| 1 | Tree-ring precipitation records from Baiyinaobao, Inner Mongolia since A.D. 1838显示文摘Correlation function analysis shows that total precipitation from April to early July (to July 10) in growing season limits the ring width of Picea Koraiensis significantly in Baiyinaobao, Xilinhot, Inner Mongolia. Thus a transfer function is designed to reconstruct the total precipitation from April to early July over the region for the last 160 years. The explained variance of the reconstruction is 49.3% (and 45.7% when adjusted for loss of degrees of freedom, N = 31, r = 0.702, F = 13.608, p < 0.0001). There are three relatively wet periods in the reconstruction which are: 1869—1875, 18861921 and 19431968, and four relatively dry periods: 1851—1868, 1876—1885, 1922—1943 and 1969—1999. Among them, the wet periods of 1886—1921 and 1943—1968, and the dry period of 1922—1943 appeared almost at the same time with those in Mt. Qilian region. Meanwhile, the dry period of 1922—1943 corresponds to high temperature and low rainfall during the 1920s and thereafter, and wetness of 1943—1968 corresponds very well to low temperature and more precipitation after the 1940s in Northwest, Northern China and the Changjiang River drainage basin. Calculation shows that the reconstruction series is significantly correlated with local dry-wet index (r = -0.234, p < 0.007, N = 143). Wavelet analysis indicates that there is a 22-a cycle in the precipitation of April to early July during 18381920, and an 11-a cycle during 1920—1952. No more cycles that reach 95% confidence limits are identified after 1953. The precipitation from April to early July quickly dropped into low during the 1920s. | 4 | Effect of acupuncture at Renying(ST 9) on gene expression profile of hypothalamus in spontaneously hypertensive rats显示文摘OBJECTIVE: To investigate changes in gene expression profiles in the hypothalamus related to the effects of acupuncture at the Renying(ST 9) acupoint in spontaneously hypertensive(SH) rats.METHODS: We randomly divided 18 SH rats into Renying(ST 9) group and model control group, 9 body weight-matched Wistar-Kyoto rats were used as blank controls. Acupuncture was performed manually for 20-min daily over 28 d in the Renying(ST 9) group. Rat Gene 2.0 array technology was used for the determination of gene expression profiles and the screened key genes were verified by real-time quantitative polymerase chain reaction(RT-PCR) analyses.RESULTS: The different groups exhibited differential gene expression: compared with the blank control group, 48 genes were up-regulated and 91 genes were down-regulated in the model group;compared with the model group, 79 genes were up-regulated and 80 genes were down-regulated in Renying(ST 9) group. The RT-PCR results of the key genes including Chi3 l1, Ephx2, Klk1, 5-HT1 A and Cbs were consistent with that of gene chip analysis.CONCLUTION: Acupuncture at Renying(ST 9)could significantly lower the blood pressure of SH rats and affect their hypothalamic gene expression profile. | 5 | Ipomoea batatas HKT1 transporter homolog mediates K^+ and Na^+ uptake in Saccharomyces cerevisiae显示文摘Soil salinity causes the negative effects on the growth and yield of crops. In this study,two sweet potato(Ipomoea batatas L.) cultivars,Xushu 28(X-28) and Okinawa 100(O-100),were examined under 50 and 100 mmol L^(–1) Na Cl stress. X-28 cultivar is relatively high salt tolerant than O-100 cultivar. Interestingly,real-time quantitative polymerase chain reaction(RT-q PCR) results indicated that sweet potato high-affinity K^+ transporter 1(IbHKT1) gene expression was highly induced by 50 and 100 mmol L^(–1) Na Cl stress in the stems of X-28 cultivar than in those of O-100 cultivar,but only slightly induced by these stresses in the leaves and fibrous roots in both cultivars. To characterize the function of IbHKT1 transporter,we performed ion-flux analysis in tobacco transient system and yeast complementation. Tobacco transient assay showed that IbHKT1 could uptake sodium(Na^+). Yeast complementation assay showed that IbHKT1 could take up K^+ in 50 mmol L^(–1) K^+ medium without the presence of Na Cl. Moreover,Na^+ uptake significantly increased in yeast overexpressing IbHKT1. These results showed that IbHKT1 transporter could have K^+-Na^+ symport function in yeast. Therefore,the modes of action of IbHKT1 in transgenic yeast could differ from the mode of action of the other HKT1 transporters in class I. | 6 | Risk factors of electrocoagulation syndrome after esophageal endoscopic submucosal dissection显示文摘AIM To investigate post endoscopic submucosal dissection electrocoagulation syndrome(PEECS) of the esophagus.METHODS We analyzed 55 consecutive cases with esophageal endoscopic submucosal dissection for superficial esophageal squamous neoplasms at a tertiary referral hospital in South Korea. Esophageal PEECS was defined as 'mild' meeting one of the following criteria without any obvious perforation: fever(≥ 37.8 ℃), leukocytosis(> 10800 cells/μl), or regional chest pain more than 5/10 points as rated on a numeric pain intensity scale. The grade of PEECS was determined as 'severe' when meet two or more of above criteria.RESULTS We included 51 cases without obvious complications in the analysis. The incidence of mild and severe esophageal PEECS was 47.1% and 17.6%, respectively. Risk factor analysis revealed that resected area, procedure time, and muscle layer exposure were significantly associated with PEECS. In multivariate analysis, a resected area larger than 6.0 cm^2(OR = 4.995, 95%CI: 1.110-22.489, P = 0.036) and muscle layer exposure(OR = 5.661, 95%CI: 1.422-22.534, P = 0.014) were independent predictors of esophageal PEECS. | 8 | Dietary energy sources and levels shift the multi-kingdom microbiota and functions in the rumen of lactating dairy cows显示文摘Background: Dietary energy source and level in lactation diets can profoundly affect milk yield and composition.Such dietary effects on lactation performance are underpinned by alteration of the rumen microbiota, of which bacteria, archaea, fungi, and protozoa may vary differently. However, few studies have examined all the four groups of rumen microbes. This study investigated the effect of both the level and source of dietary energy on rumen bacteria, archaea, fungi, and protozoa in the rumen of lactating dairy cows. A 2 × 2 factorial design resulted in four dietary treatments: low and high dietary energy levels(LE: 1.52–1.53;and HE: 1.71–1.72 Mcal/kg dry matter) and two dietary energy sources(GC: finely ground corn;and SFC: steam-flaked corn). We used a replicated 4 × 4 Latin square design using eight primiparous Chinese Holstein cows with each period lasting for 21 d. The rumen microbiota was analyzed using metataxonomics based on kingdom-specific phylogenetic markers [16 S r RNA gene for bacteria and archaea, 18 S r RNA gene for protozoa, and internally transcribed spacer 1(ITS1) for fungi] followed with subsequent functional prediction using PICRUSt2.Results: The GC resulted in a higher prokaryotic(bacterial and archaeal) species richness and Faith's phylogenetic diversity than SFC. For the eukaryotic(fungi and protozoa) microbiota, the LE diets led to significantly higher values of the above measurements than the HE diets. Among the major classified taxa, 23 genera across all the kingdoms differed in relative abundance between the two dietary energy levels, while only six genera(none being protozoal)were differentially abundant between the two energy sources. Based on prokaryotic amplicon sequence variants(ASVs) from all the samples, overall functional profiles predicted using PICRUSt2 differed significantly between LE and HE but not between the two energy sources. Fish Taco analysis identified Ruminococcus and Coprococcus as the taxa potentially contributing to the enriched KEGG pathways for biosynthesis of amino acids and to the metabolisms of pyruvate, glycerophospholipid, and nicotinate and nicotinamide in the rumen of HE-fed cows. The co-occurrence networks were also affected by the dietary treatments, especially the LE and GC diets, resulting in distinct co-occurrence networks. Several microbial genera appeared to be strongly correlated with one or more lactation traits.Conclusions: Dietary energy level affected the overall rumen multi-kingdom microbiota while little difference was noted between ground corn and steam-flaked corn. Some genera were also affected differently by the four dietary treatments, including genera that had been shown to be correlated with lactation performance or feed efficiency.The co-occurrence patterns among the genera exclusively found for each dietary treatment may suggest possible metabolic interactions specifically affected by the dietary treatment.
