1Celastrus orbiculatus extract induces mitochondrial-mediated apoptosis in human hepatocellular carcinoma cells显示文摘OBJECTIVE:To investigate the apoptotic effects and underlying molecular mechanisms of Celastrus orbiculatus(C.orbiculatus) extract in human hepatocellular carcinoma cells.METHODS:Human hepatocellular carcinoma cells(HCCLM6) were treated with C.orbiculatus extract(COE) at different nontoxic concentrations(10,20,40,80,and 160 μg/mL).The effect of COE on HC-CLM6 viability was examined using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) assays.Cellular apoptosis following COE treatment was assessed by flow cytometry and western blot analysis.RESULTS:COE significantly inhibited cell viability and induced apoptosis of HCCLM6 cells in a dose-dependent manner.Apoptosis was accompanied by increased Bax expression and decreased Bcl-2 expression.In addition,COE treatment led to the release of cytochrome c,activation of caspase-3,and cleavage of poly(ADP-ribose) polymerase(PARP).Furthermore,activation of extracellular signal-regulated kinase(ERK),p38 kinase,and c-Jun N-terminal kinase(JNK) phosphorylation,and down-regulation of Akt phosphorylation was observed.CONCLUSION:COE induces mitochondrial-mediated,caspase-dependent apoptosis in HCCLM6 cells,which might be attributed to the activation of mitogen-activated protein kinase(MAPK) and inhibition of Akt signaling pathways.These data suggest that COE may be a potential treatment for human hepatocellular carcinoma.Hua Zhang Yayun Qian Yanqing Liu Guoqing Li Pingfang Cui Yaodong Zhu Hui Ma Xue Ji Shiyu Guo Hisamits Tadashi 2012Journal of Traditional Chinese Medicine2012,32,4:18
2Effects of motilin and ursodeoxycholic acid on gastrointestinal myoelectric activity of different origins in fasted rats显示文摘AIM: To investigate gastrointestinal migrating myoelectric complex (MMC) and the effects of porcine motilin and ursodeoxycholic acid (UDCA) on MMC of gastrointestinal tract of different origins in fasted rats.METHODS: Three bipolar silver electrodes were chronically implanted on the antrum, duodenum and jejunum. Seven days later 24 experimental rats were divided into 2 groups.One group was injected with porcine motilin via sublingual vein at a dose of 20 lig/kg, the other group was perfused into stomach with UDCA. The gastrointestinal myoelectric activity was recorded 1 h before and 2 h after the test substance infusions into the rats.RESULTS: In all fasted rats a typical pattern of MMC was observed. Among the totally 68 activity fronts recorded in fasted rats under control, 67% started in duodenum, and 33% in antrum. MMC cycle duration and duration of phase III of antral origin were longer than those of duodenal origin. Administration of 20 lig/kg porcine motilin induced a premature antral phase III of antral origin. But perfusion into stomach with UDCA resulted in shorter MMC cycle duration, longer duration of phase III of duodenal origin,which were followed with shorter cycle duration and duration of antral phase III.CONCLUSION: In fasted rats, MMC could originate from antrum and duodenum respectively. The characteristics of MMC of different origins may contribute to the large variations within subjects. The mechanisms of different origins of phase III may be different. 3Whole genome re-sequencing reveals evolutionary patterns of sacred lotus (Nelumbo nucifera)显示文摘Sacred lotus(Nelumbo nucifera or lotus) is an important aquatic plant in horticulture and ecosystems. As a foundation for exploring genomic variation and evolution among different germplasms, we re-sequenced 19 individuals from three cultivated temperate lotus subgroups(rhizome,seed and flower lotus), one wild temperate lotus subgroup(wild lotus), one tropical lotus group(Thai lotus) and an outgroup(Nelumbo lutea). Through genetic diversity and polymorphism analysis by non-missing SNP sites widely distributed in the whole genome, we confirmed that wild and Thai lotus exhibited greater differentiation with a higher genomic diversity compared to cultivated lotus. Rhizome lotus had the lowest genomic diversity and a closer relationship to wild lotus, whereas the genomes of seed and flower lotus were admixed. Genes in energy metabolism process and plant immunity evolved rapidly in lotus, reflecting local adaptation.We established that candidate genes in genomic regions with significant differentiation associated with temperate and tropical lotus divergence always exhibited highly divergent expression pattern. 