| 1 | Centrophenoxine improves chronic cerebral ischemia induced cognitive deficit and neuronal degeneration in rats显示文摘AIM: To study the effects of centrophenoxine (CPH, meclofenoxate) on chronic cerebral hypoperfusion induced deficits in rats. METHODS: Chronic hypoperfusion in rats was performed by permanent bilateral ligation of the common carotid arteries. Morris water maze was used to measure spatial memory performance. Spectrophoto- metrical techniques were used to assay SOD, GPx activities, MDA content, TXB2, and 6-keto-PGF1α levels. Mor- phological change was examined by HE staining. The expression of Bax and p53 protein were assayed by immuno- histochemistry analysis. RESULTS: Chronic hypoperfusion in rats resulted in spatial memory impairments shown by longer escape latency and shorter time spent in the target quadrant. These behavioral dysfunction were accom- panied by increase in SOD and GPx activities, the content of MDA, the levels of pro-inflammatory mediators (TXB2, 6-keto-PGF1α), overexpression of Bax and P53 protein, and delayed degeneration of neurons in cortex and hippocampus. Oral administration of CPH (100 mg/kg, once per day for 37 d) markedly improved the memory impairment, reduced the increase in antioxidant enzyme activities, MDA content and the levels of pro-inflammatory mediators to their normal levels, and attenuated neuronal damage. | 2 | Single nucleotide polymorphism in DNA methyltransferase 3B promoter and its association with gastric cardiac adenocarcinoma in North China显示文摘AIM: To investigate the association between single nucleotide polymorphism (SNP)in promoter of the DNA methyltransferase 3B (DNNT3B) gene and risk for development and lymphatic metastasis of gastric cardiac adenocarcinoma (GCA).METHODS: The hospital based case-control study included 212 GCA patients and 294 control subjects without overt cancer. The DNMT3B SNP was genotyped by PCR and restriction fragment length polymorphism (RFLP) analysis. RESULTS: The C/C genotype was not detected in both GCA patients and controls. In control subjects, the frequency of T/T and C/T genotypes was 94.9% and 5.1% respectively,and that of T and C alleles was 97.4% and 2.6%, respectively.The genotype and allelotype distribution in the GCA patients was not significantly different from that in controls (P = 0.34 and 0.33, respectively). When stratified by smoking status and family history of upper gastrointestinal cancer, significant difference in the genotype distribution was not observed between GCA patients and controls. The distribution of DNMT3Bgenotypes in GCA patients with or without lymphatic metastasis did not show significant difference (P = 0.42). CONCLUSION: The distribution of DNMT3BSNP in North China is distinct from that in Caucasians. | 5 | No association of the matrix metalloproteinase 1 promoter polymorphism with susceptibility to esophageal squamous cell carcinoma and gastric cardiac adenocarcinoma in northern China显示文摘AIM: To investigate association of the 2G or 1G single nudeotide polymorphism (SNP)in matrix metalloproteinase 11(MMP1)promoter with susceptibility to esophageal squam-ous cell carcinoma (ESCC) and gastric cardiac adenocarcinoma (GCA) in a population of North China.METHODS: MMP1 promoter SNP was genotyped by polymerase-chain reaction (PCR)-restriction fragment length polymorphism (RFLP) analysis in 417 cancer patients (234ESCC and 183 GCA) and 350 healthy controls.RESULTS: The genotype frequencies of the MMP1promoter SNP in healthy controls were 55.4% (2G/2G),30% (1G/2G) and 14.6% (1G/1G), respectively. The genotype and allelotype distribution in ESCC and GCA patients was not significantly different from that in healthy controls (all Pvalues were above 0.05). Compared with the 1G/1G genotype, neither the 2G/2G nor in combination with the 1G/2G genotype significantly modified the risk of developing ESCC and GCA, the adjusted odds ratio was 1.28 (95%CI = 0.78-2.09), 1.23 (95%CI = 0.38-2.05)in ESCC and 1.39 (95%CI = 0.80-2.41), 1.34 (95%CI =0.74-2.40) in GCA, respectively. When stratified by smoking status and family history of upper gastrointestinal cancer, the 2G/2G genotype alone or in combination with the 1G/2G genotype also did not show any significant influence on the risk of ESCC and GCA development. | 6 | Effects of huperzine A on secretion of nerve growth factor in cultured rat cortical astrocytes and neurite outgrowth in rat PC12 cells显示文摘Aim: To study the effects of huperzine A (HupA) on neuritogenic activity and the expression of nerve growth factor (NGF). Methods: After being treated with 10μmol/L HupA, neurite outgrowth of PC 12 cells was observed and counted under phase-contrast microscopy. Mitogenic activity was assayed by [^3H]thymidine incorporation. Cell cytotoxicity was evaluated by lactate dehydrogenase (LDH) release. AChE activity, mRNA and protein expression were measured by the Ellman's method, RT-PCR, and Western blot, respectively. NGF mRNA and protein levels were determined by RT-PCR and ELISA assays. Results: Treatment of PC 12 cells with 10μmol/L HupA for 48 h markedly increased the number of neuritebearing cells, but caused no significant alteration in cell viability or other signs of cytotoxicity. In addition to inhibiting AChE activity, 10μmol/L HupA also increased the mRNA and protein levels of this enzyme. In addition, following 2h exposure of the astrocytes to 10μmol/L HupA, there was a significant up-regulation of mRNA for NGF and P75 low-affinity NGF receptor. The protein level of NGF was also increased after 24 h treatment with HupA. | 8 | Abnormal Effective Connectivity of the Anterior Forebrain Regions in Disorders of Consciousness显示文摘A number of studies have indicated that disorders of consciousness result from multifocal injuries as well as from the impaired functional and anatomical