1Canopy Temperature Depression as a Potential Selection Criterion for Drought Resistance in Wheat显示文摘Field studies were conducted at Bushland, Texas, USA, in 2004 to examine usefulness of canopy temperature depression(CTD), the difference of air-canopy temperature, in screening wheat (Triticum aestivum L.) genotypes for yield underdryland and irrigated. Forty winter wheat genotypes were grown under irrigation and dryland. CTDs were recorded afterheading between 1 330 and 1 530 h on 6 clear days for dryland and 9 days for irrigation. Drought susceptible index (DSI)for each genotype was calculated using mean yield under dryland and irrigated conditions. Genotypes exhibited greatdifferences in CTD under each environment. The dryland CTDs averaged 1.33°C ranging from -0.67 to 2.57°C, and theaverage irrigation CTD were 4.59°C ranging from 3.21 to 5.62°C. A low yield reduction was observed under drylandconditions relative to irrigated conditions for high-CTD genotypes. CTD values were highly negatively correlated withDSI under dryland, and genotypes of CTDs = 1.3°C in dryland condition were identified as drought resistant. For 21genotypes classified as drought resistant by DSI, their CTDs were 1.68°C for dryland and 4.35°C for irrigation on average;for 19 genotypes classified as drought susceptible by DSI, average CTD was 0.94°C in dryland and 4.85°C in irrigation.The high-yield genotypes consistently had high CTD values, and the low-yield ones had low CTD values for allmeasurements in dryland. After heading, genotypes maintained consistent ranking for CTD. Regression results for CTDand yield suggested that the best time for taking CTD measurement was 3-4 weeks after heading in irrigation but any timebefore senescence in dryland. Crop water stress index (CWSI) calculated from CTD data was highly correlated with CWSIcalculated from yield, which suggesting traditional costly CWSI measurement may be improved by using portable infraredthermometers. Most importantly, grain yield was highly correlated with CTD under dryland (R2 = 0.79-0.86) and irrigation(R2 = 0.46-0.58) conditions. 2Bile acids as endogenous etiologic agents in gastrointestinal cancer显示文摘Bile acids are implicated as etiologic agents in cancer of the gastrointestinal (GI) tract, including cancer of the esophagus, stomach, small intestine, liver, biliary tract, pancreas and colon/rectum. Deleterious effects of bile acid exposure, likely related to carcinogenesis, include: induction of reactive oxygen and reactive nitrogen species; induction of DNA damage; stimulation of mutation; induction of apoptosis in the short term, and selection for apoptosis resistance in the long term. These deleterious effects have, so far, been reported most consistently in relation to esophageal and colorectal cancer, but also to some extent in relation to cancer of other organs. In addition, evidence is reviewed for an association of increased bile acid exposure with cancer risk in human populations, in specific human genetic conditions, and in animal experiments. 4Effects of Extreme Air Temperature and Humidity on the Insecticidal Expression Level of Bt Cotton显示文摘The higher survival rates of Helicoverpa amigera larvae were usually observed after adverse climate which was related to extreme temperature(T) and relative humidity(RH) stresses in transgenic Bacillus thuringiensis(Bt) cotton.The unstable resistance of Bt cotton to bollworms has been correlated with the reduced expression of CryIAc δ-endotoxin.The objective of this study was to investigate the effects of combined temperature and relative humidity stresses on the leaf CryIAc insecticidal protein expression during critical developmental stages.The study was undertaken on two transgenic cotton cultivars that share same parental background,Sikang 1(a conventional cultivar) and Sikang 3(a hybrid cultivar),during the 2007 and 2008 growing seasons at the Yangzhou University Farm,Yangzhou,China.The study was arranged with two factors that consisted of temperature(two levels) and relative humidity(three levels).The six T/RH treatments were 37°C/95%,37°C/70%,37°C/50%,18°C/95%,18°C/70%,and 18°C/50%.In 2007,the six treatments were imposed to the plants at peak flowering stage for 24 h;in 2008,the six treatments were applied to the plants at peak square,peak flowering,and peak boll stages for 48 h.The results of the study indicated that the leaf insecticidal protein expression in CryIAc was significantly affected by extreme temperature only at peak flowering stage,and by both extreme temperature and relative humidity during boll filling stage.The greatest reductions were observed when the stresses were applied at peak boll stage.In 2008,after 48 h stress treatment,the leaf Bt endotoxin expression reduced by 25.9-36.7 and 23.6-40.5% at peak boll stage,but only by 14.9-26.5 and 12.8-24.0% at peak flowering stage for Sikang 1 and Sikang 3,respectively.The greatest reduction was found under the low temperature combined with low relative humidity condition for both years.It is believed that the temperature and relative humidity stresses may be attributed to the reduced efficacy of Bt cotton in growing conditions in China,where extreme temperatures often increase up to 35-40°C and/or decrease down to 15-20°C,and relative humidity may reach to 85-95% and/or reduce to 40-55% during the cotton growing season.CHENYuan WEN Yu-jin CHEN Yuan John Tom Cothren ZHANG Xiang WANG Yong-hui WilliamA Payne CHEN De-hua 2012Journal of Integrative Agriculture2012,11,11:10

