| 2 | Analysis of Genetic Diversity and Differentiation of Seven Stocks of Litopenaeus vannamei Using Microsatellite Markers显示文摘Seven microsatellite markers were used to evaluate the genetic diversity and differentiation of seven stocks of Litopenaeus vannamei, which were introduced from Central and South America to China. All seven microsatellite loci were polymorphic, with polymorphism information content(PIC) values ranging from 0.593 to 0.952. Totally 92 alleles were identified, and the number of alleles(Na) and effective alleles(Ne) varied between 4 and 21 and 2.7 and 14.6, respectively. Observed heterozygosity(Ho) values were lower than the expected heterozygosity(He) values(0.526–0.754), which indicated that the seven stocks possessed a rich genetic diversity. Thirty-seven tests were detected for reasonable significant deviation from Hardy-Weinberg equilibrium. Fis values were positive at five loci, suggesting that there was a relatively high degree of inbreeding within stocks. Pairwise Fst values ranged from 0.0225 to 0.151, and most of the stock pairs were moderately differentiated. Genetic distance and cluster analysis using UPGMA revealed a close genetic relationship of L. vannamei between Pop2 and Pop3. AMOVA indicated that the genetic variation among stocks(11.3%) was much lower than that within stocks(88.7%). | 3 | Identification of differential genes in the ovary relative to the testis and their expression patterns in half-smooth tongue sole (Cynoglossus semilaevis)显示文摘Half-smooth tongue sole (Cynoglossus semilaevis) is a rare marine flatfish whose mature ovary and testis greatly differ in volume and weight. The length and weight of mature females are over twice greater than those of mature males. To obtain sufficient information on gonad differentiation and the relationship between gonad development and growth in the fish, we compared gene expression between the ovary and testis using suppression subtractive hybridization (SSH). Testis cDNAs are subtracted from ovary cDNAs and are used to es- tablish an ovary testis-subtracted cDNA library. A total of 41 nonredundant clones are identified, including 20 known cDNAs, 9 unchar-acterized cDNAs (EST clones), and 12 novel sequences. For selected genes such as ZPC, RacGAP, survivin, aquaporin, CPEB, O5, O15, and O18, gene expression patterns are analyzed by reverse transcriptase-polymerase chain reaction (RT-PCR). The results confirm that these genes are only expressed in the ovary and not in the testis, or at higher levels in the ovary than in the testis. At the same time, expressions of certain genes such as ZPC, survivin, aquaporin, CPEB, and O15 are demonstrated to possess sexual dimorphism in the kidney or muscle, and spleen. | 14 | Statistical properties of kinetic-scale magnetic holes in terrestrial space显示文摘Kinetic-scale magnetic holes(KSMHs)are structures characterized by a significant magnetic depression with a length scale on the order of the proton gyroradius.These structures have been investigated in recent studies in near-Earth space,and found to be closely related to energy conversion and particle acceleration,wave-particle interactions,magnetic reconnection,and turbulence at the kineticscale.However,there are still several major issues of the KSMHs that need further study—including(a)the source of these structures(locally generated in near-Earth space,or carried by the solar wind),(b)the environmental conditions leading to their generation,and(c)their spatio-temporal characteristics.In this study,KSMHs in near-Earth space are investigated statistically using data from the Magnetospheric Multiscale mission.Approximately 200,000 events were observed from September 2015 to March 2020.Occurrence rates of such structures in the solar wind,magnetosheath,and magnetotail were obtained.We find that KSMHs occur in the magnetosheath at rates far above their occurrence in the solar wind.This indicates that most of the structures are generated locally in the magnetosheath,rather than advected with the solar wind.Moreover,KSMHs occur in the downstream region of the quasi-parallel shock at rates significantly higher than in the downstream region of the quasi-perpendicular shock,indicating a relationship with the turbulent plasma environment.Close to the magnetopause,we find that the depths of KSMHs decrease as their temporal-scale increases.We also find that the spatial-scales of the KSMHs near the subsolar magnetosheath are smaller than those in the flanks.Furthermore,their global distribution shows a significant dawn-dusk asymmetry(duskside dominating)in the magnetotail. | ShuTao Yao ZongShun Yue QuanQi Shi Alexander William Degeling HuiShan Fu AnMin Tian Hui Zhang Andrew Vu RuiLong Guo ZhongHua Yao Ji Liu Qiu-Gang Zong XuZhi Zhou JingHuan Li WenYa Li HongQiao Hu YangYang Liu WeiJie Sun | 2021 | Earth and Planetary Physics2021,5,1: | 1 |
| 15 | Tumor necrosis factor-alpha(TNF-α) in spotted halibut Verasper variegatus at the embryonic and metamorphic stages显示文摘As an aquatic fish,the spotted halibut Verasper variegatus is highly susceptible to bacterial and virus infections.Tumor necrosis factor-alpha(TNF-α)as a cytokine could control the inflammatory responses.The functions of TNF-αin many species have been widely studied,particularly in mammals.However,little is known about the TNF-αfunctions in V.variegatus.We first cloned and sequenced the TNF-αgene in V.variegatus(VvTNF-α).The two conserved cysteine residues,transmembrane sequence,Thr-Leu motif,and TNF family signature,as well as the TA-rich motifs of its proteins related to inflammatory responses had high similarity to those of the other teleost and mammalian TNF-α.The phylogenetic analysis showed that VvTNF-αwas consistent with TNF-αgenes of other vertebrates.The VvTNF-αtranscripts were extensively distributed in the peripheral blood leukocytes(PBLs),spleen,and gill,indicating that the VvTNF-αhad a role in immune function.Furthermore,treatment with pathogen-associated molecular patterns(PAMPs)could induce a rapid and significant increase of VvTNF-αin the PBLs,which reveals that VvTNF-αdoes participate in the host immune responses against bacterial and viral pathogens.We found that VvTNF-αhad an interesting expression pattern during metamorphosis,showing that the flatfish TNF-αmay have some novel functions during specific developmental stages.In addition,the 3 D structure prediction of VvTNF-αprovided an indication of how it is likely to interact with other proteins.Therefore,VvTNF-αhas multiple functions,and provides valuable information to explore novel functions of TNF-α. | LI Zan LIU Xiumei DU Xinxin ZHANG Kai CHEN Yan WANG Xubo WANG Zhigang YU Haiyang ZHANG Quanqi | 2020 | Journal of Oceanology and Limnology2020,38,2: | 1 |
