4Achalasia: A review of clinical diagnosis, epidemiology,treatment and outcomes显示文摘Achalasia is a neurodegenerative motility disorder of the oesophagus resulting in deranged oesophageal peristalsis and loss of lower oesophageal sphincter function.Historically,annual achalasia incidence rates were believed to be low,approximately 0.5-1.2 per 100000.More recent reports suggest that annual incidence rates have risen to 1.6 per 100000 in some populations.The aetiology of achalasia is still unclear but is likely to be multi-factorial.Suggested causes include environmental or viral exposures resulting in inflammation of the oesophageal myenteric plexus,which elicits an autoimmune response.Risk of achalasia may be elevated in a sub-group of genetically susceptible people.Improvement in the diagnosis of achalasia,through the introduction of high resolution manometry with pressure topography plotting,has resulted in the development of a novel classification system for achalasia.This classification system can evaluate patient prognosis and predict responsiveness to treatment.There is currently much debate over whether pneumatic dilatation is a superior method compared to the Heller’s myotomy procedure in the treatment of achalasia.A recent com-parative study found equal efficacy,suggesting that patient preference and local expertise should guide the choice.Although achalasia is a relatively rare condition,it carries a risk of complications,including aspiration pneumonia and oesophageal cancer.The risk of both squamous cell carcinoma and adenocarcinoma of the oesophagus is believed to be significantly increased in patients with achalasia,however the absolute excess risk is small.Therefore,it is currently unknown whether a surveillance programme in achalasia patients would be effective or cost-effective.Orla M O'Neill Brian T Johnston Helen G Coleman 2013World Journal of Gastroenterology2013,19,35:26

8BH3-only proteins target BCL-xL/MCL-1,not BAX/BAK,to initiate apoptosis显示文摘It has been widely accepted that mitochondria-dependent apoptosis initiates when select BH3-only proteins(BID,BIM,etc.)directly engage and allosterically activate effector proteins BAX/BAK.Here,through reconstitution of cells lacking all eight pro-apoptotic BH3-only proteins,we demonstrate that all BH3-only proteins primarily target the anti-apoptotic BCL-2 proteins BCL-xL/MCL-1,whose simultaneous suppression enables membrane-mediated spontaneous activation of BAX/BAK.BH3-only proteins*apoptotic activities correlate with affinities for BCL-xL/MCL1 instead of abilities to directly activate BAX/BAK.Further,BID and BIM do not distinguish BAX from BAK or accelerate BAX/BAK activation following inactivation of BCL-xL/MCL-1.Remarkably,death ligandinduced apoptosis in cells lacking BH3-only proteins and MCL-1 is fully restored by BID mutants capable of neutralizing BCL-xL,but not direct activation of BAX/BAK.Taken together,our findings provide a'Membrane-mediated Permissive'model,in which the BH3-only proteins only indirectly activate BAX/BAK by neutralizing the anti-apoptotic BCL-2 proteins,and thus allowing BAX/BAK to undergo unimpeded,spontaneous activation in the mitochondrial outer membrane milieu,leading to apoptosis initiation.Kai Huang Katelyn L.O'Neill Jian Li Wei Zhou Na Han Xiaming Pang Wei Wu Lucas Struble Gloria Borgstahl Zhaorui Liu Liqiana Zhang Xu Luo 2019Cell Research2019,29,11:16

