| 2 | Factors Associated with Thrombolysis Outcome in Ischemic Stroke Patients with Atrial Fibrillation显示文摘The outcome of early intravenous thrombolysis for ischemic stroke in patients with atrial fibrillation(AF)is worse than that without thrombosis. How to increase the efficacy of intravenous thrombolysis for AF-related ischemic stroke remains largely unknown. In this study, we investigated factors that influence the effect of intravenous thrombolysis in these patients. Our results showed that thrombolysis was independently associated with a favorable outcome(P \ 0.001) and did not influence the mortality of AF-related ischemic stroke, although it increased the risk of hemorrhage within 24 h after treatment. Risk factors for a poor outcome at admission were:heart failure(P = 0.045); high systolic pressure(P = 0.039); high blood glucose(P = 0.030); and a high National Institutes of Health Stroke Scale(NIHSS) score(P \ 0.001). Moreover, high systolic pressure at admission(P = 0.007), high blood glucose(P = 0.027), and a high NIHSS score(P \ 0.001) were independent risk factors for mortality at 3 months. Besides thrombolysis, a high NIHSS score(P = 0.006) and warfarin taken within 48 h before stroke onset(P = 0.032) were also independent risk factors for symptomatic hemorrhage within 24 h after treatment. | 3 | Efficacy of decoction from Jieduan Niwan formula on rat model of acute-on-chronic liver failure induced by porcine serum显示文摘OBJECTIVE:To dynamically observe the efficacy of Jieduan Niwan formula(JDNW)on a rat model of acute-on-chronic liver failure(ACLF).METHODS:Seventy Wistar rats were divided into control group(6 rats),model group(22 rats),JDNW group(21 rats),and SP600125 group(21 rats).13 weeks'porcine serum injection followed with D-galactosamine and lipopolysaccharide joint acute attack was used to establish ACLF model.Rats in JDNW group were orally given JDNW formula for 3 days before acute attack;rats in SP600125 group were injected with SP60012530 min ahead of acute attack.Rats were sacrificed respectively at 4,8 and 12 h after model established.Alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBIL),Creatinine(CR),blood urea nitrogen(BUN),prothrombin activity(PTA)were examined by biochemical process,Tumor necrosis factor-alpha(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6),interleukin-10(IL-10),transformed growth factor-beta 1(TGF-β1),High mobility group box-1(HMGB-1),CD3,CD4,CD8 were analyzed by enzyme-linked immunosorbent assay,apoptotic index(AI)was detected by terminal-deoxynucleoitidyl transferase mediated nick end labeling staining,expression of Bad,phosphorylated Jun N-terminal kinases(p-JNK)and Cytochrome C(Cyt C)were detected by immunohistochemical analysis,Bax and Bid were detected by Western blot analysis.RESULTS:In model group,the levels of ALT,AST,TBIL,CR,BUN,IL-1β,IL-6,IL-10,TGF-β1 and HMGB-1 remarkably increased and PTA decreased compared with control group(P<0.05),as time goes on,ALT,AST,TBIL,CR,BUN,continued to grow,while IL-1β,IL-6,IL-10,HMGB-1,TGF-β1 and PTA gradually decreased;massive necrosis could be seen;the levels of TNF-α,CD3,CD4,CD8,AI,p-JNK,Bax,Bad,Bid and Cyt C increased at 4 h and peaked at 8 h,but decreased at 12 h(P<0.05).JDNW group,by contrast,showed less pathological injury,increased PTA level,and reduced ALT,AST,TBIL,TNF-α,IL-1β,IL-6,IL-10,TGF-β1,HMGB-1,CD3,CD4 and CD8 levels(P<0.05),moreover,the AI and expression of p-JNK,Bax,Bad,Bid and Cyt C were lower than model group at 4 and 8 h but were higher at 12 h(P<0.05).Similar results were observed in SP600125 group.CONCLUSION:An ACLF rat model with low mortality can be established by porcine serum joint with D-galactosamine+lipopolysaccharide induction;JDNW decoction can effectively suppress the inflammatory reaction,improve the immune system,and protect the liver of ACLF rats,the mechanism might involve the inhibition of the JNK-induced mitochondrial apoptotic pathway. | Li Jinxia Zhang Qiuyun Gao Lianyin Du Yuqiong Chen Yu | 2020 | Journal of Traditional Chinese Medicine2020,40,4: | 3 |
