2Chromosomal localization of the major ribosomal RNA genes in scallop Chlamys farreri显示文摘The chromosomes of Chlamys farreri were analyzed by means of silver staining and fluorescence in situ hybridization (FISH) with 18S—28S rDNA probe. Probe was made by PCR amplification of a DNA fragment containing internal transcribed spacers ITS1 between 18S and 5.8S ribosomal RNA gene, ITS2 between 5.8S and 28S ribosomal RNA gene and 5.8S rRNA gene, and labeled by PCR incorporation of bio-16-dUTP. FISH signals were located on the short arm of subtelocentric chromosome 10. After silver-staining, nucleolus organizer regions (NORs) could be observed on the telomere of the short arm of chromosome 10. 3High-speed flowing plasmas in the Earth’s plasma sheet显示文摘A research topic of great interest to the space physics community is the observation of plasmas flowing at hundreds of kilometers per second in the Earth’s plasma sheet. Although considerable effort has been made to understand the source of fast-flowing plasmas, many questions remain unanswered about the mechanisms that produce high-speed flows and the effects they have on magnetospheric disturbances, especially their contributions to magnetospheric convection and substorms. In this paper, we discuss briefly the history of high-speed flows and review the proposed mechanisms, signatures of high-speed flows in auroras and their interaction with the background plasma. 4Assessing genetic diversity of wild populations of Japanese flounderusing AFLP markers显示文摘Amplified fragment length polymorphism (AFLP) analysis was used to evaluate the genetic diversity of four wild geographical populations of Japanese flounder (Paralichthys olivaceus). A total of 775 loci (58.32% of which was polymorphic) in the range between 100 and 1 300 base pairs were detected from 110 individuals using seven primer combinations. The percentage of polymorphic loci detected by single primer combination for each population was calculated, ranging from 19.59% to 53.33%. Genetic similarities within and among the populations were calculated from the binary matrices of presence-absence. Phylogenetic tree of four populations was constructed by using the UPGMA method using PHYLIP Version 3.5. According to intrapopulation genetic similarities, CW population displayed the highest genetic diversity value and KY population had the lowest genetic diversity value. The distance between CW and CF populations was the farthest, which was possibly resulted from the farthest distance of Weihai of Shandong and Fujian of China compared with the geographical distance between other locations of populations. The subpopulation differentiation value (G_ st ) is 0.356 5, showing a certain extent of differentiation among the four geographical populations. AFLP technology was confirmed to be an effective tool to assess within-and among-population genetic diversity of Japanese flounder. 5Enhanced anti-sunward flow near local noon during a period of horizontal IMF and high solar wind velocity V_Y显示文摘It is believed that a southward interplanetary magnetic field (IMF) is mainly responsible for the energy input from solar wind into the magnetosphere.This paper presents an unusual case of strong anti-sunward plasma flow (up to 2 km/s) in the polar cap ionosphere and large cross-polar cap potential (CPCP) during a period of horizontal IMF (|BZ| < 2 nT) observed by both ACE (at the L1 point) and Geotail (on the dusk flank of the magnetosheath).The CPCP is even higher than that under preceding BZ ≈-23 nT.Furthermore,GOES8 observed that the magnetosheath field turns northward as the anti-sunward plasma flow and CPCP start to increase,which implies that the magnetosheath field interacting with the Earth's magnetopause has significantly rotated and differs from the IMF observed by ACE and Geotail.In accordance with previous theoretical work,we suggest that the magnetic field line draping produces a southward magnetosheath field and enhances anti-sunward plasma flow and the CPCP.WEI Yong ZONG QiuGang PU ZuYin WAN WeiXing LIU JianJun FU SuiYan SHI QuanQi 2011Chinese Science Bulletin2011,56,11:7