8Cancer and age related colonic crypt deficiencies in cytochrome c oxidase Ⅰ显示文摘AIM: To investigate whether defi ciency of expressionof cytochrome c oxidase I (CcOI) in colonic crypts is associated with colon cancer.METHODS: The pattern and level of expression of CcOI in non-neoplastic colonic crypts,and in dysplastic tissues,was assessed using standard immunohis-tochemical methods.Biopsies were obtained from individuals undergoing colonoscopies for screening purposes or for a medically indicated reason.Tissue samples were also obtained from surgical colonic resections.Samples from resections were taken from colonic mucosa 1 and 10 cm from tumors and from the tumors themselves.Samples were evaluated for frequency of crypts with reduced or absent expression of CcOI.In most crypts the loss was apparent throughout the entire crypt,while in a small minority the loss was segmental.The strong immunoreactivity using this monoclonal antibody makes the scoring unambiguous.The percent of crypts with reduced or absent expression of CcOI or (infrequent) segmented loss of expression was then calculated.Data analyses were performed using SPSS statistical package 17.0.RESULTS: The average frequency of CcOI deficient crypts (CcOI-DC) is low in individuals between 20 and 39 years of age,with 0.48% ± 0.40% CcOI-DC for women and 1.80% ± 0.35% for men.CcOI-DC increases after age 40 years,so that between the ages of 40 and 44 years the average frequency of CcOI- DC goes up to 5.89% ± 0.84% in women and 2.15% ± 1.27% in men.By 80-84 years of age,the average frequency of CcOI-DC goes up in women to 15.77% ± 0.97% and in men to 22.6% ± 0.65%.The increases in CcOI-DC from ages 40-44 years compared to 80-84 years in women and men are significantly different with P < 0.01.For women over age 60 years,deficiency of CcOI expression is greater in those women who have had a cancer in their colon.The frequency of CcOI-DC,measured in men,increased in tissues adjacent to colon cancer,being 4.03% ± 0.27% in individuals free of neoplasia in the age range 55-64 yearsand 14.13% ± 0.35% in resected histologically normal tissue of men with cancer in the same age range,P < 0.001.Similar signifi cant differences were noted in older age ranges.The frequency of CcOI-DC crypts in the cecum and sigmoid colon of an individual are signifi cantly correlated,with an R2 = 0.414 for women and R2 = 0.528 for men,P < 0.001.This suggests that the factors determining the level of CcOI deficiency act throughout the colon.Most defective crypts are in clusters of two or more,a likely consequence of crypt fission.In the non-neoplastic margins of cancers,crypts are frequently defi cient for CcOI,and such crypts may appear in large clusters,some containing more than 100 defi cient crypts.CcOI defi ciency is also apparent in colon cancers and sometimes involves a large section of the tumor.Overall,CcOI deficient cells can be visualized in segments of crypts,in whole crypts that increase in frequency with age,in crypts undergoing f ission,in clusters of crypts where the clusters increase in size with age,in increased frequency near tumors,in large clusters in the intimate margins of tumors,and in the tumors themselves.There is no clear dividing line between early stages that can be considered aspects of aging and later stages that can be considered aspects of the progression to cancer.This ambiguity may re ect a rather general situation leading to adult cancer where the early stages of cellular change appear to be relatively innocuous features of the aging process but over decades may evolve into malignancy.CONCLUSION: CcOI defi cient crypts increase in frequency with age,and clusters of defi cient crypts are associated with,and may give rise to,colon cancer.Carol Bernstein Alexander Facista Huy Nguyen Beryl Zaitlin Nadia Hassounah Cristy Loustaunau Claire Margaret Payne Bhaskar Banerjee Steve Goldschmid V Liana Tsikitis Robert Krouse Harris Bernstein 2010World Journal of Gastrointestinal Oncology2010,2,12:5

12New risk assessment methodology for coal mine excavated slopes显示文摘This paper presents a new risk assessment methodology for coal mine excavated slopes. This new empirical-statistical slope stability assessment methodology(SSAM) is intended for use by geotechnical engineers at both the design review and operational stages of a mine's life to categorise the risk of an excavated coal mine slope. A likelihood of failure is determined using a new slope stability classification system for excavated coal mine slopes developed using a database of 119 intact and failed case studies sourced from open cut coal mines in Australia. Consequence of failure is based on slope height and stand-off distance at the toe of the excavated slope. Results are presented in a new risk matrix, with slope risk being divided into low, medium and high categories. The SSAM is put forward as a new risk assessment methodology to assess the potential for, and consequence of, excavated coal mine slope failure.Unlike existing classification systems, assumptions about the likely failure mode or mechanism are not required. Instead, the SSAM applies an approach which compares the conditions present within the excavated slope face, with the known past performance of slopes with similar geotechnical and geometrical conditions, to estimate the slope's propensity for failure. The SSAM is novel in that it considers the depositional history of strata in an excavated slope and how this sequence affects slope stability. It is further novel in that it does not require explicit measurements of intact rock, rock mass and/or defect strength to rapidly calculate a slope's likelihood of failure and overall risk. Ratings can be determined entirely from visual observations of the excavated slope face.
