1Tumor-expressed B7-H3 mediates the inhibition of antitumor T-cell functions in ovarian cancer insensitive to PD-1 blockade therapy显示文摘Although PD-L1/PD-1 blockade therapy has been approved to treat many types of cancers,the majority of patients with solid tumors do not respond well,but the underlying reason remains unclear.Here,we studied ovarian cancer(OvCa),a tumor type generally resistant to current immunotherapies,to investigate PD-1-independent immunosuppression.We found that PD-L1 was not highly expressed in the tumor microenvironment(TME)of human OvCa.Instead,B7-H3,another checkpoint molecule,was highly expressed by both tumor cells and tumor-infiltrating antigen-presenting cells(APCs),which correlated with T-cell exhaustion in patients.Using ID8 OvCa mouse models,we found that B7-H3 expressed on tumor cells,but not host cells,had a dominant role in suppressing antitumor immunity.Therapeutically,B7-H3 blockade,but not PD-1 blockade,prolonged the survival of ID8 tumorbearing mice.Collectively,our results demonstrate that tumor-expressed B7-H3 inhibits the function of CD8^(+)T cells and suggest that B7-H3 may be a target in patients who are not responsive to PD-L1/PD-1 inhibition,particularly OvCa patients.Dongli Cai Jiaming Li Dingfeng Liu Shanjuan Hong Qin Qiao Qinli Sun Pingping Li Nanan Lyu Tiantian Sun Shan Xie Li Guo Ling Ni Liping Jin Chen Dong 2020Cellular & Molecular Immunology2020,17,3:8

3Flexibility analysis and design of heat exchanger network for syngas-to-methanol process显示文摘The heat exchanger network(HEN)in a syngas-to-methanol process was designed and optimized based on pinch technology under stable operating conditions to balance the energy consumption and economic gain.In actual industrial processes,fluctuations in production inevitably affect the stable operation of HENs.A flexibility analysis of the HEN was carried out to minimize such disturbances using the downstream paths method.The results show that two-third of the downstream paths cannot meet flexibility requirements,indicating that the HEN does not have enough flexibility to accommodate the disturbances in actual production.A flexible HEN was then designed with the method of dividing and subsequent merging of streams,which led to 13.89%and 20.82%reductions in energy consumption and total cost,respectively.Owing to the sufficient area margin and additional alternative heat exchangers,the flexible HEN was able to resist interference and maintain production stability and safety,with the total cost increasing by just 4.08%.Jinchang Liu Pingping Zhang Qiang Xie Dingcheng Liang Lei Bai 2021International Journal of Coal Science & Technology2021,8,6:1

7A Note on Reviving the Goddard Satellite-Based Surface Turbulent Fluxes (GSSTF) Dataset显示文摘Accurate sea surface flux measurements are crucial for understanding the global water and energy cycles. The oceanic evaporation, which is a major component of the global oceanic fresh water flux, is useful for predicting oceanic circulation and transport. The global Goddard Satellite-based Surface Turbulent Fluxes Version-2 (GSSTF2; July 1987–December 2000) dateset that was o?cially released in 2001 has been widely used by scientific community for global energy and water cycle research, and regional and short period data analyses. We have recently been funded by NASA to resume processing the GSSTF dataset with an objective of continually producing a uniform dataset of sea surface turbulent fluxes, derived from remote sensing data. The dataset is to be reprocessed and brought up-to-date (GSSTF2b) using improved input datasets such as a recently upgraded NCEP/DOE sea surface temperature reanalysis, and an upgraded surface wind and microwave brightness temperature V6 dataset (Version 6) from the Special Sensor Microwave Imager (SSM/I) produced by Remote Sensing Systems (RSS). A second new product (GSSTF3) is further proposed with a finer temporal (12-h) and spatial (0.25°× 0.25°) resolution. GSSTF2b (July 1987–December 2008) and GSSTF3 (July 1999–December 2009) will be released for the research community to use by late 2009 and early 2011, respectively.Chung-Lin SHIE Long S. 9Phylogenetic and Expression Analyses of With-No-Lysine Kinase Genes Reveal Novel Gene Family Diversity in Fruit Trees显示文摘With-No-Lysine kinases(WNK) have been reported to be associated with plant growth regulation in Arabidopsis, soybean, and rice, but little is known of their roles in fruits. In this study, a total of 114 WNK genes were identified from 8 fruit trees species, and these WNK genes belonged to 2 classes(I and II) that respectively contain 4(A, B, C, D) and 2 groups(E, F). The WNK genes had variable exon-intron structures and were randomly distributed among most chromosomes of each genus. The expression levels of six of the 18 WNK genes in peach were almost undetectable in fruits, suggesting that they may not be associated with fruit development and ripening. Of the other 12 WNK genes, PpWNK.B1 exhibited stronger stability of expression levels than the best reference genes reported previously in fruits from two peach cultivars. These results suggested that PpWNK.B1 could be a reliable reference gene for gene expression studies of peach fruit. Moreover, qRT-PCR assays yielded evidence that when using single reference gene as internal control, mistakes were more possible, indicating that the use of three reference genes is necessary for gene expression studies. Based on the gene expression profiles validated by qRT-PCR, PpWNK.A1 is predicted to likely be involved in fruit ripening, while PpWNK.A2 and PpWNK.E3.1 are likely associated with early fruit development. 20Biased heteroplasmy within the mitogenomic sequences of Gigantometra gigas revealed by sanger and high-throughput methods显示文摘Few studies have explored the differences between Sanger and HTS methods in the results of mitogenome sequencing. We used a single individual of insect to study the differences between the sequences given by Sanger and PCR-free HTS methods. Here we provided evidence for biased results of sequencing due to different methods in the mitochondrial genes of atp6, atp8, cox1, cox2, cox3, Cytb, nad2, nad3, nad4, nad5, rrn S, rrnL, trnH, trn I, and control region at various degrees. Especially, in cox1, the differently sequenced nucleotides account for 2.6% of the complete length. Furthermore, the highest value of the intraspecific genetic distance based on K2 P accounts for 2.5% using a barcode fragment size of cox1(651 bp, Sanger), while the maximum distance of the corresponding cox1 fragment obtained by the two sequencing methods was 5.0%. We revealed that the methods of Sanger and HTS may give different sequencing results of mitochondrial genes, which may reflect the heteroplasmy of mitogenomes within an insect individual.
