| 2 | Chemical compound cinobufotalin potently induces FOXO1-stimulated cisplatin sensitivity by antagonizing its binding partner MYH9显示文摘In this study,we present novel molecular mechanisms by which FOXO1 functions as a tumor suppressor to prevent the pathogenesis of nasopharyngeal carcinoma(NPC).First,we observed that FOXO1 not only controlled tumor stemness and metastasis,but also sensitized NPC cells to cisplatin(DDP)in vitro and in vivo.Mechanistic studies demonstrated that FOXO1-induced miR-200b expression through the GSK3β/β-catenin/TCF4 network-mediated stimulation of ZEB1,which reduced tumor stemness and the epithelial–mesenchymal transition(EMT)signal.Furthermore,we observed FOXO1 interaction with MYH9 and suppression of MYH9 expression by modulating the PI3K/AKT/c-Myc/P53/miR-133a-3p pathway.Decreased MYH9 expression not only reduced its interactions with GSK3β,but also attenuated TRAF6 expression,which then decreased the ubiquitin-mediated degradation of GSK3βprotein.Increased GSK3βexpression stimulated theβ-catenin/TCF4/ZEB1/miR-200b network,which increased the downstream tumor stemness and EMT signals.Subsequently,we observed that chemically synthesized cinobufotalin(CB)strongly increased FOXO1-induced DDP chemosensitivity by reducing MYH9 expression,and the reduction in MYH9 modulated GSK3β/β-catenin and its downstream tumor stemness and EMT signal in NPC.In clinical samples,the combination of low FOXO1 expression and high MYH9 expression indicated the worst overall survival rates.Our studies demonstrated that CB potently induced FOXO1-mediated DDP sensitivity by antagonizing its binding partner MYH9 to modulate tumor stemness in NPC. | YongHao Li Xiong Liu Xian Lin Menyang Zhao Yanyi Xiao Chen Liu Zixi Liang Zelong Lin Renhui Yi Zibo Tang Jiahao Liu Xin Li Qingping Jiang Libo Li Yinyin Xie Zhen Liu Weiyi Fang | 2019 | Signal Transduction and Targeted Therapy2019,4,1: | 6 |
| 3 | Accuracy of space-for-time substitution for vegetation state prediction following shrub restoration显示文摘Aims:Space-for-time substitution(SFT)is often used for vegetation status estimation during the recovery process of deserts.However,the evaluated accuracy of SFT remains uncertain.An eight-year located observation was used to assess the validity of SFT for vegetation state prediction.Methods:This study analyzed a chronosequence of Caragana microphylla Lam.plantings using the located observation method to test the accuracy of SFT for vegetation state prediction in the mobile sand dunes of the Horqin Sandy Land in northeastern China from July 2005 to June 2013.Important Findings:According to SFT,simple vegetation parameters(density,coverage and biomass)were found to be unstable,while sophisticated veg-etation parameters(species diversity and evenness)were relatively stable across the experimental treatments during the study period.Conversely,both the simple and sophisticated parameters were found to be relatively stable when tested using the located obser-vation method.Furthermore,most simple vegetation parameters slightly increased,while sophisticated parameters slightly decreased after eight years of field observations.Thus,long-term restoration management facilitated improvements in the simple parameters,but may have adversely impacted the sophisticated parameters in the post-restoration community.Our results suggest that sophisticated vegetation parameter states can be predicted by SFT,while simple vegetation parameter states are not well predicted by SFT.in conclusion,located observations or other effective evaluation methods must be employed to offset the deficiency of the SFT method for the prediction of vegetation parameters. | Renhui Miao Xueli Qiu Meixia Guo Ala Musa Deming Jiang | 2018 | Journal of Plant Ecology2018,11,2: | 5 |

