1Application and prospects of butylphthalide for the treatment of neurologic diseases显示文摘Objective: The 3-N-butylphthalide (NBP) comprises one of the chemical constituents of celery oil. It has a series of pharmacologic mechanisms including reconstructing microcirculation, protecting mitochondrial function, inhibiting oxidative stress, inhibiting neuronal apoptosis, etc. Based on the complex multi-targets of pharmacologic mechanisms of NBP, the clinical application of NBP is increasing and more clinical researches and animal experiments are also focused on NBP. The aim of this review was to comprehensively and systematically summarize the application of NBP on neurologic diseases and briefly summarize its application to non-neurologic diseases. Moreover, recent progress in experimental models of NBP on animals was summarized. Data sources: Literature was collected from PubMed and Wangfang database until November 2018, using the search terms including '3-N-butylphthalide,''microcirculation,''mitochondria,''ischemic stroke,''Alzheimer disease,''vascular dementia,''Parkinson disease,''brain edema,''CO poisoning,''traumatic central nervous system injury,''autoimmune disease,''amyotrophic lateral sclerosis,''seizures,''diabetes,''diabetic cataract,' and 'atherosclerosis.' Study selection: Literature was mainly derived from English articles or articles that could be obtained with English abstracts and partly derived from Chinese articles. Article type was not limited. References were also identified from the bibliographies of identified articles and the authors’ files. Results: NBP has become an important adjunct for ischemic stroke. In vascular dementia, the clinical application of NBP to treat severe cognitive dysfunction syndrome caused by the hypoperfusion of brain tissue during cerebrovascular disease is also increasing. Evidence also suggests that NBP has a therapeutic effect for neurodegenerative diseases. Many animal experiments have found that it can also improve symptoms in other neurologic diseases such as epilepsy, cerebral edema, and decreased cognitive function caused by severe acute carbon monoxide poisoning. Moreover, NBP has therapeutic effects for diabetes, diabetes-induced cataracts, and non-neurologic diseases such as atherosclerosis. Mechanistically, NBP mainly improves microcirculation and protects mitochondria. Its broad pharmacologic effects also include inhibiting oxidative stress, nerve cell apoptosis, inflammatory responses, and anti-platelet and anti-thrombotic effects. 2Deep carbon cycles constrained by a large-scale mantle Mg isotope anomaly in eastern China显示文摘Although deep carbon recycling plays an important role in the atmospheric CO_2 budget and climate changes through geological time,the precise mechanisms remain poorly understood.Since recycled sedimentary carbonate through plate subduction is the main light-δ^(26)Mg reservoir within deep-Earth,Mg isotope variation in mantle-derived melts provides a novel perspective when investigating deep carbon cycling.Here,we show that the Late Cretaceous and Cenozoic continental basalts from 13 regions covering the whole of eastern China have low δ^(26)Mg isotopic compositions,while the Early Cretaceous basalts from the same area and the island arc basalts from circum-Paciic subduction zones have mantle-like or heavy Mg isotopic characteristics.hus,a large-scale mantle low δ^(26)Mg anomaly in eastern China has been delineated,suggesting the contribution of sedimentary carbonates recycled into the upper mantle,but limited into the lower mantle.his large-scale spatial and temporal variation of Mg isotopes in the mantle places severe constraints on deep carbon recycling via oceanic subduction.Shu-Guang Li Wei Yang Shah Ke Xunan Meng Hengci Tian Lijuan Xu Yongsheng He Jian Huang Xuan-Ce Wang Qunke Xia Weidong Sun Xiaoyong Yang Zhong-Yuan Ren Haiquan Wei Yongsheng Liu Fancong Meng Jun Yan 2017National Science Review2017,4,1:23

6Virus-induced accumulation of intracellular bile acids activates the TGR5-β-arrestin-SRC axis to enable innate antiviral immunity显示文摘The mechanisms on metabolic regulation of immune responses are still elusive. We show here that viral infection induces immediate-early NF-kB activation independent of viral nucleic acid-triggered signaling, which triggers a rapid transcriptional induction of bile acid (BA) transporter and rate-limiting biosynthesis enzymes as well as accumulation of intracellular BAs in divergent cell types. The accumulated intracellular BAs activate SRC kinase via the TGR5-GRK-卩-arrestin axis, which mediates tyrosine phosphorylation of multiple antiviral signaling components including RIG-I, VISA/MAVS, MITA/STING, TBK1 and IRF3. The tyrosine phosphorylation of these components by SRC conditions for efficient innate antiviral immune response. Consistently, TGR5 deficiency impairs innate antiviral immunity, whereas BAs exhibit potent