1De novo characterization of the root transcriptome of a traditional Chinese medicinal plant Polygonum cuspidatum显示文摘Various active components have been extracted from the root of Polygonum cuspidatum.However,the genetic basis for their activity is virtually unknown.In this study,25600002 short reads (2.3 Gb) of P.cuspidatum root transcriptome were obtained via Illumina HiSeq 2000 sequencing.A total of 86418 unigenes were assembled de novo and annotated.Twelve,18,60 and 54 unigenes were respectively mapped to the mevalonic acid (MVA),methyl-D-erythritol 4-phosphate (MEP),shikimate and resveratrol biosynthesis pathways,suggesting that they are involved in the biosynthesis of pharmaceutically important anthraquinone and resveratrol.Eighteen potential UDP-glycosyltransferase unigenes were identified as the candidates most likely to be involved in the biosynthesis of glycosides of secondary metabolites.Identification of relevant genes could be important in eventually increasing the yields of the medicinally useful constituents of the P.cuspidatum root.From the previously published transcriptome data of 19 non-model plant taxa,1127 shared orthologs were identified and characterized.This information will be very useful for future functional,phylogenetic and evolutionary studies of these plants.HAO DaCheng MA Pei MU Jun CHEN ShiLin XIAO PeiGen PENG Yong HUO Li XU LiJia SUN Chao 2012Science China(Life Sciences)2012,55,5:36

3Overexpression of potato miR482e enhanced plant sensitivity to Verticillium dahliae infection显示文摘Verticillium wilt of potato is caused by the fungus pathogen Verticillium dahliae. Present s RNA sequencing data revealed that miR482 was in response to V. dahliae infection but the function in potato is elusive. Here, we characterized potato miR482 family and its putative role resistance to Verticillium wilt. Members of the potato mi R482 superfamily are variable in sequence, but all variants target a class of disease-resistance proteins with nucleotide binding site(NBS)and leucine-rich repeat(LRR) motifs. When potato plantlets were infected with V. dahliae, the expression level of miR482e was downregulated, and that of several NBS-LRR targets of miR482e were upregulated. Transgenic potato plantlets overexpressing miR482e showed hypersensitivity to V. dahliae infection. Using s RNA and degradome datasets, we validated that miR482e targets m RNAs of NBS-LRR disease-resistance proteins and triggers the production of trans-acting(ta)si RNAs, most of which target m RNAs of defense-related proteins. Thus, the hypersensitivity of transgenic potato could be explained by enhanced miR482e and miR482e-derived tasi RNA-mediated silencing on NBS-LRR-disease-resistance proteins. 4Qualitative analysis for a Wolbachia infection model with diffusion显示文摘We consider a reaction-diffusion model which describes the spatial Wolbachia spread dynamics for a mixed population of infected and uninfected mosquitoes. By using linearization method, comparison principle and Leray-Schauder degree theory, we investigate the influence of diffusion on the Wolbachia infection dynamics.After identifying the system parameter regions in which diffusion alters the local stability of constant steadystates, we find sufficient conditions under which the system possesses inhomogeneous steady-states. 5Natural products as LSD1 inhibitors for cancer therapy显示文摘Natural products generally fall into the biologically relevant chemical space and always possess novel biological activities, thus making them a rich source of lead compounds for new drug discovery. With the recent technological advances, natural product-based drug discovery is now reaching a new era. Natural products have also shown promise in epigenetic drug discovery, some of them have advanced into clinical trials or are presently being used in clinic. The histone lysine specific demethylase1(LSD1), an important class of histone demethylases, has fundamental roles in the development of various pathological conditions. Targeting LSD1 has been recognized as a promising therapeutic option for cancer treatment. Notably, some natural products with different chemotypes including protoberberine alkaloids, flavones, polyphenols, and cyclic peptides have shown effectiveness against LSD1. These natural products provide novel scaffolds for developing new LSD1 inhibitors. In this review, we mainly discuss the identification of natural LSD1 inhibitors, analysis of the co-crystal structures of LSD1/natural product complex, antitumor activity and their modes of action. We also briefly discuss the challenges faced in this field. 