| 16 | Molecular Cloning, Expression Pattern, and 3D Structural Prediction of the Cold Inducible RNA- Binding Protein(CIRP) in Japanese Flounder(Paralichthys olivaceus)显示文摘Cold-inducible RNA-binding protein(CIRP) is a kind of RNA binding proteins that plays important roles in many physiological processes. The CIRP has been widely studied in mammals and amphibians since it was first cloned from mammals. On the contrary, there are little reports in teleosts. In this study, the Po CIRP gene of the Japanese flounder was cloned and sequenced. The genomic sequence consists of seven exons and six introns. The putative Po CIRP protein of flounder was 198 amino acid residues long containing the RNA recognition motif(RRM). Phylogenetic analysis showed that the flounder Po CIRP is highly conserved with other teleost CIRPs. The 5' flanking sequence was cloned by genome walking and many transcription factor binding sites were identified. There is a Cp Gs region located in promoter and exon I region and the methylation state is low. Quantitative real-time PCR analysis uncovered that Po CIRP gene was widely expressed in adult tissues with the highest expression level in the ovary. The m RNA of the Po CIRP was maternally deposited and the expression level of the gene was regulated up during the gastrula and neurula stages. In order to gain the information how the protein interacts with m RNA, we performed the modeling of the 3D structure of the flounder Po CIRP. The results showed a cleft existing the surface of the molecular. | 19 | Intelligent method to experimentally identify the fracture mechanism of red sandstone显示文摘Tensile and shear fractures are significant mechanisms for rock failure.Understanding the fractures that occur in rock can reveal rock failure mechanisms.Scanning electron microscopy(SEM)has been widely used to analyze tensile and shear fractures of rock on a mesoscopic scale.To quantify tensile and shear fractures,this study proposed an innovative method composed of SEM images and deep learning techniques to identify tensile and shear fractures in red sandstone.First,direct tensile and preset angle shear tests were performed for red sandstone to produce representative tensile and shear fracture surfaces,which were then observed by SEM.Second,these obtained SEM images were applied to develop deep learning models(AlexNet,VGG13,and SqueezeNet).Model evaluation showed that VGG13 was the best model,with a testing accuracy of 0.985.Third,the features of tensile and shear fractures of red sandstone learned by VGG13 were analyzed by the integrated gradient algorithm.VGG13 was then implemented to identify the distribution and proportion of tensile and shear fractures on the failure surfaces of rock fragments caused by uniaxial compression and Brazilian splitting tests.Results demonstrated the model feasibility and suggested that the proposed method can reveal rock failure mechanisms. | Zida Liu Diyuan Li Quanqi Zhu Chenxi Zhang Jinyin Ma Junjie Zhao | 2023 | International Journal of Minerals,Metallurgy and Materials2023,30,11: | 0 |
| 20 | Identification of two Major Histocompatibility(MH) Class Ⅱ A Genes and Their Association to Vibrio anguillarum Infection in Half-smooth Tongue Sole(Cynoglossus semilaevis)显示文摘Major histocompatibility complex class II antigens are important in vertebrate immune system.In the present study,the full cDNA sequence of class II A gene was synthesized by RACE-PCR from half-smooth tongue sole(Cynoglossus semilaevis),and its open reading frame(ORF) polymorphism was studied.The whole cDNA sequence was 992 bp in length,including the ORF with 717 bp.Twenty-five alleles were identified and clustered into two distinct groups according to the specific nucleotides/amino acids in specific positions.Eleven alleles belonged to Cyse-DAA while fourteen alleles belonged to Cyse-DBA.Four Cyse-DAA alleles were observed in one individual,and three to five Cyse-DBA alleles were observed in each of the three detected individuals,which indicated that at least two loci existed in each gene.Moreover,in order to study the function of the alleles in resistance to infection,200 individuals were intraperitoneally injected with Vibrio anguillarum and the first 20 dead individuals and 20 surviving ones were selected for genotype analysis.Fifty-six alleles were identified among the 40 individuals.Twenty-nine alleles belonged to Cyse-DAA and the other 27 alleles belonged to Cyse-DBA.Eighteen alleles were selected for studying their function in resistance to infection.Alleles Cyse-DAA*0201,Cyse-DAA*1101,Cyse-DBA*0401,Cyse-DBA*1102,Cyse-DBA*1801 and Cyse-DBA*2201 were identi-fied only in surviving individuals,while alleles Cyse-DAA*0901,Cyse-DBA*1101 and Cyse-DBA*1401 occurred more frequently in dead individuals.This study confirmed the existence and polymorphism of two class II A genes as well as the relationship between alleles of class II A genes and disease susceptibility/resistance in half-smooth tongue sole. | LI Chunmei WANG Xubo ZHANG Quanqi WANG Zhigang QI Jie YI Qilin LIU Zhipeng WANG Yanan YU Haiyang | 2012 | Journal of Ocean University of China2012,11,1: | 0 |