connectivity between various anterior forebrain regions.However, the specific causal mechanism linking these regions remains unclear. In this study, we used spectral dynamic causal modeling to assess how the effective connections(ECs) between various regions differ between individuals. Next, we used connectome-based predictive modeling to evaluate the performance of the ECs in predicting the clinical scores of DOC patients. We found increased ECs from the striatum to the globus pallidus as well as from the globus pallidus to the posterior cingulate cortex, and decreased ECs from the globus pallidus to the thalamus and from the medial prefrontal cortex to the striatum in DOC patients as compared to healthy controls.Prediction of the patients' outcome was effective using the negative ECs as features. In summary, the present study highlights a key role of the thalamo-basal ganglia-cortical loop in DOCs and supports the anterior forebrain mesocircuit hypothesis. | 9 | Epoxide hydrolase Tyr113 His polymorphism is not associated with susceptibility to esophageal squamous cell carcinoma in population of North China显示文摘AIM: To investigate the possible association of microsomal epoxide hydrolase ( mEH) Tyr113 His polymorphism with susceptibility to esophageal squamous cell carcinoma (ESCC) in a population of North China.METHODS: The mEH-Tyr113 His genotypes were determined by polymerase-chain reaction (PCR)-restriction fragment length polymorphism (RFLP) analysis in 257 patients with esophageal squamous cell carcinoma (ESCC) and 252 healthy subjects as a control group.RESULTS: The frequencies for Tyr and His alleles were 44.2 %, 55.8 % in ESCC patients, and 44.0 % and 56.0 %in healthy subjects, respectively. No statistic difference in allele distribution was observed between ESCC patients and controls (x2=0.008, P=0.929). The overall genotype distribution difference was not observed between cancer cases and controls (x2=2.116, P=-0.347). Compared with Tyr/Tyr genotype, neither His/His genotype nor in combination with Tyr/His genotype significantly modified the risk of the development of ESCC, the adjusted odds ratio was 1.076 (95 % CI=0.850-1.361) and 0.756 (95 % CI=0.493-1.157),respectively. | 17 | Proteomic response of Phaeocystis globosa to nitrogen limitation显示文摘Phaeocystis globosa is an important unicellular eukaryotic alga that can also form colonies.P.globosa can cause massive harmful algal blooms and plays an important role in the global carbon or sulfur cycling.Thus far,the ecophysiology of P.globosa has been investigated by numerous studies.However,the proteomic response of P.globosa to nitrogen depletion remains largely unknown.We compared four protein preparation methods of P.globosa for two-dimensional electrophoresis(2-DE)(Urea/Triton X-100 with trichloroacetic acid(TCA)/acetone precipitation;TCA/acetone precipitation;Radio Immuno Precipitation Assay(RIPA)with TCA/acetone precipitation;and Tris buffer).Results show that the combination of RIPA with TCA/acetone precipitation had a clear gel background and showed the best protein spot separation effect,based on which the proteomic response to nitrogen depletion was studied using 2-DE.In addition,we identified six differentially expressed proteins whose relative abundance increased or decreased more than 1.5-fold(P<0.05).Most proteins could not be identified,which might be attributed to the lack of genomic sequences of P.globosa.Under nitrogen limitation,replication protein-like,RNA ligase,and sn-glycerol-3-phosphate dehydrogenase were reduced,which may decrease the DNA replication level and ATP production in P.globosa cells.The increase of endonucleaseⅢand transcriptional regulator enzyme may affect the metabolic and antioxidant function of P.globosa cells and induce cell apoptosis.These findings provide a basis for further proteomic study of P.globosa and the optimization of protein preparation methods of marine microalgae. | Haisu LIU Ruiwang WEI Qiangyong LEI Lei CUI Songhui LÜ | 2024 | Journal of Oceanology and Limnology2024,42,1: | 0 |
| 18 | Foldable high-strength electrode enabled by nanosheet subunits for advanced sodium-ion batteries显示文摘Power sources with strong mechanical properties and high-energy density are highly desirable for the next-generation flexible electronics.However,the challenge arises from the current electrode structure design,which is unable to bring both satisfactory mechanical and electrochemical properties with high active materials content and mass.Herein,we reported novel flexible,highstrength,and mechanically stable TiO_(2)-based film electrodes for advanced sodium-ion batteries,achieving an ultrahigh strength(up to≈60 MPa)and commercial-level areal capacity(4.5 mAh cm^(-2)).Highly-dispersed TiO_(2) and interlaced carbon nanotube(CNT)networks are embedded in the sheet-liked cellulose to form porous,high-conductive,and high-active TiO_(2)-C nanosheets that is basic building subunits of TiO_(2)-C films,allowing the films with structural robustness and origami-level flexibility.This strategy reconciles the contradiction between mechanical properties and active material content in flexible electrodes,and the fabricated electrode with a high TiO_(2) content of>65%can be bent more than 11000 times without breaking.Meanwhile,good capacity and excellent cycle stability(0.02‰capacity-decay rate over 9000 cycles)of TiO_(2)-C film under a higher active content(75%)has well satisfied the demands of flexible energy storage devices for electrochemical performances.This TiO_(2)-C subunit assembly methodology demonstrates enormous potential in high-strength/toughness flexible electrode construction for flexible electronics. | Hanwei Wang Jinzhou Fu Chao Wang Ruiwang Zhang Yingying Li Yushan Yang Haobo Li Qingfeng Sun Huiqiao Li | 2022 | InfoMat2022,4,2: | 0 |
