4Mechanical force-driven TNFαendocytosis governs stem cell homeostasis显示文摘Mesenchymal stem cells(MSCs)closely interact with the immune system,and they are known to secrete inflammatory cytokines in response to stress stimuli.The biological function of MSC-derived inflammatory cytokines remains elusive.Here,we reveal that even under physiological conditions,MSCs produce and release a low level of tumor necrosis factor alpha(TNFα),which is unexpectedly required for preserving the self-renewal and differentiation of MSCs via autocrine/paracrine signaling.Furthermore,TNFαcritically maintains MSC function in vivo during bone homeostasis.Mechanistically,we unexpectedly discovered that physiological levels of TNFαsafeguard MSC homeostasis in a receptor-independent manner through mechanical force-driven endocytosis and that endocytosed TNFαbinds to mammalian target of rapamycin(mTOR)complex 2 and restricts mTOR signaling.Importantly,inhibition of mTOR signaling by rapamycin serves as an effective osteoanabolic therapeutic strategy to protect against TNFαdeficiency and mechanical unloading.Collectively,these findings unravel the physiological framework of the dynamic TNFαshuttlebased mTOR equilibrium that governs MSC and bone homeostasis.Wenjing Yu Chider Chen Xiaoxing Kou Bingdong Sui Tingting Yu Dawei Liu Runci Wang Jun Wang Songtao Shi 2021Bone Research2021,9,1:1

6Palaeoenvironmental reconstruction for the Permian(lower Gondwana)succession of the Godavari Valley Coalfield in southern India based on a combined palynofacies,carbon isotope,and biomarker study显示文摘The study presents integrated palynofacies,organic carbon isotope,and biomarker data to reconstruct palaeoenviromental setting for the Permian sediments(borehole MGK-6)of the Kachinapalli block of the Godavari Valley Coalfield,southern India.The palynofacies data reveal three distinct palynofacies(A-C)as follows.Palynofacies A is dominated by abundant arborescent vegetation(glossopterids and conifers)along with structured organic matter,suggesting freshwater forest swamps in proximal settings.Palynofacies B is dominated by abundant degraded organic matter and amorphous organic matter,suggesting lakeshore/flooded palaeomires in slightly dismal settings.It has a high water level in the hinterland,which enables bacterial degradation of organic matter and establishes low-oxygenation conditions within the water column.These conditions would have provided grounds for the accumulation of either degraded or amorphous organic matter.Palynofacies C is dominated by the abundance of charcoal/opaque phytoclasts,indicating prolonged transportation or postdepositional alteration,suggesting a highly-oxidizing condition in distal settings.The bulk organic δ^(13)C range(-25.1‰ to-20.9‰)in the borehole MGK-6 is in close agreement with the global Permian records.This study also shows a weak but statistically significant correlation with the major forms of palynofacies A and C.The interlinked behaviour of bulk organic δ^(13)C and palynofacies suggests that the changes in palaeobiogeography/palaeodepositional settings were most likely driven by the change in mean annual precipitation.The biomarker study shows the presence of n-alkane from C_(15) to C_(31) with unimodal and bimodal distribution patterns,revealing the source materials as vascular plants and microbially-altered organic matter.The combined palynofacies,organic carbon isotope,and biomarker data provide vital clue to salient findings for the development of environmental conditions of Godavari Valley Coalfield during lower Gondwana sedimentation.Neha Aggarwal Runcie Paul Mathews Arif Husain Ansari Biswajeet Thakur Shailesh Agrawal 2022Journal of Palaeogeography2022,11,1:1

10Apoptotic vesicles ameliorate lupus and arthritis via phosphatidylserine-mediated modulation of T cell receptor signaling显示文摘Mesenchymal stem cells(MSCs)influence T cells in health,disease and therapy through messengers of intercellular communication including extracellular vesicles(EVs).Apoptosis is a mode of cell death that tends to promote immune tolerance,and a large number of apoptotic vesicles(apoVs)are generated from MSCs during apoptosis.In an effort to characterize these apoVs and explore their immunomodulatory potential,here we show that after replenishing them systemically,the apoV deficiency in Fas mutant mice and pathological lymphoproliferation were rescued,leading to the amelioration of inflammation and lupus activity.ApoVs directly interacted with CD4^(+)T cells and inhibited CD25 expression and IL-2 production in a dose-dependent manner.A broad range of Th1/2/17 subsets and cytokines including IFNγ,IL17A and IL-10 were suppressed while Foxp3^(+)cells were maintained.Mechanistically,exposed phosphatidylserine(PtdSer/PS)on apoVs mediated the interaction with T cells to disrupt proximal T cell receptor signaling transduction.Remarkably,administration of apoVs prevented Th17 differentiation and memory formation,and ameliorated inflammation and joint erosion in murine arthritis.Collectively,our findings unveil a previously unrecognized crosstalk between MSC apoVs and CD4^(+)T cells and suggest a promising therapeutic use of apoVs for autoimmune diseases.Runci Wang Meng Hao Xiaoxing Kou Bingdong Sui Maria Laura Sanmillan Xiao Zhang Dawei Liu Jun Tian Wenjing Yu Chider Chen Ruili Yang Lingyun Sun Yi Liu Claudio Giraudo Deepak A.Rao Nan Shen Songtao Shi 2023Bioactive Materials2023,,7:1
