1Effect of chronic corticosterone-induced depression on circadian rhythms and age-related phenotypes in mice显示文摘Disrupted circadian rhythms are a recognized effect of depression,and our previous article demonstrated an association between depression and premature aging,but the underlying mechanisms are not well understood.In the present study,we used a mouse model of chronic corticosterone (CORT)-treated depression to elucidate a mechanism by which depression may be associated with the circadian clock and mediate age-related phenotypes.Mice received a daily injection of 20 mg/kg CORT for 21 consecutive days,and the depression-like behaviors of mice were identified by the sucrose intake test,tail suspension test and open field test.Our findings indicated that CORT injection may be correlatedwith the circadian clock by impairing circadian rhythms or shifting the phase values of clock genes.We also showed that CORT-treated mice exhibited a significant gradual reduction in body weight gain with increased oxidative stress, including reduced activity of antioxidant-related enzymes,reduced glutathione:glutathione disulfide ratio and cytochrome (Cyt)-C level,and elevated reactive oxygen species content.Moreover, chronic CORT injection affected inflammatory responses,the production of mitochondrial ATP and telomere shortening,which may be associated with the Sirtuin 3 (SIRT3)signaling pathway. 2Simultaneous enhancement in electrical conductivity and Seebeck coefficient by single-to double-valley transition in a Dirac-like band显示文摘SnTe possesses a single-to double-valley transition in the conduction band minimum when a compressive strain is applied.Through a tight-binding analysis,it is shown that the variation of the band structure is attributed to the strain-induced delocalization of both the Sn-5s orbitals and Te-5p orbitals with different angular momenta.This effect can largely increase the electron density of states near the band edge and thus keep the Fermi level of the compressed SnTe closer to it,where the electrons have lower scattering rates.The strain-induced double valleys lead to simultaneous increases in the electrical conductivity and the Seebeck coefficient and thereby nearly four times the enhancement of the power factor at the doping concentration of 5×10^(19) cm^(–3).This work suggests a feasible concept that can be employed to promote the power factor of a Dirac semiconductor via manipulating the valley degeneracy in the conduction band minimum.Yu Dai Wenjiang Zhou Hyun-Jung Kim Qichen Song Xin Qian Te-Huan Liu Ronggui Yang 2022npj Computational Materials2022,,1:1
3Cross-Modal Interaction and Integration Through Stimulus-Specific Adaptation in the Thalamic Reticular Nucleus of Rats显示文摘Stimulus-specific adaptation(SSA),defined as a decrease in responses to a common stimulus that only partially generalizes to other rare stimuli,is a widespread phenomenon in the brain that is believed to be related to novelty detection.Although cross-modal sensory processing is also a widespread phenomenon,the interaction between the two phenomena is not well understood.In this study,the thalamic reticular nucleus(TRN),which is regarded as a hub of the attentional system that contains multi-modal neurons,was investigated.The results showed that SSA existed in an interactive oddball stimulation,which mimics stimulation changes from one modality to another.In the bimodal integration,SSA to bimodal stimulation was stronger than to visual stimulation alone but similar to auditory stimulation alone,which indicated a limited integrative effect.Collectively,the present results provide evidence for independent cross-modal processing in bimodal TRN neurons.Yumei Gong Yuying Zhai Xinyu Du Peirun Song Haoxuan Xu Qichen Zhang Xiongjie Yu 2022Neuroscience Bulletin2022,38,7:0