6Analysis of Genetic Diversity and Differentiation of Seven Stocks of Litopenaeus vannamei Using Microsatellite Markers显示文摘Seven microsatellite markers were used to evaluate the genetic diversity and differentiation of seven stocks of Litopenaeus vannamei, which were introduced from Central and South America to China. All seven microsatellite loci were polymorphic, with polymorphism information content(PIC) values ranging from 0.593 to 0.952. Totally 92 alleles were identified, and the number of alleles(Na) and effective alleles(Ne) varied between 4 and 21 and 2.7 and 14.6, respectively. Observed heterozygosity(Ho) values were lower than the expected heterozygosity(He) values(0.526–0.754), which indicated that the seven stocks possessed a rich genetic diversity. Thirty-seven tests were detected for reasonable significant deviation from Hardy-Weinberg equilibrium. Fis values were positive at five loci, suggesting that there was a relatively high degree of inbreeding within stocks. Pairwise Fst values ranged from 0.0225 to 0.151, and most of the stock pairs were moderately differentiated. Genetic distance and cluster analysis using UPGMA revealed a close genetic relationship of L. vannamei between Pop2 and Pop3. AMOVA indicated that the genetic variation among stocks(11.3%) was much lower than that within stocks(88.7%). 7Analysis of magnetotail flux rope events by ARTEMIS observations显示文摘Three earthward flowing magnetic flux ropes observed in the duskside plasma sheet at geocentric solar magnetospheric coordinate X~–55 Re by P1 and P2 of acceleration,reconnection,turbulence and electrodynamics of moon’s interaction with the sun mission during 13:00–15:00 UT on July 3,2012,were studied.The morphologies of the flux ropes were studied in detail based on Grad-Shfranov reconstruction method and electronic pitch angle distribution data.It is found that(1)the flux rope cross-sectional dimensions are 1.0 Re×0.78 Re,1.3 Re×0.78 Re,and 2.5 Re×1.25 Re,respectively.The magnetic field lines were asymmetric about the center with field line compression on both sides of the current sheet at the leading region;(2)the electron energy flux data presented asymmetry with larger electron flux and lower temperature in the precursor region.The flux ropes were blocked by the resistance of compressed particle density in the front central plasma sheet and the enhanced magnetic field on its sides;and(3)it is found that the flux rope has a layered structure.From inside out,event 1 can be divided into three regions,namely electronic depletion core region,closed field line region,and the caudal area possible with fields connected with the ionosphere.It suggests that the flux ropes cannot merge with the tail magnetic field lines near the lunar orbit.Especially,the flux rope asymmetrical shape reflects the different reconnection processes that caused it on both sides of the magnetic structure.The events shown in this paper support the multiple X-line magnetic reconnection model for flux ropes with in situ observations.TIAN AnMin SHI QuanQi ZONG QiuGang DU Jian FU SuiYan DAI YaNan 2014Science China(Technological Sciences)2014,57,5:5
8Discrete energetic(~50-200 keV) electron events in the high-altitude cusp/polar cap/lobe显示文摘We identified 28 discrete electron events(DEEs) with enhanced fluxes at ~50-200 keV in the high-altitude cusp/polar cap/lobe,using the electron measurements by the BeiDa Image Electron Spectrometer(BD-IES) instrument onboard an inclined(55°)geosynchronous orbit(IGSO) satellite from October 2015 to January 2016. We find that among the 28 DEEs, 22 occur in the nightside and mostly in the northern cusp/polar cap/lobe, while 6 occur in the dayside and all in the southern cusp; 24 events correspond to an average interplanetary magnetic field(IMF) component B_z>0, 3 correspond to an average IMF B_z<0, and 1 has no OMNI IMF data. In these DEEs, the observed average omnidirectional electron differential flux generally fits well to a power-law spectrum, J^E^(-γ), with the spectral index y ranging from 2.6 to 4.6, while the average electron flux varies over three orders of magnitude from event to event. The spectral index of these cusp DEEs are(strongly) larger than the spectral index of solar wind superhalo electrons(radiation belt electrons) observed by the WIND 3 D Plasma & Energetic Particle instrument(the BD-IES). At^110 keV,the electron flux of DEEs in the cusp/polar cap/lobe shows a weak positive correlation with the solar wind superhalo electron flux but no obvious correlation with the radiation belt electron flux. 9Reconstruction of morningside plasma sheet compressional ULF Pc5 wave显示文摘We studied the compressional wave event in Pc5 frequency range observed in the dawn-side magnetic equator on 9 