| 4 | Phenotypic characterization of patients with activated PI3Kδ syndrome 1 presenting with features of systemic lupus erythematosus显示文摘Activated phosphoinositide 3-kinase d syndrome 1(APDS1)is a primary immunode-ficiency disease caused by gain-of-function mutations in PIK3CD.Clinical features of autoimmune disease have been reported in patients with APDS1.In this study,we reported three patients with APDS1 presenting with systemic lupus erythematosus(SLE)phenotype.The clinical manifestations included recurrent respiratory tract infection,lymphoproliferation,Coombs-positive hemolytic anemia,decreased complement fractions,positive antinuclear antibodies,renal complications related to SLE associated diseases,which met the clinical spectrum of APDS1 and the classification criteria of SLE.The immunological phenotype included an inversion in the CD4:CD8 ratio,an increase in both non-circulating Tfh CD4^(+)memory T and circulating Tfh populations,a low level of recent thymic emigrant T cells,overexpression of CD57 on T cells,and a decrease in B cells with fewer antibody class switch recombination.These phenotypes detected in patients with APDS1 presenting with SLE were resemble that in patients with APDS1 presenting without SLE.Meanwhile,we described the effect of glucocorticoids and rapamycin therapy on patients with APDS1.The phosphorylation of S6 at Ser235/236 was inhibited in patients with APDS1 who underwent glucocorticoids therapy,including two who presented with SLE phenotype.The phosphorylation of AKT at Ser473 and phosphorylation of S6 at Ser235/236 were inhibited in other patients with APDS1 who underwent rapamycin therapy.Here,we showed the coexistence of immunodeficiency and SLE phenotype in APDS1,and the inhibition of rapamycin in activated Akt-mTOR signaling pathway. | Yanping Wang Qiuyun Yang Xuemei Chen Wenjing Tang Lina Zhou Zhi Chen Yunfei An Zhiyong Zhang Xuemei Tang Xiaodong Zhao | 2021 | Genes & Diseases2021,8,6: | 3 |

| 9 | A novel manganese complex LMnAc selectively kills cancer cells by induction of ROS-triggered and mitochondrial-mediated cell death显示文摘We previously identified a novel synthesized metal compound,LMnAc([L2Mn2(Ac)(H2O)2](Ac)(L=bis(2-pyridylmethyl)amino-2-propionic acid)).This compound exhibited significant inhibition on cancer cell proliferation and was more selective against cancer cells than was the popular chemotherapeutic reagent cisplatin.In this study,we further investigated the underlying molecular mechanisms of LMnAc-induced cancer cell death.We found that LMnAc achieved its selectivity against cancer cells through the transferrin-transferrin receptor system,which is highly expressed in tumor cells.LMnAc triggered cancer cells to commit autophagy and apoptosis,which was mediated by the mitochondrial pathway.Moreover,LMnAc disrupted mitochondrial function,resulting in mitochondrial membrane potential collapse and ATP reduction.In addition,LMnAc induced intracellular Ca2+overload and reactive oxygen species generation.Interestingly,its anticancer effect was significantly reduced following pretreatment with the antioxidant N-acetyl cysteine,indicating that reactive oxygen species triggered cell death.Altogether,our data suggest that LMnAc appears to be a selectively promising anticancer drug candidate. | LI Xiang ZHAO KaiDi GUO WenJie LIU Xu LIU Jia GAO Jing CHEN QiuYun BAI YiDong | 2014 | Science China(Life Sciences)2014,57,10: | 1 |

Although various techniques were successfully employed for development of hybrid proteininorganic nanoparticles, cancer cell activated protein-inorganic nanoparticles have been challengeable.Herein, a cancer cell responsive nanoparticle(PDAMn-CuS@BSA-FA, NPs) was constructed and characterized. It is found that the quenching interaction of dyes(PDA) and central metal in NPs can be adjusted by CH_3O-PEG-phosphatide or the cancer cells, hence, NPs showed turn-on fluorescence emission with the titration of CH_3O-PEG-phosphatide. In particular, breast cancer cells lysis can switch on the green emission greatly, while normal cells show less effect. Breast cancer cells turn-on fluorescence imaging and mitochondria targeting imaging demonstrate that the NPs can sense breast cancer cells and enter mitochondria. | 16 | Reveal the mechanisms of prescriptions for liver cancer'treatment based on two illustrious senior TCM physicians显示文摘OBJECTIVE:To explore the core herbs of two illustrious senior Traditional Chinese Medicine(TCM)Physicians for the treatment of liver cancer,and to further clarify the gene targets and pathway mechanisms of liver cancer that the core prescription(CP)may regulate.METHODS:We used the patient information of two illustrious senior TCM physicians from the Affiliated Hospital of Shandong University of Traditional Chinese Medicine and the Affiliated Hospital of Capital Medical University as the database.The CP was analyzed using the community network algorithm.The pathway mechanism was analyzed using network pharmacology method.And the prognostic survival genes were identified using Single factor cox regression analysis.Integrative Pharmacology-based Research Platform of Traditional Chinese Medicine(TCMIP),Herb,Online Mendelian Inheritance in Man(OMIM),Genecards,Kyoto Encyclopedia of Genes and Genomes(KEGG),Genomic Data Commons(GDC),The Cancer Genome Atlas(TCGA),Gephi