| 1 | The development of a new bioartificial liver and its application in 12 acute liver failure patients显示文摘AIM: Bioartificial liver is a hope of supporting liver functions in acute liver failure patients. Using polysulfon fibers, a new bioartificial liver was developed. The aim of this study was to show whether this bioartificial liver could support liver functions or not.METHODS: Hepatocytes were procured from swine using Seglen′s methods. The bioartificial liver was constructed by polysulfon bioreactor and more than 10 10 hepatocytes. It was applied 14 times in 12 patients, who were divided into 7 cases of simultaneous HBAL and 5 cases of nonsimultaneous HBAL. Each BAL treatment lasted 6 hours. The general condition of the patients and the biochemical indexes were studied.RESULTS: After treatment with bioartificial liver, blood ammonia, prothrombin time and total bilirubin showed significant decrease. 2 days later, blood ammonia still showed improvment. within one month period, 1 case (1/7) in simultaneous group died while in non-simultaneous group 2 cases (2/5) died. The difference was significant. Mortality rate was 25 %.CONCLUSION: The constructed bioartificial liver can support liver functions in acute liver failure. | 2 | Molecular Characterization of Viral G Gene in Emerging and Re-emerging Areas of Rabies in China, 2007 to 2011显示文摘In recent years (2007 to 2011), although the overall number of rabies cases in China has decreased, there is evidence of emerging or re-emerging cases in regions without previous rabies cases or with low incidence of rabies. To investigate the origin and the factors affecting the spread of rabies in China, specimens were collected from 2007 to 2011 from provinces with emerging and re-emerging cases and tested for the presence of the rabies virus. Positive specimens were combined with sequences from GenBank to perform comparisons of homology and functional sites, and to carry out phylogenetic analyses. Out of these regions, five provinces had 9 positive specimens from canine and cattle, and 34 canine or human specimens were obtained from previously high-incidence provinces. Complete sequences of G gene were obtained for these samples. Homology of the sequences of these 43 specimens was 87%-100% at the nucleotide level and 93.7% -100% at the amino acid level. These G gene sequences were combined with reference sequence from GenBank and used to construct a phylogenetic tree. The results showed that 43 specimens were all assigned to China clade I and clade II, with all specimens from emerging and re-emerging areas placed within clade I. Specimens isolated from Shanxi and Inner Mongolia in 2011 were distinct from previously-isolated local strains and had closer homology to strains from Hebei, Beijing and Tianjin whereas new isolates from Shanghai were tightly clustered with strains isolated in the 1990s. Finally, Shaanxi isolates were clustered with strains from adjacent Sichuan. | 3 | Factors predicting sensory and motor recovery after the repair of upper limb peripheral nerve injuries显示文摘OBJECTIVE: To investigate the factors associated with sensory and motor recovery after the repair of upper limb peripheral nerve injuries. DATA SOURCES: The online PubMed database was searched for English articles describing outcomes after the repair of median, ulnar, radial, and digital nerve injuries in humans with a publication date between 1 January 1990 and 16 February 2011. STUDY SELECTION: The following types of article were selected:(1) clinical trials describing the repair of median, ulnar, radial, and digital nerve injuries published in English; and(2) studies that reported sufficient patient information, including age, mechanism of injury, nerve injured, injury location, defect length, repair time, repair method, and repair materials. SPSS 13.0 software was used to perform univariate and multivariate logistic regression analyses and to investigate the patient and intervention factors associated with outcomes. MAIN OUTCOME MEASURES: Sensory function was assessed using the Mackinnon-Dellon scale and motor function was assessed using the manual muscle test. Satisfactory motor recovery was defined as grade M4 or M5, and satisfactory sensory recovery was defined as grade S3+ or S4.RESULTS: Seventy-one articles were included in this study. Univariate and multivariate logistic regression analyses showed that repair time, repair materials, and nerve injured were independent predictors of outcome after the repair of nerve injuries(P < 0.05), and that the nerve injured was the main factor affecting the rate of good to excellent recovery. | 4 | Multi-drug Resistance and Characteristic of Integrons in Shigella spp.Isolated from China显示文摘Objective To investigate the characteristic of integrons and the relationship between integrons and antimicrobial resistance in Shigella spp. Methods Ninety Shigella strains (83 S. flexneri and 7 S. sonnei) were isolated from the stools of patients in China. Susceptibility to 8 antimicrobials was tested for all