6High levels of circulating GM-CSF^(+) CD4^(+) T cells are predictive of poor outcomes in sepsis patients: a prospective cohort study显示文摘Granulocyte colony-stimulating factor(GM-CSF),produced by CD4^(+)T cells,has recently been implicated in the pathogenesis of inflammatory diseases,such as multiple sclerosis and juvenile arthritis.However,the role of GM-CSF-producing CD4^(+)T cells in sepsis remains unknown.This study reports peripheral changes in GM-CSF-producing CD4^(+)T cells in septic patients and the possible underlying mechanism by which GM-CSF influences the outcome of sepsis.Forty-three septic patients,20 SIRS patients,and 20 healthy controls were enrolled in this study and followed for 28 days to assess mortality.We measured the peripheral frequency of GM-CSF^(+)CD4^(+)T cells and recorded their associated relationship with disease progression.Our data demonstrated that peripheral GM-CSF-producing CD4^(+)T cells were significantly higher in septic patients than in both SIRS patients and healthy controls.These cells exhibit a memory phenotype and impaired IFN-γ-secreting capacity in sepsis patients.Using a receiver operating curve analysis with 8.01%as a cut-off point,the percentage of GM-CSF^(+)CD4^(+)T cells could predict the outcome of septic patients.Combined with the increase in GM-CSF-producing CD4^(+)T cells,inflammatory cytokines IL-1βand IL-6 were also upregulated.Using an in vitro neutrophil model,we found that GM-CSF inhibited C3aR expression,while inducing IL-8 production.Furthermore,this effect was transferrable in plasma from sepsis patients and was attenuated by inhibition of GM-CSF using an anti-GM-CSF antibody.These results indicate that GM-CSF-producing CD4^(+)T cells may serve as a marker of sepsis severity.Thus,targeting GM-CSF overproduction may benefit sepsis patients.Huihuang Huang Siyu Wang Tianjun Jiang Rong Fan Zheng Zhang Jinsong Mu Ke Li Yonggang Wang Lei Jin Fang Lin Jie Xia Lijian Sun Biao Xu Chengcheng Ji Jing Chen Juan Chang Bo Tu Bing Song Chao Zhang Fu-Sheng Wang Ruonan Xu 2019Cellular & Molecular Immunology2019,16,6:6
7Clinical features of pediatric patients with COVID-19: a report of two family cluster cases显示文摘Background Coronovirus disease 2019 (COVID-19) has spread rapidly across the globe.People of all ages are susceptible to COVID-19.However,literature reports on pediatric patients are limited.Methods To improve the recognition of COVID-19 infection in children,we retrospectively reviewed two confirmed pediatric cases from two family clusters.Both clinical features and laboratory examination results of the children and their family members were described.Results The two confirmed children only presented with mild respiratory or gastrointestinal symptoms.Both of them had normal chest CT images.After general and symptomatic treatments,both children recovered quickly.Both families had travel histories to Hubei Province.Conclusions Pediatric patients with COVID-19 are mostly owing to family cluster or with a close contact history.Infected children have relatively milder clinical symptoms than infected adults.We should attach importance to early recognition,early diagnosis,and early treatment of infected children.Li-Na Ji Shuang Chao Yue-Jiao Wang Xue-Jun Li Xiang-Dong Mu Ming-Gui Lin Rong-Meng Jiang 2020World Journal of Pediatrics2020,16,3:5
8Simultaneous Extraction of Carboxylated Celulose Nanocrystals and Nanofibrils via Citric Acid Hydrolysis--A Sustainable Route显示文摘In this study, cellulose nanocrystals(CNC) with surface carboxylic groups were prepared from bleached softwood pulp by hydrolysis with concentrated citric acid at concentrations of 60 wt%~80 wt%. The solid residues from acid hydrolysis were collected for producing cellulose nanofibrils(CNF) via post high-pressure homogenization. Citric acid could be easily recovered after hydrolysis reactions through crystallization due to its low water solubility or through precipitation as a calcium salt followed by acidification. Several important properties of CNC and CNF, such as dimension, crystallinity, surface chemistry, thermal stability, were evaluated. Results showed that the obtained CNC and CNF surfaces contained carboxylic acid groups that facilitated functionalization and dispersion in aqueous processing. The recyclability of citric acid and the carboxylated CNC/CNF give the renewable cellulose nanomaterial huge potential for a wide range of industrial applications. Furthermore, the resultant CNC and CNF were used as reinforcing agents to make sodium carboxymethyl cellulose(CMC) films. Both CNC and CNF showed reinforcing effects in CMC composite films. The tensile strength of CMC films increased by 54.3% and 85.7% with 10 wt% inclusion of CNC and CNF, respectively. 9Substantial transition to clean household energy mix in rural China显示文摘The household energy mix has significant impacts on human health and climate,as it contributes greatly to many health-and climate-relevant air pollutants.Compared to the well-established urban energy statistical system,the rural household energy statistical system is incomplete and is often associated with high biases.Via a nationwide investigation,this study revealed high contributions to energy supply from coal and biomass fuels in the rural household energy sector,while electricity comprised∼20%.Stacking(the use of multiple sources of energy)is significant,and the average number of energy types was 2.8 per household.Compared to 2012,the consumption of biomass and coals in 2017 decreased by 45%and 12%,respectively,while the gas consumption amount increased by 204%.Increased gas and decreased coal consumptions were mainly in