antiviral activity in wild-type but not TGR5-deficient cells and mice. 7Positive effects of tree species diversity on litterfall quantity and quality along a secondary successional chronosequence in a subtropical forest显示文摘Aims Litterfall,as an important link between aboveground and belowground processes,plays a key role in forest ecosystems.Here,we test for effects of tree species richness on litter production and litter quality in subtropical forest.The study further encompasses a factorial gradient of secondary succession that resulted from human exploitation.Given that a large percentage of subtropical forests are in secondary successional stages,understanding the role of biodiversity on forest re-growth after disturbance appears critical.Methods From January 2009 to December 2014,we monitored forest litterfall in 27 Comparative Study Plots that spanned a gradient of tree species richness(3-20 species)and secondary successional ages(~20 to 120 years)in Gutianshan Natural Nature Reserve,Zhejiang Province,China.The experiment is part of the biodiversity-ecosystem functioning research platform‘BEF-China’.Tree litterfall was collected in monthly intervals using litter traps.Samples were separated into leaf and non-leaf components.Leaf litter was further sorted into dominant and other species.Community level monthly leaf litter C and N contents were analysed through a full year.General linear mixed-effects models were applied to test for effects of tree species richness and successional age on litter quantity and leaf litter C/N.Important Findings Litterfall increased with species richness among and within successional age and this effect was consistent across years.Successionally older stands had higher litterfall and this effect was related to increased tree species richness.However,species richness did not change the intra-and inter-annual temporal stability of litterfall.Increasing tree species richness increased leaf litter quality(decreased C/N),while successional age had no effect.Our study indicates that more diverse forest stands produce more leaf litter and that this litter has higher N concentrations,which could promote forest growth through accelerated nutrient re-cycling.Yuanyuan Huang Yinlei Ma Ke Zhao Pascal A.Niklaus Bernhard Schmid Jin-Sheng He 2017Journal of Plant Ecology2017,10,1:18
8Curcumin inhibits hepatitis B virus infection by downregulating ccc DNA-bound histone acetylation显示文摘AIM To investigate the potential effect of curcumin on hepatitis B virus(HBV) covalently closed circular DNA(ccc DNA) and the underlying mechanism.METHODS A Hep G2.2.15 cell line stably transfected with HBV was treated with curcumin, and HBV surface antigen(HBs Ag) and e antigen(HBe Ag) expression levels were assessed by ELISA. Intracellular HBV DNA replication intermediates and ccc DNA were detected by Southern blot and real-time PCR, respectively. The acetylation levels of histones H3 and H4 were measured by Western blot. H3/H4-bound ccc DNA was detected by chromatin immunoprecipitation(Ch IP) assays. The deacetylase inhibitors trichostatin A and sodium butyrate were used to study the mechanism of action for curcumin. Additionally, short interfering RNAs(si RNAs) targeting HBV were tested along with curcumin.RESULTS Curcumin treatment led to time-and dose-dependent reductions in HBs Ag and HBe Ag expression and significant reductions in intracellular HBV DNA replication intermediates and HBV ccc DNA. After treatment with 20 μmol/L curcumin for 2 d, HBs Ag and ccc DNA levels in Hep G2.2.15 cells were reduced by up to 57.7%(P < 0.01) and 75.5%(P < 0.01), respectively, compared with levels in non-treated cells. Meanwhile, time-and dose-dependent reductions in the histone H3 acetylation levels were also detected upon treatment with curcumin, accompanied by reductions in H3-and H4-bound ccc DNA. Furthermore, the deacetylase inhibitors trichostatin A and sodium butyrate could block the effects of curcumin. 12Characteristics and pathological mechanism on magnetic resonance diffusion-weighted imaging after chemoembolization in rabbit liver VX-2 tumor model显示文摘AIM: To investigate dynamic characteristics and pathological mechanism of signal in rabbit VX-2 tumor model on diffusion-weighted imaging (DWI) after chemoembolization. METHODS: Forty New Zealand rabbits were included in the study and forty-seven rabbit VX-2 tumor models were raised by implanting directly and intrahepatically after abdominal cavity opened. Forty VX-2 tumor models from them were divided into four groups. DWI was performed periodically and respectively for each group after chemoembolization. All VX-2 tumor samples of each group were studied by pathology. The distinction of VX-2 tumors on DWI was assessed by their apparent diffusion coefficient (ADC) values. The statistical significance between different time groups, different area groups or different b-value groups was