| 2 | MiR146a-loaded engineered exosomes released from silk fibroin patch promote diabetic wound healing by targeting IRAK1显示文摘Unhealable diabetic wounds need to be addressed with the help of newer,more efficacious strategies.Exosomes combined with biomaterials for sustained delivery of therapeutic agents are expected to bring new hope for chronic wound treatment.Here,the engineered exosomes modified for efficiently loading miR146a and attaching to silk fibroin patch(SFP)were demonstrated to promote diabetic wound healing.Silk fibroin binding peptide(SFBP)was screened through phage display,and SFBP-Gluc-MS2(SGM)and pac-miR146a-pac fusion protein were constructed.The designed exosomes(SGM-Exos,miR146a-Exos,and SGM-miR146a-Exos)were isolated from the engineered placental mesenchymal stem cells(PMSCs)transduced with SGM or/and pac-miR146a-pac protein.Gluc signals indicated SGM-Exo@SFP markedly increased the binding rate and the stability of SGM-Exo.Moreover,the loading efficiency of miR146a in SGM-miR146a-Exos was ten-fold higher than that in miR146a-Exos.Superior to untreated,SGM-miR146a-Exo-only treated,and SFP-only treated groups,SGM-miR146a-Exo@SFP drived wound healing associated with less inflammation,collagen deposition,and neovascularization.The transcriptomics analysis suggested anti-inflammatory and regenerative effects with SGM-miR146a-Exo@SFP treatment.Here,we show efficient exosome@biomaterial-based miRNA delivery systems for regenerative medicine and tissue engineering. | Qiankun Li Wenzhi Hu Qilin Huang Jie Yang Bingmin Li Kui Ma Qian Wei Yaxi Wang Jianlong Su Mengli Sun Shengnan Cui Rungong Yang Haihong Li Xiaobing Fu Cuiping Zhang | 2023 | Signal Transduction and Targeted Therapy2023,8,3: | 2 |

| 4 | DESIGN OF TERNARY COUNTER BASED ON ADIABATIC DOMINO CIRCUIT显示文摘By researching the ternary counter and low power circuit design method, a novel design of low power ternary Domino counter on switch-level is proposed. Firstly, the switch-level structure expression of ternary loop operation circuit with enable pin is derived according to the switch-signal theory, and the one bit ternary counter is obtained combining the ternary adiabatic Domino literal operation circuit and buffer. Then the switch-level structure expression of enable signal circuit is derived, and the four bits ternary counter is obtained by cascade connection. Finally, the circuit is simulated by Spice tool and the output waveforms transform in proper order indicating that the logic function is correct. | 5 | The influence of pulsed parameters on the damage of a Darlington transistor显示文摘Theoretical research on the heat accumulation effect of a Darlington transistor induced by high power microwave is conducted,and temperature variation as functions of pulse repetitive frequency(PRF)and duty cycle(DC)are studied.According to the distribution of the electronic field and the current density in the Darlington transistor,the research of the damage mechanism is carried out.The results show that for repetitive pulses with the same pulse widths and different PRFs,the value of temperature variation increases with PRF increases,and the peak temperature has almost no change when PRF is lower than 200 k Hz;while for the repetitive pulses with the same PRF and different pulse widths,the larger the pulse width is,the greater temperature variation varies.The response of the peak temperature caused by a single pulse demonstrates that there is no temperature variation when the rising time is much shorter than the falling time.In addition,the relationship between the temperature variation and the time during the rising edge time as well as that between the temperature variation and the time during the falling edge time are obtained utilizing the curve fitting method.Finally,for a certain average power,with DC increases the value of temperature variation decreases. | Qiankun Wang Changchun Chai Yuqian Liu Yintang Yang | 2018 | Journal of Semiconductors2018,39,9: | 1 |
| 6 | COPPER:a combinatorial optimization problem solver with processing-in-memory architecture显示文摘The combinatorial optimization problem(COP),which aims to find the optimal solution in discrete space,is fundamental in various fields.Unfortunately,many COPs are NP-complete,and require much more time to solve as the problem scale increases.Troubled by this,researchers may prefer fast methods even if they are not exact,so approximation algorithms,heuristic algorithms,and machine learning have been proposed.Some works proposed chaotic simulated annealing(CSA)based on the Hopfield neural network and did a good job.However,CSA is not something that current general-purpose processors can handle easily,and there is no special hardware for it.To efficiently perform CSA,we propose a software and hardware co-design.In software,we quantize the weight and output using appropriate bit widths,and then modify the calculations that are not suitable for hardware implementation.In hardware,we design a specialized processing-in-memory hardware architecture named COPPER based on the memristor.COPPER is capable of efficiently running the modified quantized CSA algorithm and supporting the pipeline further acceleration.The results show that COPPER can perform CSA remarkably well in both speed and energy. | Qiankun WANG Xingchen LI Bingzhe WU Ke YANG Wei HU Guangyu SUN Yuchao YANG | 2023 | Frontiers of Information Technology & Electronic Engineering2023,24,5: | 0 |
