2Annual report on status of cancer in China,2010显示文摘Objective:Population-based cancer registration data in 2010 were collected,evaluated and analyzed by the National Central Cancer Registry(NCCR) of China.Cancer incident new cases and cancer deaths were estimated.Methods:There were 219 cancer registries submitted cancer incidence and death data in 2010.All data were checked and evaluated on basis of the criteria of data quality from NCCR.Total 145 registries' data were qualified and accepted for cancer statistics in 2010.Pooled data were stratified by urban/rural,area,sex,age group and cancer site.Cancer incident cases and deaths were estimated using age-specific rates and national population.The top ten common cancers in different groups,proportion and cumulative rate were also calculated.Chinese census in 2000 and Segi's population were used for age-standardized incidence/ mortality rates.Results:All 145 cancer registries(63 in urban and 82 in rural) covered a total of 158,403,248 population(92,433,739 in urban and 65,969,509 in rural areas).The estimates of new cancer incident cases and cancer deaths were 3,093,039 and 1,956,622 in 2010,respectively.The morphology verified cases(MV%) accounted for 67.11% and 2.99% of incident cases were identified through death certifications only(DCO%) with mortality to incidence ratio(M/I) of 0.61.The crude incidence rate was 235.23/100,000(268.65/100,000 in males,200.21/100,000 in females),age-standardized incidence rates by Chinese standard population(ASIRC,2000) and by world standard population(ASIRW) were 184.58/100,000 and 181.49/100,000 with the cumulative incidence rate(0-74 years old) of 21.11%.The cancer incidence and ASIRC were 256.41/100,000 and 187.53/100,000 in urban areas whereas in rural areas,they were 213.71/100,000 and 181.10/100,000,respectively.The crude cancer mortality in China was 148.81/100,000(186.37/100,000 in males and 109.42/100,000 in females),age-standardized incidence rates by Chinese standard population(ASMRC,2000) and by world standard population(ASMRW) were 113.92/100,000 and 112.86/100,000,and the cumulative incidence rate(0-74 years old) was 12.78%.The cancer mortality and ASMRC were 156.14/100,000 and 109.21/100,000 in urban areas,whereas in rural areas,they were 141.35/100,000 and 119.00/100,000 respectively.Lung cancer,gastric cancer,colorectal cancer,liver cancer,esophageal cancer,pancreas cancer,encephaloma,lymphoma,female breast cancer and cervical cancer,were the most common cancers,accounting for 75% of all cancer cases in urban and rural areas.Lung cancer,gastric cancer,liver cancer,esophageal cancer,colorectal cancer,pancreatic cancer,breast cancer,encephaloma,leukemia and lymphoma accounted for 80% of all cancer deaths.Conclusions:The coverage of cancer registration population had a rapid increase and could reflect cancer burden in each area and population.As the basis of cancer control program,cancer registry plays an irreplaceable role in cancer epidemic surveillance,evaluation of cancer control programs and making anticancer strategy.China is facing serious cancer burden and prevention and control should be enhanced.Wanqing Chen Rongshou Zheng Siwei Zhang Ping Zhao Hongmei Zeng Xiaonong Zou Jie He 2014Chinese Journal of Cancer Research2014,26,1:198

4Biotransfer of heavy metals along a soil-plant-insect-chicken food chain:Field study显示文摘The accumulation and transfer of Pb,Zn,Cu,and Cd along a soil-plant-insect-chicken food chain at contaminated sites were investigated.The study site nearing the Pb/Zn mine had been contaminated by heavy metals severely.Cadmium and Pb concentrations steadily declined with increasing trophic level(p < 0.01),but concentrations of Zn and Cu slightly increased from plant to insect larva(p > 0.05).The concentrations of heavy metals were the highest in chicken muscle,with lower values in liver and blood.The bioaccumulation of Pb was observed in chicken livers.The eliminations of Pb,Zn,Cu,and Cd via insect and chicken feces avoid metal bioaccumulation in insect and chicken body.These results suggest that the accumulation of heavy metals in specific animal organ of tissues could not be neglected,although transfer of metals to chicken from plant and insect was limited.ZHUANG Ping ZOU Huiling SHU Wensheng 2009Journal of Environmental Sciences2009,21,6:31
