2Upregulation of hypoxia inducible factor 1α mRNA is associated with elevated vascular endothelial growth factor expression and excessive angiogenesis and predicts a poor prognosis in gastric carcinoma显示文摘AIM: To investigate the implication of the hypoxia inducible factor HIF-1α mRNA in gastric carcinoma and its relation to the expression of vascular endothelial growth factor (VEGF) protein, tumor angiogenesis invasion/metastasis and the patient's survival. METHODS: In situ hybridization was used to examine expression of HIF-1α mRNA, and immunohistochemical staining was used to examine expression of VEGF protein and CD34 in 118 specimens from patients with gastric carcinoma. RESULTS: The positive rates of HIF-1α mRNA and VEGF protein were 49.15% and 55.92%, respectively. Positive expressions of HIF-1α and VEGF in stage T3-T4 tumors and those with vessel invasion, lymph node metastasis and distant metastasis were dramatically stronger than stage T1-T2 cases and those without vessel invasion, lymph node metastasis and distant metastasis. The mean microvascular density (MVD) in stage T3-T4 tumors and those with vessel invasion, lymph node metastasis and distant metastasis was significantly higher than stage T1-T2 tumors and those without vessel invasion, lymph node metastasis and distant metastasis. The mean MVD in tumors with positive HIF-1α and VEGF expression was significantly higher than that in tumors with negative HIF-1α and VEGF expression. The expression of HIF- 1α was positively correlated with VEGF protein. There were positive correlations between MVD and expressionof HIF-1α and VEGF. The mean survival time and the 5-year survival rate in cases with positive expression HIF-1α and VEGF and MVD value ≥ 41.5/0.72 mm2 were significantly lower than those with negative expression of HIF-1α and VEGF and MVD value < 41.5/0.72 mm2. CONCLUSION: Overexpression of HIF-1α is found in gastric carcinoma. HIF-1α may induce the angiogenesis in gastric carcinoma by upregulating the transcription of VEGF gene, and take part in tumor invasion and metastasis. 3Grape Seed Proanthocyanidin Extract Alleviates Arsenic-induced Oxidative Reproductive Toxicity in Male Mice显示文摘Objective To determine the ability of grape seed proanthocyanidin extract(GSPE) in alleviating arsenic-induced reproductive toxicity. Methods Sixty male Kunming mice received the following treatments by gavage: normal saline solution(control); arsenic trioxide(ATO; 4 mg/kg); GSPE(400 mg/kg); ATO+GSPE(100 mg/kg); ATO+GSPE(200 mg/kg) and ATO+GSPE(400 mg/kg). Thereafter, the mice were sacrificed and weighed, and the testis was examined for pathological changes. Nuclear factor(erythroid-derived 2)-like 2(Nrf2), heme oxygenase 1(HO1), glutathione S-transferase(GST), NAD(P)H dehydrogenase, and quinone 1(NQO1) expression in the testis was detected by real-time PCR. Superoxide dismutase(SOD), glutathione(GSH), total antioxidative capability(T-AOC), malondialdehyde(MDA), 8-hydroxydeoxyguanosine(8-OHd G), and reproductive indexes were analyzed. Results ATO-treated mice showed a significantly decreased sperm count and testis somatic index and activity levels of SOD, GSH, and T-AOC than control group. Compared to the ATO-treated group, ATO +GSPE group showed recovery of the measured parameters. Mice treated with ATO+high-dose GSPE showed the highest level of m RNA expression of Nrf2, HO, NQO1, and GST. 5High-Efficiency Electromagnetic Interference Shielding of rGO@FeNi/Epoxy Composites with Regular Honeycomb Structures显示文摘With the rapid development of fifth-generation mobile com-munication technology and wearable electronic devices,electromagnetic interference and radiation pollution caused by electromagnetic waves have attracted worldwide attention.Therefore,the design and development of highly efficient EMI shielding materials are of great importance.In this work,the three-dimensional graphene oxide(GO)with regular honeycomb structure(GH)is firstly constructed by sacrificial template and freeze-dry-ing methods.Then,the amino functionalized