1Effects of Zr/Ti molar ratio in SO_4^(2-)/ZrO_(2-)TiO_2 calcined at different temperatures on its surface properties and glucose reactivity in near-critical methanol显示文摘Effects of Zr/Ti molar ratio in SO42-/ZrO2-TiO2 solid acid catalyst calcined at different temperatures on its surface properties and catalytic activity were thoroughly investigated in this paper. The physicochemical characteristics of prepared samples were determined by N2 adsorptiondesorption, XRD, NH3-TPD and XPS techniques, respectively. It was found that the crystallization temperature of the samples increased after the combination of ZrO2 and TiO2; and phase transformations from the anatase to the rutile of TiO2 species and the tetragonal to the monoclinic of ZrO2 species were effectively suppressed at higher temperature. The sample with a Zr/Ti molar ratio of 3/1 calcined at 450℃ showed the highest surface area and the most acid sites among all the tested samples. The acid site densities of samples were relatively closed to each other if they were calcined at the same temperature, however, decreased with the calcination temperature. The result indicates that the sulfur content in samples is a crucial factor to control the acid site density. Calcining the sample at 650℃ and higher temperatures resulted in a significant desorption of sulfate ion on the samples. The synthesized samples were evaluated as a potential catalyst for glucose conversion under the near-critical methanol conditions (200℃/4 MPa). The results suggested that the relatively weaker acid sites of the catalyst were more favorable for the accumulation of methyl glucosides, while the moderate acid sites were responsible for the formation of methyl levulinate. The catalytic activity for methyl levulinate production almost increases linearly with the catalyst acid site density. The catalyst deactivation is due to the loss of sulfate ion and the two catalysts with Zr/Ti molar ratios of 3/1 and 1/3 could effectively alleviate the deactivation caused by sulfate solution in the reaction medium and can be reused after calcination with the reuse rate of over 90% in terms of the methyl levulinate selectivity.Lincai Peng Junping Zhuang Lu Lin 2012Journal of Natural Gas Chemistry2012,21,2:3
2Determining antimicrobial resistance profiles and identifying novel mutations of Neisseria gonorrhoeae genomes obtained by multiplexed MinION sequencing显示文摘Gonorrhea is one of the most common sexually transmitted diseases worldwide. To cure infection and prevent transmission,timely and appropriate antimicrobial therapy is necessary. Unfortunately, Neisseria gonorrhoeae, the etiological agent of gonorrhea, has acquired nearly all known mechanisms of antimicrobial resistance(AMR), thereby compromising the efficacy of antimicrobial therapy. Treatment failure resulting from AMR has become a global public health concern. Whole-genome sequencing is an effective method to determine the AMR characteristics of N. gonorrhoeae. Compared with next-generation sequencing, the MinION sequencer(Oxford Nanopore Technologies(ONT)) has the advantages of long read length and portability. Based on a pilot study using MinION to sequence the genome of N. gonorrhoeae, we optimized the workflow of sequencing and data analysis in the current study. Here we sequenced nine isolates within one flow cell using a multiplexed sequencing strategy. After hybrid assembly with Illumina reads, nine integral circular chromosomes were obtained. By using the online tool Pathogenwatch and a BLAST-based workflow, we acquired complete AMR profiles related to seven classes of antibiotics. We also evaluated the performance of ONT-only assemblies. Most AMR determinants identified by ONT-only assemblies were the same as those identified by hybrid assemblies. Moreover, one of the nine assemblies indicated a potentially novel antimicrobial-related mutation located in mtrR which results in a frame-shift, premature stop codon, and truncated peptide.In addition, this is the first study using the MinION sequencer to obtain complete genome sequences of N. gonorrhoeae strains which are epidemic in China. This study shows that complete genome sequences and antimicrobial characteristics of N.gonorrhoeae can be obtained using the MinION sequencer in a simple and cost-effective manner, with hardly any knowledge of bioinformatics required. 4Research progress in Shigella in the postgenomic era显示文摘Gram-negative,facultative intracellular anaerobes of the genus Shigella,the principal etiologic agents of shigellosis,continue to pose a threat to public health. Shigellosis causes 1.1 million deaths with over 164 million annual cases. The Shigella spp. can be divided into four serogroups:Shigella dysenteriae,Shigella flexneri, Shigella boydii and Shigella sonnei. 