March 1998 by Grad-Shafranov(GS) reconstruction method for the first time. To test the effectiveness of application of GS method on Pc5 compressional wave, we benchmarked our procedure by applying it to a one-dimensional current sheet model first. Excluding the left-hand corners, the average error magnitude was less than 10%. The reconstruction of actual data showed that we obtained the 2-D map of compressional wave without suffering model constraints for the first time. The magnetic filed lines density cyclical changed, and the wavelength was about 2-4 times earth radius. The reconstructed magnetic topology had a shape very similar to the empirical 2-dimensional standing wave model proposed by the former workers. 10THEMIS observation of a magnetotail current sheet flapping wave显示文摘A flapping wave was observed by THEMIS-B(P1)and THEMIS-C(P2)probes on the dawn side of the magnetotail,while the solar wind was generally stable.The magnetic activity was quite weak,suggesting that this flapping wave was generated by an internal instability,which normally occurs during magnetic quiet times.Our analysis shows that the flapping wave was propagating downward with a tail-aligned scale of at least 3.7 R E and did not show much change in shape during its propagation from P1 to P2.Correlation analysis employed to estimate the time lag between the corresponding half waveforms of P1 and P2 shows that the propagating velocities along the current sheet normal directions were close to each other in the beginning,but increased linearly later on.The average wavelength of the flapping wave is approximately 4 R E.Theoretical analysis suggests that the ballooning type wave model may not be the mechanism for the observed flapping wave,but that the magnetic double-gradient instability model is a more plausible candidate.Weijie Sun Suiyan Fu Quanqi Shi Qiugang Zong Zhonghua Yao Ting Xiao George Parks 2014Chinese Science Bulletin2014,59,2:3
11Braking of high-speed flows in the magnetotail:THEMIS joint observations显示文摘The motion and deceleration processes of plasma sheet high-speed flows have great significance to magnetospheric particle acceleration,magnetic field perturbation,magnetic flux transport,triggering of substorm,and the current system formation in the magnetotail.From February to April 2009,two satellites of the Time History of Events and Macroscale Interactions during Substorms mission,THA and THE,were often separated largely in Z direction,but had small X and Y separations.Such special configuration allows simultaneous observations of highspeed flows at the center and boundary of the plasma sheet.Based on selected case study and statistical analysis,it is found that for about 89%of the events we selected,the probe further away from the neutral sheet observed the high-speed flow earlier than the one close to the center,and the flow is mainly field aligned.And for about 95%events the probe further away from the neutral sheet observed higher X component of the plasma flow.With the hypothesis that parallel flow keeps the same speed during its earthward propagation while central plasma sheet stream uniformly or suddenly brakes on its way to the earth,we deduced the position where the deceleration begins to be between 13 Re and 17 Re downtail,where thenear-earth reconnection is supposed to occur.In addition,our statistical results show that dipolarization fronts observed in the central plasma sheet are more prominent than those observed in the plasma sheet boundary layer ahead of the high-speed flow.Wensai Shang Zhonghua Yao Quanqi Shi Weijie Sun Suiyan Fu Jiang Liu Anmin Tian Qiugang Zong Zuyin Pu Ting Xiao Vassilis Angelopoulos 2014Chinese Science Bulletin2014,59,3:3
12Identification of differential genes in the ovary relative to the testis and their expression patterns in half-smooth tongue sole (Cynoglossus semilaevis)显示文摘Half-smooth tongue sole (Cynoglossus semilaevis) is a rare marine flatfish whose mature ovary and testis greatly differ in volume and weight. The length and weight of mature females are over twice greater than those of mature males. To obtain sufficient information on gonad differentiation and the relationship between gonad development and growth in the fish, we compared gene expression between the ovary and testis using suppression subtractive hybridization (SSH). Testis cDNAs are subtracted from ovary cDNAs and are used to es- tablish an ovary testis-subtracted cDNA library. A total of 41 nonredundant clones are identified, including 20 known cDNAs, 9 unchar-acterized cDNAs (EST clones), and 12 novel sequences. For selected genes such as ZPC, RacGAP, survivin, aquaporin, CPEB, O5, O15, and O18, gene expression patterns are analyzed by reverse transcriptase-polymerase chain reaction (RT-PCR). The results confirm that these genes are only expressed in the ovary and not in the testis, or at higher levels in the ovary than in the testis. At the same time, expressions of certain genes such as ZPC, survivin, aquaporin, CPEB, and O15 are demonstrated to possess sexual dimorphism in the kidney or muscle, and spleen. 