FeNi alloy particles(f-FeNi)are loaded on the GH skeleton followed by in-situ reduction to prepare rGH@FeNi aerogel.Finally,the rGH@FeNi/epoxy EMI shielding com-posites with regular honeycomb structure is obtained by vacuum-assisted impregnation of epoxy resin.Benefitting from the construction of regular honeycomb structure and electromagnetic synergistic effect,the rGH@FeNi/epoxy composites with a low rGH@FeNi mass fraction of 2.1 wt%(rGH and f-FeNi are 1.2 and 0.9 wt%,respectively)exhibit a high EMI shielding effectiveness(EMI SE)of 46 dB,which is 5.8 times of that(8 dB)for rGO/FeNi/epoxy composites with the same rGO/FeNi mass fraction.At the same time,the rGH@FeNi/epoxy composites also possess excellent thermal stability(heat-resistance index and temperature at the maximum decomposition rate are 179.1 and 389.0°C respectively)and mechanical properties(storage modulus is 8296.2 MPa).Ping Song Zhonglei Ma Hua Qiu Yifan Ru Junwei Gu 2022Nano-Micro Letters2022,14,3:13
6Effects of water salinity and N application rate on water- and N-use efficiency of cotton under drip irrigation显示文摘In arid and semi-arid regions, freshwater scarcity and high water salinity are serious and chronic problems for crop production and sustainable agriculture development. We conducted a field experiment to evaluate the effect of irrigation water salinity and nitrogen(N) application rate on soil salinity and cotton yield under drip irrigation during the 2011 and 2012 growing seasons. The experimental design was a 3×4 factorial with three irrigation water salinity levels(0.35, 4.61 and 8.04 dS/m) and four N application rates(0, 240, 360 and 480 kg N/hm2). Results showed that soil water content increased as the salinity of the irrigation water increased, but decreased as the N application rate increased. Soil salinity increased as the salinity of the irrigation water increased. Specifically, soil salinity measured in 1:5 soil:water extracts was 218% higher in the 4.61 dS/m treatment and 347% higher in the 8.04 dS/m treatment than in the 0.35 dS/m treatment. Nitrogen fertilizer application had relatively little effect on soil salinity, increasing salinity by only 3%–9% compared with the unfertilized treatment. Cotton biomass, cotton yield and evapotranspiration(ET) decreased significantly in both years as the salinity of irrigation water increased, and increased as the N application rate increased regardless of irrigation water salinity; however, the positive effects of N application were reduced when the salinity of the irrigation water was 8.04 dS/m. Water use efficiency(WUE) was significantly higher by 11% in the 0.35 dS/m treatment than in the 8.04 dS/m treatment. There was no significant difference in WUE between the 0.35 dS/m treatment and the 4.61 dS/m treatment. The WUE was also significantly affected by the N application rate. The WUE was highest in the 480 kg N/hm2 treatment, being 31% higher than that in the 0 kg N/hm2 treatment and 12% higher than that in the 240 kg N/hm2 treatment. There was no significant difference between the 360 and 480 kg N/hm2 treatments. The N use efficiency(NUE) was significantly lower in the 8.04 dS/m treatment than in either the 4.61 dS/m or the 0.35 dS/m treatment. There was no significant difference in NUE between the latter two treatments. These results suggest that irrigation water with salinity <4.61 dS/m does not have an obvious negative effect on cotton production, WUE or NUE under the experimental conditions. 