| 1 | Meiotic nuclear divisions 1(MND1)fuels cell cycle progression by activating a KLF6/E2F1 positive feedback loop in lung adenocarcinoma显示文摘Background:Considering the increase in the proportion of lung adenocarcinoma(LUAD)cases among all lung cancers and its considerable contribution to cancer-related deaths worldwide,we sought to identify novel oncogenes to provide potential targets and facilitate a better understanding of the malignant progression of LUAD.Methods:The results from the screening of transcriptome and survival analyses according to the integrated Gene Expression Omnibus(GEO)datasets and The Cancer Genome Atlas(TCGA)data were combined,and a promising risk biomarker called meiotic nuclear divisions 1(MND1)was selectively acquired.Cell viability assays and subcutaneous xenograftmodelswere used to validate the oncogenic role ofMND1 in LUADcell proliferation and tumor growth.Aseries of assays,including mass spectrometry,co-immunoprecipitation(Co-IP),and chromatin immunoprecipitation(ChIP),were performed to explore the underlying mechanism.Results:MND1 up-regulation was identified to be an independent risk factor for overall survival in LUAD patients evaluated by both tissue microarray staining and third party data analysis.In vivo and in vitro assays showed that MND1 promoted LUAD cell proliferation by regulating cell cycle.The results of the Co-IP,ChIP and dual-luciferase reporter assays validated that MND1 competitively bound to tumor suppressor Kruppel-like factor 6(KLF6),and thereby protecting E2F transcription factor 1(E2F1)from KLF6-induced transcriptional repression.Luciferase reporter and ChIP assays found that E2F1 activated MND1 transcription by binding to its promoter in a feedback manner.Conclusions:MND1,KLF6,and E2F1 form a positive feedback loop to regulate cell cycle and confer DDP resistance in LUAD.MND1 is crucial for malignant progression and may be a potential therapeutic target in LUAD patients. | Quanli Zhang Run Shi Yongkang Bai Lijuan Meng Jingwen Hu Hongyu Zhu Tongyan Liu Xiaomeng De Siwei Wang Jie Wang Lin Xu Guoren Zhou Rong Yin | 2021 | Cancer Communications2021,41,6: | 2 |
| 2 | Effects of V and Zr Additions on Microstructures and Mechanical Properties of Nb-Ti-Al-Base Alloys显示文摘Microstructures and mechanical properties of Nb-32Ti-7Al alloys containing different V and Zr contents were investigated. The microstructures were characterized using optical microscopy and scanning electron microscopy (SEM). The alloy with V and Zr presents single phase Nb solid solution (Nbss). Tensile testing was carried out at room temperature and 1373 K. The results show that these alloys have good ductility at room temperature. | 3 | Preferentially released miR-122 from cyclodextrin-based star copolymer nanoparticle enhances hepatoma chemotherapy by apoptosis induction and cytotoxics efflux inhibition显示文摘Chemotherapy,as one of the most commonly used treatment modalities for cancer therapy,provides limited benefits to hepatoma patients,owing to its inefficient delivery as well as the intrinsic chemo-resistance of hepatoma.Bioinformatic analysis identified the therapeutic role of a liver-specific microRNA-miR-122 for enhancing chemo-therapeutic efficacy in hepatoma.Herein,a cyclodextrin-cored star copolymer nanoparticle system(sCDP/DOX/miR-122)is constructed to co-deliver miR-122 with doxorubicin(DOX)for hepatoma therapy.In this nanosystem,miR-122 is condensed by the outer cationic poly(2-(dimethylamino)ethyl methacrylate)chains of sCDP while DOX is accommodated in the inner hydrophobic cyclodextrin cavities,endowing a sequential release manner of miR-122 and DOX.The preferentially released miR-122 not only directly induces cell apoptosis by down regulation of Bcl-w and enhanced p53 activity,but also increases DOX accumulation through inhibiting cytotoxic efflux transporter expression,which realizes synergistic performance on cell inhibition.Moreover,sCDP/DOX/miR-122 displays remarkably increased anti-tumor efficacy in vivo compared to free DOX and sCDP/DOX alone,indicating its great promising in hepatoma therapy. | Qingqing Xiong Yang Bai Run Shi Jian Wang Weiguo Xu Mingming Zhang Tianqiang Song | 2021 | Bioactive Materials2021,6,11: | 2 |

