| 1 | Efficacy and safety of metformin and sitagliptin based triple antihyperglycemic therapy(STRATEGY):a multicenter,randomized,controlled,non-inferiority clinical trial显示文摘Despite the current guideline's recommendation of a timely stepwise intensification therapy,the 'clinical inertia',termed as the delayed treatment intensification,commonly exists in the real world,which may be partly due to the relatively little substantial evidence and no clear consensus regarding the efficacy and safety of triple oral agents in patients inadequately controlled with dual therapy.In this clinical trial performed in 237 centers in China,5,535 type 2 diabetic patients inadequately controlled by previous therapies were treated with a stable metformin/sitagliptin dual therapy for 20 weeks.The patients who did not reach the glycated hemoglobin A1c(HbA1c) goal were then further randomized into glimepiride,gliclazide,repaglinide,or acarbose group for an additional 24-week triple therapy.A mean HbAlc reduction of 0.85%was observed when sitagliptin was added to the patients inadequately controlled with metformin in 16 weeks.Further HbAlc reductions in the 24-week triple therapy stage were 0.65%in glimepiride group,0.70%in gliclazide group,0.61%in repaglinide group,and 0.45%in acarbose group.The non-inferiority criterion for primary hypotheses was met for gliclazide and repaglinide,but not for acarbose,compared with glimepiride,when added to metformin/sitagliptin dual therapy.The incidences of adverse events(AEs) were 29.2%in the dual therapy stage and30.3%in the triple therapy stage.Metformin/sitagliptin as baseline therapy,with the addition of a third oral antihyperglycemic agent,including glimepiride,gliclazide,repaglinide,or acarbose,was effective,safe and well-tolerated for achieving an HbAlc<7.0%goal in type 2 diabetic patients inadequately controlled with previous therapies.The timely augmentation of up to three oral antihyperglycemic agents is valid and of important clinical benefit to prevent patients from exposure to unnecessarily prolonged hyperglycemia. | Wen Xu Yiming Mu Jiajun Zhao Dalong Zhu Qiuhe Ji Zhiguang Zhou Bin Yao Anhua Mao Samuel S.Engel Bin Zhao Yan Bi Longyi Zeng Xingwu Ran Juming Lu Linong Ji Wenying Yang Weiping Jia Jianping Weng | 2017 | Science China(Life Sciences)2017,60,3: | 19 |
| 2 | The levels, sources and reactivity of volatile organic compounds in a typical urban area of Northeast China显示文摘Air concentrations of volatile organic compounds(VOCs) were continually measured at a monitoring site in Shenyang from 20 August to 16 September 2017. The average concentrations of alkanes, alkenes, aromatics and carbonyls were 28.54, 6.30, 5.59 and9.78 ppbv, respectively. Seven sources were identified by the Positive Matrix Factorization model based on the measurement data of VOCs and CO. Vehicle exhaust contributed the most(36.15%) to the total propene-equivalent concentration of the measured VOCs,followed by combustion emission(16.92%), vegetation emission and secondary formation(14.33%), solvent usage(10.59%), petrochemical industry emission(9.89%), petrol evaporation(6.28%), and liquefied petroleum gas(LPG) usage(5.84%). Vehicle exhaust, solvent usage and combustion emission were found to be the top three VOC sources for O_3 formation potential, accounting for 34.52%, 16.55% and 11.94%, respectively. The diurnal variation of the total VOCs from each source could be well explained by their emission characteristics,e.g., the two peaks of VOC concentrations from LPG usage were in line with the cooking times for breakfast and lunch. Wind rose plots of the VOCs from each source could reveal the possible distribution of the sources around the monitoring site. | 5 | Effects of antioxidants on homocysteine thiolactone-induced apoptosis in human umbilical vein endothelial cells显示文摘Background and objectives Hyperhomocysteinemia is an independent risk factor for cardiovascular disease. Homocysteine thiolactone (HcyT), one of the homocysteine metabolites in vivo, is toxic both in vivo and in vitro. The aim of this study was to investigate the effect of HcyT on apoptotic damage in human umbilical vein endothelial cells (HUVECs) and the role of antioxidants in the reduction of HcyT-induced apoptosis. Methods HUVECs were cultured in DMEM supplemented with 20% heat inactivated fetal bovine serum cell cultures were maintained in a humidified 5% CO2 atmosphere at 37 ℃. Cytotoxicity was determined by MTT assay,which consists of hypodiploid cells with propidium iodide labeling and intracellular reactive oxygen species levels using 2',7'-dichlorofluorescein diacetate as the probe by flow cytometry. Results HcyT (250-2000μM) induced HUVECs apoptosis in a time- and concentration-dependent manner. Reactive oxygen species levels rose in response to increasing HcyT concentrations at 24-h incubation.The