7Gut microbiota dysbiosis-induced activation of the intrarenal renin-angiotensin system is involved in kidney injuries in rat diabetic nephropathy显示文摘Some studies have shown that gut microbiota along with its metabolites is closely associated with diabetic mellitus(DM).ln this study we explored the relationship between gut microbiota and kidney injuries of early diabetic nephropathy(DN)and its underlying mechanisms.Male SD rats were intraperitoneally injected with streptozotocin to induce DM.DM rats were orally administered compound broad-spectrum antibiotics for 8 wieeks.After the rats were sacrificed,their blood,urine,feces,and renal tssues were harvested for analyses.We found that compared with the control rats,DM rats had abnormal intestinalmicroflora,increased plasma acetate levels,increased proteinuria,thickened glomerular basement membrane,and podocyte foot process effacement in the kidneys.Furthermore,the protein levels of angiotensinⅡ,angiotensin-converting enzyme,and angiotensinⅡtype 1 receptor in the kidneys of DM rats were significantly increased.Administration of broad-spectrum antbiotics in DM rats not only completely killed most intestinal microflora,but also significanty lowered the plasma acetate levels,inhibited intrarenal RAS activation,and attenuated kidney damage.Finally,we showed that plasma acetate levels were positively correlated with intrarenal angiotensinⅡprotein expression(r=0.969,P<0.001).In conclusion,excessive acetate produced by disturbed gut microbiota might be involved in the kidney injuries of early DN through activating intrarenal RAS.Chen-chen Lu Ze-bo Hu Ru Wang Ze-hui Hong Jian Lu Pei-pei Chen Jia-xiu Zhang Xue-qi Li Ben-yin Yuan Si-jia Huang Xiong-zhong Ruan Bi-cheng Liu Kun-ling Ma 2020Acta Pharmacologica Sinica2020,41,8:12
8Active greeting technique: a mother-and-child catheter based technique to facilitate retrograde wire externalization in recanalization of coronary chronic total occlusion显示文摘Although retrograde approach has greatly improved the success rate of percutaneous coronary intervention(PCI) for coronary chronic total occlusion(CTO), retrograde wire externalization still remains challenging and time-consuming in some cases. Cases utilizing ‘‘Active Greeting Technique(AGT)', a mother-and-child catheter based technique to facilitate retrograde wire externalization, were extracted from Chronic Total Occlusion Club, China(CTOCC) database. AGT was performed by deep intubation a mother-and-child catheter(GuidezillaTMextension, 4 or 5 Fr inner catheter, and etc.) in combination with either reverse controlled antegrade or retrograde subintimal tracking(CART) technique or retrograde wire crossing technique. A total of 111 patients with 112 CTO lesions treated with this technique were retrospectively analyzed. Reverse CART technique and retrograde wire crossing technique were performed in 90.2% and 9.8% of all procedures. The utilization of GuidezillaTMextension, 4 Fr, and 5 Fr inner catheter accounted for 94.6%, 3.6%, and 1.8%, respectively. Externalization of retrograde wire was successful in all cases. No procedural complications were adjudicated to AGT. Complications independent of AGT included two target vessel perforations and two collateral perforations. No in-hospital major adverse cardiac events were found. 9BLT1 in dendritic cells promotes Th1/Th17 differentiation and its deficiency ameliorates TNBS-induced colitis显示文摘Leukotriene B4(LTB4)synthesis is enhanced in the colonic mucosa in patients with inflammatory bowel disease(IBD).BLT1,a highaffinity receptor for LTB4,exhibits no effect on the progression of dextran sodium sulfate(DSS)-induced colitis,which mostly relies on innate immunity.Here,we reported that BLT1 regulates trinitrobenzene sulfonic acid(TNBS)-induced colitis,which reflects CD4+T-cell-dependent adaptive immune mechanisms of IBD.We found that BLT1 signaling enhanced the progression of colitis through controlling the production of proinflammatory cytokines by dendritic cells(DCs)and modulating the differentiation of Th1 and Th17.BLT1−/−mice displayed an alleviated severity of TNBS-induced colitis with reduced body weight loss and infiltrating cells in the lamina propria.BLT1 deficiency in DCs led to reduced production of proinflammatory cytokines,including IL-6,TNF-α,and IL-12,and these results were further confirmed via treatment with a BLT1 antagonist.The impaired cytokine production by BLT1−/−DCs subsequently led to reduced Th1 and Th17 differentiation both in vitro and in vivo.We further performed a conditional DC reconstitution experiment to assess whether BLT1 in