cooking,while decreased biomass was in both cooking(41%)and heating(59%).The time-sharing fraction of electricity and gases(E&G)for daily cooking grew,reaching 69%in 2017,but for space heating,traditional solid fuels were still dominant,with the national average shared fraction of E&G being only 20%.The non-uniform spatial distribution and the non-linear increase in the fraction of E&G indicated challenges to achieving universal access to modern cooking energy by 2030,particularly in less-developed rural and mountainous areas.In some non-typical heating zones,the increased share of E&G for heating was significant and largely driven by income growth,but in typical heating zones,the time-sharing fraction was<5%and was not significantly increased,except in areas with policy intervention.The intervention policy not only led to dramatic increases in the clean energy fraction for heating but also accelerated the clean cooking transition.Higher income,higher education,younger age,less energy/stove stacking and smaller family size positively impacted the clean energy transition.Guofeng Shen Rui Xiong Yanlin Tian Zhihan Luo Bahabaike Jiangtulu Wenjun Meng Wei Du Jing Meng Yuanchen Chen Bing Xue Bin Wang Yonghong Duan Jia Duo Fenggui Fan Lei Huang Tianzhen Ju Fenggui Liu Shunxin Li Xianli Liu Yungui Li Mu Wang Ying Nan Bo Pan Yanfang Pan Lizhi Wang Eddy Zeng Chao Zhan Yilin Chen Huizhong Shen Hefa Cheng Shu Tao 2022National Science Review2022,9,7:4
10Cratering process and morphological features of the Xiuyan impact crater in Northeast China显示文摘By using crater impact morphological theory and mathematics modeling,the paper studies the cratering process and morphological features of the Xiuyan Crater in Northeast China based on remote sensing imagery,digital elevation model(DEM)and drilling and measuring data.The results show that:1)Simulated calculation indicates the diameter of the transient crater was1406±12 m and the depth was 497±4 m.The diameter of the final crater was 1758±15 m and the depth was 374.5±3.5 m;the thickness of the breccia lens was 188.5±0.5 m.The data is basically the same as previous drilling data.2)Preliminary determination of the size of Xiuyan impactor:The diameter at about 55 m for an iron meteorite,and about 115 m for a stony meteorite.3)The depth-to-diameter ratio is 0.143,similar to the typical simple crater.The circularity index of the Xiuyan Crater is0.884,indicating erosion and degradation.4)The distribution pattern of radial fractures has priority in NW-SE and WNW-ESE and these fractures control the development and evolution of drainage inside the crater.WANG XinYuan LUO Lei GUO HuaDong MU LingLi LI Chao JI Wei CAI Heng 2013Science China Earth Sciences2013,56,10:4
11GmCRY1s modulate gibberellin metabolism to regulate soybean shade avoidance in response to reduced blue light显示文摘Soybean is an important legume crop that displays the classic shade avoidance syndrome(SAS),including exaggerated stem elongation,which leads to lodging and yield reduction under density farming conditions.Here,we compared the effects of two shade signals,low red light to far-red light ratio(R:FR)and low blue light(LBL),on soybean status and revealed that LBL predominantly induces excessive stem elongation.We used CRISPR-Cas9-engineered Gmcry mutants to investigate the functions of seven cryptochromes(GmCRYs)in soybean and found that the four GmCRY1s overlap in mediating LBL-induced SAS.Lightactivated GmCRY1s increase the abundance of the bZlP transcription factors STF1 and STF2,which directly upregulate the expression of genes encoding GA2 oxidases to deactivate GA1 and repress stem elongation.Notably,GmCRY1b overexpression lines displayed multiple agronomic advantages over the wild-type control under both dense planting and intercropping conditions.Our study demonstrates the integration of GmCRY1-mediated signals with the GA metabolic pathway in the regulation of LBL-induced SAS in soybean.It also provides a promising option for breeding lodging-resistant,high-yield soybean cultivars in the future.Xiangguang Lyu Qican Cheng Chao Qin Yinghui Li Xinying Xu Ronghuan Ji Ruolan Mu Hongyu Li Tao Zhao Jun Liu Yonggang Zhou Haiyan Li Guodong Yang Qingshan Chen Bin Liu 2021Molecular Plant2021,14,2:4

16Detection of Penicillin via Surface Plasmon Resonance Biosensor显示文摘A method of using Au colloid to capture the decomposed product of penicillin, penicillamine, on a surface plasmon resonance(SPR) biosensor for the quantitative determination of penicillin was developed. Based on the de-composition of penicillin to generate penicillamine and penilloaldehyde, a high sensitive biosensor for detecting penicillin was also developed. In our experiment, it was penicillamine rather than penicillin that has been measured. This is because penicillamine contains a functional group that makes it self-assembling on Au colloid to increase the molecular weight so as to improve the surface plasmon resonance signal. On a UV-Vis spectrophotometer, a high concentration of penicilliamine-Au complex was determined, indicating that penicillamine was already well combined with Au colloid.