| 7 | Scutellarin prevents acute alcohol-induced liver injury via inhibiting oxidative stress by regulating the Nrf2/HO-1 pathway and inhibiting inflammation by regulating the AKT,p38 MAPK/NF-κB pathways显示文摘Alcoholic liver disease(ALD)is the most frequent liver disease worldwide,resulting in severe harm to personal health and posing a serious burden to public health.Based on the reported antioxidant and anti-inflammatory capacities of scutellarin(SCU),this study investigated its protective role in male BALB/c mice with acute alcoholic liver injury after oral administration(10,25,and 50 mg/kg).The results indicated that SCU could lessen serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)levels and improve the histopathological changes in acute alcoholic liver;it reduced alcohol-induced malondialdehyde(MDA)content and increased glutathione peroxidase(GSH-Px),catalase(CAT),and superoxide dismutase(SOD)activity.Furthermore,SCU decreased tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),and IL-1βmessenger RNA(mRNA)expression levels,weakened inducible nitric oxide synthase(iNOS)activity,and inhibited nucleotide-binding oligomerization domain(NOD)-like receptor protein 3(NLRP3)inflammasome activation.Mechanistically,SCU suppressed cytochrome P450 family 2 subfamily E member 1(CYP2E1)upregulation triggered by alcohol,increased the expression of oxidative stress-related nuclear factor erythroid 2-related factor 2(Nrf2)and heme oxygenase-1(HO-1)pathways,and suppressed the inflammation-related degradation of inhibitor of nuclear factor-κB(NF-κB)-α(IκBα)as well as activation of NF-κB by mediating the protein kinase B(AKT)and p38 mitogen-activated protein kinase(MAPK)pathways.These findings demonstrate that SCU protects against acute alcoholic liver injury via inhibiting oxidative stress by regulating the Nrf2/HO-1 pathway and suppressing inflammation by regulating the AKT,p38 MAPK/NF-κB pathways. | Xiao ZHANG Zhicheng DONG Hui FAN Qiankun YANG Guili YU Enzhuang PAN Nana HE Xueqing LI Panpan ZHAO Mian FU Jingquan DONG | 2023 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,7: | 0 |

| 11 | NLRP3 inflammasome signal pathway involves in Vibrio harveyi-induced inflammatory response in murine peritoneal macrophages in vitro显示文摘Vibrio harveyi,an important zoonotic pathogen,can infect wounds and cause inflammatory response.Understanding the inflammatory response pathways could facilitate the exploration of molecular mechanisms for treating V.harveyi infection.NLR family pyrin domain-containing 3(NLRP3)inflammasome is involved in the interaction between hosts&pathogenic microorganisms&could be sensed by various pathogen-associated molecular patterns(PAMPs)or damage-associated molecular patterns(DAMPs).Nonetheless,the function of NLRP3 inflammasome in V.harveyi infection remains unclear.In the present study,we established a V.harveyi infection model using murine peritoneal macrophages(PMs).Various techniques,including western blot analysis,enzyme-linked immunosorbent assay(ELISA),RT-qPCR,immunofluorescence,&inhibition assays,were used to explore the molecular mechanism of V.harveyi-induced inflammation.The results showed that many inflammatory cytokines participated in V.harveyi infection,with interleukin(IL)-1βbeing the most abundant.Pan-caspase inhibitor pretreatment significantly decreased the secretion of IL-1βin murine PMs.Moreover,the identification of V.harveyi involved a large number of NLR molecules,especially the NLRP3 receptor,&further studies revealed that NLPR3 inflammasome was activated by V.harveyi infection,as evidenced by puncta-like NLRP3 surrounding cell nuclear,ASC specks in the nucleus&cytoplasm,&ASC oligomerization.Inhibition of NLRP3 inflammasome impaired the release of mature IL-1βin V.harveyi-infected murine PMs.Furthermore,blocking the secretion of mature IL-1βcould markedly decrease the release of other proinflammatory cytokines,including IL-6,IL-12,&tumor necrosis factor-α.Overall,these data indicated that NLRP3 inflammasome was activated in response to V.harveyi infection and enhanced inflammatory response by promoting IL-1βsecretion in murine PMs. | Guili Yu Jinxin Wang Wei Zhang Qiankun Yang Gang Liu Lei Wang Babatunde Kazeem Bello Xiao Zhang Tianmeng Zhang Hui Fan Panpan Zhao Wei Liang Jingquan Dong | 2021 | Acta Biochimica et Biophysica Sinica2021,53,12: | 0 |