13Shape and position of Earth's bow shock near-lunar orbit based on ARTEMIS data显示文摘Earth's bow shock is the result of interaction between the supersonic solar wind and Earth's magnetopause. However, data limitations mean the model of the shape and position of the bow shock are based largely on near-Earth satellite data. The model of the bow shock in the distant magnetotail and other factors that affect the bow shock, such as the interplanetary magnetic field(IMF) B_y, remain unclear. Here, based on the bow shock crossings of ARTEMIS from January 2011 to January 2015, new coefficients of the tail-flaring angle a of the Chao model(one of the most accurate models currently available) were obtained by fitting data from the middle-distance magnetotail(near-lunar orbit, geocentric distance -20R_E>X>-50R_E). In addition, the effects of the IMF B_y on the flaring angle a were analyzed. Our results showed that:(1) the new fitting coefficients of the Chao model in the middle-distance magnetotail are more consistent with the observed results;(2) the tail-flaring angle a of the bow shock increases as the absolute value of the IMF B_y increases. Moreover, positive IMF B_y has a greater effect than negative IMF B_y on flaring angle. 14Weighted Correlation Network Analysis(WGCNA) of Japanese Flounder(Paralichthys olivaceus) Embryo Transcriptome Provides Crucial Gene Sets for Understanding Haploid Syndrome and Rescue by Diploidization显示文摘Artificial gynogenesis is of great research value in fish genetics and breeding technology. However, existing studies did not explain the mechanism of some interesting phenomena. Severe developmental defects in gynogenetic haploids can lead to death during hatching. After diploidization of chromosomes, gynogenetic diploids may dispense from the remarkable malformation and restore the viability, although the development time is longer and the survival rate is lower compared with normal diploids. The aim of this study was to reveal key mechanism in haploid syndrome of Japanese flounder, a commercially important marine teleost in East Asia. We measured genome-scale gene expression of flounder haploid, gynogenetic diploid and normal diploid embryos using RNA-Seq, constructed a module-centric co-expression network based on weighted correlation network analysis(WGCNA) and analyzed the biological functions of correlated modules. Module gene content analysis revealed that the formation of gynogenetic haploids was closely related to the abnormality of plasma proteins, and the up-regulation of p53 signaling pathway might rescue gynogenetic embryos from haploid syndrome via regulating cell cycle arrest, apoptosis and DNA repair. Moreover, normal diploid has more robust nervous system. 15Sex Determination Mechanisms in Fish显示文摘In fish,sex determination(SD) system shows high variation. The SD mechanisms include environmental and genetic regulation. The research on SD system and related genes in intensively studied fish species was reviewed. Although some genes have been described as sex-related,only DMRT1bY can be considered as a master sex determination gene and none of them has been utilized in aquaculture. The variation of fish SD system,the importance of sex-related genes in evolution research and the relations between environmental factors and sex-related genes were also discussed. The fish sex determination mechanism remains largely unknown. 