| 1 | BMAL1 regulates mitochondrial fission and mitophagy through mitochondrial protein BNIP3 and is critical in the development of dilated cardiomyopathy显示文摘Dysregulation of circadian rhythms associates with cardiovascular disorders.It is known that deletion of the core circadian gene Bma/1 in mice causes dilated car-diomyopathy.However,the biological rhythm regulation system in mouse is very different from that of humans.Whether BMAL1 plays a role in regulating human heart function remains unclear.Here we generated a BMAL1 knockout human embryonic stem cell(hESC)model and further derived human BMAL1 deficient cardiomy-ocytes.We show that BMAL1 deficient hESC-derived cardiomyocytes exhibited typical phenotypes of dilated cardiomyopathy including attenuated contractility,cal-cium dysregulation,and disorganized myofilaments.In addition,mitochondrial fission and mitophagy were suppressed in BMAL1 deficient hESC-cardiomyocytes,which resulted in significantly attenuated mitochondrial oxidative phosphorylation and compromised cardiomy-ocyte function.We also found that BMAL1 binds to the E-box element in the promoter region of BNIP3 gene and specifically controls BNIP3 protein expression.BMAL1 knockout directly reduced BNIP3 protein level,causing compromised mitophagy and mitochondria dysfunction and thereby leading to compromised cardiomyocyte function.Our data indicated that the core circadian gene S/VMLf is critical for normal mitochondria activities and cardiac function.Circadian rhythm disruption may directly link to compromised heart function and dilated cardiomyopathy in humans. | Ermin Li Xiuya Li Jie Huang Chen Xu Qianqian Liang Kehan Ren Aobing Bai Chao Lu Ruizhe Qian Ning Sun | 2020 | Protein & Cell2020,11,9: | 17 |

| 8 | Fluorine in drinking water and urine in the urban and rural areas of northwestern China——Its determination by a fluoride ion selective electrode显示文摘In this study, the author determined fluorine in drinking water and urine of residents who are divided into four age groups (5, 12, 35-44, 65-74 aged), living in Huangling City, Shaanxi Province and at 6 villages of Qin'an County, Gansu Province, P.R.China. Some residents are living in fluorine exposure areas. A total of 929 residents (463 females and 466 males) involved in the study were selected from 7 tap water systems. Drinking water samples were collected from each area and analyzed using the fluoride ion-selective method. No positive correlation relationship was found between fluorine concentrations in urine and those in drinking water in the area where the fluorine concentrations of drinking water are within the range of 1-2 mg/L. The fluorine absorbed by resistents of different ages is different in amount. With the same concentrations of fluorine in drinking water, more fluorine would be absorbed by young residents than old residents. | 13 | Cellulose-based Polymeric Liquid Crystals as a Biomimetic Modifier for Suppressing Protein Adsorption显示文摘A novel biomimetic protein-resistant modifier based on cellulose-based polymeric liquid crystals was described(PLCs). Two types of PLCs of propyl hydroxypropyl cellulose ester(PPC) and octyl hydroxypropyl cellulose ester(OPC) were prepared by esterification from hydroxypropyl cellulose, and then were mixed with polyvinyl chloride and polyurethane to obtain composite films by solution casting, respectively. The surface morphology of PLCs and their composite films were characterized by polarized optical microscopy(POM) and scanning electron microscopy(SEM), suggesting the existence of microdomain separation with fingerprint texture in PLC composite films. Water contact angle measurement results indicated that hydrophilicity of PLC/polymer composite films was dependent on the type and content of PLC as well as the type of matrix due to their interaction. Using bovine serum albumin(BSA) as a model protein, protein adsorption results revealed that PLCs with protein-resistant property can obviously suppress protein adsorption on their composite films, probably due to their flexible LC state. | 15 | Mitochondria-localized iridium(Ⅲ) complexes with anthraquinone groups as effective photosensitizers for photodynamic therapy under hypoxia显示文摘Photodynamic therapy(PDT)is a potential way for the tumor treatment.However,it notably suffers the limitation of hypoxia in solid tumors.Thus,it is significant to develop effective photosensitizers which can exhibit excellent therapeutic performance under both normoxia and hypoxia.Herein,we reported four ionic iridium(Ⅲ)complexes(Irl-Ir4)with anthraquinone groups which can regulate their excited state energy levels effectively.Among them,the energy gap of Ir1 was between 1.63 and 2.21 eV,which can match well with that of O2,and the HOMO energy of Ir1 is less than-5.51 eV.Compared with Ir2-Ir4,the luminescent quantum efficiency of Irl was the highest.Particularly,Ir1 can specifically target the mitochondria of the tumor cells.Meanwhile,Ir1 showed high singlet oxygen quantum yields(Ф△)in both solutions and living cells with low cytotoxicity.The results of PDT experiments revealed that Irl,as a photosensitizer,exhibited excellent therapeutic effect not only in normoxia but also in hypoxia condition.We believe that this work is meaningful for developing excellent PDT agents based on cyclometalated Ir(III)complexes via rational ligand modification. | Song Guo Meiping Han Ruizhe Chen Yanling Zhuang Liang Zou Shujuan Liu Wei Huang Qiang Zhao | 2019 | Science China Chemistry2019,62,12: | 1 |