reduction of cell apoptosis by N-acetylcysteine, vitamin E, or pyrrolidine dithiocarbamate, occurred simultaneously with a significant decrease in intracellular reactive oxygen species levels. Conclusion HcyT exerts its cytotoxic effects on endothelial cells through an apoptotic mechanism involving cellular reactive oxygen species production. The capacity of N-acetylcysteine, vitamin E, and pyrrolidine dithiocarbamate to scavenge HcyT-induced cellular reactive oxygen species correlates well with their efficiency to protect against HcyT-promoted apoptotic damage. | 6 | Ginsenoside Rb1 relieves glucose fluctuationinduced oxidative stress and apoptosis in Schwann cells显示文摘Cultured Schwann cells were treated with 5.6 mM and 50 mM glucose alternating every 8 hours to simulate intermittent high glucose. The present study analyzed the neuroprotective effects of 1, 10 and 100 μM ginsenoside Rb1 on oxidative damage and apoptosis in Schwann cells induced by intermittent high glucose. Flow cytometry demonstrated that ginsenoside Rb1 reduced intermittent high glucose-mediated reactive oxygen species production. Enzyme linked immunosorbent assay showed that 8-hydroxy-2-deoxy guanosine levels in Schwann cells decreased following ginsenoside Rb1 treatment. Quantitative real-time reverse transcription-PCR and western blot assay results revealed that ginsenoside Rb1 inhibited intermittent high glucose-upregulated Bax expression, but antagonized intermittent high glucose-downregulated Bcl-2 expression in Schwann cells. These effects were most pronounced with 100 μM ginsenoside Rb1. | 7 | Pure bulk orbital and spin photocurrent in two-dimensional ferroelectric materials显示文摘We elucidate a bias-free light-induced orbital and spin current through nonlinear response theory,which generalizes the wellknown bulk photovoltaic effect in centrosymmetric broken materials from charge to the spin and orbital degrees of freedom.We use two-dimensional nonmagnetic ferroelectric materials(such as GeS and its analogs)to illustrate this bulk orbital/spin photovoltaic effect,through first-principles calculations.These materials possess a vertical mirror symmetry and time-reversal symmetry but lack of inversion symmetry.We reveal that in addition to the conventional photocurrent that propagates parallel to the mirror plane(under linearly polarized light),the symmetric forbidden photocurrent perpendicular to the mirror actually contains electrons flow,which carries angular momentum information and move oppositely.This generates a pure orbital moment current with zero electric charge current.Such hidden photo-induced pure orbital current could lead to a pure spin current via spin–orbit coupling interactions.Therefore,a four-terminal device can be designed to detect and measure photo-induced charge,orbital,and spin currents simultaneously.All these currents couple with electric polarization P,hence their amplitude and direction can be manipulated through ferroelectric phase transition.Our work provides a route to generalizing nanoscale devices from their photo-induced electronics to orbitronics and spintronics. | Xingchi Mu Yiming Pan Jian Zhou | 2021 | npj Computational Materials2021,,1: | 2 |
| 8 | Preparation,structural characterization and bioactivities of polysaccharides from mulberry(Mori Fructus)显示文摘Mulberry(Mori Fructus),a highly promising functional food,has long exhibited multiple therapeutic effects as a tonic in traditional Chinese food and pharmacology.Polysaccharides have been regarded as the pivotal pharmacological ingredient in mulberry.In this study,structural characterization and bioactivities of polysaccharides from mulberry(PFM-3)were determined.The results showed that PFM-3 was a pyranose containing carboxyl group,which was mainly composed of glucose(Glc)and a small amount of mannose(Man)and fucose(Fuc).The molecular weight of PFM-3 was 96.17 kDa,and the surface of PFM-3 was compact and uniform crack.PFM-3 markedly decreased lipid accumulation,triglyceride(TG)and low density lipoprotein cholesterin(LDL-C)on hyperlipidemia human hepatocellular carcinoma(HepG2)cells.PFM-3 recovered nuclear morphometry and mitochondrial membrane potential(MMP),enhanced the activity of glutathione peroxidase(GPx)and decreased cell arrest in G2 phase on oxidative stress HepG2 cells.In addition,PFM-3 promoted the decline of tumor necrosis factor-α(TNF-α)and the augment of interleukin-10(IL-10)secretion on inflammatory mouse leukemia cells of monocyte macrophage(RAW 264.7).This work provides a certificate that PFM-3 could markedly improve cellular lipid accumulation,endogenous antioxidant defense capacity and inflammation levels to further use for treatment and amelioration of related diseases. | Yumeng Liu Yiming Liu Delun Mu Hui Yang Ying Feng Ruiqin Ji Rina Wu Junrui Wu | 2022 | Food Bioscience2022,46,2: | 1 |
