| 1 | Expression of circulating microRNA-20a and let-7a in esophageal squamous cell carcinoma显示文摘AIM: To investigate the expressions of micro RNA-20a(mi R-20a) and let-7a in esophageal squamous cell carcinoma(ESCC) and their diagnostic value. METHODS: Seventy patients with ESCC and 40 healthy subjects were enrolled to investigate the expression of mi R-20 a and let-7a using quantitative real-time PCR. The expression of mi R-20 a and let-7a was compared between ESCC patients and healthy subjects. The plasma levels of mi R-20 a and let-7a in relation to patient clinicopathologic parameters, the receiver operating characteristic(ROC) curve, and the sensitivity and specificity of mi R-20 a and let-7a in ESCC diagnosis were analyzed.RESULTS: Plasma levels of mi R-20 a were significantly higher in ESCC patients than in healthy controls, and plasma levels of let-7 were lower in ESCC patients than in healthy controls(both P < 0.05). The area under the ROC curve of mi R-20 a was 0.767(95%CI: 0.677-0.857; P < 0.001), when the cut-off value was set at 4.77, the sensitivity and specificity were 64.3% and 75.0%, respectively. The area under the ROC curve of let-7a was 0.829(95%CI: 0.754-0.904; P < 0.001), when the cut-off value was set at 6.22, the sensitivity and specificity were 74.3% and 85.0%, respectively. Thus, the sensitivity and specificity of let-7a were higher than those of mi R-20 a. The median relative plasma expression of let-7a in clinical stage Ⅲ/Ⅳ(0.24) was lower than that in stage Ⅰ/Ⅱ(0.42), while the expression of mi R-20 a according to stage was not statistically different. | 2 | Polymorphism in the upstream regulatory region of human papilloma virus type 16 from the cervical cancer biopsies in Xinjiang Uygur women显示文摘To investigate the mutations in the upstream regulatory region (URR) of human papillomavirus type 16 (HPV-16) from the cervical cancer biopsies in Xinjiang Uygur women and its relationship to the high incidence of cervical cancer in the southern Xinjiang, the tissue DNA was extracted from the cervical cancer biopsies, and the URR segment of HPV-16 DNA was amplified, sequenced and analyzed. Thereafter, the polymorphism of URR in HPV-16 was then analyzed. It was demonstrated that the positive rate detected for the presence of URR in HPV-16 was 89.47% (17/19). Compared with the previously published sequence in URR of prototype HPV-16, some mutations were detected in the sequence of URR. The mutations in 17 URR fragments of HPV-16 could be divided into 11 patterns (XJU-1 to XJU-11) at nucleic acid level, in which each of XJU-1 and XJU-4 accounted for 23.53% (4/17), and other patterns of mutation accounted for 5.88% (1/17) . In comparison with the URR of prototype HPV-16, the DNA identity of these patterns was 98.50%-99.68% . In these 17 URR fragments, two point mutations occurred at position 7192 (G to T) and position 7520 (G to A) and they appeared to be constant in Xinjiang area. These two mutations were ubiquitous in the Asia-American type and conferred strong infection activity and carcinogenicity of this virus. In addition, the mutations at position 7729 (A to C), position 7843 (A to G) and position 7792 (C to T) could enhance its transcription activity considerably. | 3 | Network pharmacology used to decode potential active ingredients in Ferula assafoetida and mechanisms for the application to Alzheimer’s disease显示文摘Background:Alzheimer’s disease(AD)is a neurodegenerative disease that primarily manifests as progressive memory loss and cognitive impairment.Traditional herbal medicines may be helpful in the discovery of new anti-AD drugs.Studies have shown that Ferula assafoetida has neuroprotective and memory-enhancing effects,which may be beneficial for the treatment of AD.However,the combination of active ingredients and their mechanisms remain unclear.Therefore,we aimed to identify potential active ingredients in F.assafoetida and their mechanisms of action against AD by using network pharmacology.Methods:In our study,an integrated network pharmacological approach,that included adsorption,distribution,metabolism and excretion screening,target identification,network construction,topological analysis,gene ontology and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis,and molecular docking,was used to predict the pharmacological material basis and potential mechanisms through which these ingredients may treat and prevent AD.Results:The results indicated that 12 key active ingredients,obtained by topological analysis(including farnesiferol a,conferol,farnesiferol b,ferulic acid,etc.),may be the primary pharmacological components that may ameliorate AD.The 2 key significant pathways identified are the cholinergic synapse signaling pathway(critical targets include