| 11 | Yiqi Qingre Xiaozheng formula protects against diabetic nephropathy by restoring autophagy in mice显示文摘Objective:To evaluate the protective effect of Yiqi Qingre Xiaozheng formula(YQQRXZF)via the regulation of autophagy in diabetic nephropathy(DN)mice induced by injection of streptozotocin(STZ)after high fat diet(HFD).Methods:The composition of YQQRXZF was analyzed by high performance liquid chromatographyelectrospray ionization/mass spectrometry(HPLC-ESI/MSn).The DN model was induced by intraperitoneally injection of 50 mg/kg STZ within 5 days,followed by HFD feeding for 8 weeks in C57BL/6J mice.Mice were randomly separated into DN group,YQQRXZF group,irbesartan group,and control group.Blood glucose was calculated,and body weight was measured every 2 weeks.An enzyme-linked immunosorbent assay was used to measure the urinary albumin-to-creatinine ratio(uACR)before and after treatment,and the serum concentrations of total cholesterol(TC),low-density lipoprotein(LDL),triglycerides(TG),high-density lipoprotein(HDL),blood urea nitrogen(BUN),and serum creatinine(Scr)were measured.In addition,hematoxylin-eosin(H&E)staining,periodic acid-Schiff(PAS)staining,Masson's trichrome staining,and transmission electron microscopy(TEM)were used to observe pathological changes in renal tissue.Autophagy levels were determined by immunofluorescence staining.Results:In this study,21 dominant chemical constituents were identified in YQQRXZF.The treatment group reduced u ACR in a more significant way than the DN group(P=.018).The treatment group reduced TC,TG,and LDL concentrations after YQQRXFF treatment(P=.021,P=.014 and P=.026,respectively).H&E,PAS,and Masson staining showed that pathological damage to mice kidneys improved,the volume of renal glomeruli was reduced,and glomerular sclerosis and fibrosis were reduced.Immunofluorescence analysis revealed that expressions of LC3-Ⅱprotein increased in the treatment group(P=.013).In contrast,the protein expressions of P62 were reduced after treatment(P=.025). | Na Sun Nan Yang Jingwei Zhou Ying He Jian Wang Yingnan Liang Run Dai Jiajia Bai Zhenjie Chen | 2023 | Journal of Traditional Chinese Medical Sciences2023,10,3: | 0 |
| 12 | Construction and validation of a severity prediction model for metabolic associated fatty liver disease显示文摘Objective:To analyze the independent risk factors for the occurrence of moderate-to-severe metabolic-associated fatty liver disease(MAFLD),to construct a prediction model for moderate-to-severe MAFLD,and to verify the validity of the model.Methods:In the first part,278 medical examiners who were diagnosed with MAFLD in Medical Examination Center at the Second Affiliated Hospital of Hainan University from January to May 2022 were taken as the study subjects(training set),and they were divided into mild MAFLD group(200)and moderate-severe MAFLD group(78)based on ultrasound results.Demographic data and laboratory indexes were collected,and risk factors were screened by univariate and multifactor analysis.In the second part,a dichotomous logistic regression equation was used to construct a prediction model for moderate-to-severe MAFLD,and the model was visualized in a line graph.In the third part,the MAFLD population(200 people in the external validation set)from our physical examination center from November to December 2022 was collected as the moderate-to-severe MAFLD prediction model,and the risk factors in both groups were compared.The receiver operating characteristic(ROC)curves,calibration curves,and clinical applicability of the model were plotted to represent model discrimination for internal and external validation.Results:The risk factors of moderate-to-severe MAFLD were fasting glucose(FPG),blood uric acid(UA),triglycerides(TG),triglyceride glucose index(TyG),total cholesterol(CHOL),and high-density lipoprotein(HDL-C).UA[OR=1.021,95%CI(1.015,1.027),P<0.001]and FPG[OR=1.575,95%CI(1.158,2.143),P=0.004]were independent risk factors for people with moderate to severe MAFLD.The visualized line graph model showed that UA was the factor contributing more to the risk of moderate to severe MAFLD in this model.The ROC curves showed AUC values of 0.8701,0.8686 and 0.7991 for the training set,internal validation set and external validation set,respectively.The curves almost coincided with the reference line after calibration of the model calibration degree with P>0.05 in Hosmer-Lemeshow test.The decision curve analysis(DCA)plotted by the clinical applicability of the model was higher than the two extreme curves,predicting that patients with moderate to severe MAFLD would benefit from the prediction model.Conclusion:The prediction model constructed by combining FPG with UA has higher accuracy and better clinical applicability,and can be used for clinical diagnosis. | ZHANG Da‑ya CHEN Shi‑ju CHEN Run‑xiang ZHANG Xiao‑dong HUANG Shi‑mei ZENG Fan CHEN Chen LI Da BAI Fei‑hu | 2023 | Journal of Hainan Medical University2023,29,8: | 0 |
