| 1 | Expression and function of classical protein kinase C isoenzymes in gastric cancer cell line and its drugresistant sublines显示文摘AIM: To investigate the expression and function of classicalprotein kinase C (PKC) isoenzymes in inducing MDRphenotype in gastric cancer cells.METHODS: Two cell lines were used in the study: gastriccancer cell SGC7901 and its drug-resistant cell SGC7901/VCRstepwise-selected by vincristine 0.3, 0. 7 and 1.0 mg@ L-1 ,respectively. The expression of classical PKC (cPKC)isoenzymes in SGC7901 cells and SGC7901/VCR cells weredetected using immunofluorescent cytochemistry, laserconfocal scanning microscope and Wsstern blot. The effectsof anti-PKC isoenzymes antibody of adriamycinaccumulation in SGC7901/VCR cells were determined usingflow cytometric analysis.RESULTS: (1) SGC7901 cells exhibited positive staining ofPKC-α. SGC7901/VCR cells exhibited stronger staining ofPKC-α than SGC7901 cells. The higher dosage vincristineselected, the much stronger staining of PKC-α was observedon SGC7901/VCR cells. (2) Both SGC7901 and SGC7901/VCRcells exhibited positive staining of PKC-βⅠ and PKC-βⅡ withno significant difference. ( 3 ) Compared with SGC7901,SGC7901/VCR cells had decreased adriamycin accumulationand retention. Accumulation of adriamycin in SGC7901 was5.21 + 2.56 mg@ L-1, in SGC7901/VCR 0.3 was 0.85 + 0.29 mg@L-1 , in SGC7901/VCR 0.7 was 0.81 + 0.32 og@ L-1 , and inSGC7901NCR 1.0 was 0.80 + 0.33 mg @ L-1; Retention ofadriamycin in SGC 7901 was 2.51 + 1.23 mg@L-1, in SGC7901/VCR 0.3 was 0.47 + 0.14 mg@ L-1 , in SGC7901/VCR 0.7 was 0.44 + 0.15 mg@ L-1, and in SGC 7901/VCR 1.0 was 0.41 + 0.1 1mg @ L-1 . | 3 | Protective effects of a novel drug RC28-E blocking both VEGF and FGF2 on early diabetic rat retina显示文摘AIM: To investigate protective effects of a novel recombinant decoy receptor drug RC28-E on retinal damage in early diabetic rats.METHODS: The streptozotocin(STZ)-induced diabetic rats were randomly divided into 6 groups: diabetes mellitus(DM) group(saline, 3 μL/eye); RC28-E at low(0.33 μg/μL, 3 μL), medium(1 μg/μL, 3 μL), and high(3 μg/μL,3 μL) dose groups; vascular endothelial growth factor(VEGF) Trap group(1 μg/μL, 3 μL); fibroblast growth factor(FGF) Trap group(1 μg/μL, 3 μL). Normal control group was included. At week 1 and 4 following diabetic induction,the rats were intravitreally injected with the corresponding solutions. At week 6 following the induction, apoptosis in retinal vessels was detected by TUNEL staining.Glial fibrillary acidic protein(GFAP) expression was examined by immunofluorescence. Blood-retinal barrier(BRB) breakdown was assessed by Evans blue assay.Ultrastructural changes in choroidal and retinal vessels were analyzed by transmission electron microscopy(TEM).Content of VEGF and FGF proteins in retina was measured by enzyme linked immunosorbent assay(ELISA). The retinal expression of intercellular cell adhesion molecule-1(ICAM-1), tumor necrosis factor-α(TNF-α), VEGF and FGF genes was examined by quantitative polymerase chain reaction(qPCR).RESULTS: TUNEL staining showed that the aberrantly increased apoptotic cells death in diabetic retinal vascular network was significantly reduced by treatments of medium and high dose RC28-E, VEGF Trap, and FGF Trap(all P<0.05), the effects of medium and high dose RC28-E or FGF Trap were greater than VEGF Trap(P<0.01). GFAP staining suggested that reactive gliosis was substantially inhibited in all RC28-E and VEGF Trap groups, but the inhibition in FGF Trap group was not as prominent.Evans blue assay demonstrated that only high dose RC28-E could significantly reduce vascular leakage in early diabetic retina(P<0.01). TEM revealed that the ultrastructures in choroidal and retinal vessels were damaged in early diabetic retina, which was ameliorated to differential extents by each drug. The expression of VEGF and FGF2 proteins was significantly upregulated in early diabetic retina, and normalized by RC28-E at all dosages and by the corresponding Traps. | 5 | In Situ HRTEM Observation of Electron-Irradiation-Induced Amorphization and Dissolution of the E(Al_(18)Cr_2Mg_3) Phase in 7475 Al Alloy显示文摘Electron irradiation effects on phase stability of the E(Al18Cr2Mg3) phase have been investigated by highangle annular dark-field scanning transmission electron microscopy and high-resolution transmission electron microscopy(HRTEM). The in situ HRTEM observations show that the Al18Cr2Mg3 particles with different thickness undergo amorphization and dissolution under 300 ke V electron irradiation at 25 °C. The results indicate that the intermetallic compound Al18Cr2Mg3 is unstable under electron irradiation, and structural changes mainly depend on the thickness of particles. Amorphization in the thick particles is caused by a combination of chemical disordering and an increase in point defect concentration. | 9 | EQR SiPM with P-on-N diode configuration显示文摘The silicon photomultiplier(SiPM) with epitaxial quenching resistor(EQR) is an emerging and developing technology that has recently attracted the interest from the research community. It has characteristics of a continuous low-resistance cap layer and integrated quenching resisters in epitaxial silicon layer, which makes it possible to increase microcell density or reduce microcell size, thus obtaining large dynamic range and high photon detection efficiency(PDE) simultaneously. Results published show that the EQR SiPM with N-on-P diode configuration had relatively low PDE at peak wavelength of 480 nm as 16%. This paper reported the EQR SiPM with P-on-N diode configuration having active area of 3 × 3 mm^2 and cell density of 10,000/mm^2(total 90,000 pixels). It was characterized with gain of 2E5, dark count rate of 7 MHz, crosstalk of 7%, dynamic range of 85,000 pixels, overall recovery time of 32 ns at room temperature and over-voltage of 3.5 V. | 13 | Effect of Pd doping on the acetone-sensing properties of NdFeO_3显示文摘The acetone-sensing properties of the undoped and Pd doped perovskite-type oxides NdFeO3 were investigated from room temperature to 400°C. The perovskite-type NdFeO3 was synthesized by a sol-gel method, and the dopants Pd with the content from 1wt% to 5wt% were implanted into NdFeO3 nanoparticles by thermal diffusion. X-ray diffraction (XRD) and scanning electron microscopy (SEM) techniques show that NdFeO3 is an orthorhombic structure with the average particle size of about 40 nm. A giant acetone-sensing response of 675.7 is observed when the Pd content in NdFeO3 powders is about 3wt%. The response and recovery time of the sensor to the 5×10-4 acetone gas are 16 and 1 s, respectively.