16Analysis of New Microsatellite Markers Developed From Reported Sequences of Japanese Flounder Paralichthys olivaceus显示文摘The expressed sequence tags (ESTs) of Japanese flounder, Paralichthys olivaceus, were selected from GenBank to iden-tify simple sequence repeats (SSRs) or microsatellites.A bioinformatic analysis of 11111 ESTs identified 751 SSR-containing ESTs, including 440 dinucleotide, 254 trinucleotide, 53 tetranucleotide, 95 pentanucleotide and 40 hexanucleotide microsatellites respec-tively.The CA/TG and GA/TC repeats were the most abundant microsatellites.AT-rich types were predominant among trinucleotide and tetranucleotide microsatellites.PCR primers were designed to amplify 10 identified microsatellites loci.The PCR results from eight pairs of primers showed polymorphisms in wild populations.In 30 wild individuals, the mean observed and expected het-erozygosities of these 8 polymorphic SSRs were 0.71 and 0.83 respectively and the average PIC value was 0.8.These microsatellite markers should prove to be a useful addition to the microsatellite markers that are now available for this species.YU Haiyang JIANG Liming CHEN Wei WANG Xubo WANG Zhigang ZHANG Quanqi 2010Journal of Ocean University of China2010,9,4:2
17Tissue Expression and Stock Variation of Isozymes of Stone Flounder(Kareius bicoloratus)显示文摘Tissue expression and stock variation of isozymes of stone flounder (Kareius bicoloratus) were analyzed with horizontal starch gel electrophoresis. For the fourteen enzymes assayed, 31 loci were recorded. The results indicated that all the isozymes ex- amined were obviously tissue-specific. The expressions of SOD*, GDH*, G3PDH-2* and ADH-2* were detected only in liver, SDH-1*, MDH-1* and ADH-1* only in muscle, and LDH-B* and LDH-C* only in eyes. In comparison, MDH-2*, GPI-3* and SDH-2* were detected in all tissues examined. Other loci examined were detected in a variety of tissues. Muscle and liver were se- lected to detect the isozyme variation of the two geographic stocks of Qingdao and Weihai, Shandong Province, China. The percent- ages of polymorphic loci (P0.99) were 29.17% and 25.00%, the observed heterozygosities (H0) were 0.028 ± 0.014 and 0.040 ± 0.019, and the expected heterozygosities (He) were 0.039 ± 0.017 and 0.052 ± 0.022 in Qingdao and Weihai stock, respectively. The coeffi- cient of gene differentiation (Fst) and genetic distance (D) between the two stocks was 0.012 and 0.0011, respectively, indicating that the genetic differentiation is low between them. 18Comparative Studies on the Molecular Genetic Diversities among Haliotis discus hannai, H. discus discus and Their Hybrids显示文摘The hybrid (H. discus hannai♀× H. discus discus♂) shows strong heterosis in both growth and survival rates during aquaculture. In order to better understand the genetic basis of heterosis, AFLP markers are adopted to compare the genetic diversities of the two parents and their hybrids. Six primer combinations reveal 552 loci, among which 88 loci show significant difference between the two parent populations (P<0.01). Analysis of molecular variance (AMOVA) indicates the genetic variance between them is significantly different (P<0.001). Thus there should be genetic basis of heterosis for their hybrids. In contrast to parents, more loci with lower frequency are amplified in hybrids than those in parents, whereas the loci with 0% and 100% frequency are less in hybrids than those in parents. Moreover, the genetic diversities of hybrids increase since the similarity indexes are lower and heterozygosities are higher in hybrids than those in parents. 19A Microsatellite Genetic Linkage Map of Black Rockfish(Sebastes schlegeli)显示文摘Ovoviviparous black rockfish(Sebastes schlegeli) is an important marine fish species for aquaculture and fisheries in China. Genetic information of this species is scarce because of the lack of microsatellite markers. In this study, a large number of microsatellite markers of black rockfish were isolated by constructing microsatellite-enriched libraries. Female- and male-specific genetic linkage maps were constructed using 435 microsatellite markers genotyped in a full-sib family of the fish species. The female linkage map contained 140 microsatellite markers, in which 23 linkage groups had a total genetic length of 1334.1 c M and average inter-marker space of 13.3 c M. The male linkage map contained 156 microsatellite markers, in which 25 linkage groups had a total genetic length of 1359.6 c M and average inter-marker distance of 12.4 c M. The genome coverage of the female and male linkage maps was 68.6% and 69.3%, respectively. The female-to-male ratio of the recombination rate was approximately 1.07:1 in adjacent microsatellite markers. This paper presents the first genetic linkage map of microsatellites in black rockfish.