and R were used to mine CP,building a pathway network diagram.All the analyses were visualized.RESULTS:We found that the CP consistes of Huangqi(Radix Astragali Mongolici),Danshen(Radix Salviae Miltiorrhizae),Shuihonghuazi(Polygoni Orientalis Fructus),Baihuasheshecao(Herba Hedyotdis),Banzhilian(Herba Scutellariae Barbatae),and Ezhu(Rhizoma Curcumae Phaeocaulis),which were attributed to the two physicians respectively.The CP played an anticancer role through such pathways as signal transduction,energy metabolism,immune system and other pathways,covering a total of 112 pathways and 176 herb-diseaserelated genes.Fourteen genes have a significant impact on the prognosis and survival of liver cancer.CONCLUSION:Based on the liver cancer cases of two illustrious senior TCM physicians,we obtained the CP through data mining.The CP may mainly exert anticancer effects by inhibiting inflammatory response,angiogenesis,and enhancing the body’s immune response.We screened out 14 genes in the CP that may be related to the prognosis of liver cancer,and these genes may play an important regulatory role in the prognosis of liver cancer. | WANG Bochuan ZHANG Yong ZHANG Qiuyun ZHANG Zhiqiang LUO Changyong WANG Zhendong BAI Chen WANG Yuhan GE Xueyi QIAN Ying YU He GU Xiaohong | 2023 | Journal of Traditional Chinese Medicine2023,43,1: | 0 |

| 19 | Thioredoxin domain-containing protein 9 protects cells against UV-B-provoked apoptosis via NF-κB/p65 activation in cutaneous squamous cell carcinoma显示文摘Cutaneous squamous cell carcinoma(cSCC),a type of non-melanoma skin cancer(NMSC),is the most common malignancy worldwide.Thioredoxin(TXN)domain-containing protein 9(TXNDC9)is a member of the TXN family that is important in cell differentiation.However,the biological function of this protein in cancer,particularly cSCC,is still unknown.In the present study,our experiments revealed the protective effects of TXNDC9 on UV-B-irritated cSCC cells.The initial findings showed that TXNDC9 is significantly upregulated in cSCC tissue and cells compared to normal skin tissue and keratinocytes.UV-B radiation robustly induces the expression of TXNDC9,and UV-B-induced cSCC cell death is boosted by TXNDC9 deficiency.Moreover,cSCC cells lacking TXNDC9 displayed attenuated activation of the NF-κB pathway.Additional studies by inhibiting TXNDC9 confirmed this finding,as TXNDC9 deficiency attenuated UV-B radiation-induced translocation of NF-κB p65 from the cytoplasm to the nucleus of cSCC.In conclusion,our work demonstrates the biological roles of TXNDC9 in cSCC progression and may provide a novel therapeutic target to treat cSCC in the future. | ZHIXUN XIAO QIUYUN XU HAIQING WANG XIAOTONG ZHOU YANTING ZHU CHENGBEI BAO LIHONG CHEN PENG ZHANG MIN LIN CHAO JI TING GONG | 2023 | Oncology Research2023,31,1: | 0 |
| 20 | The Relationship between Mental Disorders and Personality of Outpatients in a Psychiatric Clinic in Nanjing, China显示文摘Psychosis has increasingly become a social problem,emphasizing the need to understand the relationship between mental disorders and personality.This study aimed to investigate the relationship between mental disorders and personality among psychiatric outpatients based on real-world data.Symptom Checklist 90(SCL-90)and Eysenck Personality Questionnaire(EPQ)were used to evaluate the personality and psychopathological symptoms of patients(n=8409)in the Psychiatric Outpatient Department at Nanjing Drum Tower Hospital.t-test was used to compare scores between patients and national norms.Pearson’s correlation coefficient and path analysis were used to explore the relationship between mental health status and personality.The correlation coefficient between the neuroticism(N)score and each factor score of the SCL-90 test,as well as the correlation between psychoticism(P)and hostility and paranoia,exceeded 0.4.Path analysis revealed that the standardized path coefficients of N score and SCL-90 were all higher than 0.4.In addition,the standardized path coefficient of hostility and paranoia on P score were 0.313 and 0.280,respectively.Interpersonal sensitivity,depression and obsessive-compulsive symptoms were affected by extraversion(E)score,with standardized path coefficients of-0.149,-0.138,and-0.105,respectively.The path analysis also showed the direct and indirect effects of age,gender,education,and marital status on SCL-90.Patients characterized as melancholic had higher scores in all factors of SCL-90.In conclusion,mental health was related to personality traits of neuroticism,psychoticism and introversion. | Yiteng Zang Biyun Xu Sizhen Chen Grace Mutale Qiuyun Cao Bingwei Chen | 2023 | International Journal of Mental Health Promotion2023,25,12: | 0 |