5Lime and Phosphate Could Reduce Cadmium Uptake by Five Vegetables Commonly Grown in South China显示文摘A pot experiment was conducted in artificially Cd-contaminated (5 mg Cd kg-1) soils to investigate the feasibility of using lime (3 g kg-1) or phosphate (80 mg P kg-1) to mitigate uptake of Cd by vegetables.Five common vegetables in South China,including lettuce (Lactuca sativa L.var.ramosa Hort.),Chinese cabbage [Brassica rapa L.subsp.Chinensis (L.) var.parachinensis (L.H.Bailey) Hanect],Chinese broccoli (Brassica oleracea L.var.albiflora Kuntze),white amaranth (Amaranthus tricolor L.) and purslane (Amaranthus viridis L.),were grown in the soils and harvested after 60 d.The results showed that liming significantly reduced Cd uptake by most vegetables by 40%-50% (or a maximum of 70%),mainly due to immobilization of soil Cd.Increased availability of Ca in the soil might also contribute to the Cd uptake reduction as a result of absorption competition between Ca and Cd.Liming caused biomass reduction in white amaranth and purslane,but did not influence growth of the other vegetables.Phosphate decreased Cd uptake by vegetables by 12%-23%.Compared with lime,phosphate decreased,to a smaller extent,the bioavailability of Cd in most cases.Phosphate markedly promoted growth of vegetables.Changes in soil chemistry by adding lime or phosphate did not markedly influence nutrient uptake of vegetables except that lime increased Ca content and phosphate increased P content in shoots of the vegetables.The results suggested that a proper application of lime could be effective in reducing Cd uptake of vegetables,and phosphate could promote growth of the vegetables as well as alleviate the toxicity of Cd.TAN Wan-Neng LI Zhi-An QIU Jing ZOU Bi LI Ning-Yu ZHUANG Ping WANG Gang 2011Pedosphere2011,21,2:34
6Berberine inhibits hepatic gluconeogenesis via the LKB1-AMPK-TORC2 signaling pathway in streptozotocin-induced diabetic rats显示文摘AIM: To investigate the molecular mechanisms of berberine inhibition of hepatic gluconeogenesis in a diabetic rat model.METHODS: The 40 rats were randomly divided into five groups. One group was selected as the normal group. In the remaining groups(n = 8 each), the rats were fed on a high-fat diet for 1 mo and received intravenous injection of streptozotocin for induction of the diabetic models. Berberine(156 mg/kg per day)(berberine group) or metformin(184 mg/kg per day)(metformin group) was intragastrically administered to the diabetic rats and 5-aminoimidazole-4-carboxamide1-β-D-ribofuranoside(AICAR)(0.5 mg/kg per day)(AICAR group) was subcutaneously injected to the diabetic rats for 12 wk. The remaining eight diabetic rats served as the model group. Fasting plasma glucose and insulin levels as well as lipid profile were tested.The expressions of proteins were examined by western blotting. The nuclear translocation of CREB-regulated transcription co-activator(TORC)2 was observed by immunohistochemical staining. RESULTS: Berberine improved impaired glucose tolerance and decreased plasma hyperlipidemia. Moreover, berberine decreased fasting plasma insulin and homeostasis model assessment of insulin resistance(HOMA-IR). Berberine upregulated protein expression of liver kinase(LK)B1, AMP-activated protein kinase(AMPK) and phosphorylated AMPK(p-AMPK). The level of phophorylated TORC2(p-TORC2) protein in the cytoplasm was higher in the berberine group than in the model group, and no significant difference in total TORC2 protein level was observed. Immunohistochemical staining revealed that more TORC2 was localized in the cytoplasm of the berberine group than in the model group. Moreover, berberine treatment downregulated protein expression of the key gluconeogenic enzymes(phosphoenolpyruvate carboxykinase and glucose-6-phosphatase) in the liver tissues. 8Heavy Metal Contamination in Soil and Soybean near the Dabaoshan Mine, South China显示文摘Concentrations of Pb, Cd, Cu, Zn, Cr and Ni in soybean (Glycine max L.) grown near the Dabaoshan Mine were investigated, and their potential risk to the health of inhabitants was estimated. In the Fandong (FD) and Zhongxin (ZX) villages, which are near the Dabaoshan mineral deposit, concentrations of Pb (0.34 mg kg^(-1) for FD), Cd (0.23 mg kg^(-1) for ZX) and Cr (1.14 and 1.75 mg kg^(-1) for FD and ZX, respectively) in the seeds of soybean exceeded the tolerance limit set by Chinese standards. The estimated daily intakes (EDIs) from consumption of soybean seeds for FD inhabitants were 0.570, 0.170, 38.550, 142.400, 1.910 and 14.530 μg d^(-1) kg^(-1) boby weight for Pb, Cd, Cu, Zn, Cr and Ni, respectively. 