calculated by using SPSS12.0 software. RESULTS: Under b-value of 100 s/mm2, ADC values were lowest at 16 h after chemoembolization in area of VX-2 tumor periphery, central, and normal liver parenchyma around tumor, but turned to increase with further elongation of chemoembolization treatment. The distinction of ADC between different time groups was significant respectively (F = 7.325, P < 0.001; F = 2.496, P < 0.048; F = 6.856, P < 0.001). Cellular edema in the area of VX-2 tumor periphery or normal liver parenchyma around tumor, increased quickly in sixteen h after chemoembolization but, from the 16th h to the 48th h, cellular edema in the area of normal liver parenchyma around tumor decreased gradually and that in the area of VX-2 tumor periphery decreased lightly at, and then increased continually. After chemoembolization, Cellular necrosis in the area of VX-2 tumor periphery was more significantly high than that before chemoembolization. The areas of dead cells in VX-2 tumors manifested low signal and high ADC value, while the areas of viable cells manifested high signal and low ADC value. CONCLUSION: DWI is able to detect and differentiate tumor necrotic areas from viable cellular areas before and after chemoembolization. 13Projected drought conditions in Northwest China with CMIP6 models under combined SSPs and RCPs for 2015—2099显示文摘Northwest China is one of the most arid regions in the world and has experienced intriguing climate warming and humidification.Nonetheless,future climate conditions in Northwest China still remain uncertain.In this study,we applied an ensemble of the 12 latest model simulations of the Coupled Model Intercomparison Project Phase 6(CMIP6)to assess future drought conditions until 2099 in Northwest China,as inferred from the Palmer Drought Severity Index(PDSI).Future drought conditions were projected under three climate change scenarios through the combination of shared socioeconomic pathways(SSPs)and representative concentration pathways(RCPs),namely,SSP126(SSP1+RCP2.6,a green development pathway),SSP245(SSP2+RCP4.5,an intermediate development pathway),and SSP585(SSP5+RCP8.5,a high development pathway).For 2015-2099,drought severity showed no trend under SSP126,in contrast,for the SSP245 and SSP585 scenarios,a rapid increase during 2015-2099 was observed,especially under SSP585.We also found that the drought frequency in Northwest China under SSP585 was generally lower than that under SSP126 and SSP245,although the drought duration under SSP585 tended to be longer.These findings suggest the green development pathway in drought mitigation and adaptation strategies in Northwest China,an arid and agricultural region along the Silk Road.LI Su-Yuan MIAO Li-Juan JIANG Zhi-Hong WANG Guo-Jie GNYAWALI Kaushal Raj ZHANG Jing ZHANG Hui FANG Ke HE Yu LI Chun 2020Advances in Climate Change Research2020,11,3:14
14Precision screening for esophageal squamous cell carcinoma in China显示文摘Esophageal squamous cell carcinoma(ESCC)is the predominant subtype of esophageal cancer in China,and this neoplasm is associated with high morbidity and mortality as well as clear geographical heterogeneity.Since primary prevention for ESCC lacks a clear intervention target,secondary prevention,also known as screening and early diagnosis and early treatment,has become the mainstay of ESCC prevention and control in China.ESCC screening in China has been subject to decades of evaluation and practice.However,the ESCC screening strategy currently adopted in China has encountered a developmental bottleneck.In this review,we have summarized studies and significant findings for ESCC screening and proposed advancement of screening strategies as follows:1)evidence from randomized controlled trials is needed to support the effectiveness and health economic value of endoscopic screening for ESCC;2)the current traditional screening and surveillance strategies warrant reform,and a risk-prediction-based precision strategy should be established;and 3)a deeper understanding of the value of opportunistic screening in the prevention and control of ESCC in China is called for.Due to the low absolute prevalence of precancerous lesions,substantial investment of resources and nonnegligible risks of invasive screening techniques,precision and individualization should be the main direction of cancer screening programs for the future.We advocate cooperation on the part of Chinese scientists to solve this major China-specific health problem in the next decades.Zhonghu He Yang Ke 2020Chinese Journal of Cancer Research2020,32,6:14