4Novel gold nanoparticles targeting somatostatin receptor subtype two with near-infrared light for neuroendocrine tumour therapy显示文摘Neuroendocrine tumours(NETs)are rare cancers with positive somatostatin receptor 2(SSTR2)expression,and treatment strategies for NETs are not satisfactory.Nanomaterial-mediated therapy targeting SSTR2 in NETs is very promising.This study firstly combined mesoporous silica-coated gold nanorods(AuNRs@mSiO_(2))and targeting-SSTR2 dodecane tetraacetic acidtyrosine3-octreotate(DOTA-TATE)into AuNRs@mSiO_(2)@DOTA-TATE to investigate NETs inhibition under near-infrared light.AuNRs@mSiO_(2)@DOTA-TATE showed good photothermal conversion efficiency.In vitro,under light irradiation,the cell viability significantly decreased with increasing AuNR@mSiO_(2)@DOTA-TATE concentration;in two successfully established neuroendocrine tumour organoids with SSTR2 expression,AuNRs@mSiO_(2)@DOTA-TATE with light inhibited tumours significantly better than AuNRs@mSiO_(2) with light.In vivo,the SSTR2-targeting ability and biodistribution of AuNRs@mSiO_(2)@DOTA-TATE were confirmed with AuNRs@mSiO_(2)@64Cu-DOTA-TATE under micro-positron emission tomography/computed tomography(micro-PET/CT);in the AuNRs@mSiO_(2)@DOTA-TATE with laser group,the tumour surface temperature increased rapidly,with tumour volumes similar to those in the octreotide group and significantly lower than those in other groups.There was no significant difference in mice body weight between the AuNRs@mSiO_(2)@DOTA-TATE with laser group and other groups.No significant inflammatory lesions or cell necrosis was found in the main organs.In summary,we presented a feasible strategy to construct AuNRs@mSiO_(2)@DOTA-TATE with good photothermal conversion efficiency,targetingSSTR2 ability,significant antitumour effects,and good biocompatibility,warranting further explorations of AuNRs@mSiO_(2)@DOTA-TATE for NETs therapy applications.Qichen Chen Zilin Li Jiangyuan Yu Qing Xie Haizhen Lu Yiqiao Deng Jinghua Chen Wenjia Zhu Li Huo Yizhou Zhang Wei Song Jianqiang Lan Jianqiang Cai Zhen Huang Zixi Wang Hong Zhao 2022Nano Research2022,15,10:0
5RING finger 138 deregulation distorts NF-κB signaling and facilities colitis switch to aggressive malignancy显示文摘Prolonged activation of nuclear factor(NF)-кB signaling significantly contributes to the development of colorectal cancer(CRC).New therapeutic opportunities are emerging from targeting this distorted cell signaling transduction.Here,we discovered the critical role of RING finger 138(RNF138)in CRC tumorigenesis through regulating the NF-кB signaling,which is independent of its Ubiquitin-E3 ligase activity involved in DNA damage response.RNF138^(−/−) mice were hyper-susceptible to the switch from colitis to aggressive malignancy,which coincided with sustained aberrant NF-кB signaling in the colonic cells.Furthermore,RNF138 suppresses the activation of NF-кB signaling pathway through preventing the translocation of NIK and IKK-Beta Binding Protein(NIBP)to the cytoplasm,which requires the ubiquitin interaction motif(UIM)domain.More importantly,we uncovered a significant correlation between poor prognosis and the downregulation of RNF138 associated with reinforced NF-кB signaling in clinical settings,raising the possibility of RNF138 dysregulation as an indicator for the therapeutic intervention targeting NF-кB signaling.Using the xenograft models built upon either RNF138-dificient CRC cells or the cells derived from the RNF138-dysregulated CRC patients,we demonstrated that the inhibition of NF-кB signaling effectively hampered tumor growth.Overall,our work defined the pathogenic role of aberrant NF-кB signaling due to RNF138 downregulation in the cascade events from the colitis switch to colonic neoplastic transformation and progression,and also highlights the possibility of targeting the NF-кB signaling in treating specific subtypes of CRC indicated by RNF138-ablation.Yalan Lu Rong Huang Jianming Ying Xingchen Li Tao Jiao Lei Guo Haitao Zhou Han Wang Amannisa Tuersuntuoheti Jianmei Liu Qichen Chen Yanhong Wang Luying Su Changyuan Guo Fu Xu Ziyi Wang Yan Lu Kai Li Junbo Liang Zhen Huang Xiao Chen Jinjie Yao Hanjie Hu Xiaowen Cheng Yufeng Wan Xinyan Chen Ning Zhang Shiying Miao Jianqiang Cai Linfang Wang Changzheng Liu Wei Song Hong Zhao 2022Signal Transduction and Targeted Therapy2022,7,7:0