11Submucosal tunneling endoscopic resection: An effective and safe therapy for upper gastrointestinal submucosal tumors originating from the muscularis propria layer显示文摘AIM To evaluate the effectiveness and safety of submucosal tunneling endoscopic resection(STER) and compare its outcomes in esophageal and cardial submucosal tumors(SMTs) of the muscularis propria(MP) layer.METHODS From May 2012 to November 2017, 173 consecutive patients with upper gastrointestinal(GI) SMTs of the MP layer underwent STER. Overall, 165 patients were included, and 8 were excluded. The baseline characteristics of the patients and SMTs were recorded. The en bloc resection rate, complete resection rate,residual rate, and recurrence rate were calculated to evaluate the effectiveness of STER, and the complication rate was recorded to evaluate its safety. Effectiveness and safety outcomes were compared between esophageal and cardial SMTs.RESULTS One hundred and twelve men and 53 women with a mean age of 46.9 ± 10.8 years were included. The mean tumor size was 22.6 ± 13.6 mm. Eleven SMTs were located in the upper esophagus(6.7%), 49 in the middle esophagus(29.7%), 46 in the lower esophagus(27.9%), and 59 in the cardia(35.7%). Irregular lesions accounted for 48.5% of all lesions. STER achieved an en bloc resection rate of78.7%(128/165) for GI SMTs with an overall complication rate of 21.2%(35/165).All complications resolved without intervention or were treated conservatively without the need for surgery. The en bloc resection rates of esophageal and cardial SMTs were 81.1%(86/106) and 72.1%(42/59), respectively(P = 0.142), and the complication rates were 19.8%(21/106) and 23.7%(14/59), respectively,(P =0.555). The most common complications for esophageal SMTs were gas-related complications and fever, while mucosal injury was the most common for cardial SMTs.CONCLUSION STER is an effective and safe therapy for GI SMTs of the MP layer. 13Effect and mechanism of Irbesartan on occurrence of ventricular arrhythmias in rats with myocardial ischemia through connexin43(cx43)显示文摘Objective: To explore the effect and mechanism of angiotensin Ⅱ receptor blockers-Irbesartan on occurrence of ventricular arrhythmias in rats with myocardial ischemia. Methods: Rats with embryonic cardiomyocytes-H9c2 were randomly divided into control group, ischemia group, Irbesartan group and Irbesartan+ischemia group. The cell viability of rats in each group was tested using MTT. Real-time PCR was employed to detect the expression of connexin43(Cx43) mR NA and western blot to detect the expression of Cx43 and phosphorylated Cx43. SD rats were randomly divided into the sham-operation group(SO), myocardial infarction group(MI), Irbesartan group and MI+ Irbesartan group, with 10 rats in each group. HE staining was employed to observe the change in the pathomorphology of left ventricular tissue and TUNEL method to analyze the cell apoptosis in the tissue. The immunofluorescence was adopted to observe the expression and distribution of Cx43 in the left ventricular myocardium and study the change in the expression of Cx43 in the cardiac muscular tissue at mR NA and protein level. Results: The intervention of Irbesartan in the condition of ischemia indicated the significant decrease in the number of necrotic cells. The expression of Cx43 was significantly decreased under the culture of ischemia(P<0.05), but in the presence of Irbesartan, the expression of Cx43 was increased compared with the ischemia group(P<0.01). The results of WB assay showed the similar trend of change at mRNA level. There was the significant difference in the score of ventricular arerythmia between MI group and SO group(P<0.01). The incidence of ventricular tachycardia or ventricular fibrillation was significantly increased compared with the one in SO group(P<0.05). There was the significant difference in the overall score between MI+Irbesartan group and MI group(P<0.05). The expression of Cx43 in the cardiac muscular tissue in MI group was significantly decreased(P<0.01 vs SO group). But the expression of Cx43 was increased after the treatment with Irbesartan. Conclusions: Irbesartan can inhibit the injury of H9c2 cardiomyocytes and the decreased expression of Cx43 that are induced by the ischemic myocardial infarction. 