PCC 6803 (cyanobacterium) photosystem Ⅱ to pyrene stress显示文摘In order to explore the mechanism of acute toxicity for pyrene to cyanobacterial organisms,the responses of Synechocystis sp.PCC 6803 photosystem II(PS II) under pyrene stress were studied.The results showed there was no significant difference about the oxygen evolution under 0.125 mg/L pyrene stress when compared with control,but it was significantly lower than control at 0.625 mg/L pyrene.Polyphasic chlorophyll-a fluorescence transients in cells of Synechocystis sp.PCC 6803 exhibited a typical increase including O,J,I,and P phases.Fluorescence yield at phases J,I and P declined slightly at 0.125 and 0.625 mg/L pyrene,and significantly lower than control at 3.125 mg/L.According to the parameters deviated from JIP-test,no modification was induced by pyrene both at the donor side and at the acceptor side of PS II,and the reaction centre of PS II is the primary damaging target.Based on the expressing of four key genes(psbA,psbB,psbC and psbO) of PS II,only psbA showed significant difference at 3.125 mg/L pyrene when compared with control. | Jihai Shao Gongliang Yu Zhongxing Wu Xin Peng Renhui Li | 2010 | Journal of Environmental Sciences2010,22,7: | 2 |

| 6 | Effects of warming on soil respiration during the non-growing seasons in a semiarid temperate steppe显示文摘尽管许多研究认为增温对陆地生态系统非生长季土壤呼吸的影响非常重要,但是关于这一问题仍缺乏充分的理解。本论文研究了非生长季增温对温带半干旱草原土壤呼吸的影响,并探讨了其影响机制。本实验位于温带半干旱典型草原,包括10对样地,每对样地包括一个对照和模拟增温处理。模拟增温样地自2014年10月开始,采用开顶箱(OTC)进行。2014年10月到2015年4月,2015年10月到2016年4月期间每月测定几次土壤呼吸、土壤温度与土壤湿度;并测定0-20 cm微生物量碳(MBC)、微生物量氮(MBN)和土壤速效氮。采用重复测量方差和成对t检验方法分析增温、时间及其交互作用对土壤呼吸的影响。采用简单回归确定各指标之间的因果关系。结果表明,对照样地非生长季平均土壤呼吸速率为0.24μmol m^(-2) s^(-1),约占当地生长季碳通量的14.4%。模拟增温在两个非生长季使土壤温度和土壤呼吸分别比对照样地提高了1.48℃(P<0.001)和42.1%(P<0.01)。2015-2016年非生长季模拟增温降低了0.66%的土壤湿度,降低幅度不显著。模拟增温处理下2015-2016年非生长季MBC和MBN分别显著提高了19.72%和20.99%(P<0.05)。此外,研究结果还表明,土壤温度、MBC和MBN的变化调控了土壤呼吸对模拟增温的响应。本结果揭示非生长季土壤呼吸变化能影响碳循环的其它组分,并可以有效促进气候变化背景下陆地生态系统碳循环的模型预测。 | Yuan Miao Mengzhou Liu Juan Xuan Wei Xu Shilin Wang Renhui Miao Dong Wang Wei Wu Yinzhan Liu Shijie Han | 2020 | Journal of Plant Ecology2020,13,3: | 2 |
| 7 | Molecular characterization and toxin quantification of Microcystis panniformis: A microcystin producer in Lake Taihu,China显示文摘Microcystis panniformis is a bloom forming species with flat panniform-like colonies. This species was recently found in Lake Taihu, China. To specifically characterize M. panniformis based on isolated strains, morphological examination on colonial transition and genetic examination are needed. Three M. panniformis strains isolated from a water bloom sample in Lake Taihu were characterized by molecular analysis and toxin quantification. Phylogenetic analysis based on both 16S rRNA gene and internal transcribed spacer(ITS) between 16S and 23S rRNA genes were performed and compared to facilitate easy identification of the species.Relatively high similarities(98%–99%) were shown in 16S rDNA sequences between the strains of M. panniformis and those of other Microcystis species, whereas the similarities for ITS sequences were 88%–95%. In the phylogenetic tree based on the 16S rDNA sequences, the M. panniformis and M. aeruginosa strains were intermixed together with no clear division,whereas all of the M. panniformis strains were clustered together in a single clade based on the ITS sequences based phylogenyetic tree. The mcyE gene was detected in all three strains, and microcystin was determined by high-performance liquid chromatography.