isolated strains. PCR, RFLP and sequencing analysis of integrons were applied to all of them. Results High prevalence of multi‐drug resistance (95.6%) was identified. Of the isolates 79 (87.8%) carried integrase genes of class 1 integron (3.3%), class 2 integron (10.0%) or both (74.4%). No intI3 was detected in the tested isolates. The prevalence of intI2 was significantly higher in isolates with multi‐drug resistance to at least 3 antibiotics than that in isolates with resistance to 2 and less antibiotics (P<0.05). Gene cassettes dfrA17‐aadA5, dfrA12‐orfF‐aadA2 of class 1 integron and dfrA1‐sat1‐aadA1 of class 2 integron were identified. | 5 | The Full-length Genome Analysis of a Street Rabies Virus Strain Isolated in Yunnan Province of China显示文摘The epidemic of rabies has rapidly increased and expanded in Yunnan province in recent years. In order to further analyze and understand the etiological reasons for the rapid expansion of rabies in Yunnan, a strain of rabies virus CYN1009D in Yunnan was isolated, and the complete genomic sequencing was carried out, and the bioimfomative analysis on genes/encoded proteins and phylogeny with reference to sequences in GenBank was performed. The complete genome of CYN1009D was 11923 nt in length and belonged to genotype I. The genes encoding different structural proteins were all conserved in their lengths, in comparison to other strains in China. The amino acid sequence was conserved at different antigen sites of NP, but the variation was detected at the secondary phosphorylation site of position 375; variations were also detected in the phosphorylation sites at positions 63-63 and 162 of PP; the sites playing important roles in virus synthesis, budding and viral morphology in MP were conserved; two glycosylation sites were detected at Asn37 and Asn319 in GP, the neutralizing antigen sites in GP were conserved; the initial amino acid of LP (ML) was different from that of most of the strains in China (MM); the variations in G-L region in the intergenic region were significant. | 7 | Growth Characteristics and Reinforcing Behavior of In-situ NbCp in Laser Cladded Fe-based Composite Coating显示文摘Over the past decade,researchers demonstrated much interest in laser cladded metal matrix composite coatings for its good wear resistance.In this paper,in-situ NbCp reinforced Fe-based wear-resistance coatings with different designed NbC contents were produced by laser cladding.The formation mechanism of NbC particle was analyzed.The effects of NbC content on the microstructure and mechanical properties of coatings were investigated.It was revealed that α-(Fe,Cr),(Fe,Cr)_7C_3,NbC existed in all coatings.NbC particles formed by the chemical reaction of Nb and C dissolved in the molten pool.The increase of designed NbC content led to the growth of the porosity amount,and the increase of the size and the area ratio of the particle,as well as the transformation of the NbCp morphology from quadrangle to petaloid shape.Moreover,the micro-hardness and the wear resistance of the coating were improved with the increase of NbC.When NbCp content was 30 wt%,the mechanical properties decreased slightly. | Qingtang Li Yongping Lei Hanguang Fu | 2015 | Journal of Materials Science & Technology2015,31,7: | 4 |
| 8 | A simple model of radial nerve injury in the rhesus monkey to evaluate peripheral nerve repair显示文摘Current research on bone marrow stem cell transplantation and autologous or xenogenic nerve transplantation for peripheral nerve regeneration has mainly focused on the repair of peripheral nerve defects in rodents.In this study,we established a standardized experimental model of radial nerve defects in primates and evaluated the effect of repair on peripheral nerve injury.We repaired 2.5-cm lesions in the radial nerve of rhesus monkeys by transplantation of autografts,acellular allografts,or acellular allografts seeded with autologous bone marrow stem cells.Five months after surgery,regenerated nerve tissue was assessed for function,electrophysiology,and histomorphometry.Postoperative functional recovery was evaluated by the wrist-extension test.Compared with the simple autografts,the acellular allografts and allografts seeded with bone marrow stem cells facilitated remarkable recovery of the wrist-extension functions in the rhesus monkeys.This functional improvement was coupled with radial nerve distal axon growth,a higher percentage of neuron survival,increased nerve fiber density and diameter,increased myelin sheath thickness,and increased nerve conduction velocities and peak amplitudes of compound motor action potentials.Furthermore,the quality of nerve regeneration in the bone marrow stem cells-laden allografts group was comparable to that achieved with autografts.The wrist-extension test is a simple behavioral method for objective quantification of peripheral nerve regeneration. | Dong Wang Xijun Huang Guo Fu Liqiang Gu Xiaolin Liu Honggang Wang Jun Hu Jianhua Yi Xiaofeng Niu Qingtang Zhu | 2014 | Neural Regeneration Research2014,9,10: | 3 |
