10Cytotoxicity of Quartz and Montomrillointe in Human Lung Epthlial Cell显示文摘In this study A549 cell viability, extracellular activities of lactate dehydrogenase (LDH), tumor necrosis factor (TNF)–α and interleukin (IL)-6 levels were investgated after incubation with quartz (KWC-Q4 and KWC-Q3), Nano-SiO2, and KWC-M, the micronucleaus test and comet assay was carried out to evaluate the genotoxicity. The results showed that, there were significant difference in the cell death rate and extracellular LDH activity compared with the control group, and appeared a good linear relationship in certain concentration range. All mineral particle tested can induce the increase of TNF-α after incubation with mineral powders at 200 μg/mL for 3h merely, and significant increase of IL-6 for 24h, the results indicated the inflammatory reaction can be triggered by the exposure of KWC-Q4, KWC-Q3, Nano-SiO2, and KWC-M. The micronucleus test result showed the MNF (Micronucleus frequency) listed as Nano-SiO2>KWC-Q3>KWC-Q4. There is no significant increase of KWC-M compared with the control group, maybe resulted from its high cell death rate at low concentration. 11Gestational dexamethasone exposure impacts hippocampal excitatory synaptic transmission and learning and memory function with transgenerational effects显示文摘The formation of learning and memory is regulated by synaptic plasticity in hippocampal neurons.Here we explored how gestational exposure to dexamethasone,a synthetic glucocorticoid commonly used in clinical practice,has lasting effects on offspring's learning and memory.Adult offspring rats of prenatal dexamethasone exposure(PDE)displayed significant impairments in novelty recognition and spatial learning memory,with some phenotypes maintained transgenerationally.PDE impaired synaptic transmission of hippocampal excitatory neurons in offspring of F1 to F3 generations,and abnormalities of neurotransmitters and receptors would impair synaptic plasticity and lead to impaired learning and memory,but these changes failed to carry over to offspring of F5 and F7 generations.Mechanistically,altered hippocampal miR-133a-3p-SIRT1-CDK5-NR2B signaling axis in PDE multigeneration caused inhibition of excitatory synaptic transmission,which might be related to oocyte-specific high expression and transmission of miR-133a-3p.Together,PDE affects hippocampal excitatory synaptic transmission,with lasting consequences across generations,and CDK5 in offspring's peripheral blood might be used as an early-warning marker for fetal-originated learning and memory impairment.Mingcui Luo Yiwen Yi Songqiang Huang Shiyun Dai Lulu Xie Kexin Liu Shuai Zhang Tao Jiang Tingting Wang Baozhen Yao Hui Wang Dan Xu 2023Acta Pharmaceutica Sinica B2023,13,9:0
12Chinese herbal compound Jinsiwei(金思维)improves synaptic plasticity in mice with sporadic Alzheimer’s disease induced by streptozotocin显示文摘OBJECTIVE:To investigate the efficacy of Jinsiwei(a patented Chinese herbal compound)on learning and memory impairment,the number of synapses and synaptic plasticity-related structural and functional protein expression in mice with sporadic Alzheimer’s disease(SAD)induced by streptozotocin.METHODS:Seventy-five C57/BL6J male mice were intracerebroventricularly injected with streptozotocin to establish the animal model of SAD.Mice were randomly divided into the model group(MG),donepezil group(DG),and the Jinsiwei high,medium,and low-dose groups(JH,JM,JL).Another fifteen C57/BL6J male mice were injected with artificial cerebrospinal fluid as the control group(CG).The intervention groups were intragastrically administrated with corresponding medicine,while the CG and MG were given 0.5%carboxymethyl cellulose by gavage.After 3 months,the Morris Water Maze test and step-down passive avoidance test were used to assess the learning and memory ability of mice.Synapses in hippocampal CA1 were observed by transmission electron microscope.Immunohistochemistry and western blotting were used to assess the distribution and expression levels of synaptic plasticity-related structural and functional proteins involving drebrin,cofilin,synapsin(syn),and N-methyl