10Effect of Pregnancy on TMJ Nociception in Rats显示文摘Objective:Does TMJ nociception,as measured by a behavioral assay,differ during pregnancy,potentially due to the effect of sex steroids?Methods:Thirty-two Sprague Dawley rats were randomly allocated to either mid-(E11)or late-(E17)pregnancy groups.The animals within each group were further allocated to a saline or CFA treatment.At E11 or E17,the animals were injected bilaterally into TMJ with either saline(15μL)or 1μg/μL CFA(15μL).Nociception was measured with a feeding assay(pellet recording)and analyzed by comparing intra-meal rates using a distance-based permutation method.Results:CFA injection resulted in the animals eating longer in both the mid-pregnant and late-pregnant groups.Plasma estradiol was higher in the late-pregnant group versus the mid-pregnant group.Importantly,the CFA injected late-pregnant group ate for a shorter amount of time(i.e.,shorter meal rate)than the CFA injected rats at mid-pregnancy.Conclusion:The reduced nociceptive response(shorter meal rate)in the CFA injected late-pregnant group may be due to greater estradiol versus the CFA injected mid-pregnancy rats.Thus,one potential reason why women report fewer TMJ symptoms during pregnancy is the higher level of circulating estradiol.Mikhail Umorin Larry L.Bellinger Phillip R.Kramer 2018口腔医学研究2018,34,3:0
11Is activation of GDNF/RET signaling the answer for successful treatment of Parkinson's disease?A discussion of data from the culture dish to the clinic显示文摘The neurotrophic signaling of glial cell line-derived neurotrophic factor(GDNF)with its canonical receptor,the receptor tyrosine kinase RET,coupled together with the GDNF family receptor alpha 1 is important for dopaminergic neuron survival and physiology in cell culture experiments and animal models.This prompted the idea to try GDNF/RET signaling as a therapeutic approach to treat Parkinson’s disease with the hallmark of dopaminergic cell death in the substantia nigra of the midbrain.Despite several clinical trials with GDNF in Parkinson’s disease patients,which mainly focused on optimizing the GDNF delivery technique,benefits were only seen in a few patients.In general,the endpoints did not show significant improvements.This suggests that it will be helpful to learn more about the basic biology of this fascinating but complicated GDNF/RET signaling system in the dopaminergic midbrain and about recent developments in the field to facilitate its use in the clinic.Here we will refer to the latest publications and point out important open questions in the field.James A.Conway Edgar R.Kramer 2022Neural Regeneration Research2022,17,7:0
