| 1 | MicroRNA in TLR signaling and endotoxin tolerance显示文摘Toll-like receptors(TLRs)in innate immune cells are the prime cellular sensors for microbial components.TLR activation leads to the production of proinflammatory mediators and thus TLR signaling must be properly regulated by various mechanisms to maintain homeostasis.TLR4-ligand lipopolysaccharide(LPS)-induced tolerance or cross-tolerance is one such mechanism,and it plays an important role in innate immunity.Tolerance is established and sustained by the activity of the microRNA miR-146a,which is known to target key elements of the myeloid differentiation factor 88(MyD88)signaling pathway,including IL-1 receptor-associated kinase(IRAK1),IRAK2 and tumor-necrosis factor(TNF)receptor-associated factor 6(TRAF6).In this review,we comprehensively examine the TLR signaling involved in innate immunity,with special focus on LPS-induced tolerance.The function of TLR ligand-induced microRNAs,including miR-146a,miR-155 and miR-132,in regulating inflammatory mediators,and their impact on the immune system and human diseases,are discussed.Modulation of these microRNAs may affect TLR pathway activation and help to develop therapeutics against inflammatory diseases. | Md A Nahid Minoru Satoh Edward KL Chan | 2011 | Cellular & Molecular Immunology2011,8,5: | 34 |

| 4 | Exercise preconditioning exhibits neuroprotective effects on hippocampal CA1 neuronal damage after cerebral ischemia显示文摘Recent evidence has suggested the neuroprotective effects of physical exercise on cerebral ischemic injury. However, the role of physical exercise in cerebral ischemia-induced hippocampal damage remains controversial. The aim of the present study was to evaluate the effects of pre-ischemia treadmill training on hippocampal CA1 neuronal damage after cerebral ischemia. Male adult rats were randomly divided into control, ischemia and exercise + ischemia groups. In the exercise + ischemia group, rats were subjected to running on a treadmill in a designated time schedule(5 days per week for 4 weeks). Then rats underwent cerebral ischemia induction th rough occlusion of common carotids followed by reperfusion. At 4 days after cerebral ischemia, rat learning and memory abilities were evaluated using passive avoidance memory test and rat hippocampal neuronal damage was detected using Nissl and TUNEL staining. Pre-ischemic exercise significantly reduced the number of TUNEL-positive cells and necrotic cell death in the hippocampal CA1 region as compared to the ischemia group. Moreover, pre-ischemic exercise significantly prevented ischemia-induced memory dysfunction. Clinically significant changes in central nervous system function are observed following brain ischemia and reperfusion. Stroke patients exhibit behavioral, cognitive,emotional, affective and electrophysiological changes during recovery phase. Brain injury by transient complete global brain ischemia or by transient incomplete brain ischemia afflicts a very large number of patients in the world with death or permanent disability. In order to reduce this damage, we must sufficiently understand the mechanisms involved in brain ischemia and reperfusion and repair to design clinically effective therapy.Cerebral ischemia and reperfusion is known to induce the generation of reactive oxygen species that can lead to oxidative damage of proteins, membrane lipids and nucleic acids.A decrease in tissue antioxidant capacity, an increase in lipid peroxidation as well as an increase in lipid peroxidation inhibitors have been demonstrated in several models of brain ischemia. This paper reviews the number of commonly used types of herbal medicines effective for the treatment of stroke. | 11 | Biopsy-driven diagnosis in infants with cholestatic jaundice in Iran显示文摘AIM:To determine the frequencies of diagnoses confirmed by liver biopsy in infants with cholestasis in an Iranian pediatric hospital.METHODS:This was a retrospective study conducted in a tertiary referral children’s hospital in Iran.We retrieved all pathology reports of liver biopsies from children less than two years of age who had presented for evaluation of cholestatic jaundice from March 2001to March 2011.Additional specimen samples obtained from archived pathology blocks were reviewed by a pathologist blinded to the final diagnosis.These results were compared with the pathology reports from chart records to ensure consensus and eliminate any inconsistencies in final diagnoses.A structured checklist was used to gather information on multiple variables including age,sex,gestational age at birth,birth weight,age at which hyperbilirubinemia manifested,presence and identification of associated anomalies,clinical manifestations,and histological findings from liver biopsies.The baseline data are reported using descriptive statistics,and differences between groups were assessed by Fisher’s exact test and Student’s t test when indicated.RESULTS:Fifty-five cases(28 females;27 males)of infantile cholestasis(IC)were included in this study.The mean serum total bilirubin and direct bilirubin at presentation were 13.6±5.9 and 7.3±3.4,respectively.Forty cases(72.7%)were the product of term pregnancies.Common associated clinical findings were acholic stool in 33 cases(60.0%),hepatomegaly in30 cases(54.5%),and dark-colored urine in 21 cases(38.2%).Biliary atresia(BA)was the most frequent diagnosis,found in 32 cases(58.2%),followed by intrahepatic bile duct paucity found in 6 cases(10.9%),metabolic disease in 6 cases(10.9%),idiopathic neonatal hepatitis in 5 cases(9.1%),choledochal cyst in2 cases(3.6%),liver cirrhosis in 2 cases(3.6%),and progressive familial intrahepatic cholestasis and portal fibrosis each in 1 case(1.8%).The mean times for jaundice onset and liver biopsy were 43.8 and 102.0 d,respectively.In BA,the mean age at jaundice presentation was 21 d and for liver biopsy was 87.5 d,representing a mean delay of 66.5 d.CONCLUSION:A significant delay was found between IC presentation and liver biopsy,which is detrimental in conditions that can cause irreversible liver damage,such as BA. | Elham Talachian Ali Bidari Mitra Mehrazma Nahid Nick-khah | 2014 | World Journal of Gastroenterology2014,20,4: | 3 |
| 12 | Cardio- and hepato-protective potential of methanolic extract of Syzygium cumini(L.) Skeels seeds:A diabetic rat model study显示文摘Objective: To evaluate the effect of methanolic extract of Syzygium cumini(L.) Skeels(S. cumini) seeds on the major organs in an animal model of diabetes through biochemical and histopathological studies.Methods: The methanolic extracts of S. cumini seeds(100 and 200 mg/kg body weight) were administered to alloxan-induced diabetic rats daily, with fasting blood glucose levels being measured by glucometry at one-day interval for a duration of two weeks. Biochemical assays to evaluate changes in the functions of the heart, liver,pancreas and kidney were carried out. Histopathological changes in the diabetic rat organs(pancreas, liver, heart, kidney and spleen) were also observed after the 14 days of treatment with the extracts.Results: Oral administration of methanolic extracts of S. cumini seeds(100 and 200 mg/kg body weight), with gliclazide as a positive control(25 mg/kg), showed beneficial effects including lowering blood glucose levels(P < 0.001), improved heart and liver functions, and hyperlipidemia due to diabetes.