10Targeting immunometabolism as an anti-inflammatory strategy显示文摘The growing field of immunometabolism has taught us how metabolic cellular reactions and processes not only provide a means to generate ATP and biosynthetic precursors,but are also a way of controlling immunity and inflammation.Metabolic reprogramming of immune cells is essential for both inflammatory as well as anti-inflammatory responses.Four anti-inflammatory therapies,DMF,Metformin,Methotrexate and Rapamycin all work by affecting metabolism and/or regulating or mimicking endogenous metabolites with anti-inflammatory effects.Evidence is emerging for the targeting of specific metabolic events as a strategy to limit inflammation in different contexts.Here we discuss these recent developments and speculate on the prospect of targeting immunometabolism in the effort to develop novel anti-inflammatory therapeutics.As accumulating evidence for roles of an intricate and elaborate network of metabolic processes,including lipid,amino acid and nucleotide metabolism provides key focal points for developing new therapies,we here turn our attention to glycolysis and the TCA cycle to provide examples of how metabolic intermediates and enzymes can provide potential novel therapeutic targets.Eva M.Palsson-McDermott Luke A.J.O'Neill 2020Cell Research2020,30,4:12
11Mitogen-activated protein kinases mediate the oxidative burst and saponin synthesis induced by chitosan in cell cultures of Panax ginseng显示文摘Chitosan (CHN) specially induced the activities of 39 kD and 42 kD protein kinases in ginseng cells, which could be suppressed by an inhibitor of mitogen-activated protein kinase (MAPK) pathway, PD98059. The immunoprecipitation (IP) using MAPK antibody or kinase assay in vitro also showed that CHN-induced 42 kD and 39 kD protein kinases belonged to the MAPK family. PD98059 suppressed CHN-induced transcriptions of ginseng squalene synthase and ginseng squalene epoxidase genes (gss and gse), CHN-induced accumulation of b-Amyrin synthase (b-AS) and synthesis of saponin. These results showed that CHN-induced activities of MAPKs were necessary for the CHN-induced saponin synthesis. EGTA and LaCl3 suppressed CHN-induced 39 kD and 42 kD MAPK activities. Ruthenium red (RR) could suppress CHN-induced 39 kD activity. All of them suppressed CHN-induced saponin synthesis. These results indicated that CHN-induced increment of cytosolic calcium was necessary for CHN-induced saponin synthesis. PD98059 also suppressed CHN-induced oxidative burst (in-cluding the increment of activity of plasma membrane NADPH oxidase and production of H2O2), but diphenylene iodonium (DPI), dimethylthiourea (DMTU) and 2,5-dihydroxycinnamic acid methyl ester (DHC) could not suppress CHN-induced MAPK activities, which indicated that MAPK was possibly function upstream of CHN-induced oxidative burst.Steven J. 13Performance characterization of Ni60-WC coating on steel processed with supersonic laser deposition显示文摘Ni60-WC particles are used to improve the wear resistance of hard-facing steel due to their high hardness. An emerging technology that combines laser with cold spraying to deposit the hard-facing coatings is known as supersonic laser deposition. In this study, Ni60-WC is deposited on low-carbon steel using SLD. The microstructure and performance of the coatings are investigated through SEM, optical microscopy, EDS, XRD, microhardness and pin-on-disc wear tests. 16Hardfacing steel with nanostructured coatings of Stellite-6 by supersonic laser deposition显示文摘The ability to manufacture coatings is critical to engineering design. Many components require the application of additional layers toenhance mechanical properties and protect against hostile environments. Supersonic laser deposition (SLD) is a novel coating methodbased on cold spray (CS) principles. In this technique, the deposition velocities can be significantly lower than those required foreffective bonding in CS applications. The addition of laser heat energy permits a change in the thermodynamic experience of impactingparticles, thereby offering a greater opportunity for efficient bonding at lower velocities as compared with the CS process. The workreported in this paper demonstrates the ability of the SLD process to deliver hardfacing materials to engineering surfaces. Stellite-6 hasbeen deposited on low-carbon steel tubes over a range of process parameters to establish the appropriate target power and traversespeeds for the coating formation. The coating properties and parameters were examined to determine the primary characteristics andgrain structure size. Their morphology and performance were studied through optical microscopy, scanning electron microscope(SEM), X-ray diffraction, hardness measurements and wear testing. 