14CYP2E1-dependent hepatotoxicity and oxidative damage after ethanol administration in human primary hepatocytes显示文摘AIM: To observe the relationship between ethanol-induced oxidative damage in human primary cultured hepatocytes and cytochrome P450 2E1 (CYP2E1) activity, in order to address if inhibition of CYP2E1 could attenuate ethanolinduced cellular damage. METHODS: The dose-dependent (25-100 mmol/L) and time-dependent (0-24 h) exposures of primary human cultured hepatocytes to ethanol were carried out. CYP2E1 activity and protein expression were detected by spectrophotometer and Western blot analysis respectively. Hepatotoxicity was investigated by determination of Iactate dehydrogenase (LDH) and aspartate transaminase (AST) level in hepatocyte culture supernatants, as well as the intracellular formation of malondialdehyde (MDA). RESULTS: A dose-and time-dependent response between ethanol exposure and CYP2E1 activity in human hepatocytes was demonstrated. Moreover, there was a time-dependent increase of CYP2E1 protein after 100 mmol/L ethanol exposure. Meanwhile, ethanol exposure of hepatocytes caused a time-dependent increase of cellular MDA level, LDH, and AST activities in supernatants. Furthermore, the inhibitor of CYP2E1, diallyl sulfide (DAS) could partly attenuate the increases of MDA, LDH, and AST in human hepatocytes. 17Solution structure of the RNA recognition domain of METTL3-METTL14 N^6-methyladenosine methyltransferase显示文摘N^6-methyladenosine(m6A),a ubiquitous RNA modification,is installed by METTL3-METTL14 complex.The structure of the heterodimeric complex between the methyltransferase domains(MTDs)of METTL3 and METTL14 has been previously determined.However,the MTDs alone possess no enzymatic activity.Here we present the solution structure for the zinc finger domain(ZFD)of METTL3,the inclusion of which fulfills the methyltransferase activity of METTL3-METTL14.We show that the ZFD specifically binds to an RNA containing 5'-GGACU-3'consensus sequence,but does not to one without.The ZFD thus serves as the target recognition domain,a structural feature previously shown for DNA methyltransferases,and cooperates with the MTDs of METTL3-METTL14 for catalysis.However,the interaction between the ZFD and the specific RNA is extremely weak,with the binding affinity at several hundred micromolar under physiological conditions.The ZFD contains two CCCH-type zinc fingers connected by an anti-parallel P-sheet.Mutational analysis and NMR titrations have mapped the functional interface to a contiguous surface.As a division of labor,the RNA-binding interface comprises basic residues from zinc finger 1 and hydrophobic residues fromβ-sheet and zinc finger 2.Further we show that the linker between the ZFD and MTD of METTL3 is flexible but partially folded,which may permit the cooperation between the two domains during catalysis.Together,the structural characterization of METTL3 ZFD paves the way to elucidate the atomic details of the entire process of RNA m6A modification.Jinbo Huang Xu Dong Zhou Gong Ling-Yun Qin Shuai Yang Yue-Ling Zhu Xiang Wang Delin Zhang Tingting Zou Ping Yin Chun Tang 2019Protein & Cell2019,10,4:11

20Constructing a Core Collection for Maize (Zea mays L.) Landrace from Wuling Mountain Region in China显示文摘Based on the genetic clustering from 42 microsatellite (SSR) markers with a combination of their geographic origin and germplasm characteristics, 124 maize landraces from Wuling Mountain region in China were used for constructing a core collection. Four evaluating parameters for maize landrace core collection, including mean difference percentage (MD), variance difference percentage (VD), coincidence rate of range (CR), and variable rate of coefficient of variation (VR), were assessed with 20 quantitative traits. It was shown that genetic relationships among landraces in Wuling Mountain region had the tendency to associate with their geographic origins. The 124 landraces were clustered into 18 subgroups when the coefficient of genetic similarity (GS) is 0.28. Eighteen landraces, each of which was from one subgroup, were applied to construct the core collection with a sampling percentage of 15%. Comparison of the initial and core collection indicated that there existed no significant differences in most quantitative traits. An average of 6.3 and 6.5 alleles were detected in the initial and core collection, respectively. Mean polymorphism information content in the core collection (0.75) was higher than that in the initial one (0.72). MD was lesser than 20% and CR was more than 80%.