DCs plays a major role in regulating the pathogenesis of TNBS-induced colitis,and the results indicate that BLT1 deficiency in DCs also significantly reduces disease severity.The mechanistic study demonstrated that BLT1-regulated proinflammatory cytokine production through the Gαiβγsubunit-phospholipase Cβ(PLCβ)-PKC pathway.Notably,we found that treatment with the BLT1 antagonist also reduced the production of proinflammatory cytokines by human peripheral blood DCs.Our findings reveal the critical role of BLT1 in regulating adaptive immunity and TNBS-induced colitis,which further supports BLT1 as a potential drug target for adaptive immunity-mediated IBD.Jinfeng Zhou Weiming Lai Wanjie Yang Juping Pan Hu Shen Yingying Cai Cuixia Yang Ningjia Ma Yue Zhang Ru Zhang Xin Xie Zhongjun Dong Yuan Gao Changsheng Du 2018Cellular & Molecular Immunology2018,15,12:9

11Synthesis of Li_4Ti_5O_(12) nanostructural anode materials with high charge–discharge capability显示文摘This study demonstrates a facile and efficient hydrothermal method to prepare spindle titanate(Li4Ti5O12 denoted as LTO)and/or carbon-LTO nanocomposites(CLTO),in which the LTO or C-LTO microspheres have diameters of a few micrometers,composed of numerous nanosheets with thickness of*30 nm and edge length of hundreds of nanometers.The morphology and size control of these nanoparticles could be achieved by varying experimental parameters including concentration of titanium butoxide,lithium hydroxide,and cetyltrimethylammonium bromide,as well as reaction temperature and time.These micro-nanostructures were characterized by several advanced techniques,such as transmission electron microscopy,scanning electron microscopy,X-ray diffraction,energy dispersive spectroscopic analysis,surface area,and electrochemical measurements.The LTO and C-LTO microstructures were examined in the charge–discharge capacity at a rate of 50 C,as well as the stability after 100 cycles at a rate of 10 C.The excellent capability may be attributed to good conductivity,large surface area,and stable assembly structure of such micro-nanostructures,which could be explored as a promising anode material for lithium-ion batteries.Ru Wen Jeffrey Yue Zifeng Ma Wuming Chen Xuchuan Jiang Aibing Yu 2014Chinese Science Bulletin2014,59,18:8
12LDHs derived nanoparticle-stacked metal nitride as interlayer for long-life lithium sulfur batteries显示文摘Shuttle effect is one of the most serious disadvantages in lithium-sulfur battery which results in poor cycle performance and hinders the commercialization of Li-S battery.To reduce the dissolution of polysulfides into the electrolyte and prolong the cycling stability,nanoparticle-stacked metal nitride derived from layered double hydroxides(LDHs)as an interlayer was inserted between sulfur cathode and separator to confine polysulfides by physical and chemical interactions.Meanwhile,the surface of metal nitride will form an oxide passivation layer.The passivation layer possesses hydrophilic metal-O group and provides a polar surface for strong binding with polysulfide.What’s more,the nanoparticlesstacked structure could immerge and retain electrolyte well,which could enhance the ability of promoting the electron exchange rate.The sulfur electrode with nanoparticle-stacked metal nitride interlayer has an excellent cycle performance owing to the interactions between metal nitride and polysulfides.The battery delivered an initial capacity of 764.6 m Ahg^(-1) and still possesses a capacity of 477.5 mAhg^(-1) with the retention of 62.4% after 800 cycles.Zhen Li Zhaoling Ma Yanyong Wang Ru Chen Zhenjun Wu Shuangyin Wang 2018Science Bulletin2018,63,3:8