6Construction, detection and microarray analysis on Shigella dysenteriae A1 IroN, ShuA single, double mutants显示文摘In this study, we constructed single mutants MTS-1, MTS-2 of IroN and ShuA gene and double mutant MTS of them in Shigella dysenteriae A1 strain 51197 by insert and absence. The functional detection of every mutant was performed at the level of culture medium and cell experiment. The gene expression profiles of the mutants and the wild-type strains under iron- enriched and iron-limited conditions were analyzed by the SD51197 whole genomic microarray. The results showed that all the mutants grew obviously less well than the wild-type strains in L broth appending iron chelator DIP. The addition of iron to the cultures can stimulate the growth of mutants back to wild-type levels. In either the experiments on the ability of intracellular multiplication or the cell-to-cell spread in HeLa and U937 cell lines, mutants showed no obvious change in virulence compared with the parental strain SD51197. However when DIP was added to the cultured HeLa cells, the ability of intracellular multiplication of MTS-1, MTS-2, MTS has reduced about 23.4%, 25.2%, 43.6% respectively. The analysis of expression profiles under the iron-limited condition showed that the mutants were more sensitive for the changes of iron deficiency than the wild-type strains, many genes have been altered. Up-regulated genes mainly involved genes of transcription, coenzyme metabolism, amino acid transport and metabolism, and unknown functional genes, while down-regulated genes mainly involved genes of energy and carbohydrate metabolism and unknown function genes; the expression levels of known iron-transport associated genes generally showed up-regulated. 8Using Micro-Level Data to Evaluate Infrastructure of the Walking Environment Around Bus Rapid Transit Stations:A Case Study of Xiamen,China显示文摘China has experienced rapid car-oriented suburbanization during the past decades and is facing emerging challenges such as traffic congestion,air pollution,and even health threats.Learning from North American cities,Chinese cities have started adopting and implementing a Transit-Oriented Development(TOD) strategy,which encourages the transit ridership and walking trips by creating a pedestrian-friendly urban environment.Although much research has examined the walking environment in various neighborhoods or communities,few existing studies have examined the walking environment around transit stations,especially using micro-level data,such as walking infrastructure variables.This research chooses several infrastructure variables that are critical for pedestrians to investigate the walking environment around stations of the Bus Rapid Transit(BRT) No.1 Line in Xiamen City,China.This study defines a half mile buffer centered on each station as the spatial unit of analysis.The Geographic Information System(GIS) and field audits are employed to measure the characteristics of the infrastructure for pedestrians in each spatial unit of analysis.The results of this analysis can help assess the existing conditions of the infrastructure for pedestrians in each station area,which might need to be added or improved.The findings will not only provide empirical support for improving TODs in Xiamen,but also offer a lesson on how areas around transit stations could be retrofitted to improve the pedestrian environment.Xu Junping Wang Peng Kenneth Joh Bi Bo 2016China City Planning Review2016,25,2:1

12A multiplex method for detection of SARS-CoV-2 variants based on MALDI-TOF mass spectrometry显示文摘The recent outbreak of the coronavirus disease 2019(COVID-19)pandemic and the continuous evolution of the severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)have highlighted the significance of new detection methods for global monitoring and prevention.Although quantitative reverse transcription PCR(RT-qPCR),the current gold standard for diagnosis,performs excellently in genetic testing,its multiplexing capability is limited because of the signal crosstalk of various fluorophores.Herein,we present a highly effi-cient platform which combines 17-plex assays with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF MS),enabling the targeting of 14 different mutation sites of the spike gene.Diagnosis using a set of 324 nasopharyngeal swabs or sputum clinical samples with SARS-CoV-2 MS method was identical to that with the RT-qPCR.The detection consistency of mutation sites was 97.9%(47/48)com-pared to Sanger sequencing without cross-reaction with other respiratory-related pathogens.Therefore,the MS method is highly potent to track and assess SARS-CoV-2 changes in a timely manner,thereby aiding the continuous response to viral variation and prevention of further transmission.Ziyuan Zhao Liying Sun Liqin Wang Xiaodong Li Junping Peng 2023Biosafety and Health2023,5,2:0
13Nanoscale superconductivity of ?-Ga islands grown by molecular beam epitaxy显示文摘We report on the preparation and superconductivity of metastable γ-Ga islands on Si(111) substrate. The Ga grows in a typical Volmer-Weber mode at a low temperature of 110 K, resulting in formation of Ga nanoislands at various sizes with the identical γ-phase. In-situ low temperature scanning tunneling spectra reveal quantized electronic states in ultrathin Ga islands. It is found that both the lateral size and thickness of the Ga islands strongly suppress the superconductivity. 15The first principles investigation of ferrite magnetic response with mismatch stress显示文摘The quasi-ferrite model is proposed and an appropriate PBE exchange functional with the spin density functional theory(SDFT) is selected for the calculation of the relation between magnetic moment and residual stress in ferrite using a quantum mechanics code. The relationship between ferrite magnetism and the carbon content is determined,and then a ferrite interstitial solid solution(ISS) model in a low carbon concentration state is replaced with an α- Fe model in the case of majority magnetic calculation. The band structure of the loaded-Fe is compared with that of the unloaded α-Fe. The comparison shows that the energy of Fe atomic 3d orbital changes a little,while the energy of electron orbital of iron core below 3d almost keeps unchanged. The relationship between the magnetic moment and the stress appears intermittent due to the Bragg total reflection.