15Differential mercury volatilization by tobacco organs expressing a modi-fied bacterial merA gene显示文摘Mercury pollution is a major environmental problem accompanying industrial activities. Most of the mercury released ends up and retained in the soil as complexes of the toxic ionic mercury (Hg2+), which then can be converted by microbes into the even more toxic methylmercury which tends to bioaccumulate. Mercury detoxification of the soil can also occur by microbes converting the ionic mercury into the least toxic metallic mercury (Hg0) form, which then evaporates. The remediation potentia1 of transgenic plants carrying the MerA gene from E. coLi encoding mercuric ion reductase could be evaluated. A modified version of the gene, optimized for plant codon preferences (merApe9, Rugh et al. 1996), was introduced into tobacco by Agrobacterium-mediated leaf disk transformation. Transgenic seeds were resistant to HgCl2 at 50 μM and some of them (10-20% ) could germinate on media containing as much as 350 μM HgCl2, while the control plants were fully inhibited or died on 50 μM HgCl2. The rate of elemental mercury evolution from Hg2+ (added as HgCl2) was 5-8 times higher for transgenic plants than the control. Mercury volatilization by isolated organs standardized for fresh weight was higher (up to 5 times) in the roots than in shoots or the leaves. The data suggest that it is the root system of the transgenic plants that volatilizes most of the reduced mercury (Hg0). It also suggests that much of the mercury need not enter the vascular system to be transported to the leaves for volatilization. 16Intracorporeal esophagojejunostomy after totally laparoscopic total gastrectomy: A single-center 7-year experience显示文摘AIM: To assess the efficacy and safety of intracorporeal esophagojejunostomy in patients undergoing laparoscopic total gastrectomy(LTG) for gastric cancer.METHODS: A retrospective review of 81 consecutive patients who underwent LTG with the same surgical team between November 2007 and July 2014 was performed. Four types of intracorporeal esophagojejunostomy using staplers or hand-sewn suturing were performed after LTG. Data on clinicopatholgoical characteristics, occurrence of complications, postoperative recovery, anastomotic time, and operation time among the surgical groups were obtained through medical records.RESULTS: The average operation time was 288.7 min, the average anastomotic time was 54.3 min, and the average estimated blood loss was 82.7 m L. There were no cases of conversion to open surgery. The first flatus was observed around 3.7 d, while the liquid diet was started, on average, from 4.9 d. The average postoperative hospital stay was 10.1 d. Postoperative complications occurred in 14 patients, nearly 17.3%.However, there were no cases of postoperative death.CONCLUSION: LTG performed with intracorporeal esophagojejunostomy using laparoscopic staplers or hand-sewn suturing is feasible and safe. 17Study on β-TCP Coated Porous Mg as a Bone Tissue Engineering Scaffold Material显示文摘Three-dimensional honeycomb-structured magnesium (Mg) scaffolds with interconnected pores of accurately controlled pore size and porosity were fabricated by laser perforation technique. Biodegradable and bioactive β-tricalcium phosphate (β-TCP) coatings were prepared on the porous Mg to further improve its biocompatibility, and the biodegradation mechanism was simply evaluated in vitro. It was found that the mechanical properties of this type of porous Mg significantly depended on its porosity. Elastic modulus and compressive strength similar to human bones could be obtained for the porous Mg with porosity of 42.6%-51%. 20Unequal-thickness billet optimization in transitional region during isothermal local loading forming of Ti-alloy rib-web component using response surface method显示文摘Avoiding the folding defect and improving the die filling capability in the transitional region are desired in isothermal local loading forming of a large-scale Ti-alloy rib-web component(LTRC). To achieve a high-precision LTRC, the folding evolution and die filling process in the transitional region were investigated by 3 D finite element simulation and experiment using an equal-thickness billet(ETB). It is found that the initial volume distribution in the second-loading region can greatly affect the amount of material transferred into the first-loading region during the second-loading step, and thus lead to the folding defect. Besides, an improper initial volume distribution results in non-concurrent die filling in the cavities of ribs after the second-loading step, and then causes die underfilling. To this end, an unequal-thickness billet(UTB) was employed with the initial volume distribution optimized by the response surface method(RSM). For a certain eigenstructure, the critical value of the percentage of transferred material determined by the ETB was taken as a constraint condition for avoiding the folding defect in the UTB optimization process,and the die underfilling rate was considered as the optimization objective. Then, based on the RSM models of the percentage of transferred material and the die underfilling rate, non-folding parameter combinations and optimum die filling were achieved.