13Disulfide-stabilized single-chain antibody-targeted superantigen: Construction of a prokaryotic expression system and its functional analysis显示文摘AIM: To construct the expression vector of B3 (scdsFv)-SEA (D227A) and to identify its binding and cytotoxic ability to B3 antigen positive carcinoma cell lines.METHODS: This fusion protein was produced by a bacterial expression system in this study. It was expressed mainly in the inclusion body. The gene product was solubilized by guanidine hydrochloride, refolded by conventional dilution method, and purified using SP-sepharose cation chromatography.RESULTS: The expression vector B3 (scdsFv)-SEA-PETwas constructed, the expression product existed mainly in the inclusion body, the refolding product retained the binding ability of the single-chain antibody and had cytotoxic effect on HT-29 colon carcinoma cells. 14Distribution and Ecological Risk Assessment of HCHs and DDTs in Surface Seawater and Sediment of the Mariculture Area of Jincheng Bay,China显示文摘The distribution of hexachlorocyclohexanes(HCHs) and dichlorodiphenyltrichloroethanes(DDTs) in the surface seawater and sediment of Jincheng Bay mariculture area were investigated in the present study. The concentration of total HCHs and DDTs ranged from 2.98 to 14.87 ng L-1 and were < 0.032 ng L-1, respectively, in surface seawater, and ranged from 5.52 to 9.43 and from 4.11 to 6.72 ng g-1, respectively, in surface sediment. It was deduced from the composition profile of HCH isomers and DDT congeners that HCH residues derived from a mixture of technical-grade HCH and lindane whereas the DDT residues derived from technical-grade DDT and dicofol. Moreover, both HCH and DDT residues may mainly originate from historical inputs. The hazard quotient of α-HCH, β-HCH, γ-HCH and δ-HCH to marine species was 0.030, 0.157, 3.008 and 0.008, respectively. It was estimated that the overall probability of adverse biological effect from HCHs was less than 5%, indicating that its risk to seawater column species was low. The threshold effect concentration exceeding frequency of γ-HCH, p,p'-DDD, p,p'-DDE and p,p'-DDT in sediment ranged from 8.3% to 100%, and the relative concentration of the HCH and DDT mixture exceeded their probable effect level in sediment. 16Simultaneous quantification of ginsenoside Rg1 and its metabolites by HPLC–MS/MS:Rg1 excretion in rat bile, urine and feces显示文摘Ginsenoside Rg1(Rg1), the major effective component of ginseng, has been shown to have multiple bioactivities, but low oral bioavailability. The aim of this study was to develop a simple, sensitive and rapid high performance liquid chromatography–tandem mass spectrometry(LC–MS/MS) method, which could be used to validate and quantify the concentrations of Rg1 and its metabolites in Sprague-Dawley rat bile,urine, and feces after oral administration(25 mg/kg). Calibration curves offered satisfactory linearity(r40.995)within the determined ranges. Both intra-day and inter-day variances were less than 15%, and the accuracy was within 80–120%. The excretion recoveries of Rg1, ginsenoside Rh1(Rh1), and protopanaxatriol(Ppt) in bile,urine, and feces combined were all greater than 70%. The fecal excretion recoveries of Rg1, Rh1, and Ppt were40.11%, 22.19%, and 22.88%, respectively, whereas 6.88% of Rg1 and 0.09% of Rh1 were excreted in bile.Urinary excretion accounted for only 0.04% of Rg1. 18Generation of electricity from CO_2 mineralization: Principle and realization显示文摘Current CO2 reduction and utilization technologies suffer from high energy consuming. Thus, an energy favourable route is in urgent demanding. CO2 mineralization is theoretically an energy releasing process for CO2 reduction and utilization, but an approach to recovery this energy has so far remained elusive. For the first time, here we proposed the principle of harvesting electrical energy directly from CO2 mineralization, and realized an energy output strategy for CO2 utilization and reduction via a CO2-mineralization fuel cell(CMFC) system. In this system CO2 and industrial alkaline wastes were used as feedstock, and industrial valuable Na HCO3 was produced concomitantly during the electricity generation. The highest power density of this system reached 5.5 W/m2, higher than many microbial fuel cells. The maximum open circuit voltage reached 0.452 V. Moreover, this system was demonstrated viable to low concentration CO2(10%) and other carbonation process.
