occlusion or thrombosis of the hepatic vein,and communicating vessels in the liver were found in 8/11 patients,which were absent in PA-HSOS patients.Conclusions:The Nanjing criteria and simplified Nanjing criteria exhibit excellent performance in diagnosing PA-HSOS.Thus,both could be valuable diagnostic tools in clinical practice.Wei Zhang Lu Liu Ming Zhang Feng Zhang Chunyan Peng Bin Zhang Jun Chen Lin Li Jian He Jiangqiang Xiao Yanhong Feng Xunjiang Wang Aizhen Xiong Li Yang Xiaoping Zou Yuecheng Yu Yuzheng Zhuge 2021Journal of Clinical and Translational Hepatology2021,9,3:4
5mRNA profiles of cytokine receptors in unstimulated peripheral blood mononuclear cells from patients with chronic idiopathic urticaria显示文摘This present study was aimed to investigate the roles of the receptors of Th1/Th2 cytokines and chemokines in the pathogenesis of chronic idiopathic urticaria (CIU).Thirty patients with CIU,30 patients with dermographism and 30 healthy controls were randomly enrolled.Reverse transcription-PCR (RT-PCR) was used to analyze the mRNA of cytokine receptors in peripheral blood mononuclear cells (PBMCs).The mRNA levels of tumor necrosis factor receptor (TNFR),interferon-γ receptor (IFN-γR),and interleukin-10 receptor (IL-10R) were statistically increased in the CIU group (P < 0.05),while IL-2R,IL-4R,IL-6R,and IL-13R showed no significant differences between the CIU and other groups.The mRNA levels of CCR3 and CCR6 were statistically increased in the CIU group (P < 0.05).The toll-like receptor 2 (TLR2) mRNA level was significantly lower in the CIU group than the healthy control group (P < 0.05).These findings indicate that the regulation of mRNA of TNFR,IFN-γR,IL-10R,CCR3,CCR6 and TLR2 may be involved in the pathogenesis of CIU.Jianming Gao Aizhen Yang Min Chen Ansheng Li Xu Yao Yumei Li Shihai Xie Xueyuan Yang Liansheng Zhong Zhiqiang Chen 2011The Journal of Biomedical Research2011,25,2:3
6Aggregate-related microbial communities and nutrient stoichiometry under different croplands显示文摘Ecological stoichiometry is an important indicator presenting multiple elements balance in agro-ecosystems.However,information on microbial communities and nutrient stoichiometry in soil aggregate fractions under different croplands(rice,maize,and soybean fields)remains limited.Thus,this study investigated water-stable aggregate structure and their internal nutrient stoichiometry under different croplands and ascertain their interaction mechanism with microbial communities.The results showed that no significant difference on the carbon-to-nitrogen ratio(C:N)in soil aggregate fractions was observed,while the carbon-to-phosphorus ratio(C:P)and the nitrogen-to-phosphorus ratio(N:P)were ranked as rice field>maize field>soybean field,and were higher in mega-aggregates(ME,>1 mm).General fatty acid methyl ester(FAME),Gram-positive bacteria(G+),and Gramnegative bacteria(G−)were predominant microbial communities in all croplands and tented to condense into coarse-aggregates.Redundancy analysis(RDA)demonstrated that N:P ratio was primary environmental controls on the distribution of soil microorganisms.In the Sanjiang Plain,N was the nutrient element limiting agro-ecosystem productivity,and rice cultivation is expected to improve the N-limited nutrient status.Hu Cui Yang Ou Duian Lv Lixia Wang Aizhen Liang Baixing Yan Yingxin Li 2020Ecological Processes2020,9,1:2

8Phytochemical Contents and Antioxidant Properties of Ganoderma lucidum Polysaccharides in Relation to Growth Stage and Culture Substrate显示文摘[Objectives]To determine physicochemical characteristics and antioxidant activity of Ganoderma lucidum polysaccharides(GLP)in different growth stages and culture media.[Methods]Five polysaccharides(GLP1,GLP2,GLP3,GLPW and GLPB)were extracted and purified from the fruiting body of G.lucidum at three growth stages and culture substrates(wood culture and bag culture).The chemical components and antioxidant activity of the five polysaccharides were determined.[Results]GLP1 contained the highest content of neutral sugar(87.65%)and GLP3 had the highest uronic acid content.All the samples mainly comprised glucose,galactose,mannose,xylose and arabose with different ratios.Moreover,their antioxidant activities were investigated on the basis of DPPH radical,ABTS radical,SOD-like activity,and antihaemolytic activity.Results indicated in all three growth stages GLP2 had the greatest antioxidant properties.In addition,the antioxidant activity of GLPW was significantly higher than that of GLPB.[Conclusions]Overall,by comparison,G.lucidum in growth stage two may have potential health benefits,and wood culture may be a superior choice of artificial cultivation due to their abundance of active polysaccharides.Guoying LU Aizhen HE Jinrong CHEN Zhouwei XU Chaolian HE Jianfei CHEN Zuofa ZHANG 2023Medicinal Plant2023,14,3:1
