1Structure of human NTCP reveals the basis of recognition and sodium-driven transport of bile salts into the liver显示文摘Dear Editor,The sodium taurocholate(TC)co-transporting polypeptide NTCP(SLC10A1)is a secondary active membrane transport protein that mediates the uptake of bile salts from the portal blood plasma into the cytoplasm of liver cells.NTCP was originally cloned from rat and demonstrated to be localized in the basolateral membrane.1 Its activity is a key component of bile acid homeostasis and enterohepatic circulation of bile salts(Fig.1a),a process that includes at least three additional transporter proteins,namely BSEP,ASBT and OSTα/β.2,3 In addition to its role in mediating bile salt uptake,NTCP also serves as the receptor of the human hepatitis B and D viruses.4 The mechanism by which NTCP recognizes and transports bile salts is not understood,in part because no structural insight into the interaction of NTCP with its substrates is available.While recent studies reported structures of NTCP bound to inhibitory nanobodies5 or Fab fragments,6,7 no substrate-binding pockets were identified.Hongtao Liu Rossitza N.Irobalieva Rose Bang-Sørensen Kamil Nosol Somnath Mukherjee Parth Agrawal Bruno Stieger Anthony A.Kossiakoff Kaspar P.Locher 2022Cell Research2022,32,8:2

14Inverse problem for adaptive SIR model:Application to COVID-19 in Latin America显示文摘This work presents a method for solving an Adaptive Susceptible-Infected-Removed(ASIR)epidemic model with time-dependent transmission and removal rates.Available COVID-19 data as of March 2021 are used for identifying the rates from an inverse problem.The estimated rates are used to solve the adaptive SIR system for the spread of the infectious disease.This method simultaneously solves the problem for the time-dependent rates and the unknown functions of the A-SIR system.Presented results show the spread of COVID-19 in the World,Argentina,Brazil,Colombia,Dominican Republic,and Honduras.Comparisons of the reported affected by the disease individuals from the available real data and the values obtained with the A-SIR model demonstrate how well the model simulates the dynamic of the infectious disease.Tchavdar T.Marinov Rossitza S.Marinova 2022Infectious Disease Modelling2022,7,1:1

17What could microRNA expression tell us more about colorectal serrated pathway carcinogenesis?显示文摘In the last two decades,the vision of a unique carcinogenesis model for colorectal carcinoma(CRC)has completely changed.In addition to the adenoma to carcinoma transition,colorectal carcinogenesis can also occur via the serrated pathway.Small non-coding RNA,known as microRNAs(miRNAs),were also shown to be involved in progression towards malignancy.Furthermore,increased expression of certain miRNAs in premalignant sessile serrated lesions(SSLs)was found,emphasizing their role in the serrated pathway progression towards colon cancer.Since miRNAs function as post-transcriptional gene regulators,they have enormous potential to be used as useful biomarkers for CRC and screening in patients with SSLs particularly.In this review,we have summarized the most relevant information about the specific role of miRNAs and their relevant signaling pathways among different serrated lesions and polyps as well as in serrated adenocarcinoma.Additional focus is put on the correlation between gut immunity and miRNA expression in the serrated pathway,which remains unstudied.Milena Peruhova Monika Peshevska-Sekulovska Boris Krastev Gabriela Panayotova Viktoriya Georgieva Rossitza Konakchieva Georgi Nikolaev Tsvetelina Veselinova Velikova 2020World Journal of Gastroenterology2020,26,42:1