D-aspartate receptor subtype 2B(NR2B).RESULTS:The Morris Water Maze results showed that the escape latency in the Jinsiwei intervention groups was significantly shorter than that of the MG.Results of the step-down passive-avoidance test showed that the error times in the Jinsiwei intervention groups were significantly reduced compared with the MG.Transmission electron microscope results showed that the number of synapses in hippocampal CA1 was obviously increased in the Jinsiwei intervention groups compared with the MG.Immunohistochemical and western blotting results revealed that the positive cells and expression levels of drebrin,syn,and NR2B were significantly decreased in the MG and meanwhile cofilin significantly increased,while these changes were reversed after the Jinsiwei treatment.CONCLUSIONS:Jinsiwei can alleviate learning and memory impairments in a mouse model of SAD,increase the number of synapses and enhance synaptic plasticity via rescuing the expression of drebrin,syn,and NR2B and inhibiting cofilin expression.TIAN Meijing HE Yannan ZHENG Mingcui QIN Gaofeng GONG Zhuoyan HUANG Shuaiyang WANG Pengwen 2023Journal of Traditional Chinese Medicine2023,43,1:0

14Raf激酶抑制蛋白增强宫颈癌细胞的化疗敏感性(英文)显示文摘Objective:The purpose of the study is to investigate the effects of up-regulation of Raf kinase inhibitor protein(RKlP) on the chemosensitivity of cervical cancer Hela cells.Methods:Eukaryotic expression plasmid pcDNA3.1(+)-ssRKIP containing human overall length RKIPcDNA was transfected into cervical cancer Hela cell by lipofectin assay,establishing a stable cell line containing a target gene by G418.Expression of RKIP in Hela cells was measured by Western blot analysis.After treatment with cisplatin of different concentrations and intervals of time,the effect of RKIP on the proliferation of Hela cells was evaluated by MTT method.The flow cytometry was used to investigate whether the RKIP could inhibit apoptosis in Hela cells induced by cisplatin.Results:The expression of RKIP in Hela cells transfected with pcDNA3.1-ssRKIP was increased obviously.After different concentrations of cisplatin treatment cells for 24,48 and 72 h,the growth inhibition rate in Hela cells transfected with pcDNA3.1-ssRKIP was significantly higher than in control cells(P < 0.05).With 5 μg/mL cisplatin treatment for 24 h,pcDNA3.1-ssRKIP-transfected Hela cells had an obviously higher percentage of apoptosis(23.2 ± 0.24)% than non-transfected cells(12.4 ± 0.31)% and empty vector-transfected cells(13.4 ± 0.47)%.Without treatment of cisplatin,the percentage of apoptosis for Hela cells transfected with pcDNA3.1-ssRKIP was(5.7 ± 0.12)%,which was still higher than those of the non-transfected cells(2.9 ± 0.21)% and empty vector-transfected cells(3 ± 0.08)%.Conclusion:Higher expression of RKIP gene can improve chemosensitivitv of cervical cancer Hela cells to cisplatin.Xiao Chu Xinqiang Ji Mingcui Wang Wenqing Zhang Hui Ou Chong Li 2014The Chinese-German Journal of Clinical Oncology2014,13,3:0

17Quantum coherence transfer between an optical cavity and mechanical resonators显示文摘This study highlights the theoretical investigation of quantum coherence in mechanical oscillators and its transfer between the cavity and mechanical modes of an optomechanical system comprising an optical cavity and two mechanical oscillators that,in this study,were simultaneously coupled to the optical cavity at different optomechanical coupling strengths.The quantum coherence transfer between the optical and mechanical modes is found to depend strongly on the relative magnitude of the two optomechanical couplings.The laser power,decay rates of the cavity and mechanical oscillators,environmental temperature,and frequency of the mechanical oscillator are observed to significantly influence the investigated quantum coherences.Moreover,quantum coherence generation in the optomechanical system is restricted by the system's stability condition,which helps sustain high and stable quantum coherence in the optomechanical system.GuoYao Li WenJie Nie XiYun Li MingCui Li AiXi Chen YueHeng Lan 2019Science China(Physics,Mechanics & Astronomy)2019,62,10:0