17Macrophage inhibitory cytokine-1/growth differentiation factor-15 in premalignant and neoplastic tumours in a high-risk pancreatic cancer cohort显示文摘BACKGROUND Pancreatic cancer(PC)is a leading cause of cancer related mortality worldwide,with poor survival due to late diagnosis.Currently,biomarkers have limited use in early diagnosis of PC.Macrophage inhibitory cytokine-1 or growth differentiation factor-15(MIC-1/GDF15)has been implicated as a potential serum biomarker in PC and other malignancies.AIM To determine the role of MIC-1/GDF15 in detecting pre-malignant pancreatic lesions and neoplastic tumours in an asymptomatic high-risk cohort part of Australian Pancreatic Cancer Screening Program.METHODS A feasibility prospective single centre cohort study was performed.Participants recruited for yearly surveillance with endoscopic ultrasound(EUS)had serial fasting blood samples collected before EUS for MIC-1/GDF15,C-reactive protein and carbohydrate antigen 19-9.Patients were stratified into five groups based on EUS findings:Normal;pancreatic cysts,branch-duct intraductal papillary mucinous neoplasm;diffuse non-specific abnormalities;and neoplastic tumours.MIC-1/GDF15 serum levels were quantified using ELISA.Participants in whom EUS demonstrated abnormalities but not malignancy were closely followed up with magnetic resonance imaging(MRI)or computed tomography.RESULTS One hundred twenty participants were prospectively recruited from 2011-2018.Forty-seven participants(39.2%)had an abnormal EUS and five participants(4.2%)were diagnosed with neoplastic tumours,three by EUS(two pancreatic and one liver)and two by MRI/computed tomography(breast cancer,bladder cancer),which were performed for follow up of abnormal EUS.Baseline serum MIC-1/GDF15 was a significant predictor of neoplastic tumours on receiver operator characteristic curve analysis[area under curve(AUC)=0.814,P=0.023].Baseline serum MIC-1/GDF15 had moderate predictive capacity for branch-duct intraductal papillary mucinous neoplasm(AUC=0.644)and neoplastic tumours noted on EUS(AUC=0.793),however this was not significant(P=0.188 and 0.081 respectively).Serial serum MIC-1/GDF15 did not demonstrate a significant percentage change between a normal and abnormal EUS(P=0.213).Median baseline MIC-1/GDF15 was greater in those with neoplastic tumours(Median=1039.6,interquartile range=727.0-1977.7)compared to those diagnosed with a benign lesion(Median=570.1,interquartile range=460.7-865.2)on EUS and MRI(P=0.012).CONCLUSION In this pilot study MIC-1/GDF15 has predictive capacity for neoplastic tumours in asymptomatic individuals with a genetic predisposition for PC.Further imagining may be warranted in patients with abnormal EUS and raised serum MIC-1/GDF15.Larger multicentric prospective studies are required to further define the role of MIC-1/GDF15 as a serological biomarker in pre-malignant pancreatic lesions and neoplastic tumours.Robert Sean O’Neill Sam Emmanuel David Williams Alina Stoita 2020World Journal of Gastroenterology2020,26,14:8
18胶北粉子山群石榴云母片岩的三叠纪独居石年龄及其地质意义显示文摘胶北地块粉子山群石榴云母片岩中石榴石变斑晶内包裹物迹线明显,保留了岩石形成过程中的多期变质变形信息。电子探针成分面扫描图显示石榴石成分环带明显,可分为核部、幔部和边部。石榴石中MgO、FeO、MnO和CaO含量变化特征表明其核部到边部温度先升高后降低,对应进变质及退变质过程。根据原位独居石Y元素成分面扫描图显示,部分独居石颗粒由核部到边部Y含量呈现逐渐降低趋势,说明测得的232.6±1.1Ma^229.5±3.7Ma的独居石U-Pb年龄,对应石榴石的进变质生长过程。结合1869±72Ma的锆石U-Pb年龄数据,可推断粉子山群石榴云母片岩至少经历了古元古代及三叠纪两期变质事件的改造。粉子山群石榴云母片岩卷入了苏鲁超高压变质带的俯冲碰撞造山事件。电子探针成分分析结果表明粉子山群石榴云母片岩中的石榴石属于铁铝榴石,反映出经受中级区域变质作用的特征。说明粉子山群石榴云母片岩虽然参与了三叠纪苏鲁超高压变质带的俯冲碰撞造山过程,但俯冲深度较浅。这可用大陆俯冲过程中上盘的俯冲剥蚀来解释,并可为陆-陆碰撞俯冲剥蚀模式提出的扬子板片在240~220Ma的深俯冲作用过程中拽动胶北地块向下俯冲又折返的运动过程提供佐证,但胶北地块是否经历了深俯冲超高压变质作用,还需要进一步验证。曹汇 VERVOORT Jeff 王达 李广旭 NEILL Owen 2016岩石学报2016,32,12:7