4Synthesis of magnetically modified palygorskite composite for immobilization of Candida sp. 99–125 lipase via adsorption显示文摘Magnetically modified palygorskite composites were synthesized with γ-Fe2O3 dispersing on the external surface of clay mineral. The magnetic clay was characterized with Fourier transform infrared, X-ray diffraction, transmission electron microscopy, and vibrating sample magnetometer. Candida sp. 99–125 lipase was immobilized on magnetic palygorskite composites by physical adsorption with enzyme loading of41.5 mg·g-1support and enzyme activity of 2631.6 U·(g support)-1. 5Attenuated phenotypes and analysis of a herpes simplex virus 1strain with partial deletion of the UL7, UL41 and LAT genes显示文摘We previously constructed a herpes simplex virus 1(HSV-1) UL7 mutant virus(M1) and showed that a partial deletion mutation of the UL7 gene led to a lower proliferative rate and an attenuated phenotype. Using the M1 mutant, we further modified the UL41 gene, which encodes another tegument protein, and the latency-associated transcript(LAT) gene. Observations of the resulting mutants with modified UL7 and UL41(M2) or UL7, UL41 and LAT(M3) genes indicated attenuated phenotypes, with lower proliferative ratios in various cells, non-lethal infections in mice and lower viral loads in nervous tissues compared with the wild-type strain. Furthermore, no LAT stable intron could be detected in the trigeminal ganglion of M3-infected animals. The results obtained with the three HSV-1 mutants indicate that the M3 mutant is an attenuated strain with low pathogenicity during both acute and latent infections. 6A comparative study of the characteristics of two Coxsackie A virus type 16 strains (genotype B)显示文摘Coxsackie A virus is one of the major pathogens associated with hand, foot and mouth disease (HFMD). The etiological characteristics of Coxsackie A virus type 16 (CA16) are thought to correlate with the pathological process of its infection. Two CA16 strains that were isolated from a severe HFMD patient presented with different plaque forms. This observation, along with biological analysis, indicated that the differences in the strains' biological characteristics, such as proliferation kinetics and immunogenicity, correlate with differences in their pathogenicity toward neonatal mice. Furthermore, these differences are thought to be associated with the sequence of the 5′ non-coding region of the viral genome and the VP1 structural region sequence. 7Fundamental CRISPR-Cas9 tools and current applications in microbial systems显示文摘Derived from the bacterial adaptive immune system,CRISPR technology has revolutionized conventional genetic engineering methods and unprecedentedly facilitated strain engineering.In this review,we outline the fundamental CRISPR tools that have been employed for strain optimization.These tools include CRISPR editing,CRISPR interference,CRISPR activation and protein imaging.To further characterize the CRISPR technology,we present current applications of these tools in microbial systems,including model-and non-model industrial microorganisms.Specially,we point out the major challenges of the CRISPR tools when utilized for multiplex genome editing and sophisticated expression regulation.To address these challenges,we came up with strategies that place emphasis on the amelioration of DNA repair efficiency through CRISPR-Cas9-assisted recombineering.Lastly,multiple promising research directions were proposed,mainly focusing on CRISPR-based construction of microbial ecosystems toward high production of desired chemicals.Pingfang Tian Jia Wang Xiaolin Shen Justin Forrest Rey Qipeng Yuan Yajun Yan 2017Synthetic and Systems Biotechnology2017,2,3:4