ACHE,CHRM1,CHRM2,MAPK1,PIK3CA,PIK3CB,PIK3CD,and PIK3CG)and the AD signaling pathway(critical targets include APP,BACE1,GSK3B,MAPK1,NCSTN,NOS1,PSEN1).These critical targets are closely related to the regulation of three typical pathological features of AD[central nervous system(CNS)cholinergic hypofunction,amyloid-β(Aβ)plaques,and hyperphosphorylated tau proteins].Finally,14 critical targets in the 2 key significant pathways were validated by molecular docking analysis.Conclusion:F.assafoetida may be effective for alleviating AD symptoms,through multi-component,multi-target,and multi-pathway synergistic effects,associated with the multiple pathogenesis hypotheses of AD.Our study may provide certain clues for the further development and utilization of this natural herbal medicine. | Meng Xu Lili Zhang Pin Li Chunguo Wang Yuanyuan Shi | 2020 | Journal of Traditional Chinese Medical Sciences2020,7,2: | 1 |

| 5 | Enhancing the cycling stability of Na-ion batteries by bonding MoS2 on assembled carbon-based materials显示文摘Room temperature Na-ion batteries(SIBs) show great potential for use as renewable energy storage systems.However, the large-scale application of SIBs has been hindered by the lack of an ideal SIBs anode material. We synthesized MoS2 on carbonized graphene-chitosan(G-C) using the hydrothermal method. The strong interaction between the MoS2 and the G-C greatly improved the electron transport rate and maintained the structural stability of the electrode, which lead to both an excellent rate capability and long cycle stability. Me2SnCl2, Bu2SnCl2, Et2Sn(phen)Cl2 and Et2SnCl2 can bind to dGMP mainly via phosphate; Cp2TiCl2 binds to dGMP mainly via phosphate and N7. The binding constant for (CH3)2SnCl2 binding to phos-phate of dGMP exceeds 2.71×104. The binding constant for Cp2TiCl2 to phosphate is even greater than that of Sn(IV). Cis-platin has high affinity for both N7 and phosphate, but mainly for N7. Binding of the probe to N7 of dGMP reduces the binding affinity for phosphate of the same dGMP molecule by a factor of 5 to 6. | 10 | Polycephalomycetaceae,a new family of clavicipitoid fungi segregates from Ophiocordycipitaceae显示文摘Clavicipitoid fungi comprise three families,namely Clavicipitaceae,Cordycipitaceae,and Ophiocordycipitaceae.They are found worldwide and are specialized pathogens of invertebrate,plant and fungal hosts.Over the last decade,morphology-and phylogeny-based studies on clavicipitoid fungi have increased.The latter have revealed that Polycephalomyces,Perennicordyceps and Pleurocordyceps consistently cluster together.These genera are currently considered as members of Ophiocordycipitaceae.Nonetheless,information with regard to their diversity and ecology remains sparse.To fill this gap,we collected 29 fresh specimens from insect and fungal substrates from tropical and subtropical evergreen forests in Thailand and southwestern China.We performed detailed morphological analyses and constructed photoplates for all isolated fungi.We used extensive taxon sampling and a dataset comprising internal transcribed spacer gene region(ITS),small subunit ribosomal RNA gene region(SSU),large subunit rRNA gene region(LSU),translation elongation factor 1-alpha gene region(TEF-1α),RNA polymerase II largest subunit gene region(RPB1)and RNA polymerase II second largest subunit(RPB2)to infer order-,family and genus-level phylogenetic trees.Based on these biphasic analyses,we segregate Polycephalomyces,Perennicordyceps,and Pleurocordyceps from Ophiocordycipitaceae and introduce the new family Polycephalomycetaceae to accomodate these three genera.The majority of species in this family have a vast range of insect and fungal hosts.The sexual morph of Polycephalomycetaceae has stromatic ascomata,long stipes,thick peridium,and cylindrical secondary spores.The asexual morph is characterized by colonies on the host surface or synnemata with stipes on the host,one or two types of phialides,and cylindrical to fusiform conidia.We expand the number of taxa in the new family by introducing seven new species(Polycephalomyces albiramus,Perennicordyceps lutea,Pleurocordyceps parvicapitata,Pleurocordyceps lanceolatus,Pleurocordyceps nutansis,Pleurocordyceps heilongtanensis,Pleurocordyceps vitellina),nine new hosts,and one new combination(Perennicordyceps elaphomyceticola).The results herein hint at a high level of diversity for Polycephalomycetaceae.Future investigations focusing on obtaining additional collections and specimens from different geographical areas would help to reveal not only the extent of the group’s diversity,but also resolve its deeper phylogenetic placement. | Yuan‑Pin Xiao Yuan Bing Wang Kevin D.Hyde Gentekaki Eleni Jing‑zu Sun Yu Yang Juan Meng Hong Yu Ting‑Chi Wen | 2023 | Fungal Diversity2023,,3: | 0 |