8Gene Expression Profile Changes in Germinating Rice显示文摘Water absorption is a prerequisite for seed germination.During imbibition,water influx causes the resumption of many physiological and metabolic processes in growing seed.In order to obtain more complete knowledge about the mechanism of seed germination,two-dimensional gel electrophoresis was applied to investigate the protein profile changes of rice seed during the first 48 h of imbibition.Thirtynine differentially expressed proteins were identified,including 19 down-regulated and 20 up-regulated proteins.Storage proteins and some seed development-and desiccation-associated proteins were down regulated.The changed patterns of these proteins indicated extensive mobilization of seed reserves.By contrast,catabolism-associated proteins were up regulated upon imbibition.Semi-quantitative real time polymerase chain reaction analysis showed that most of the genes encoding the down-or upregulated proteins were also down or up regulated at mRNA level.The expression of these genes was largely consistent at mRNA and protein levels.In providing additional information concerning gene regulation in early plant life,this study will facilitate understanding of the molecular mechanisms of seed germination.Dongli He Chao Han Pingfang Yang 2011Journal of Integrative Plant Biology2011,53,10:3

10A Recommendation Mechanism for Web Publishing Based on Sentiment Analysis of Microblog显示文摘Microblog is a social platform with huge user community and mass data. We propose a semantic recommendation mechanism based on sentiment analysis for microblog. Firstly, the keywords and sensibility words in this mechanism are extracted by natural language processing including segmentation, lexical analysis and strategy selection. Then, we query the background knowledge base based on linked open data(LOD) with the basic information of users. The experiment result shows that the accuracy of recommendation is within the range of 70%-89% with sentiment analysis and semantic query. 11A Review of Co-processing of Kitchen Waste and Municipal Residual Sludge显示文摘In this paper,the methods of kitchen waste disposal in recent years were summed up,and the advantages and disadvantages of the current treatment methods were analyzed. At the same time,it has been found that the co-processing of kitchen waste and residual sludge not only has dealt with both kitchen waste and residual sludge,but also the carbon and nitrogen ratio and moisture content of the mixture are balanced,and increases the processing efficiency significantly. It has a certain processing advantage. At present,most research is about the anaerobic digestion of kitchen waste and residual sludge,and the process is more mature,but the study of VFAs accumulation is still insufficient. Aerobic composting is still in the early stage of development,and there are few studies on it at home and abroad. 17Identifying RNA Modifications by Direct RNA Sequencing Reveals Complexity of Epitranscriptomic Dynamics in Rice显示文摘Transcriptome analysis based on high-throughput sequencing of a cDNA library has been widely applied to functional genomic studies.However,the cDNA dependence of most RNA sequencing techniques constrains their ability to detect base modifications on RNA,which is an important element for the post-transcriptional regulation of gene expression.To comprehensively profile the N^(6)-methyladenosine(m^(6)A)and N^(5)-methylcytosine(m5 C)modifications on RNA,direct RNA sequencing(DRS)using the latest Oxford Nanopore Technology was applied to analyze the transcriptome of six tissues in rice.Approximately 94 million reads were generated,with an average length ranging from 619 nt to 1013 nt,and a total of 45,707 transcripts across 34,763 genes were detected.Expression profiles of transcripts at the isoform level were quantified among tissues.Transcriptome-wide mapping of m^(6)A and m5 C demonstrated that both modifications exhibited tissue-specific characteristics.The transcripts with m^(6)A modifications tended to be modified by m5 C,and the transcripts with modifications presented higher expression levels along with shorter poly(A)tails than transcripts without modifications,suggesting the complexity of gene expression regulation.Gene Ontology analysis demonstrated that m^(6)A-and m5 C-modified transcripts were involved in central metabolic pathways related to the life cycle,with modifications on the target genes selected in a tissue-specific manner.Furthermore,most modified sites were located within quantitative trait loci that control important agronomic traits,highlighting the value of cloning functional loci.The results provide new insights into the expression regulation complexity and data resource of the transcriptome and epitranscriptome,improving our understanding of the rice genome.Feng Yu Huanhuan Qi Li Gao Sen Luo Rebecca Njeri Damaris Yinggen Ke Wenhua Wu Pingfang Yang 2023Genomics, Proteomics & Bioinformatics2023,21,4:1
